Abstract:Over the past few years, progress in electrochemical biosensors for DNA analysis is outstanding. This article briefly reviewed the principals of such biosensors, followed by a selection of topics that are of particular current interest. Selectivity and sensitivity are the two major challenges in DNA analysis. The former can be accomplished using peptide nucleic acids as probes, or relying on the DNA-mediated electron transfer. Special emphasis has been given to the efforts for high sensitivity, involving combination with polymerase chain reaction techniques, enzyme-labeled methods, direct label-free detection and nano-based techniques.

Abstract:Electrochemiluminescence (ECL) detection is widely applied in many fields, including chemical measurement, biological analysis, and clinic tests, due to its high sensitivity. Currently, the fast development of many new electrochemical luminophores is continuously improving the ECL-based detection ability. Besides the enhancement of luminescence emission for a high detection sensitivity, minimizing the effect of co-reactants on ECL detection and achieving multiple analysis in one sample are also the main directions in this field. This review focuses on a summary of recently prepared new luminophores to achieve the three aims mentioned above. Especially, the review is composed by three parts, focusing on the luminophores or materials with high ECL efficiency, self-enhancing properties, and multi-color ECL luminophores. The fabrication of biosensors using these molecules is also reviewed to exhibit the advances in biological applications.

Abstract:Electrochemiluminescence (ECL) is one of the most powerful techniques that meet the needs of analysis and detection in a variety of scenarios, because of its highly analytical sensitivity and excellent spatiotemporal controllability. ECL combined with microscopy (ECLM) offers a promising approach for quantifying and mapping a wide range of analytes. To date, ECLM has been widely used to image biological entities and processes, such as cells, subcellular structures, proteins and membrane transport properties. In this review, we first introduced the mechanisms of several classic ECL systems, then highlighted the progress of visual biosensing and bioimaging by ECLM in the last decade. Finally, the characteristics of ECLM were summarized, as well as some of the current challenges. The future research interests and potential directions for the application of ECLM were also outlooked.

Abstract:The recent progress in electrochemiluminescent (ECL) assay is reviewed. This review begins with the fundamental researches in ECL, including the discovery of new ECL-active species, such as biochemical, organic and metallorganic materials, digital modeling of ECL process, the flow cells used in ECL assay, and electrochemiluminescent sensor. The application of ECL in environmental analysis, immunoassay, nucleotide acid hybridization sensor. The applications of ECL in environmental analysis, immunooassay, nucleic acid hybridization assay, and other aspects are reviewed with the latest references in detail. Finally, the main problems in the further investigation are outlooked, so are its prospects.

Abstract:Inspired by dual-wavelength fluorescence ratiometric method which could reduce the influence from the environmental change, here, we present a novel dual-potential electrochemiluminescence (ECL) ratiometric sensing approach. CdS nanocrystal (NC) and luminol as two different ECL emitters are employed. ECL from CdS NCs coated on glassy carbon electrode at -1.25 V (vs SCE) could be quenched by closely contacted Pt nanoparticles (NPs) via a biological binding event, while ECL from luminol at +0.45 V (vs SCE) could be enhanced by the same Pt NPs, in the presence of their common coreactant of H₂O₂. Thus, the quenching of ECL from CdS NCs and the enhancement of ECL from luminol could indicate the same biological binding event. With the mp53 oncogene as a model DNA molecule, a molecular beacon (MB) containing a 20-base loop, which is complementary with the mp53 oncogene, is immobilized on CdS NCs/GCE first; Pt NPs are then captured on CdS NCs surface by DNA hybridization between the MB and mp53 oncogene labeled on Pt NPs. By measuring the ratio of ECL intensities at two excitation potentials, this approach could sensitively detect the concentration of target DNA in a wide range from 5.0 fM to 1.0 pM. The sensing scheme is general and can be utilized for many other biological binding events.

