Abstract:AIM: To investigate the effect of advanced glycation end products(AGEs) on autophagy in human umbilical endothelial cells(HUVECs) and to identify the role of autophagy in advanced glycation end product-induced cell apoptosis.METHODS: HUVECs were cultured and treated with AGEs or bovine serum albumin.The protein expression was detected by Western blotting.Autophagosomes were observed under electron microscope.The cell apoptotic rate was determined by flow cytometry.The cell viability was quantified by MTT assay.RESULTS: After treated with AGEs,the level of autophagy-associated protein LC3-II in HUVECs was up-regulated,and the number of autophagosomes was increased.Compared with control group,the apoptotic rate of HUVECs increased and the viability of HUVECs was decreased in AGEs treatment group.Furthermore,pretreating the cells with an autophagy inhibitor 3-methyladenine aggravated these effects.The levels of phospho-protein kinase B(Akt) and phospho-mammalian target of rapamycin(mTOR) in HUVECs were also decreased by treatment with AGEs.Pretreatment with Akt activator insulin-like growth factor 1(IGF-1) increased Akt phosphorylation and suppressed the AGE-induced LC3-II expression.CONCLUSION: AGEs induce autophagy in HUVECs through PI3K/Akt/mTOR signal pathway.Autophagy plays a protective role in AGE-induced apoptosis in HUVECs.
Autophagy and Apoptosis of Retinal Müller Cells Induced by High Glucose

Abstract:Objective::To investigate whether high glucose (HG) can regulate the autophagy and apoptosis in Müller cells.Methods::In this experimental research, primary SD rat retinal Müller cells were exposed to normal glucose (NG) or 3 h, 6 h, 12 h, 24 h, 36 h of high glucose. And Müller cells were exposed to 5.5, 20, 30, 40, 50 mmol/L for 24 h. LC3Ⅰ/LC3Ⅱ, P62, and Beclin-1 protein expression was examined by Western blot analysis in the various experimental groups. And in fluorescence microscopy experiments, autophagy was evaluated by the autophagy markers LC3Ⅰ/LC3Ⅱ and P62. The formation of autophagosomes and autolysosomes were examined by electron microscopy. TUNEL assay was used to detect apoptosis in high glucose Müller cells. One-way analysis of variance was used to compare data between different groups.Results::The relative expression levels of Beclin-1, P62 and LC3Ⅰ/LC3Ⅱ in Müller cytoplasm of the normal glucose group, and 20 mmol/L, 30 mmol/L, 40 mmol/L and 50 mmol/L high glucose groups were compared. There were statistically significant differences ( F=131.21, P=0.029; F=197.25, P=0.012; F=100.02, P=0.045). Compared to the normal group, Beclin-1 and LC3Ⅰ/LC3Ⅱ protein in Müller cytoplasm of the high glucose groups significantly decreased ( P<0.05). The relative expression of P62 protein significantly increased ( P<0.001). The relative expression levels of autophagy related protein Beclin-1, P62 and LC3Ⅰ/LC3 in Müller cytoplasm of the normal group and high glucose groups at 6 h, 12 h, 24 h and 36 h were statistically significant ( F=98.46, P=0.015; F=109.48, P=0.004; F=99.27, P=0.032). LC3Ⅰ/LC3 and Beclin-1 protein significantly increased in the 6 h high glucose group ( P=0.002, 0.015); LC3Ⅰ/LC3Ⅱ in and Beclin-1 protein significantly decreased in 12 h, 24 h and 36 h compared to the normal group ( P<0.05), but the P62 protein significantly increased ( P<0.001). There were statistically significant differences in the relative number of GFP-LC3-transfected positive particles and autophagosomes in the normal group, and high glucose 6 h and 24 h retinal Müller cells ( F=240.49, P=0.014; F=198.98, P=0.001). Compared to the normal group, the relative number of GFP-LC3-transfected positive particles and autophagosomes in the 6h group increased. There were statistically significant difference ( P=0.002, 0.029) and there were significant decreases in 24 h ( P=0.019, 0.036). The apoptosis indexes of retinal Müller cells were 3.14±1.01, 15.34±3.87, 25.82±4.96 and 24.79±4.01 in the normal group, 6 h and 24 h groups and 3MA group, respectively, and the overall difference was statistically significant ( F=234.12, P=0.003). Compared to the normal group, the TUNEL positive cells increased in the 6 h and 24 h groups ( P=0.022, 0.039), and the TUNEL positive cells further increased in the 6 h and 24 h groups ( P=0.017). Conclusions::HG can induce early autophagy and inhibit apoptosis in cultured retinal Müller cells, and HG can inhibit autophagy and inrease apoptosis over time. This study suggests that high glucose inhibition of autophagy may be an important factor in promoting the apoptosis of Müller cells.
Epigallocatechin-3-gallate Stimulates Autophagy and Reduces Apoptosis Levels in Retinal Müller Cells under High-Glucose Conditions.

