Machine Learning-Based QSAR Models and Structural Alerts for Prediction of Mitochondrial Dysfunction

Abstract:Mitochondria are cellular organelles critical for human health. Mitochondrial dysfunction is associated with adverse outcomes such as enhanced cellular oxidative stress, bioenergetic imbalance and cell apoptosis. Thus, it is a driving imperative to identify chemicals that can cause mitochondrial dysfunction. However, it is difficult to screen all chemicals for mitochondrial dysfunction using experimental methods, as such methods are expensive and time-consuming. Computational toxicology approaches such as quantitative structure–activity relationship (QSAR) modeling and identification of structural alerts are promising alternative methods to identify chemicals associated with mitochondrial dysfunction. In this chapter, recent QSAR models and structural alerts on mitochondrial dysfunction are reviewed. The first section describes the importance of developing computational toxicology models on mitochondrial dysfunction. Then datasets and machine learning methods used to build QSAR models as well as methods to identify structural alerts are briefly introduced. The last section presents research progress on QSAR models and structural alerts for mitochondrial dysfunction.
