Copper Regulates the Expressions of Mdlac7 and Mdccr1 to Promote the Biosynthesis of Lignin in Apple Peel

Abstract:The excessive accumulation of copper (Cu) in apple peel severely affects the appearance quality and commerciality of fruits; However, research on the relationship between Cu and lignin biosynthesis in apple peel is rare.To analyze the effects of Cu on lignin biosynthesis, 6-year-old apple trees were fed 100 μM/L CuSO4, and their relationships were analyzed using transcriptome sequencing, gene overexpression, and CRISPR/Cas9 knockout techniques. ​The present study showed that Cu significantly induced the thickening of the peel and resulted in an increase in lignin content. Transcriptome data analysis showed that 2315 and 1391 genes were significantly upregulated and downregulated in the peel of the stalk cavity after Cu treatment, respectively. In addition, the phenylpropane metabolic pathway was identified as a key pathway in response to Cu treatment. In this pathway, MdCCR1 and MdLAC7 genes of lignin biosynthesis were significantly upregulated after Cu treatment. Subsequently, CRISPR/Cas9 knockout of these two rate-limiting enzymes was performed, which significantly reduced the expressions of MdLAC7 and MdCCR1 genes in apple calli and further inhibited the biosynthesis of lignin. These results provide a theoretical basis and technical support for solving the degradation of apple quality caused by Cu pollution.
