Reactive Oxygen Species Promote the Formation of Mtb-Induced Foamy Macrophages by Downregulating the Expression of Peroxisome Proliferator-Activated Receptor-Gamma

Abstract:Inducing the formation of foamy macrophages is one of the important mechanisms for Mycobacterium tuberculosis (MTB) to escape immune killing and achieve its long-term survival in macrophage. Our previous studies found that MTB can induce the formation of foamy macrophage by inhibiting the expression of PPAR γ and ABCG1, but the involved mechanisms have yet to be elucidated. In this study, we found that the formation of foam macrophages induced by MTB infection is accompanied by the production of high-level reactive oxygen species (ROS) in macrophages. And, the accumulation level of lipids in macrophages induced by MTB infection can be manipulated by regulating the level of ROS. Moreover, regulating the levels of ROS in macrophages can change the expression of PPAR in MTB-infected macrophages. Furthermore, we confirmed that the role of ROS in inducing the formation of foam macrophages is PPAR γ dependent. Combined with our previous research, this study suggests a new mechanism for MTB to induce the formation of foamy macrophage by ROS- PPARγ-ABCG1 pathway.