Abstract:Retinal neurodegeneration is an early feature in the pathogenesis of diabetic retinopathy (DR). Autophagy is an intracellular catabolic process involved in protein and organelle degradation that has been linked in DR. Epigallocatechin gallate (EGCG) is a major polyphenol in green tea that has beneficial effects in diabetic. however, it is not currently known whether EGCG can regulate Müller cell autophagy. Here, we showed that EGCG increased autophagy by promoting the formation of autophagosomes, increasing lysosomal acidification, and stimulating autophagic flux in Müller cells, while high glucose (HG) induced a decrease in autophagy and an increase in apoptosis. However, retinal Müller cells in HG treated with EGCG showed autophagy machinery activation and reestablishment of cargo degradation, protecting the cells from apoptosis. EGCG could increase the ability of cells to proliferate by increasing autophagy. In an experimental model of diabetic retinopathy, EGCG reduced the reactive gliosis of Müller cells and decreased retinal damage. These results highlight that HG downregulates autophagy and accumulates P62 cargo due to lysosomal dysfunction and then increases apoptosis, which was reversed by treatment with EGCG. This finding might be valuable for developing a novel therapeutic strategy to treat DR.
High glucose increases podocyte autophagy through PI3K-AKT-mTOR signaling pathway

Abstract:Objective To evaluate the effects of high glucose on autophagy and apoptosis of podocyte and explore the signaling pathway in high glucose-induce podocyte autophagy.Methods Differentiated mouse podocytes were exposed to high glucose(30 mmol/L) or rapamycin (autophagy enhancer,1 μg/L) or LY294002 (a selective PI3K inhibitor,50 mmol/L) for 24 h.The formations of autophagy were observed by electron microscopy and acridine orange staining.Apoptosis was evaluated by flow cytometry.The expression of autophagy protein LC3-Ⅱ/Ⅰ and Beclin-1 as well as the phosphorylation of AKT and mTOR were examined by Western blotting analysis.Results High glucose induced podocytes apoptosis,increased autophagy and the expression of autophagy-associated proteins (all P ＜ 0.05).Rapamycin further increased the expression of LC3-Ⅱ and Beclin-1 protein (all P ＜ 0.05),but LY294002 inhibited partialiy the protein expression of LC3-Ⅱ and Beclin-1 induced by high glucose (both P ＜ 0.05).Treatment with rapamycin increased the phosphorylation of AKT,but reduced that of mTOR in podocytes.Moreover,LY294002 inhibited phosphorylation of both AKT and mTOR (both P ＜ 0.05).Conclusions High glucose promotes podocyte autophagy and apoptosis.High glucose-induced autophagy is mediated partly through PI3K-AKT-mTOR signaling pathway.
Elevated Cell Proliferation and VEGF Production by High-Glucose Conditions in Müller Cells Involve XIAP

