Abstract:Carbonate rhizoliths and related features are environmental and ecological indicators of not only palaeosols, but also modern soils. They designate subaerial vadose and pedogenic diagenesis environments. Rhizoliths are commonly used as a tool for paleoenvironmental reconstructions, but uncertainty remains regarding their formation mechanisms. Field characteristics of the dune rhizoliths in the Badain Jaran Desert, NW China were analyzed to investigate formation mechanisms and interpret paleoenvironmental significance. Systematic field sampling and intensive examination showed that the tube-like rhizoliths without morphological structures of plant roots have been placed at the dune soil surface following erosion and weathering but the underground rhizoliths are still connected to the dead Artemisia roots. Rhizoliths occurred only along the windward, long gentle slopes of mega-dunes. Their distribution patterns of both types in connection with plant distribution and landscapes have been studied. Their petrological and mineralogical characters were performed in detail using microscopic and spectroscopic techniques (cathodoluminescence, scanning electronic microscopy and energy dispersive X-ray spectroscopy) in laboratory. Altogether, field and laboratory analyses showed that rhizolith cement is calcium carbonate and despite the high porosity of soil and high aridness of climate, soil moisture plays an important role in sustaining Artemisia growth and rhizolith formation. Desert rhizoliths can therefore be used as a proxy to infer local soil moisture and plant communities. Moreover, as soil moisture content is related to climate and landscape position, rhizoliths could indirectly be indicators of aboveground environmental conditions, e.g. annual precipitation or humidity.

Abstract:Desert rhizoliths are generally found as weathered, broken and scattered samples on dune field surface, but rarely in-situ in their initial states buried under the soil of desert in the Badain Jaran Desert, northwest China. This study offers an assessment of the morphological, mineralogical, and chemical properties of intact and in-situ rhizoliths found in soils of swales and depressions among dune chains. The characteristics of these rare and precious objects were assessed using optical polarizing microscopy, cathodoluminescence, scanning electronic microscopy, radiocarbon dating, and stable isotopic analyses, providing the opportunity for discussion of the rhizolith formation mechanisms and associated environmental conditions. Field and laboratory investigations showed that the in-situ intact rhizoliths were formed only in the places where Artemisia shrubs are living, and the remaining root relicts within rhizoliths belong to this species. The spatial distribution of rhizoliths also suggested that low topographic positions on a landscape provided soil moisture, and redox environments favored rhizolith formation. A semi-closed redox environment in the subsoil at swales and depressions, where water is always present, along with the sandy soil texture, facilitated fast water percolation to deeper depths and condensation. Such a soil environment not only provides water for Artemisia growth, but also for the weathering of minerals such as felspars and calcite from primary carbonates, and for the decomposition of root relicts. Furthermore, harsh climatic conditions, such as strong winds and solar radiation, led to water evaporation through dead root channels and triggered the calcification along the root relicts. The entrapped lithogenic carbonates and to a lesser extent the decomposition of Artemisia roots provided the carbon sources for the rhizoliths formation, while the weathering of soil minerals, particularly feldspars and carbonates, was the main source of Ca. Rhizoliths in the Badain Jaran desert formed relatively quickly, probably over a few soil drying episodes. This led to the entrapment of a large quantity of lithogenic carbonates (more than 90% of carbon) within rhizolith cement. The re-dissolution of the entrapped lithogenic carbonates in rhizolith tubes should be taken into account in the paleoenvironmental interpretation of 14 C ages, the latter suggesting that rhizoliths formed during the Holocene (~ 2053 years cal BP, based on root organic relicts).

