Abstract:Objective To explore the relationship between forkhead box P3(Fox P3)gene polymorphism and susceptibility to gastric cancer in Chinese Han population.Methods From January 2014 to December 2016,a total of 312 patients with primary gastric cancer and 548 age and gender matched controls were recruited.Polymorphisms of Fox P3 at rs3761548 and rs2232365 loci were analyzed by polymerase chain reaction-restriction fragment length polymorphism techniques(PCR-RFLP).Odd ratio(OR)and 95% confidence interval(CI)were used to analyze the correlation between different genotype and susceptibility to gastric cancer,clinical pathological characteristics of patients with gastric cancer.Results The frequency of AA,AC,and CC genotype of rs3761548 in gastric cancer group and control group were 22.8%(71/312),44.2%(138/312),33.0%(103/312)and 14.8%(81/548),48.5%(266/548),36.7%(201/548),respectively.AA genotype could increase the risk of gastric cancer(OR=1.711,95% CI 1.150 to 2.545,P=0.008).The frequency of A and G allele of gastric cancer group and control group were 44.9%(280/624),55.1%(344/624)and 39.1%(428/1 096),60.9%(668/1 096), respectively.A allele could increase the risk of gastric cancer(OR=1.270,95% CI 1.041 to 1.550,P=0.018).The frequency of AA,AG,and GG genotype of rs2232365 in gastric cancer group and control group were 19.2%(60/312),46.5%(145/312),34.3%(107/312)and 14.1%(77/548),48.7%(267/548),37.2%(204/548),respectively.In recessive model(AA vs AG+GG),AA genotype could increase the risk of gastric cancer(OR=1.456,95% CI 1.005 to 2.110,P=0.046).A allele of rs3761548 was associated with the depth of invasion in gastric wall(χ2 =12.710,P=0.001)and Helicobacter pylori infection(χ2 = 5.613,P= 0.018).A genotype could increase the risk of lymphatic metastasis(χ2 =4.878,P=0.027).Conclusion Fox P3 gene polymorphism is associated with the susceptibility to gastric cancer in Chinese Han population,and can be molecular-maker of poor prognosis of gastric cancer.
Association between H-RAS T81C genetic polymorphism and gastrointestinal cancer risk: A population based case-control study in China

Abstract:Background Gastrointestinal cancer, such as gastric, colon and rectal cancer, is a major medical and economic burden worldwide. However, the exact mechanism of gastrointestinal cancer development still remains unclear. RAS genes have been elucidated as major participants in the development and progression of a series of human tumours and the single nucleotide polymorphism at H-RAS cDNA position 81 was demonstrated to contribute to the risks of bladder, oral and thyroid carcinoma. Therefore, we hypothesized that this polymorphisms in H-RAS could influence susceptibility to gastrointestinal cancer as well, and we conducted this study to test the hypothesis in Chinese population. Methods A population based case-control study, including 296 cases with gastrointestinal cancer and 448 healthy controls selected from a Chinese population was conducted. H-RAS T81C polymorphism was genotyped by Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) assay. Results In the healthy controls, the TT, TC and CC genotypes frequencies of H-RAS T81C polymorphism, were 79.24%, 19.87% and 0.89%, respectively, and the C allele frequency was 10.83%. Compared with TT genotype, the TC genotype was significantly associated with an increased risk of gastric cancer (adjusted OR = 3.67, 95%CI = 2.21–6.08), while the CC genotype showed an increased risk as well (adjusted OR = 3.29, 95%CI = 0.54–19.86), but it was not statistically significant. In contrast, the frequency of TC genotype was not significantly increased in colon cancer and rectal cancer patients. Further analysis was performed by combining TC and CC genotypes compared against TT genotype. As a result, a statistically significant risk with adjusted OR of 3.65 (95%CI, 2.22–6.00) was found in gastric cancer, while no significant association of H-RAS T81C polymorphism with colon cancer and rectal cancer was observed. Conclusion These findings indicate, for the first time, that there is an H-RAS T81C polymorphism existing in Chinese population, and this SNP might be a low penetrance gene predisposition factor for gastric cancer.
Polymorphisms of Inflammation-Related Genes and Colorectal Cancer Risk: a Population-Based Case-Control Study in China.