Abstract:Purpose Müller cells have important roles in the pathogenesis of diabetic retinopathy by promoting cell proliferation and inducing the production of vascular endothelial growth factor (VEGF) under hyperglycemic conditions. The objective of this study was to determine the potential mechanism of Müller cell proliferation and VEGF production due to high-glucose conditions. Methods Primary cultured rat Müller cells were incubated with medium containing variable concentrations of glucose and/or embelin, a specific inhibitor of X-linked inhibitor of apoptosis protein (XIAP), for 72 h. The proliferation of Müller cells was assessed by the MTT assay. The expression and/or phosphorylation of 146 proteins were assessed using protein pathway array. Results High concentrations of glucose-induced Müller cell proliferation and altered expression and/or phosphorylation of 47 proteins that have been identified to have key roles in several important signaling pathways (XIAP, VEGF, HIF1 α , NF κ B, etc) and are involved in the regulation of cell survival, proliferation, or apoptosis. However, Müller cell alterations induced by high-glucose conditions were counteracted by the XIAP inhibitor embelin, and 26 proteins/phosphorylations (out of 47) were restored to their normal levels. Nine proteins, including NF κ B p65, p-p38, tumor necrosis factor- α , urokinase-type plasminogen activator, CREB, IL-1 β , HCAM, estrogen receptor- α , and p-Stat3, were involved in regulatory networks between XIAP and VEGF. Conclusions The current study suggests that XIAP may be a potential regulator that can mediate a series of pathological changes induced by high-glucose conditions in Müller cells. Therefore, embelin could be a potential agent for the prevention and treatment of diabetic retinopathy.

Abstract:Tubular atrophy and dysfunction is a critical process underlying diabetic nephropathy (DN). Understanding the mechanisms underlying renal tubular epithelial cell survival is important for the prevention of kidney failure associated with glucotoxicity. Autophagy is a cellular pathway involved in protein and organelle degradation. It is associated with many types of cellular homeostasis and human diseases. To date, little is known of the association between high concentrations of glucose and autophagy in renal tubular cells. In the present study, we investigated high glucose-induced toxicity in renal tubular epithelial cells by means of several complementary assays, including cell viability, cell death assays and changes in ultrastructure in an immortalized human kidney cell line, HK-2 cells. The extent of apoptosis was significantly increased in the HK-2 cells following treatment with high levels of glucose. In addition, in in vivo experiments using diabetic rats, high glucose exerted harmful effects on the tissue structure of the kidneys in the diabetic rats. Chronic exposure of the HK-2 cells and tubular epithelial cells of nephritic rats to high levels of glucose induced autophagy. Liraglutide inhibited these effects; however, treatment witht a glucagon-like peptide-1 receptor (GLP-1R) antagonist enhanced these effects. Our results also indicated that the exposure of the renal tubular epithelial cells to high glucose concentrations in vitro led to the downregulation of GLP-1R expression. Liraglutide reversed this effect, while the GLP-1R antagonist promoted it, promoting autophagy, suggesting that liraglutide exerts a renoprotective effect in the presence of high glucose, at least in part, by inhibiting autophagy and increasing GLP-1R expression in the HK-2 cells and kidneys of diabetic rats.