Abstract:Electrochemiluminescence (ECL) is a light-emitting process triggered by the high energy redox between electrochemically oxidized and reduced luminophores or some coreactive intermediate radicals, representing a blooming hot topic over decades with a wide variety of bioanalytical applications. Due to the superb sensitivity, ultralow background noise, specificity, ease of integration, and real-time and in situ analysis, ECL has been developed as a convenient and versatile technique for immunodiagnostics, nucleic acid analysis, and bioimaging. Discovering highly-efficient ECL emitters has been a promising subject that will benefit the development of sensitive bioanalytical methods with prominent potential prospects. To date, the interdisciplinary integrations of electrochemistry, spectroscopy, and nanoscience have brought up the continuous emergences of novel nanomaterials which can be flexibly conjugated with specific bio-recognition elements as functional ECL emitters for bioassays. Therefore, a critical overview of recent advances in developing highly-efficient ECL emitters for ultrasensitive detection of protein biomarkers is presented in this review, where six kinds of the most promising ECL nanomaterials for biosensing and imaging of various disease-related protein biomarkers are separately introduced with references to representative works. Finally, this review discusses the ongoing opportunities and challenges of ECL emitters in developing advanced bioassays for single-molecule analysis and spatiotemporally resolved imaging of protein biomarkers with future perspectives.

Abstract:Understanding biological mechanisms operating in cells is one of the major goals of biology. Since heterogeneity is the fundamental property of cellular systems, single cell measurements can provide more accurate information about the composition, dynamics, and regulatory circuits of cells than population-averaged assays. Electrochemiluminescence (ECL), the light emission triggered by electrochemical reactions, is an emerging approach for single cell analysis. Numerous analytes, ranging from small biomolecules such as glucose and cholesterol, proteins and nucleic acids to subcellular structures, have been determined in single cells by ECL, which yields new insights into cellular functions. This review aims to provide an overview of research progress on ECL principles and systems for single cell analysis in recent years. The ECL reaction mechanisms are briefly introduced, and then the advances and representative works in ECL single cell analysis are summarized. Finally, outlooks and challenges in this field are addressed.

Abstract:Electrochemiluminescence (ECL) is a widely used analytical technique with the advantages of high sensitivity and low background signal. The recent and rapid development of electrochemical materials, luminophores, and optical elements significantly increases the ECL signals and, thus, ECL imaging with enhanced spatial and temporal resolutions is realized. Currently, ECL imaging is successfully applied to high-throughput bioanalysis and to visualize the distribution of molecules at single cells. Compared with other optical bioassays, no optical excitation is involved in imaging, so the approach avoids a background signal from illumination and increases the detection sensitivity. This review highlights some of the most exciting developments in this field, including the mechanisms, electrode designs, and the applications of ECL imaging in bioanalysis and at single cells and particles.

Abstract:Sensitive and accurate detection of biological analytes, such as proteins, genes, small molecules, ions, cells, etc., has been a significant project in life science. Signal amplification is one of the most effective approaches to improve the sensitivity of bioanalysis. Taking advantage of specific base pairing, programmable operation, and predictable assembly, DNA is flexible and suitable to perform the signal amplification procedure. In recent years, signal amplification strategies by means of DNA technology have been widely integrated into the construction of electrochemiluminescence (ECL) biosensors, achieving desirable analytical performance in clinical diagnosis, biomedical research, and drug development. To the best of our knowledge, these DNA signal amplification technologies mainly include classical polymerase chain reaction, and various amplification approaches conducted under mild conditions, such as rolling circle amplification (RCA) or hyperbranched RCA, cleaving enzyme-assisted amplification, DNAzyme-involved amplification, toehold-mediated DNA strand displacement amplification without enzyme participation, and so on. This review overviews the recent advancements of DNA signal amplification strategies for bioanalysis in the ECL realm, sketching the creative trajectory from strategies design to ultrasensitive ECL platform construction and resulting applications.

Abstract:Electrochemiluminescence (ECL) is a luminous phenomenon in which the excited state luminophore is generated by dark electrochemical reactions in solutions. Due to its near-zero background, high sensitivity, good spatiotemporal controllability, fast detection and wide dynamic range, ECL has manifested itself to be one of the most successful techniques in in vitro diagnosis and clinical detection. In addition, ECL imaging possesses the unique advantages such as high-throughput and visualization. As a powerful surface analysis technology, ECL imaging has been successfully employed in material surface/interface analysis and bioanalysis. Given that the ECL intensity is highly dependent on the properties of electrode surface, ECL imaging can be used to investigate electron-transfer properties and the distribution of electrochemical activity of chemically-modified electrode. In the first part of this review, we briefly introduce the background of ECL technology and describe the generation mechanisms of ECL systems. Then we focus on the recent research progress of imaging analysis based on ECL microscopy, including mechanism rationalization, latent fingerprint visualization and single cell analysis. Finally, some perspectives and future directions of ECL are presented.