Abstract:The previous studies found that chronic inflammation related to an increased risk of colorectal cancer (CRC). This study aims to explore the associations of polymorphisms in inflammation-related genes (IL10, IL10RA, IL6R, TNFRSF1A, TNFRSF1B, LTA and IL4) and their interactions with the risk of colorectal cancer among Chinese population. A population-based case-control study including 299 cases and 296 controls was conducted from January 2001 to December 2009. Multivariate unconditional logistic regression was used to analyse the association of nine SNPs in inflammation-related genes with the risk of CRC, colon cancer and rectal cancer, respectively. Generalized multifactor dimensionality reduction (GMDR) was implemented to explore the gene-gene interactions among all SNPs on CRC. A decreased risk of colorectal cancer in subjects with rs1800872 AC genotype of IL10 (OR = 0.643, 95%CI = 0.453, 0.912) or AC/CC genotype (OR = 0.636, 95%CI = 0.457, 0.885) was observed, compared with those with AA genotype. Meanwhile, similar associations were observed between rs1800872 and rectal cancer. Additionally, in rs1061624 of TNFRSF1B gene, AG genotype (OR=0.566; 95% CI= 0.362, 0.885) and AG/GG genotype (OR=0.638; 95% CI=0.420, 0.971) were significantly associated with a decreased risk of rectal cancer, respectively. Our findings indicated that mutants in IL10 and TNFRSF1B genes may change the CRC risk. However, there is no interaction between inflammation-related genes on CRC risk.
Case–control Study on TP73 Rs1801173 C > T Gene Polymorphism and Susceptibility to Gastric Cancer in a Chinese Han Population

Abstract:Abstract Background This study investigated the role of TP73 gene polymorphism, rs1801173on risk of gastric cancer. Methods We conducted a case-controlled study including 577 primary gastric cancer and 678 normal control cases. The target gene fragment was amplified using PCR using blood samples collected from patients. Allele analysis and genotyping were performed using snapshot method. Results The findings showed that the control group had consistent genotype frequency distribution and presented Hardy–Weinberg equilibrium. The results showed no significant differences in sex, drinking history and age distributions between subjects with the polymorphism and subjects in the control group. Smoking status was correlated with incidence of gastric cancer (P = 0.006). The rs1801173 locus of TP73 gene contained 3 genotypes including: TT, CT, and CT. Logistic regression analysis showed that distribution of recessive model and dominant model was comparable between the two groups before (P = 0.688; 0.937) or after (P = 0.703; 0.990) adjusting for confounders. The distribution frequency in case group was not significantly different relative to that of the control group (P = 0.763). Conclusion Smoking can independently influence the risk of gastric cancer. TP73 gene rs1801173 polymorphism was not significantly correlated with risk of gastric cancer.
Polymorphisms Rs2073618 and Rs3102735 in TNFRSF11B with the Susceptibility to Gastric Cancer

Abstract:Purpose This study aims to explore the potential association between TNFRSF11B gene polymorphisms (rs2073618 and rs3102735) and the risk of gastric cancer in the Chinese Han population. Methods A case-control study was conducted involving 577 individuals diagnosed with primary gastric cancer and 678 healthy volunteers. Genomic DNA was extracted from whole blood samples, and the target gene fragment was amplified via PCR. Genotyping and allele analysis were subsequently performed using the snapshot technique. Results In this retrospective research, it was discovered that the TNFRSF11B rs3102735 T > C polymorphism is significantly correlated with a heightened risk of gastric cancer. Conversely, the variant allele of the rs2073618 polymorphism demonstrated no significant association with susceptibility to gastric cancer. During the subgroup analysis of the rs3102735 polymorphism, a notably heightened risk of gastric cancer was observed in the group aged above 62 years. Stratified analyses indicated that the TNFRSF11B rs2073618 polymorphism didn’t exhibit an association with gastric cancer susceptibility across varying ages, genders, smoking and alcohol drinking. Conclusion Our research indicated a link between smoking and the incidence of gastric cancer. Additionally, we identified a link between gastric cancer and the TNFRSF11B rs3102735 polymorphism, but not with the rs2073618 polymorphism. TNFRSF11B rs3102735 CC mutated homozygote genotype may be an indicator of gastric cancer.
Correlation Between CCND1 Gene Polymorphism and Genetic Susceptibility to Gastric Cancer in Chinese Population

