Abstract:Research related to lignin valorization into fine chemicals is an extremely demanding task due to the wide distribution and low economic value of depolymerization products. Benzoic acid derivatives (BADs) are the final products of the selective oxidation of the lignin side chain (i.e., p-hydroxybenzoic acid, vanillic acid, syringic acid, and benzoic acid), with favorable processibility functionality, showing great potential in the synthesis of active pharmaceutical ingredients (APIs) containing ester and carbonyl groups. Herein, this tutorial review presents our views on lignin utilization, especially using lignin-based benzoic acid derivatives (LBADs) as raw materials for the synthesis of APIs, with the aim of providing a greener, more eco-friendly approach for providing a sustainable route for the production of APIs. We first introduce the conversion of lignin or lignin platform compounds via the oxidative cleavage of C-C and C-O bonds to LBADs. Subsequently, focusing on the functional group modification strategy for the conversion of LBADs, we raise a "Demand Orientation" concept and several typical API synthesis routes. This tutorial review provides green production routes from LBADs in six instances (i.e., trimebutine, aniracetam, diethylstilbestrol, dopamine, acetaminophen, oxybuprocaine). Finally, several others APIs from LBADs via theoretically feasible and sustainable routes are proposed. Some personal perspectives are provided to highlight the opportunities within this attractive field.
In Vitro Enzymatic Depolymerization of Lignin with Release of Syringyl, Guaiacyl, and Tricin Units.

Abstract:ABSTRACT New environmentally sound technologies are needed to derive valuable compounds from renewable resources. Lignin, an abundant polymer in terrestrial plants comprised predominantly of guaiacyl and syringyl monoaromatic phenylpropanoid units, is a potential natural source of aromatic compounds. In addition, the plant secondary metabolite tricin is a recently discovered and moderately abundant flavonoid in grasses. The most prevalent interunit linkage between guaiacyl, syringyl, and tricin units is the β-ether linkage. Previous studies have shown that bacterial β-etherase pathway enzymes catalyze glutathione-dependent cleavage of β-ether bonds in dimeric β-ether lignin model compounds. To date, however, it remains unclear whether the known β-etherase enzymes are active on lignin polymers. Here we report on enzymes that catalyze β-ether cleavage from bona fide lignin, under conditions that recycle the cosubstrates NAD+ and glutathione. Guaiacyl, syringyl, and tricin derivatives were identified as reaction products when different model compounds or lignin fractions were used as substrates. These results demonstrate an in vitro enzymatic system that can recycle cosubstrates while releasing aromatic monomers from model compounds as well as natural and engineered lignin oligomers. These findings can improve the ability to produce valuable aromatic compounds from a renewable resource like lignin. IMPORTANCE Many bacteria are predicted to contain enzymes that could convert renewable carbon sources into substitutes for compounds that are derived from petroleum. The β-etherase pathway present in sphingomonad bacteria could cleave the abundant β–O–4-aryl ether bonds in plant lignin, releasing a biobased source of aromatic compounds for the chemical industry. However, the activity of these enzymes on the complex aromatic oligomers found in plant lignin is unknown. Here we demonstrate biodegradation of lignin polymers using a minimal set of β-etherase pathway enzymes, the ability to recycle needed cofactors (glutathione and NAD+) in vitro, and the release of guaiacyl, syringyl, and tricin as depolymerized products from lignin. These observations provide critical evidence for the use and future optimization of these bacterial β-etherase pathway enzymes for industrial-level biotechnological applications designed to derive high-value monomeric aromatic compounds from lignin.

Abstract:Dopamine is not only a widely used commodity pharmaceutical for treating neurological diseases but also a highly attractive base for advanced carbon materials. Lignin, the waste from the lignocellulosic biomass industry, is the richest source of renewable aromatics on earth. Efficient production of dopamine direct from lignin is a highly desirable target but extremely challenging. Here, we report an innovative strategy for the sustainable production of dopamine hydrochloride from softwood lignin with a mass yield of 6.4 wt.%. Significantly, the solid dopamine hydrochloride is obtained by a simple filtration process in purity of 98.0%, which avoids the tedious separation and purification steps. The approach begins with the acid-catalyzed depolymerization, followed by deprotection, hydrogen-borrowing amination, and hydrolysis of methoxy group, transforming lignin into dopamine hydrochloride. The technical economic analysis predicts that this process is an economically competitive production process. This study fulfills the unexplored potential of dopamine hydrochloride synthesis from lignin.

