Mechanistic Study of the Synergistic Antibacterial Activity of Combined Tetracycline and Copper Oxide Nanoparticles

Abstract:The expanding use of tetracycline (TET) and Copper oxide nanoparticles (CuO NPs) raised concerns about their potential influences on environment. However, joint impacts of TET and CuO NPs on activated sludge (AS) process were rarely documented. This study investigated variations of flocculation, sedimentation, dewaterability, as well as nitrogen and COD removal of AS under individual and combined exposure (60 d) of TET (1 mg/L) and CuO NPs (2 mg/L). The results showed that individual and combined exposures of TET and CuO NPs inhibited the flocculation ability, induced the non-filamentous bulking, and destroyed the dewatering performance of the sludge, and these phenomena were more serious under combined exposure, which could be attributed to greater promotion on sludge EPS secretion (especially LB-EPS) at this time. On the other hand, TET exposure alone didn’t affect TN removal, while CuO NPs exposure alone inhibited TN removal due to destroying denitrification process (such as reduced NR, NIR and ETS activity), and this inhibition would be enhanced when CuO NPs was combined with TET by bring higher copper ions. 16S rRNA sequencing found that after long-term exposure, compared to the control and individual exposure groups, the co-presence of TET and CuO NPs resulted in a significant decrease in microbial community richness, diversity, and OTU numbers. Microbial community structure were reformed. According to the results of mechanism study, it is implied that under the joint effect of TET and CuO NPs, there was a stronger selective pressure on the microorganisms for copper ions, resulting in a more toxic composite exposure.