Abstract:Abstract Objective: The relationship between Cyclin D1 gene (CCND1) rs9344 polymorphism and susceptibility to gastric cancer (GC) is investigated.Methods: In a case-control study, we selected 577 cases of GC from The People's Hospital Affiliated to Jiangsu University in China along with 678 normal controls. Blood DNA was extracted and PCR amplified, gene polymorphism was determined using Snapshot method.Results: Analysis reveals significant difference in smoking between GC and control groups (P=0.006), however, not on polymorphism (P＞0.05). Conclusion: Smoking is associated with gastric cancer, whereas CCND1 rs9344 polymorphism does not implicate susceptibility of gastric cancer.
Association of lncRNA EIF3J-AS1 with target gene IGFB3 polymorphisms in susceptibility of gastric cancer

Abstract:LncRNA EIF3J-AS1 has shown to play an important regulatory role in a variety of tumor tissues, IGFB3 Gene is the target gene of LncRNA EIF3J-AS1, experimental studies shows that IGFB3 gene risk typing is associated with an increased risk of gastric cancer susceptibility of gastric cancer(GC),the relationship between LncRNA EIF3J-AS1 target IGFB3 Gene polymorphisms expression in GC patients is related. Methods : A case-controlled study was conducted, including 490 primary gastric cancers and 1476 normal controls. Targetscan, miRanda and other software are used to target and predict the genes and mirnas that lncRNA can bind to, and the expression correlation analysis is conducted, so as to construct the ceRNA network and speculate its regulation of gastric cancer expression. The target gene IGFB3 fragment was amplified in blood samples using PCR. Genotyping was performed using the snapshot method. Results : This rearch shows a signal pathway LncRNA EIF3J-AS1-IGFBP3, in IGFBP3 gene variants GA and GA + AA models, castric cancer can be decreased when in male subgroup and ≥ 61 years old subgroup, but during smoking and alcohol groups, gastic cancer risk is encreased. IGFBP3 gene is regulated by LncRNA EIF3J-AS1-miRNAs-IGFBP3 network, which could provide a potential drug target biomarker.

Abstract:Background Gastric cancer is one of the most common malignancies afflicting the Chinese population. Polymorphisms in interleukin-1B ( IL - 1B ) and interleukin-1 receptor antagonist ( IL - 1RN ) genes have been associated with increased gastric cancer risk. Aims A case–control study enrolled 392 gastric cancer patients and 508 healthy were carried out to investigate the association between polymorphisms in IL-1B and IL-1RN and gastric cancer risk. Methods Polymerase chain reaction (PCR)-restriction fragment length polymorphism was used for detection of two potentially functional polymorphisms ( IL - 1B -31 and IL - 1B -511) in the IL - 1B gene promoter and PCR was used for detection of the variable tandem repeat in the second intron of IL - 1RN . Results The data showed that the IL - 1B -31CC genotype increased gastric cancer risk to an adjusted odd of 2.27 (95% CI, 1.49–3.46), IL - 1B -31CT to 1.48 (95% CI, 1.01–2.16) and IL - 1B -31CT/CC to 1.68 (95% CI, 1.17–2.40), while IL - 1B -51TT genotype associated with increased gastric cancer risk to an adjusted odd of 2.53 (95% CI, 1.67–3.84), IL - 1B -511TC to 1.45 (95% CI, 1.02–2.06), and IL-1B -511TC TT/TC to 1.72 (95% CI, 1.23, 2.39). Furthermore, IL - 1RN heterogeneity genotype ( IL - 1RN 2L) was associated with gastric cancer risk to an adjusted odd of 1.70 (95% CI, 1.05–2.74) compared to the wild-type homozygote ( IL - 1RN LL). In addition, H. pylori infection enhanced gastric cancer risk through these SNPs. Conclusions The data from the current study demonstrated that the genotype CC or CT of IL - 1B -31, TT or CT of IL - 1B -511, and 2L of IL - 1RN increased risk of gastric cancer in this Chinese population and the risk was further enhanced by H. pylori.