Abstract:Lignin, a major component of lignocellulose, is the largest source of aromatic building blocks on the planet and harbors great potential to serve as starting material for the production of biobased products. Despite the initial challenges associated with the robust and irregular structure of lignin, the valorization of this intriguing aromatic biopolymer has come a long way: recently, many creative strategies emerged that deliver defined products via catalytic or biocatalytic depolymerization in good yields. The purpose of this review is to provide insight into these novel approaches and the potential application of such emerging new structures for the synthesis of biobased polymers or pharmacologically active molecules. Existing strategies for functionalization or defunctionalization of lignin-based compounds are also summarized. Following the whole value chain from raw lignocellulose through depolymerization to application whenever possible, specific lignin-based compounds emerge that could be in the future considered as potential lignin-derived platform chemicals.

Abstract:Value-added utilization of lignin waste streams is vital to fully sustainable and economically viable biorefineries. However, deriving substantial value from its main constituents is seriously hindered by the constant requirement for expensive coenzymes. Herein, we devised a coenzyme-free biocatalyst that could transform lignin-derived aromatics into various attractive pharmaceutical and polymer building blocks. At the center of our strategy is the integrated use of new mining phenolic acid decarboxylase and aromatic dioxygenase with extremely high catalytic efficiency, which realizes the value-added utilization of lignin in a coenzyme-independent manner. Notably, a new temperature/pH-directed strategy was proposed to eliminate the highly redundant activities of endogenous alcohol dehydrogenases. The major components of lignin were simultaneously converted to vanillin and 4-vinylphenol. Since the versatile biocatalyst could efficiently convert many other renewable lignin-related aromatics to valuable chemicals, this green route paves the way for enhancing the entire efficiency of biorefineries.

Abstract:Inherently complex, lignin-derived aromatic monomers comprising valuable structural moieties present in many pharmaceuticals would serve as ideal substrates for the construction of biologically active molecules. Here, we describe a strategy that incorporates all intrinsic functional groups present in platform chemicals obtained by lignin depolymerization into value-added amines, using sustainable catalytic methods and benign solvents. Our strikingly efficient protocol provides access to libraries of aminoalkyl-phenol derivatives and seven-membered N-heterocycles directly from wood in two, respectively three, waste-free steps. Several molecules in these libraries have shown promising antibacterial or anticancer activities, emphasizing the advantage of this modular synthetic strategy and the potential for drug discovery. The sustainable catalytic pathways presented here can lead to significant benefits for the pharmaceutical industry where reduction of hazardous waste is a prime concern, and the described strategies that lead to high-value products from non-edible biomass waste streams also markedly increase the economic feasibility of lignocellulosic biorefineries.We developed an efficient and sustainable catalytic method for the synthesis of biologically active tetrahydro-2-benzazepines from renewable resources using green solvents in three waste-free steps.

Abstract:The conversion of lignin into bioactive compounds through selective organic synthesis methods represents a promising frontier in the pursuit of sustainable raw materials and green chemistry. This review explores the versatility of lignin-derived bioactive compounds, ranging from their application in drug discovery to their role in the development of biodegradable materials. Despite notable advancements, the synthesis routes and yields of highly bioactive molecules from lignin still require further exploration and improvement. This review provides an in-depth examination of the progress made in understanding the complex structure of lignin and developing innovative approaches to exploit its potential. Specifically, the types of lignins covered include softwood Kraft lignin, hardwood organosolv lignin, and soda lignin. This work is divided into three parts: first, the transformation of lignin into bioactive molecules with chemically active centres and functionalised hydroxyl groups through depolymerisation; second, kinetic modelling techniques essential for understanding the chemical kinetics of lignin and enabling significant scaling up in the conversion of organic molecules; third, efficient catalytic pathways for synthesising molecules with anticancer and antibacterial properties. In conclusion, this comprehensive review spurs further investigations into lignin-derived bioactive compounds, their applications, and the advancement of sustainable processes.

Abstract:Abstract Benzene is a widely used commodity chemical, which is currently produced from fossil resources. Lignin, a waste from lignocellulosic biomass industry, is the most abundant renewable source of benzene ring in nature. Efficient production of benzene from lignin, which requires total transformation of C sp2 -C sp3 /C sp2 -O into C-H bonds without side hydrogenation, is of great importance, but has not been realized. Here, we report that high-silica HY zeolite supported RuW alloy catalyst enables in situ refining of lignin, exclusively to benzene via coupling Bronsted acid catalyzed transformation of the C sp2 -C sp3 bonds on the local structure of lignin molecule and RuW catalyzed hydrogenolysis of the C sp2 -O bonds using the locally abstracted hydrogen from lignin molecule, affording a benzene yield of 18.8% on lignin weight basis in water system. The reaction mechanism is elucidated in detail by combination of control experiments and density functional theory calculations. The high-performance protocol can be readily scaled up to produce 8.5 g of benzene product from 50.0 g lignin without any saturation byproducts. This work opens the way to produce benzene using lignin as the feedstock efficiently.