Pristine carbonate rhizoliths with co-existing plant root relicts in the Tengeri Desert, NW China were studied, with a combination of intensive field observations and laboratory methods such as microscopy, scanning electronic microscopy, energy dispersive X-ray spectra, radiocarbon dating, and isotope mass spectrometer. The field observations revealed that the pristine rhizoliths are only present at the sites where Artemisia sphaerocephala Krasch are growing i.e. in swales among sand dunes. Soil moisture of the swales is the main controlling factor of rhizoliths formation. It is in turn affected by the soil physical properties, landscape position, and climate variability, consistent with the locations of sampled rhizoliths in the swales where Artemisia plants are mostly distributed. The 14C AMS dating indicated that the rhizoliths are much older (4000–5000 years) than their co-existing modern plant root relicts in agreement with field observations. Morphological, mineralogical and isotopic analyses revealed that carbon sources used for the rhizoliths formation were partially derived from decomposing plant roots but with significant contribution from dissolution of lithogenic carbonates. The calcium sources were suggested to be the in situ weathering of minerals (mostly lithogenic carbonates) and the pressure-dissolution of carbonates. Enough CO2 from the root decomposition have triggered carbonate accumulation around the root to form rhizoliths. Other minor chemical components of the root are S, N, P, which produce acidic water with the negative ions of SO42−, NO3–, PO43-, have also favored acidic soil environment and enhanced carbonate dissolution and mineral weathering. Redox environment around Artemisia roots were also observed to be a key factor for the pristine rhizolith formation. The pristine rhizoliths were preferentially formed in semi-closed redox condition with water nearly always available at intermediate depths. In addition, they were formed through carbonate epidiagenesis in shallow soils of the desert. Altogether, our results showed that the formation of the pristine rhizoliths was affected by the combination of several environmental factors. This led us to propose a conceptual model of rhizolith formation in desert soils.

Abstract:Abstract Root zone microbial structure is particularly complex in plants with rhizosheaths, and greater understanding of the rhizosheath may play an important role in the future development of sustainable agricultural practices. However, one important reason to focus study on rhizosheath microbial structure is that there is no definite method for rhizosheath separation. The aim of this study was to explore rhizosheath isolation methods and the diversity characteristics of microorganisms around the rhizosphere. In this study, we isolated the rhizosheath of Stipa grandis , a dominant species in desert steppe, and the microorganisms in the roots, root epidermis, rhizosheath and rhizosphere soil were extracted and sequenced by 16S rRNA and ITS. The alpha diversity index of bacteria in Stipa grandis rhizosphere soil was the greatest, followed by rhizosheath, and the alpha diversity index of endophytic bacteria in root system was the smallest. The alpha diversity index of fungi in the rhizosheath and rhizosphere soil were significantly higher than that in the root epidermis and root system. There were significant differences in bacterial community structure between the root epidermis, endophytic bacteria, rhizosheath and rhizosphere soil. Unlike bacterial community structure, the community structure of fungi in the root epidermis was similar that of endophytic fungi, but significantly different from those in rhizosheath and rhizosphere soil. This study demonstrated a feasible method for separating plant rhizosheath and root epidermis. We suggest that the root epidermis can act as the interface between the host plant root and the external soil environment. We will have to re-examine the biological and ecological significance of rhizosheath and microorganisms in rhizosheath, as well as the mechanism explaining the close relationship of the rhizosheath and the plant root epidermis. This study provides theoretical and technical guidance for the isolation of the plant rhizosheath and the study of microorganisms in plant rhizosheath.

Dynamics of soil biota and nutrients at varied depths in a Tamarix ramosissima-dominated natural desert ecosystem: Implications for nutrient cycling and desertification management

Abstract:The underground community of soil organisms, known as soil biota, plays a critical role in terrestrial ecosystems. Different ecosystems exhibit varied responses of soil organisms to soil physical and chemical properties (SPCPs). However, our understanding of how soil biota react to different soil depths in naturally established population of salinity tolerant Tamarix ramosissima in desert ecosystems, remains limited. To address this, we employed High-Throughput Illumina HiSeq Sequencing to examine the population dynamics of soil bacteria, fungi, archaea, protists, and metazoa at six different soil depths (0-100 cm) in the naturally occurring T. ramosissima dominant zone within the Taklimakan desert of China. Our observations reveal that the alpha diversity of bacteria, fungi, metazoa, and protists displayed a linear decrease with the increase of soil depth, whereas archaea exhibited an inverse pattern. The beta diversity of soil biota, particularly metazoa, bacteria, and protists, demonstrated noteworthy associations with soil depths through Non-Metric Dimensional Scaling analysis. Among the most abundant classes of soil organisms, we observed Actinobacteria, Sordariomycetes, Halobacteria, Spirotrichea, and Nematoda for bacteria, fungi, archaea, protists, and metazoa, respectively. Additionally, we identified associations between the vertical distribution of dominant biotic communities and SPCPs. Bacterial changes were mainly influenced by total potassium, available phosphorus (AP), and soil water content (SWC), while fungi were impacted by nitrate (NO3-) and available potassium (AK). Archaea showed correlations with total carbon (TC) and AK thus suggesting their role in methanogenesis and methane oxidation, protists with AP and SWC, and metazoa with AP and pH. These correlations underscore potential connections to nutrient cycling and the production and consumption of greenhouse gases (GhGs). This insight establishes a solid foundation for devising strategies to mitigate nutrient cycling and GHG emissions in desert soils, thereby playing a pivotal role in the advancement of comprehensive approaches to sustainable desert ecosystem management.