Abstract:AimsTo explore associations between polymorphisms of IGF2-related genes including H19, IGF2, IGF2BP2 and IGF2R and Metabolic syndrome (MetS) susceptibility in the Chinese Han population.Methods66 subjects with MetS and 257 control subjects were collected for inclusion in a case-control study. PCR-RFLP was used to investigate polymorphisms in the H19, IGF2, IGF2BP2 and IGF2R genes. Elisa was used to detect the serum IGF2 concentrations.ResultsFemales carrying the GG and AG genotypes of rs680 (IGF2) exhibited a lower risk of MetS, compared with those harboring AA (adjusted OR = 0.388, p = 0.027), while GG and AG genotypes were associated with lower fasting glucose and HbA1c. In males, the Waist-to-Hip Ratio (WHR) and the level of TG were significantly higher in GG and AG genotypes than in the AA genotype of rs680 in IGF2. Levels of HDL-c were lower in men with GG and AG genotypes compared with those carrying the AA genotype. Serum IGF2 concentrations did not change among different genotypes. Finally, multifactor dimensionality reduction (MDR) analysis identified interactions between four polymorphisms: rs3741279 (H19), rs680 (IGF2), rs1470579 (IGF2BP2) and rs629849 (IGF2R).ConclusionsOur study suggests that IGF2-related genes including H19, IGF2, IGF2BP2 and IGF2R genes may play pivotal roles in the development of MetS.
Case-Control Study on TNFRSF6B Gene Polymorphism and Susceptibility to Gastric Cancer in a Chinese Han Population

Abstract:Purpose To explore the relationship between rs2297440 and rs2297441 polymorphisms of TNFRSF6B gene and susceptibility to gastric cancer. Methods A hospital-based case-control study was conducted. A total of 577 gastric cancer cases and 678 normal controls were recruited. Their genotypes were determined using the SnapShot method. Results The smoking rate in the case group (34.49%) was higher than that in the control group (27.29%). For TNFRSF6B rs2297440, among people <62 years old, the risk of gastric cancer in TC people was 1.84 times that in TT people. Among the non-drinking people, the risk of gastric cancer in the CC type was 0.66 times that in the TT+TC type. Among the drinking population, the risk of gastric cancer in the TC type was 1.67 times that in the TT type, and the risk in the TC+CC type was 1.70 times that in the TT type. As for TNFRSF6B rs2297441, in males and non-drinkers, the risk of gastric cancer in the AG type was less than that in the GG type. No matter how old the patient is, the risk of gastric cancer in the AA type was less than that in the AG+GG type. Conclusion A correlation exists between smoking and gastric cancer. For TNFRSF6B rs2297440, the TC genotype may be a risk factor for gastric cancer in people <62 years old. In the non-drinking population, the homozygous mutant of CC may be a protective factor for gastric cancer. In the drinking population, TC type may be a risk factor, whereas the TC+CC type dominated by C may be a protective factor. For TNFRSF6B rs2297441, the AG genotype may be a risk factor for gastric cancer in males and non-drinkers. The AA homozygous mutant may be a protective factor for gastric cancer.
[Correlation Between Gene Polymorphisms of Wnt Signalling Pathway Related Components and Risk of Gastric Carcinoma: a Case-Control Study].