Abstract:Lignin is a cross linked aromatic polymer and it is abundant in nature. It is regarded as a potential substitute of fossil resource and industrial raw materials due to its highly aromatic character via lignin valorization and commercialization. Lignin depolymerization is an effective solution to achieve lignin valorization by yielding value-added fine chemicals (such as monophenols and biofuels). Due to their stable structure, complex spatial configurations and electronic effect, lignin depolymerization often suffered from serious condensation and complexity of products. Commonly, this results in low yield and selectivity that limit large scale of lignin depolymerization. A challenge exists, however, on how to develop high selective catalytic strategies/reactors, which would enable efficient and selective catalysis by controlling the bond breaking mode and inhibiting the condensation of active intermedia. In this review, we begin by introducing the potential of lignin as an emerging star for lignin commercialization. And then catalytic depolymerization strategy (e.g., catalytic pyrolysis, catalytic hydrolysis, catalytic oxidation, catalytic reduction and two-step redox) and also their selective bond breaking mechanism are reviewed. Particularly, we report latest vision about lignin highly efficient valorization targeting at reaching market, by controlling of reaction condensation and using continuous-flow reactor depolymerization. Most importantly, the limitations and potential solutions for lignin depolymerization are proposed to present opportunities for future industrialization applications.

Abstract:Background Lignin, nature’s dominant aromatic polymer, is found in most terrestrial plants in the approximate range of 15 to 40% dry weight and provides structural integrity. Traditionally, most large-scale industrial processes that use plant polysaccharides have burned lignin to generate the power needed to productively transform biomass. The advent of biorefineries that convert cellulosic biomass into liquid transportation fuels will generate substantially more lignin than necessary to power the operation, and therefore efforts are underway to transform it to value-added products. Advances Bioengineering to modify lignin structure and/or incorporate atypical components has shown promise toward facilitating recovery and chemical transformation of lignin under biorefinery conditions. The flexibility in lignin monomer composition has proven useful for enhancing extraction efficiency. Both the mining of genetic variants in native populations of bioenergy crops and direct genetic manipulation of biosynthesis pathways have produced lignin feedstocks with unique properties for coproduct development. Advances in analytical chemistry and computational modeling detail the structure of the modified lignin and direct bioengineering strategies for targeted properties. Refinement of biomass pretreatment technologies has further facilitated lignin recovery and enables catalytic modifications for desired chemical and physical properties. Outlook Potential high-value products from isolated lignin include low-cost carbon fiber, engineering plastics and thermoplastic elastomers, polymeric foams and membranes, and a variety of fuels and chemicals all currently sourced from petroleum. These lignin coproducts must be low cost and perform as well as petroleum-derived counterparts. Each product stream has its own distinct challenges. Development of renewable lignin-based polymers requires improved processing technologies coupled to tailored bioenergy crops incorporating lignin with the desired chemical and physical properties. For fuels and chemicals, multiple strategies have emerged for lignin depolymerization and upgrading, including thermochemical treatments and homogeneous and heterogeneous catalysis. The multifunctional nature of lignin has historically yielded multiple product streams, which require extensive separation and purification procedures, but engineering plant feedstocks for greater structural homogeneity and tailored functionality reduces this challenge.

Abstract:Lignin is the only large-volume renewable source of aromatic chemicals. Efficient depolymerization and deoxygenation of lignin while retaining the aromatic functionality are attractive but extremely challenging. Here we report the selective production of arenes via direct hydrodeoxygenation of organosolv lignin over a porous Ru/Nb 2 O 5 catalyst that enabled the complete removal of the oxygen content from lignin. The conversion of birch lignin to monomer C 7 –C 9 hydrocarbons is nearly quantitative based on its monomer content, with a total mass yield of 35.5 wt% and an exceptional arene selectivity of 71 wt%. Inelastic neutron scattering and DFT calculations confirm that the Nb 2 O 5 support is catalytically unique compared with other traditional oxide supports, and the disassociation energy of C aromatic –OH bonds in phenolics is significantly reduced upon adsorption on Nb 2 O 5 , resulting in its distinct selectivity to arenes. This one-pot process provides a promising approach for improved lignin valorization with general applicability.
Towards bioresource-based aggregation-induced emission luminogens from lignin β-O-4 motifs as renewable resources