Abstract:Soil microorganisms play important roles in vegetation establishment and soil biogeochemical cycling. Ammodendron bifolium is a dominant sand-fixing and endangered plant in Takeermohuer Desert, and bacterial community associated with this plant rhizosphere is still unclear. In this study, we studied the composition and diversity of bacterial community from A. bifolium rhizosphere and bulk soil at different soil depths (i.e., 0–40 cm, 40–80 cm, 80–120 cm) using traditional bacterial isolation and high-throughput sequencing approaches, and preliminarily analyzed the edaphic factors influencing the structure of bacterial communities. Results showed that Takeermohuer Desert with high salinity has been an oligotrophic environment, while the rhizosphere exhibited eutrophication resulting from high content SOM (soil organic matter) and SAN (soil alkaline nitrogen) compared with bulk soil. The dominant bacterial groups in the desert were Actinobacteria (39.8%), Proteobacteria (17.4%), Acidobacteria (10.2%), Bacteroidetes (6.3%), Firmicutes (6.3%), Chloroflexi (5.6%), and Planctomycetes (5.0%) at the phyla level. However, the relative abundances of Proteobacteria (20.2%) and Planctomycetes (6.1%) were higher in eutrophic rhizosphere, and Firmicutes (9.8%) and Chloroflexi (6.9%) relatively higher in barren bulk soil. A large number of Actinobacteria were detected in all soil samples, of which the most abundant genus was Streptomyces (5.4%) and Actinomadura (8.2%) in the bulk soil and rhizosphere, respectively. The Chao1 and PD indexes in rhizosphere were significantly higher than those in bulk soil at the same soil depth, and tended to decrease with increasing soil depth. Co-occurrence network analyses showed that the keystone species in Takeermohuer Desert were Actinobacteria, Acidobacteria, Proteobacteria, and Chlorofexi. Furthermore, the major environmental factors affecting rhizosphere bacterial community were EC (electrical conductivity), SOM, STN (soil total nitrogen), SAN, and SAK (soil available potassium), while bulk soil were distance and C/N (STC/STN). We concluded that A. bifolium rhizosphere bacterial community is different from non-rhizosphere in composition, distribution, and environmental influencing factors, which will have important significances for understanding their ecological functions and maintaining biodiversity.

Abstract:The soil organisms are extremely important for the land-based ecosystem. There is a growing interest in studying the variety and composition of the entire underground soil organism community at a large ecological scale. Soil organisms show different patterns in relation to soil physiochemical properties (SPPs) in various ecosystems. However, there is limited knowledge regarding their response to soil vertical profiles (SVPs) in monoculture of Alhagi sparsifolia, which is the primary shrub in the deserts of China, and is well-known for its contributions to sand dune stabilization, traditional Chinese medicine, and forage. Here, we investigated the population dynamics of soil bacteria, fungi, archaea, protists and metazoa across six different SVPs ranging from 0 to 100 cm in monoculture of A. sparsifolia, in its natural desert ecosystem. Our findings indicate that the soil biota communities displayed a declining pattern in the alpha diversity of bacteria, protists, and metazoa with an increase in soil depth. However, the opposite trend was observed for fungi and archaea. The beta diversity of soil biota was significantly affected by SVPs, particularly for metazoa, fungi and protists as revealed by Non-Metric Dimensional Scaling. The most prevalent soil bacterial, fungal, archaeal, protist, and metazoa classes were Actinobacteria, Sordariomycetes, Nitrososphaeria, Filosa-Sarcomonadea, and Nematoda, respectively. The correlation among vertical distribution of the most abundant biotic communities and variations in SPPs exhibited that the variations in total carbon (TC) and total nitrogen (TN) had the most significant influence on bacterial changes, while available potassium (AK) had an impact on fungi. Archaea were affected by TC and pH, protists by the C/N-Ratio and TP, and metazoa by TN, AK, and soil water capacity (SWC). Collectively, our findings provide a new perspective on the vertical distribution and distinct response patterns of soil biota in A. sparsifolia monoculture under natural desert ecosystem of China.