Abstract:Objective:To investigate the expression of glia maturation factor-β (GMFB) in Müller cells under 25 mmol/L D-glucose treatment and its effect on Müller cells.Methods:(1) Müller cells were treated with 25 mmol/L Dglucose for 0 h,1 h,2 h,4 h and 6 h and GMFB expression level was examined by Western Blot.(2) Müller cells were treated with 1 μg/ml GMFB for 0 h,2 h,4 h,8 h and 12 h after infected with LC3 detection system(mRFP-GFP-LC3).We used immunofluorescence and Western Blot to explore the development of autophagy.(3) Müller cells were treated with 1 μg/ml GMFB for 24 h as experimental group and total RNA was extracted.Both the experimental group and control group were used for RNAseq assay.By comparing the differentially expressed profile of two groups,we selected the genes for GO enrichment analysis and KEGG analysis.Results:(1) In the Müller cells under the treatments of different time of 25 mmol/L D-glucose,GMFB expression level was significantly up-regulated.(2) Recombinant GMFB can initiate transient autophagy at 4 h treatment.(3) RNAseq analysis showed that 147 genes expression patterns significantly altered in experimental group,103 of which were up-regulated and 44 were down-regulated.GO analysis indicated that GMFB was involved in cellular metabolism and cellular process.KEGG analysis further indicated that GMFB was involved in cell adhesion molecules (CAMs),axon guidance and cytosolic DNA-sensing pathway.Furthermore,inflammation related gene IL-33 was also down-regulated.Conclusion:High glucose induced the up-regulation of GMFB in Müller cells.The effects of GMFB on Müller cells were related with cell-cell binding and inflammation pathway.GMFB is involved in important cellular process in Müller cells under hyperglycemia.The present work provides helpful clues to understanding the mechanism involved in diabetic retinopathy.The exact mechanism of GMFB in DR needs further investigation.
Role of Autophagy in Angiogenesis Induced by a High-Glucose Condition in RF/6A Cells.

Abstract:PURPOSE:To study the effect of autophagy on vitality, migration, and tube formation of RF/6A cells under the condition of D-glucose.METHODS:Cultured RF/6A cells were randomly divided into 4 groups (control, low glucose, high glucose, and high glucose with 3-methyladenine [3-MA]). Autophagy-related proteins (Atg7, p62, and LC3) were monitored. Cell vitality, cell migration, tube formation, reactive oxygen species (ROS) production, and apoptosis were assessed.RESULTS:Cell vitality significantly decreased and cell migration and tube formation significantly increased in the high-glucose group (p < 0.05). Pretreatment with 3-MA significantly increased cell viability and inhibited cell migration and tube formation (p < 0.05). ROS production increased in the high-glucose group and decreased in the high-glucose with N-acetylcysteine (NAC) group (p < 0.05). The level of apoptosis increased in the high-glucose group, while it was reduced in the high-glucose with 3-MA group.CONCLUSION:Autophagy maybe participates in the formation of retinal neovascularization induced by high glucose.
High glucose induced apoptosis in H9c2 cells by promoting autophagy dysfunction

Abstract:Objective To investigate the effect of autophagy on high glucose (HG)-induced cardiomyocytes H9c2 cells apoptosis. Methods The viability of H9c2 cells was detected by MTT assay. The apoptotic cells were observed by hoechst33258 staining. The expression of pro-apoptotic proteins Bax and autophagy-related proteins (Beclin-1 and P62) in H9c2 cells were detected by Western Blot. The factorial design analysis of variance was used for the OD value and the protein band gray value of each group, and the difference between the groups was analyzed by single factor ANOVA. Results High glucose induced the decrease in viability of H9c2 cells: The viability of high glucose cells at 12, 24, and 48 mmol/L was [(79.5±2.23)%] (t = 3.143, P = 0.043), [(54.6±3.08)%] (t = 12.425, P = 0.000) and [(37.2±2.59)%] (t = 13.761, P = 0.000) of the control group (100%) . Compared with the control group (100%), the cell viability of the mannitol isotonic control group was [(101.0±1.27)%] (t = 0.012, P = 0.094). High glucose induced the increase of hoechst 33258 positive cells in H9c2 cells and increased the expression of pro-apoptotic protein Bax: Compared with the control group, the gray levels of apoptotic protein Bax/β-actin in the high glucose treatment groups of 12, 24, and 48 mmol/L were (1.29±0.25, t = 2.32, P = 0.045), (1.42±0.23, t = 10.247, P = 0.000) and (1.81±0.29, t = 16.324, P = 0.000). High glucose induced autophagy dysfunction: Compared with the control group, the gray values of autophagy-related protein Beclin-1/β-actin were (0.82±0.16, t = 4.243, P = 0.032), (0.78±0.19, t = 11.341, P = 0.000), and (0.62±0.11, t = 13.455, P = 0.000), and the gray levels of P62 protein/β-actin protein were (1.29±0.25, t = 4.442, P = 0.014), (1.42±0.23, t = 13.341, P = 0.000), and (1.81±0.29, t = 15.851, P = 0.000). The autophagy inducer rapamycin reversed high glucose-induced increase in hoechst33258-positive cells and the high-glucose-induced increase in Bax expression: Compared with the control group, the Bax /β-actin gray value of the high glucose group, high glucose and rapamycin co-treatment group, and rapamycin group rapamycin group were (1.51±0.31, t = 14.342, P = 0.000), (1.42±0.23, t = 9.621, P = 0.004), and (1.81±0.12), t = 0.172, P = 0.124). Conclusion High glucose may promote apoptosis and induces autophagy dysfunction in cardiomyocytes H9c2 cells. The application of autophagy inducer may reverse the effect of high glucose on apoptosis of H9c2 cells, indicating that autophagy is an important mechanism of high glucose-induced apoptosis in H9c2 cells. High Level of Glucose Induced Apoptosis in Human Endothelial Cells and Its Mechanism of JNK and AKT Signaling Pathways