Abstract:One-pot synthesis of heterocyclic aromatics with good optical properties from phenolic β-O-4 lignin segments is of high importance to meet high value added biorefinery demands. However, executing this process remains a huge challenge due to the incompatible reaction conditions of the depolymerization of lignin β-O-4 segments containing γ-OH functionalities and bioresource-based aggregation-induced emission luminogens (BioAIEgens) formation with the desired properties. In this work, benzannulation reactions starting from lignin β-O-4 moieties with 3-alkenylated indoles catalyzed by vanadium-based complexes have been successfully developed, affording a wide range of functionalized carbazoles with up to 92% yield. Experiments and density functional theory calculations suggest that the reaction pathway involves the selective cleavage of double C-O bonds/Diels-Alder cycloaddition/dehydrogenative aromatization. Photophysical investigations show that these carbazole products represent a class of BioAIEgens with twisted intramolecular charge transfer. Distinctions of emission behavior were revealed based on unique acceptor-donor-acceptor-type molecular conformations as well as molecular packings. This work features lignin β-O-4 motifs with γ-OH functionalities as renewable substrates, without the need to apply external oxidant/reductant systems. Here, we show a concise and sustainable route to functional carbazoles with AIE properties, building a bridge between lignin and BioAIE materials.

Abstract:Heteroatom-participated lignin depolymerization for heterocyclic aromatic compounds production is of great importance to expanding the product portfolio and meeting value-added biorefinery demand, but it is also particularly challenging. In this work, the synthesis of pyrimidines from lignin β-O-4 model compounds, the most abundant segment in lignin, mediated by NaOH through a one-pot multi-component cascade reaction is reported. Mechanism study suggests that the transformation starts by NaOH-induced deprotonation of Cα-H bond in β-O-4 model compounds, and involves highly coupled sequential cleavage of C-O bonds, alcohol dehydrogenation, aldol condensation, and dehydrogenative aromatization. This strategy features transition-metal free catalysis, a sustainable universal approach, no need of external oxidant/reductant, and an efficient one-pot process, thus providing an unprecedented opportunity for N-containing aromatic heterocyclic compounds synthesis from biorenewable feedstock. With this protocol, an important marine alkaloid meridianin derivative can be synthesized, emphasizing the application feasibility in pharmaceutical synthesis.

Abstract:Lignin valorization presents significant opportunities for advancing a sustainable bioeconomy and advancing carbon neutrality goals. Biological conversion of lignin offers promising pathways for producing aromatic fine chemicals, effectively addressing the challenges associated with lignin's heterogeneous and macromolecular structure. However, the pathways for converting lignin derivatives into aromatic chemicals remain underdeveloped, and the conversion efficiency remains unsatisfactory. This work aims to prospect bioconversion pathways of lignin toward valuable aromatics, which aligns with the principles of atom-economic concept. A comprehensive overview of the critical pathways for converting lignin into aromatic chemicals is performed. A thorough discussion on advanced technologies essential for enhancing lignin bioconversion and aromatic synthesis is presented. Furthermore, existing challenges and emerging strategies are explored. The findings of this study are expected to offer valuable insights into recent advancements and future directions in the biological conversion of lignin. These insights can facilitate lignin valorization and promote the development of a lignin-based bioeconomy.

Abstract:Lignosulfonate is a by-product of the cooking process by sulfite pulping for paper manufacturing. The treatment of wood chips by various salts of sulfurous acid solubilizes lignin to produce a cellulose-rich wood pulp. Developing a technique for the conversion of lignosulfonate by-product to high value materials has an important industrial utility. Sphingobium sp. strain SYK-6, which was isolated from pulping wastewater, is one of the best enzymatically or genetically characterized bacteria for degrading lignin-derived aromatics. We have previously established a system for the production of 2-pyrone-4,6-dicarboxylic acid (PDC), a novel platform chemical that can produce a variety of bio-based polymers, by introducing of ligA, ligB, and ligC genes from SYK-6 into a mutant strain of Pseudomonas putida PpY1100. In this study, extracts from lignosulfonates, which were desulphonated and depolymerized by alkaline oxidation, were evaluated as substrates for microbiological conversion to PDC by the transgenic bacteria.
Fast and Selective Production of Aromatics via Efficient Lignin Depolymerization: Critical Factors and Mechanism Studies