Abstract:Qinghai‐Tibet Plateau is facing a serious environmental and ecological problem of Meadow degradation. Toxic weed invasion is a typical characteristic of grassland degradation. Soil microbial community composition is sensitive to environmental changes; however, the effects of poisonous weed expansion on soil bacterial communities are unclear. Here, we investigated the effects of Stellera chamaejasme L. expansion on the rhizosphere soil bacterial community structure and function using high‐throughput sequencing. The results showed that expansion of Stellera chamaejasme L. changed soil nitrogen(e.g., total nitrogen [TN, −39.02%], available nitrogen [AN, −32.95%]) and other soil nutrients. This study investigates intricate dynamics of soil animal communities within this desert ecosystem, with a particular focus on vertical profile variations beneath four dominant shrub species (AS-Alhagi sparsifolia, KC-Karelinia caspia, TR- Tamarix ramosissima, CC- Calligonum caput-medusae). Utilizing comprehensive soil sampling and metagenomics techniques, we reveal the diversity and distribution patterns of soil animal communities from the soil surface down to deeper layers (0-100 cm). Our research outcomes have unveiled that Nematoda and Arthropoda emerge as the most predominant classes of soil animals across all studied shrubs. Specifically, Nematoda exhibited notably high abundance in the KC area, while Arthropoda thrived predominantly in the TR region. We also observed a linear decrease in Nematoda populations as soil depth increased, consistent among all shrub species. Moreover, the highest Shannon diversity within soil animal communities was recorded in the KC area, underscoring a trend of declining alpha diversity in the AS region and an increase in other shrub areas as soil depth increased. Notably, the zones dominated by CC and TR displayed the highest levels of beta diversity. Our correlation analysis of soil animals and environmental factors has pinpointed soil water content, available phosphorus, and available potassium as the most influential drivers of variations in the top-classified soil animal communities. This study provides insights into soil animals in deserts, supporting future research to preserve these fragile deserts and enhance our understanding of life below the surface in challenging ecosystems.
Response of Three Dominant Shrubs to Soil Water and Groundwater along the Oasis-Desert Ecotone in Northwest China

Abstract:The ecotone from oasis to desert is an important area for combating sandy desertification. Three dominant desert shrubs (Nitraria tangutorum, Calligonuum mongolicum, Haloxylon ammodendrori) were selected in Minqin Oasis, Northwest China, to determine the groundwater level; soil water potential; and change of the three shrubs in density, coverage, and biomass along the natural and seminatural oasis-desert ecotone (ODE), respectively. The results indicated that traits of desert plant interaction with the topsoil water and groundwater depth along the ODE play an important role in generating complex desert vegetation spatial dynamics. Some natural desert plant species with shallow root systems will distribute themselves according to distribution of topsoil water. Thus, the distribution of Nitraria tangutorum had a decreasing trend in distribution along ODE. Calligonuum mongolicum occurs in different trends in natural and seminatural ODE due to utilizing groundwater as well as topsoil water. Some plant species with deep roots, such as Haloxylon ammodendron, will show more degradation near oases and will exhibit an ascending trend along ODE. Therefore, it is of primary importance to protect the integrity of groundwater depth in order to protect the stability of the oasis-desert ecotones.