Abstract:In recent years, increasing attention has been paid to DNA mediated chemiluminescence (CL) bioassays due to the merits provided by both of CL and functional DNAs. CL exhibits the advantages of low background, high sensitivity and simple equipment. Meanwhile, various functional DNAs facilitate the target recognition, signal transduction and signal amplification of the CL bioassays. However, a comprehensive summarization focused on the DNA-mediated CL bioassays is still lacking now. In this review, the CL systems for biosensing and approaches of DNA-based target recognition and signal transduction for CL bioassays were summarized. The strategies and mechanisms of signal amplification and background reducing for ultrasensitive CL assays were also discussed in detail. The applications for the detection of various analytes including nucleic acids, proteins, toxins, environmental pollutants, and metal ions were systematically reviewed as well. Finally, the further prospects and challenges were highlighted to promote the advancement of DNA-mediated CL bioassays.

Abstract:In this work, we firstly enunciated the presence of synergy effect between the electron and energy transfer in an electrochemiluminescence (ECL) system involving the CdS NCs and Au NPs, based on which an amplified ECL biosensor was constructed for sensitive detection of DNA-protein interactions. Specifically, Au NPs modified on electrode, following by assembled probe DNA. Then the modified GCE was hybridized with SiO(2)@CdS/DNA conjugates. With DNA duplex as a rigid spacer, Au NPs could not only accelerate the electron transfer but also produce the surface plasma resonance upon excited ECL of CdS NCs. However, the enhanced ECL could be greatly suppressed by the binding of target protein. The prepared biosensor possesses excellent analytical performance with the linear range of target protein from 0.015 to 150 nM.

Abstract:Electrochemiluminescence (ECL) has received considerable interest for broad applications due to the potential remarkably high sensitivity and extremely wide dynamic range. In recent years, due to the unique physical (structural, electronic, magnetic and optical) and chemical (catalytic) properties of nanomaterials (NMs), great efforts have been made to investigate their application in ECL. In this review, we present a general description of ECL related to NMs as the emitter. It mainly focuses on the basic mechanisms and the development of an ECL sensing platform fabricated from semiconductor nanocrystals and metal nanoclusters. Finally, we conclude with a look at the future challenges and prospects of the development of ECL.

Abstract:High-performance nanomaterials have been seen as a new generation of electrochemiluminescence (ECL) probes or emitters for their finely tunable structure and concomitant remarkable properties, guaranteeing the prospective applications in the analysis and diagnosis devices with superior performances. The structure–activity relationships of ECL nanoprobes in nanoscale are presenting milestone in understanding of the ECL microscopic behaviors and mechanisms, and guide the exploitation of novel ECL probes. In this mini-review, we summarized the recent development of novel ECL probes based on the nanomaterials. The mechanism and relationships between their structure as well as the active sites and functionality were revealed. In addition, the design and regulation of the ECL nanoprobes were emphasized for the biosensing and imaging application. Finally, the potential prospect of the ECL nanoprobes, design, and their applications were discussed.

Abstract:Electrochemiluminescence (ECL) analysis has become a powerful tool in recent biomarker detection and clinic diagnosis due to its high sensitivity and broad linear range. To improve the analytical performance of ECL biosensors, various advanced nanomaterials have been introduced to regulate the ECL signal such as graphene, gold nanomaterials, and quantum dots. Among these nanomaterials, some plasmonic nanostructures play important roles in the fabrication of ECL biosensors. The plasmon effect for the ECL signal includes ECL quenching by resonant energy transfer, ECL enhancement by surface plasmon resonance enhancement, and a change in the polarized angle of ECL emission. The influence can be regulated by the distance between ECL emitters and plasmonic materials, and the characteristics of polarization angle-dependent surface plasmon coupling. This paper outlines the recent advances of plasmonic based ECL biosensors involving various plasmonic materials including noble metals and semiconductor nanomaterials. The detection targets in these biosensors range from small molecules, proteins, nucleic acids, and cells thanks to the plasmonic effect. In addition to ECL biosensors, ECL microscopy analysis with plasmonic materials is also highlighted because of the enhanced ECL image quality by the plasmonic effect. Finally, the future opportunities and challenges are discussed if more plasmonic effects are introduced into the ECL realm.