Abstract:Efficient conversion of renewable lignin into value-added chemicals and biofuel is of great importance for the sustainable development of biorefineries. However, lignin valorization is highly restricted by its structural variation and complexity. In this regard, we have produced a series of lignins extracted from grass, hardwood, and softwood and focused on scrutinizing their structural variability and key characteristics to provide structural insights into fast and selective production of aromatics. As an energy-efficient approach, microwave-assisted hydrothermal liquefaction without introducing an external oxygen was performed to facilitate the base-catalyzed depolymerization of lignin under moderate conditions. The general applicability of this approach was proved on five different lignins. High yields (58.5–78.6%) of low-molecular-weight bio-oil from all lignins were obtained only after 1 min at 220 °C. In particular, a high ratio of cleavable β–O–4 linkage of grass lignin enabled a fast monomer production, while its specific p-coumaric acid moieties allowed a high monomer selectivity to 4-vinylphenol of 66%. A difference in the product distribution was systematically investigated through a comprehensive analysis of lignin characteristics (e.g., interlinkage, S/G/H unit, and molecular weight) and stability studies of main monomeric products. These results revealed the fast degradation of reactive intermediates (aldehydes and S-type products) and how the monomer’s stability/reactivity affects the selective production. Alongside reaction parameters, several critical factors governing the lignin conversion were further identified by calculating the Pearson correlation coefficient between lignin characteristics and the conversion efficiency. The lignin structure–property–degradability relationships and reaction mechanisms are expounded and updated to foster efficient lignin utilization.

Abstract:The selective transformation of lignin to value‐added biochemicals in high yields is incredibly challenging due to its structural complexity and the vast number of possible reaction pathways. Phenolic acids (PAs) are key building blocks for various aromatic polymers, but the isolation of PAs from lignin is below 5 wt.% and requires harsh reaction conditions. Herein, we demonstrate an effective route to selectively convert lignin into isolated PAs in a high yield (up to 20 wt.% of lignin) using a low‐cost graphene oxide‐urea hydrogen peroxide (GO‐UHP) catalyst system under mild conditions (<120 °C, ambient pressure). The lignin conversion yield is up to 95%, and the remaining low molecular weight organic oils can work as aviation fuel precursors to complete lignin utilization. Mechanistic studies demonstrate that pre‐acetylation allows the selective depolymerization of lignin to aromatic aldehydes with a decent yield by GO through the Ca activation of β‐O‐4 cleavage. A UHP oxidative process is followed to effectively transform aldehydes in the depolymerized product to PAs, by avoiding undesired Dakin side reaction due to the electron‐withdrawing effect of the acetyl group. This work opens a new way to selectively cleave lignin side chains to isolated biochemicals under mild reaction conditions.

Abstract:The directional production of benzene is achieved by the current-enhanced catalytic conversion of lignin. The synergistic effect between catalyst and current promotes the depolymerization of lignin and the selective recombinant of the functional groups in the aromatic monomers. A high benzene yield of 175 gbenzene/kglignin was obtained with an excellent selectivity of 92.9 C-mol%. The process potentially provides a promising route for the production of basic petrochemical materials or high value-added chemicals using renewable biomass.

Abstract:Lignocellulose is mainly composed of hydrophobic lignin and hydrophilic polysaccharide polymers, contributing to an indispensable carbon resource for green biorefineries 1,2 . When chemically treated, lignin is compromised owing to detrimental intra- and intermolecular crosslinking that hampers downstream process 3,4 . The current valorization paradigms aim to avoid the formation of new C–C bonds, referred to as condensation, by blocking or stabilizing the vulnerable moieties of lignin 5,6,7 . Although there have been efforts to enhance biomass utilization through the incorporation of phenolic additives 8,9 , exploiting lignin's proclivity towards condensation remains unproven for valorizing both lignin and carbohydrates to high-value products. Here we leverage the proclivity by directing the C–C bond formation in a catalytic arylation pathway using lignin-derived phenols with high nucleophilicity. The selectively condensed lignin, isolated in near-quantitative yields while preserving its prominent cleavable β-ether units, can be unlocked in a tandem catalytic process involving aryl migration and transfer hydrogenation. Lignin in wood is thereby converted to benign bisphenols (34–48 wt%) that represent performance-advantaged replacements for their fossil-based counterparts. Delignified pulp from cellulose and xylose from xylan are co-produced for textile fibres and renewable chemicals. This condensation-driven strategy represents a key advancement complementary to other promising monophenol-oriented approaches targeting valuable platform chemicals and materials, thereby contributing to holistic biomass valorization.
