Drinking Synthetic Alkaline Mineral Water Protects Early-Life Maternal Separation-Induced Gut Barrier Disruption

Abstract:Background: Exposure to stress in early-life (infancy) causes long-term impact on the structure and function of the intestine, leading to barrier integrity loss and an increased risk of irritable bowel syndrome in adulthood. Drinking alkaline mineral water (AMW) exerts a potential therapeutic effect in gastrointestinal disorders, but the supportive evidence from animal studies and the mechanism involved remain unreported.Methods: Here, a synthesized alkaline mineral complex (AMC) water was employed in a maternally separation (MS) piglet model. The scanning electron microscope (SEM) and Hematoxylin and Eosin (HE) staining were employed to analyze the effects of AMW on small intestinal surface damage and morphology in MS piglets. We examined the levels of inflammatory factors and the inflammatory signaling pathway (NF-κB) in the gut. Meanwhile, 16S-rRNA PacBio SMRT Gene Full-Length Sequencing analyses was employed for the cecal microbes in this study. Then, we evaluated the effects of AMC water on intestinal mucus barrier and epithelial barrier in MS piglets under weaning stress. Finally, the interaction between the microflora and gut was illustrated.Findings: MS stress causes significant surface injury to villi and decreases villous height in the gut, and this morphology injury induced by MS is alleviated by AMC water. Importantly, drinking synthetic AMC water ameliorates early-life MS-induced gut barrier disruption in MS piglets by controlling the gut inflammation, improving microflora composition and functional profiles. Mechanistically, AMC water, as a microbe regulator, targeted inhibition of NF-κB signaling by increasing gut beneficial bacteria s_prausnitzii abundance, thereby reducing the pro-inflammatory factors level, and increasing the number of goblet cells and mucins secretion, and suppressing MLCK/p-MLC signaling to improve tight junction and adherens junction function.Interpretation: These findings reveal drinking synthetic AMW prevents early-life MS stress-induced gut barrier disruption in piglets by maintaining the benign interaction between microflora and gut, which may provide a potential therapy for mammals at risk of intestinal inflammation and barrier disruption.Funding Information: This study has received assistance from National Natural Science Foundation of China (No. 32172932), Key Program of Natural Science Foundation of Heilongjiang Province of China (No. ZD2021C003), China Agriculture Research System of MOF and MARA (No. CARS-35), Distinguished Professor of Longjiang Scholars Support Project (No. T201908) and Heilongjiang Touyan Innovation Team ProgramDeclaration of Interests: The authors declare that they have no conflict of interestEthical Approval: The present study was approved by the Animal Use and Care Committee of Northeast Agricultural University (NEAU).
