MiR-760 is a Key Regulator of Glioma Proliferation Migration and Invasion by Controlling MMP2 Expression

Abstract:Abstract Background: Glioma is the most common kind of brain tumor and has a poor prognosis. As available evidence indicates, miRNAs are involved in the origin and development of glioma malignancies. MiR-760 has been reported to the development of tumors, but miR-760 has not been well studied in gliomas. Methods: In this study, we used database of GSE25631 to survey the abnormal expression of microRNA, and the expression of miR-760 validated decreased in glioblastoma. We sought to identify the microRNA-760 (miR-760) expression pattern and investigate its prognostic significance in glioma. We used quantitative real-time polymerase chain reaction to determine the relative expression of miR-760 and MMP2 in glioma cell lines. The effect of miR-760 on cell proliferation, migration, and invasion was determined using the Cell Counting Kit-8 (CCK-8), the 5-ethynyl-2'-deoxyuridine (EdU) assay, and the transwell assay. Bioinformatics analysis confirmed that the downstream target gene of miR-760. Additionally, a luciferase reporter experiment was used to identify the miR-760 target gene MMP2. Western blot and immunofluorescence was used to determine MMP2 protein levels. Result: Our data demonstrated that miR-760 expression was significantly decreased in glioma tissues and cell lines. Increased miR-760 expression induced proliferation, migration, and invasion in glioma cell lines. Bioinformatics analysis showed that miR-760 binds and inhibits MMP2 expression. These results suggest that miR-760 inhibits glioma growth by up-regulating the expression of miR-760 along the miR-760 /MMP2 axis. Conclusion: The finding of the miR-760/MMP2 axis will expand our insights into the molecular mechanisms underlying glioma growth and will identify potential therapeutics for glioma treatment.