Abstract:The continuous emergence of novel luminophores has been leading the development of electrochemiluminescence (ECL) analysis since the ECL phenomenon of silicon nanoparticles was first reported in 2002. Nanomaterial-based luminophores termed as nanoemitters exhibit great application potential in ECL bioanalysis. In this review, we systematically summarize the research progress of various types of ECL nanoemitters, such as semiconductor quantum dots (QDs), carbon dots (CDs), polymer dots (Pdots), aggregation-induced emission nanoparticles (AIE dots), and other nanoemitters including luminophor-edoped silica nanoparticles, metal-organic frameworks, covalent organic frameworks, and metal nanoclusters. We mainly emphasize the characteristic of these nanoemitters and their applications in ECL biosensing and bioimaging by using them as labels, and highlight some impressive cases in the last five years. Finally, we discuss current challenges in the field of ECL nanoemitters and point out their prospects.

Abstract:This review presents a novel kind of electrochemical biosensors based on DNA configuration switch. Special DNA probes (stem-loop or aptamers etc.) fixed on an electrode surface dramatically change their configuration on the combination of the target molecule giving birth to a detectable electrochemical signal. This strategy attracts enormous research interest because of its easier operation and higher specificity. To this day, its analysis performance has been markedly developed with an expanding application. We here summarize the development of these sensors.

Abstract:Electrochemiluminescence (ECL), also called electrogenerated chemiluminescence, is luminescence that is produced by chemical reactions triggered by electrochemical method. ECL imaging is a novel imaging technique, which can simultaneously provide two kinds of signals of both electrochemistry and optical image. Over the past two decades, ECL imaging has been not only a powerful tool for investigating the fundamental scientific questions such as ECL mechanisms and reaction kinetics, but also a versatile analytical technique for detection of a wide range of analytes including small molecules, DNA, proteins, and cells. In the first part of this review, we briefly describe the reaction mechanisms of ECL generation. Then the review focuses on the research progress on the ECL imaging approach. It is basically introduced from the following five aspects: (i) visualization of electroactive sites on surfaces, (ii) imaging analysis at the single-bead and single-cell levels, (iii) array bioanalysis, (iv) multi-color ECL imaging, and (v) paper chip based on ECL imaging. Finally, some perspectives and future directions in this active research area are presented.

Abstract:Abstract Electrochemiluminescence (ECL) as an analytical technology with a perfect combination of electrochemistry and spectroscopy has received considerable attention in bioanalysis due to its high sensitivity and broad dynamic range. Given the selectivity of bio‐recognition elements and the high sensitivity of the ECL analysis technique, ECL biosensors are powerful platforms for the sensitive detection of biomarkers, achieving the accurate prognosis and diagnosis of diseases. MicroRNAs (miRNAs) are crucial biomarkers involved in a variety of physiological and pathological processes, whose aberrant expression is often related to serious diseases, especially cancers. ECL biosensors can fulfill the highly sensitive and selective requirements for accurate miRNA detection, prompting this review. The ECL mechanisms are initially introduced and subsequently categorize the ECL biosensors for miRNA detection in terms of the quenching agents. Furthermore, the work highlights the signal amplification strategies for enhancing ECL signal to improve the sensitivity of miRNA detection and finally concludes by looking at the challenges and opportunities in ECL biosensors for miRNA detection.

Abstract:Electrochemiluminescence (ECL) is the chemiluminescence triggered by electrochemical reactions. Due to the unique excitation mode and inherent low background, ECL has been a powerful analytical technique to be widely used in biosensing and imaging. As an emerging ECL luminophore, semiconductor quantum dots (QDs) have apparent advantages over traditional molecular luminophores in terms of luminescence efficiency and signal modulation ability. Therefore, the development of an efficient ECL system with QDs as luminophores is of great significance to improve the sensitivity and detection flux of ECL biosensors. In this review, we give a comprehensive summary of recent advances in ECL using semiconductor QDs as luminophores. The luminescence process and ECL mechanism of semiconductor QDs with various coreactants are discussed first. Specifically, the influence of surface defects on ECL performance of semiconductor QDs is emphasized and several typical ECL enhancement strategies are summarized. Then, the applications of semiconductor QDs in ECL biosensing are overviewed, including immunoassay, nucleic acid analysis and the detection of small molecules. Finally, the challenges and prospects of semiconductor QDs as ECL luminophores in biosensing are featured.