Abstract:Objective To investigate the signaling pathway of high level of glucose-induced apoptosis in human umbilical vein endothelial cells (HUVECs). Methods HUVECs were incubated with physiological concentration of glucose (5 mmol/L) (NG), high glucose (30 mmol/L)(HG) and high glucose combined with SP600125(HG+I) for 72 hours. Apoptosis of HUVECs was observed in a fluorescence microscope by Hoechst 33258 stain, and quantitied by flow cytometry using annexin V-FITC stain. The level of p-JNK, p-c-JUN and p-AKT was evaluated by western blot. Results Treatment of HUVECs with high glucose resulted in a significant increase in apoptotic cells comparing to a physiological concentration (13.31% vs 5.69%, P0.01). Incubation of HUVECs with high concentration of glucose for 72 hours increased the phosphorylation of JNK and c-JUN significantly (P0.05, vs NG), while decreased the level of p-AKT markedly(P0.01, vs NG). After treated with SP600125, the level of p-JNK and p-c-JUN (P0.05, vs HG) decreased, while the level of p-AKT increased (P0.01, vs HG),and the apoptosis of HUVECs induced by high glucose was inhibited (8.38%, P0.01, vs HG). Conclusion High concentration of glucose could induce HUVECs apoptosis, probably through increasing the activation of JNK, the downstream c-jun, and inhibition of the activation of AKT. The activation JNK could regulate AKT pathway.

Abstract:Autophagy is a cellular pathway involved in protein and organelle degradation. It is relevant to many types of cellular homeostasis and human diseases. High level of glucose is known to inflict podocyte injury, but little is reported about the relationship between high concentrations of glucose and autophagy in these cells. The present study demonstrates that high glucose promotes autophagy in podocytes. Rapamycin further enhances this effect, but 3-methyadenine inhibits it. The proautophagic effect of high glucose manifested in the form of enhanced podocyte expression of LC3-2 and beclin-1; interestingly, antioxidants such as NAC were found to inhibit high glucose-induced autophagy. High glucose induced the generation of ROS by podocytes in a time-dependent manner. High glucose also enhanced podocyte expression of MnSOD and catalase. These findings indicate that high glucose-induced autophagy is mediated through podocyte ROS generation.
Autophagy Functions to Prevent Methylglyoxal-Induced Apoptosis in HK-2 Cells

