A Novel Guinea Pig Glaucoma Model Induced by Magnetic Microsphere Injection

Abstract:A reliable animal model can recapitulate the anatomical features of glaucoma to a great extent and enhance our understanding of its pathologic changes. Due to the limitations of existing primates and rodents, we aim to develop and validate the feasibility and effectiveness of a novel glaucoma model in guinea pigs. In this study, guinea pigs were intracamerally injected with magnetic microspheres to induce chronic ocular hypertension (COH). The intraocular pressure (IOP) was recorded for 8 consecutive weeks. Retinal ganglion cells (RGCs) loss and the degeneration of the optic nerve were assessed. After modeling, 73.33% of guinea pigs were successfully induced with high IOP. The mean IOP raised from baseline (18.04 ± 1.33 mmHg) to the highest at week 3 (36.31 ± 6.13 mmHg) and remained elevated for at least 7 weeks. TUNEL-positive cells in the ganglion cell layer (GCL) in the COH group were significantly more than those in the control group at weeks 4 and 8, and retinal flat mount staining indicated that the density of RGCs in the COH group (1090 ± 46.17 cells/mm2) was significantly lower than that in the control group (1574 ± 63.74 cells/mm2). The optic disc excavated and the lamina cribrosa (LC) was significantly depressed and thinned. The number of optic nerve axons in the COH group gradually decreased from baseline (44553 ± 3608 /mm2) to week 4 (28687 ± 2071 /mm2) and week 8 (17977 ± 3697 /mm2), and degenerative axons and macrophages were observed engulfing the myelin debris. In conclusion, intracameral injection of magnetic microspheres successfully induced COH in guinea pigs that showed typical glaucomatous changes including robust IOP elevation, loss of RGCs, thinning of the LC, and axonal degeneration. This novel glaucoma model has the potential to facilitate the study of glaucoma, and investigate the role of LC in the pathogenesis of glaucoma.