Abstract:OBJECTIVE:To investigate correlation between polymorphisms of rs3755557 and rs1880481 located in glycogen synthase kinase 3beta gene and beta-catenin gene respectively, the products of which are components of Wnt signalling pathway, and risk of gastric carcinoma.METHODS:PCR and denaturing high-performance liquid chromatography combining with DNA sequencing were used to analyse genotype polymorphism of rs3755557 and rs1880481 of the subjects including 26 patients of gastric carcinoma and 33 patients of chronic superficial gastritis.RESULTS:Chi-square analysis revealed that there was no significant difference in the frequencies of alleles and genotypes of rs3755557 polymorphic site between gastric carcinoma group and control group. As to the rs1880481 polymorphic site, there was no significant difference in the frequencies of alleles between gastric carcinoma group and the corresponding control group. The frequency of heterozygous genotype in male control group was 68.18% and it was significantly higher than 26.67% in male gastric carcinoma group, OR=5.893, 95%CI: 1.377-25.226 (P=0.013); but the frequencies of AA genotype of the site in male control group and male gastric carcinoma group were 9.09% and 40.00% respectively. There was statistical significance, OR=6.667, 95% CI: 1.121-39.660 (P=0.025).CONCLUSION:The above results suggest that the genotypes and alleles of rs3755557 site do not contribute to the risk of gastric carcinoma. Low level of the heterozygous genotype or high level of AA genotype of rs1880481 polymorphic site in male patients might cause a higher risk of developing gastric carcinoma.

Abstract:Long non-coding RNA (lncRNA) H19 is involved in tumor development, progression, and metastasis. This case-control study assessed the association between H19 genetic variants and susceptibility to gastric cancer (GC) in a Chinese Han population. We genotyped four lncRNA H19 single nucleotide polymorphisms (SNPs) (rs217727 C > T, rs2839698 C > T, rs3741216 A > T, rs3741219 T > C) in 500 GC patients and 500 healthy controls. Carriers of variant rs217727T and rs2839698T alleles showed increased GC risk (P = 0.008 and 0.011, respectively). Compared with the common genotype, CT + TT rs217727 and CT + TT rs2839698 genotypes were associated with significantly increased GC risk (P = 0.040, adjusted odds ratio [OR] = 1.32, 95% confidence interval [CI] = 1.01-1.71; P = 0.033, adjusted OR = 1.31, 95% CI = 1.02-1.69, respectively). Further stratified analyses revealed that the association between GC risk and variant genotypes of rs217727 was more profound in younger individuals (≤59 years) and non-smokers, while the association between risk and the rare rs2839698 genotype persisted in men and rural subjects. rs2839698 CT and TT genotypes were also associated with higher serum H19 mRNA levels compared with the CC genotype. These findings suggest that lncRNA H19 SNPs may contribute to susceptibility to GC.
Risk Prediction for Early-Onset Gastric Carcinoma: a Case-Control Study of Polygenic Gastric Cancer in Han Chinese with Hereditary Background.

Abstract:Recent genomewide studies have identified several germline variations associated with gastric cancer. The aim of the present study was to identify, in a Chinese Han population, the individual and combined effects of those single nucleotide polymorphisms (SNPs) that increase the risk of early-onset gastric cancer. We conducted a case-control study comprising 116 patients with gastric cancer as well as 102 sex- and age-matched controls and confirmed that the SNPs MUC1 (mucin 1) rs9841504 and ZBTB20 (zinc finger and BTB domain containing 20) rs4072037 were associated with an increased gastric cancer risk. Of the 116 patients diagnosed with cancer, 65 had at least 1 direct lineal relative with carcinoma of the digestive system or breast/ovarian cancer. These 65 had another 4 SNPs associated with gastric cancer susceptibility: PSCA (prostate stem cell antigen) rs2294008, PLCE1 (phospholipase C epsilon 1) rs2274223, PTGER4/PRKAA1 (prostaglandin E receptor 4/ protein kinase AMP-activated catalytic subunit alpha 1) rs13361707, and TYMS (thymidylate synthetase) rs2790. However, each of these low-penetrance susceptibility polymorphisms alone is not considered influential enough to predict the absolute risk of early-onset gastric cancer. Thus we decided to study different combinations of polygenes as they affected for our population. Those subjects with both the risk alleles MUC1 rs9841504 and ZBTB20 rs4072037 had a greater than 3-fold increased risk of gastric cancer. Also those with a hereditary background including the risk alleles PLCE1 rs2274223 and PTGER4/PRKAA1 rs13361707 were 3 times more susceptible to cardia cancer than those without. These findings show that the study of combined polymorphisms, instead of single low-penetrance variations in susceptibility, may lead to a high-risk classification for a specific population.
Risk of gastric ulcer contributed by genetic polymorphisms of PSCA: A case-control study based on Chinese Han population