Abstract:Methylglyoxal (MGO), a reactive carbonyl species, causes cellular damage and is closely related to kidney disease, particularly diabetic nephropathy. Although MGO has been reported to induce autophagy and apoptosis, the relationships between the two pathways are unclear. Here, we evaluated whether autophagy may be the underlying mechanism inhibiting MGO-induced apoptosis. MGO treatment induced concentration- and time-dependent apoptosis in HK-2 cells. Moreover, MGO upregulated the autophagy markers p62 and LC3-II. Apoptosis caused by MGO was increased in ATG5-knockdown cells compared to that in wild-type cells. In contrast, autophagy activation by 5-aminoimidazole-4-carboxamide ribonucleotide resulted in reduced apoptosis, suggesting that autophagy played a role in protecting against MGO-induced cell death. To examine the mechanisms through which autophagy occurred following MGO stimulation, we investigated changes in AKT/mammalian target of rapamycin (mTOR) signaling. Autophagy induction by MGO treatment was not related to AKT/mTOR signaling; however, it did involve autophagy-related gene expression promoted by AMP-activated protein kinase-mediated transcription factors, such as forkhead box 1. Overall, our findings indicate that MGO-induced cellular damage can be mitigated by autophagy, suggesting that autophagy may be a potential therapeutic target for diseases such as diabetic nephropathy.

Abstract:Diabetes (DB) is a risk factor for osteoarthritis progression. High glucose (HG) is one of the key pathological features of DB and has been demonstrated to induce apoptosis and senescence in chondrocytes. Autophagy is an endogenous mechanism that can protect cells against apoptosis and senescence. The effects of HG on autophagy in cells including chondrocytes have been studied; however, the results have been inconsistent. The current study aimed to elucidate the underlying mechanisms, which could be associated with the contrasting outcomes. The present study revealed that HG can induce apoptosis and senescence in chondrocytes, in addition to regulating autophagy dynamically. The present study demonstrated that HG can cause oxidative stress in chondrocytes and suppress the AMPK pathway in a dose-dependent manner. Elimination of oxidative stress by Acetylcysteine, also called N -acetyl cysteine (NAC), downregulated autophagy and alleviated HG-stimulated apoptosis and senescence, while activation of the AMPK signaling pathway by AICAR not only upregulated autophagy but also alleviated HG-stimulated apoptosis and senescence. A combined treatment of NAC and AICAR was superior to treatment with either NAC or AICAR. The study has demonstrated that HG can suppress autophagy through the AMPK pathway and induce autophagy via oxidative stress in chondrocytes.

Abstract:Objective To investigate the role of autophagy in high glucose-induced podocyte lipid droplet metabolism.Methods (1) Cultured,conditionally immortalized human podocytes (HPC)were divided into normal control group,high glucose group and mannitol group.Oil red O staining and oil red O staining extraction assay was used to observe the degree of lipid accumulation;Protein level of SREBP-1 was analyzed by Western blotting.(2) HPC were cultured and divided into normal control group,high glucose group,high glucose+3-methyladenine (3-MA) group,and mannitol group.Acridine orange staining was used to observe the formation of autophagosomes.Western blotting was used to detect the protein levels of beclin-1 and LC3-Ⅱ/LC3-Ⅰ.Oil red O staining and oil red O staining extraction assay was used to observe the degree of lipid accumulation;Western blotting was used to analyze the expression of SREBP-1.Results (1) Compared with the normal control group,the lipid accumulation in the high glucose group was increased and the lipid metabolism related molecule SREBP-1 was up-regulated (P ＜ 0.05);There was no significant difference between the normal control group and the mannitol group in lipid accumulation (P ＞ 0.05).(2) Compared with the normal group,the number of autophagosomes was increased and autophagy-related proteins beclin-1 and LC3-Ⅱ/LC3-Ⅰ were up-regulated in high glucose group (all P ＜ 0.05).After intervened with 3-methyladenine,a significant decrease in autophagosomes was observed;Protein levels of autophagy-related proteinsbeclin-1 and LC3-Ⅱ/LC3-Ⅰ were decreased (all P ＜ 0.05);The lipid droplets in the high glucose+3-MA group was decreased and lipid metabolism related molecule SREBP-1 was down-regulated (all P ＜0.05).Conclusion Autophagy may be involved in the process of high-glucose-induced podocyte lipid accumulation by affecting SREBP-1 expression,and inhibition of autophagy can alleviate the high-glucose-induced podocyte lipid accumulation.

