Lactic Acid Bacteria Isolation and Characterization from Yogurts in Qinghai Province of China

Abstract:In the present study, the probiotic potential of lactic acid bacteria (LAB) isolated from nomadic herders in the Qinghai province of China was investigated. A total of 68 yogurt samples were randomly collected from Qinghai yak and yellow buffalo yogurt, and 209 lactobacilli were isolated. Of these, 15 isolates were confirmed as lactic acid bacteria by conventional methods and identified through 16s RNA gene sequencing. The identified LAB species included Lactobacillus delbrueckii subsp. Streptococcus thermophilus, Enterococcus durans, Enterococcus hirae, and Enterococcus faecium. Enterococcus hirae and Enterococcus faecium demonstrated an evident resistance to acid and bile salts tolerance. After pH neutralization and catalase treatment, the antibiotic susceptibility profiles and antimicrobial activities suggested that bacteriocin-like inhibitory substances may play a key role in LAB antimicrobial activity, that makes the LAB a good candidate for probiotic potential. In addition, LABs were investigated for adhesion to human intestinal Caco-2 cells and antioxidant activity. In summary, LAB isolates from yak and yellow buffalo yogurts can be considered good candidates for potential probiotics. Therefore, these LAB could be developed and applied as bio preservatives to control microbial contamination in food products.