Abstract:AIM: To investigate the relationship between insulin receptor substrate-2 (IRS-2) G1057D polymorphism and the risk of gastric cancer (GC) in a Chinese population. METHODS: A case-control study with 197 GC patients and 156 age-and sex-matched control subjects was conducted. The genotypes of polymorphism were assessed by polymerase chain reaction-restriction fragment length polymorphism. RESULTS: The genotype frequencies of IRS-2 G1057D polymorphism in cases were obviously different from those in the control group (P = 0.031). Compared with GG genotype carriers, the risk for GC was significantly higher (adjusted odds ratio = 2.32, 95% CI: 1.03-5.23, P = 0.042) in the individuals with the IRS-2 DD genotype. Furthermore, stratified analysis was performed based on age, sex, smoking status and residence, but no significant difference between the two groups was found. In addition, no significant association between genotypes and clinicopathological features was observed either. CONCLUSION: This study demonstrates that IRS-2 G1057D is involved in susceptibility to GC, although further large-sample studies are still needed. (C) 2012 Baishideng. All rights reserved.

Abstract:Previous studies have reported that XPC gene polymorphisms may modify the individual susceptibility to gastric cancer. In this case-control study with a total of 1,142 cases and 1,173 controls, four potentially functional polymorphisms were genotyped in the XPC gene (rs2228001 A>C, rs2228000 C>T, rs2607775 C>G, and rs1870134 G>C) by Taqman assays and their associations were analyzed with the risk of gastric cancer in a Southern Chinese population. No significant association between any of XPC polymorphisms and gastric cancer risk was detected except for a borderline association with the rs2228000 CT/TT genotype (crude odds ratio =0.86, 95% confidence interval =0.73-1.02, P=0.088) when compared to the rs2228000 CC genotype. Further stratified analysis revealed that the protective effect of rs2228000 CT/TT on the risk of gastric cancer was only significant among subjects older than 58 years. In summary, results indicated that genetic variations in XPC gene may play a weak effect on gastric cancer susceptibility in Southern Chinese population, which warrants further confirmation in larger prospective studies with different ethnic populations.
IGFBP3 A-202C polymorphism and breast cancer susceptibility: a meta-analysis involving 33,557 cases and 45,254 controls

Abstract:Published data on the association between insulin-like growth factor binding protein 3 (IGFBP3) A-202C polymorphism and breast cancer risk are inconclusive. To derive a more precise estimation of the relationship, a meta-analysis was performed. Crude ORs with 95% CIs were used to assess the strength of association between them. A total of 27 studies including 33,557 cases and 45,254 controls were involved in this meta-analysis. Overall, significantly elevated breast cancer risk was associated with IGFBP3 C allele when all studies were pooled into the meta-analysis (CC vs. AA: OR = 1.06, 95% CI = 1.02–1.11; dominant model: OR = 1.04, 95% CI = 1.00–1.07). In the subgroup analysis by ethnicity, significantly increased risk was found for Caucasians (AC vs. AA: OR = 1.04, 95% CI = 1.00–1.08; CC vs. AA: OR = 1.05, 95% CI = 1.01–1.10; dominant model: OR = 1.04, 95% CI = 1.00–1.08) and Asians (CC vs. AA: OR = 1.35, 95% CI = 1.02–1.78; recessive model: OR = 1.38, 95% CI = 1.05–1.82). When stratified by study design, statistically significantly elevated risk was found among population-based studies (CC vs. AA: OR = 1.06, 95% CI = 1.01–1.11; dominant model: OR = 1.03, 95% CI = 1.00–1.07). In the subgroup analysis by menopausal status, no statistically significantly increased risk was found among premenopausal or postmenopausal women. In conclusion, this meta-analysis suggests that the IGFBP3 C allele is a low-penetrant risk factor for developing breast cancer.
Association between PPARγ, PPARGC1A, and PPARGC1B genetic variants and susceptibility of gastric cancer in an Eastern Chinese population