Abstract:High glucose-induced endothelial cell dysfunction is considered to be the main cause of the development of vascular complications in patients with diabetes. In this study, we examined the effect of high glucose (HG) on apoptosis of human umbilical vein endothelial cells (HUVEC) and investigated the molecular mechanisms involved. HUVEC was exposed to HG and apoptosis was assessed by Death ELISA. Our data showed that HG induced significant apoptosis in HUVEC cells after exposing to HG for 5 days (O.D. value = 0.36±0.07 in HG group vs. 0.13±0.05 in control, p<0.05). HG induced apoptosis in HUVEC is associated with decreased of insulin-like growth factor receptor (IGF-1R) mRNA (4.1±0.6 fold at 72 h, p<0.05) as assessed by real-time PCR and IGF-1R protein (3. 1±0.5 fold at 96 h, p<0.05) as assessed by western blot, which were dose- and time-dependent effects (25 mM of glucose rather than 5 mM at 72 h and 96 h). The effects of HG on reduced expression of IGF-1R and apoptosis were blocked by silencing histone deacetylase 1 (HDAC1) with small interference RNA (siRNA-HDAC1) compared to non-targeting scrambled siRNA. Moreover, HG negatively regulated IGF-1R promoter activity as determined by ChIP (chromatin immunoprecipitation) analysis, which was dependent on HDAC1 since siRNA-HDAC1 attenuated the effects of HG on IGF-1R promoter activity. HG also increased the association of IGF-1R promoter with HDAC1, and decreased the association of IGF-1R promoter with acetylated histone-4. Furthermore, trichostatin A, a specific HDAC inhibitor, relieved the repression of IGF-1R and prevented apoptosis of HUVEC following HG state. These results suggest that HG-induced repression of IGF-1R is mediated by the association of HDAC1 with the IGF-1R promoter, and by the subsequent reduced recruitment of chromatin-modifying proteins, such as histone 4 to the IGF-1R promoter. In conclusion, our data demonstrate that high glucose exposure decreases expression of IGF-1R and decreases the association of acetylated histone-4 with the IGF-1R promoter, and suggest that enhancing acetylation of histone 4 and IGF-1R expression may be a useful strategy to prevent diabetic-induced apoptosis of endothelial cells. Abstract:BACKGROUND:Autophagy is a self-degradative process that is important for balancing sources of energy at critical times in development and in response to nutrient stress. Retinal pigment epithelium (RPE) works as the outer blood retina barrier and is vulnerable to energy stress-induced injury. However, the effect of high glucose treatment on autophagy is still unclear in RPE.METHODS:Transmission electron microscopy was used to detect the generation of autophagosome. Small interfering RNA (siRNA) and MTT was used to determine the effect of autophagy on cell viability. Western blots and immunohistochemistry were used to detect the expression pattern of autophagic markers, including LC3 and p62.RESULTS:High glucose treatment results in a significant increase in the generation of autophagosome and altered expression of LC3 and p62. High glucose-induced autophagy is independent of mTOR signaling, but is mainly regulated via ROS-mediated ER stress signaling.CONCLUSION:In the scenario of high glucose-induced oxidative stress, autophagy may be required for the removal of damaged proteins, and provide a default mechanism to prevent high glucose-induced injury in RPE.
HMGB1 downregulation in retinal pigment epithelial cells protects against diabetic retinopathy through the autophagy-lysosome pathway

Abstract:<span>Diabetic retinopathy (DR) is a serious complication of diabetes mellitus and currently one of the major causes of blindness. Several previous studies have demonstrated that autophagy, which is regulated by HMGB1 (high mobility group box 1), is involved in DR development. However, the role of autophagy in DR is quite complicated in that it promotes pericyte survival in early DR, whereas excessive autophagy causes excess stress and leads to necrosis. Therefore, this study aimed to investigate the relationship between HMGB1, the macroautophagy/autophagy-lysosome pathway, and DR, as well as their underlying molecular mechanisms. In brief, the relationship between high glucose (HG) and the autophagy-lysosome pathway was examined in retinal pigment epithelial (RPE) cells. The relationship was studied by detecting classical autophagic features, and siRNAs targeting HMGB1 and pharmacological regulators were used to explore the role of the autophagy-lysosome pathway in DR development. The results demonstrated that HG inhibited autophagy and diminished the degradative capacity of autophagy due to lysosome membrane permeabilization (LMP). In addition, HMGB1 was found to be involved in LMP via the CTSB (cathepsin B)-dependent pathway, but not the CTSL (cathepsin L)-dependent pathway. Knockdown of HMGB1 expression rescued LMP, restored the degradative capacity of autophagy, decreased the expression of inflammatory factors and VEGF (vascular endothelial growth factor), and protected against apoptosis in RPE cells in the early stages of DR.</span>
High Glucose: Activating Autophagy and Affecting the Biological Behavior of Human Lens Epithelial Cells

Abstract:AIM: To clarify the effect of autophagy on human lens epithelial cells (HLECs) under high glucose conditions. METHODS: HLECs were cultured with different concentrations of glucose and 3-methyladenine (3-MA); the expression of autophagy-related protein LC3B was detected by Western blotting and immunofluorescence histochemistry. The migration of HLECs was quantified by scratch wound assay and the expression of transforming growth factor-β (TGF-β) was measured by real-time polymerase chain reaction. RESULTS: Compared with 5 mmol/L normal glucose treatment, 40 mmol/L glucose treatment can significantly increase the generation of autophagosome in HLECs, which could be inhibited by 0.375 mmol/L 3-MA treatment. The migration of HLECs and the expression of TGF-β in HLECs induced by high glucose were significantly suppressed by 0.375 mmol/L 3-MA treatment. CONCLUSION: Autophagy promotes HLECs cell migration and increases the expression of TGF-β after exposed to high glucose, which may relate to the development of diabetic cataract.

Abstract:Cardiac microvascular endothelial cells (CMECs) dysfunction is an important pathophysiological event in the cardiovascular complications induced by diabetes. However, the underlying mechanism is not fully clarified. Autophagy is involved in programmed cell death. Here we investigated the potential role of autophagy on the CMECs injury induced by high glucose. CMECs were cultured in normal or high glucose medium for 6, 12 and 24 h respectively. The autophagy of CMECs was measured by green fluorescence protein (GFP)-LC3 plasmid transfection. Moreover, the apoptosis of CMEC was determined by flow cytometry. Furthermore, 3-Methyladenine (3MA), ATG7 siRNA and rapamycin were administrated to regulate the autophagy state. Moreover, Western blotting assay was performed to measure the expressions of Akt, mTOR, LC3 and p62. High glucose stress decreased the autophagy, whereas increased the apoptosis in CMECs time dependently. Meanwhile, high glucose stress activated the Akt/mTOR signal pathway. Furthermore, autophagy inhibitor, 3-MA and ATG7 siRNA impaired the autophagy and increased the apoptosis in CMECs induced by high glucose stress. Conversely, rapamycin up-regulated the autophagy and decreased the apoptosis in CMECs under high glucose condition. Our data provide evidence that high glucose directly inhibits autophagy, as a beneficial adaptive response to protect CMECs against apoptosis. Furthermore, the autophagy was mediated, at least in part, by mTOR signaling.