Abstract:Purpose: Previous studies showed that peroxisome proliferator-activated receptor gamma (PPARγ) and PPARγ coactivator1 family (PPARGC1A and PPARGC1B) gene single nucleotide variants (SNVs)were strongly associated with cancer susceptibility. The purpose of this study was to investigate the association of PPARγ, PPARGC1A, and PPARGC1B variants with the risk of gastric cancer (GC). Patients and methods: We performed a case-control study of 490 GC cases and 1,476 healthy controls from eastern China. PPARγ rs1801282 C > G, rs3856806 C > T, PPARGC1A rs2970847 C > T, rs8192678 C > T and PPARGC1B rs7732671 G > C, rs17572019 G > A SNVs were selected to investigate the association between these SNVs and GC susceptibility. Genotypes of the SNVs were assessed by multiplex fluorescent PCR using a custom-by-design 48-Plex SNPscantm Kit. Results: The PPARγ rs1801282 SNV was associated with a decreased risk for GC (GC vs. CC: odds ratio (OR) = 0.62, 95% confidence interval (95%CI) = 0.42-0.93, adjusted P = 0.019; GC + GG vs. GG: OR = 0.63 95%CI = 0.42-0.93, adjusted P = 0.019; respectively). In addition, stratified analysis revealed that the PPARγ rs1801282 SNV was correlated with the risk of GC in subgroups of age ≥ 61, no smoking, and no alcohol consuming. We also confirmed that the PPARγ rs3856806 C > T SNV promoted the risk of GC in women. The PPARGC1A rs8192678 TT genotype decreased the susceptibility of GC in men. The PPARGC1A rs2970847 C > T SNV decreased the susceptibility of GC in the subgroup of BMI ≥ 24 kg/m2. The PPARGC1B rs7732671 G > C and rs17572019 G > A SNVs promoted the risk of GC in the subgroup of BMI ≥ 24 kg/m2. Conclusion: This study indicates that the PPARγ, PPARGC1A, and PPARGC1B SNVs may be associated with the susceptibility of GC in eastern Chinese population. Future studies with larger populations, detailed H. pylori infection status for subgroup analysis, and functional study are needed to further clarify the relationship between these SNVs and GC risk.
Effect of Xeroderma pigmentosum complementation group F polymorphisms on gastric cancer risk and associations with H.pylori infection

Abstract:We conducted a hospital case-control study by genotyping four potential functional single nucleotide polymorphisms (SNPs) to assess the association of Xeroderma pigmentosum complementation group F (XPF) with gastric cancer susceptibility, and role of XPF polymorphisms in combination with H.pylori infection in risk definition. A total of 331 patients with gastric cancer and 355 controls were collected. Four SNPs of XPF, rs180067, rs1799801, rs2276466 and rs744154, were genotyped by Taqman real-time PCR method with a 7900 HT sequence detector system. The gastric cancer patients were more likely to have smoking habit, a family history of cancer and H.pylori infection. We did not find any significant difference in the genotype distributions of XPF rs180067, rs1799801, rs2276466 and rs744154 between cases and controls. However, multivariate logistic analysis showed a non-significant decreased risk in patients carrying rs180067 G allele, rs1799801 T allele or rs2276466 T allele genotypes. A non-significant increased risk of gastric cancer was found in individuals carrying the rs744154 GG genotype. Stratification by H.pylori infection and smoking was not significantly different in polymorphisms of XPF rs180067, rs1799801, rs2276466 and rs744154. The four XPF SNPs did not show significant interaction with H.pylori infection and smoking status (P for interaction was 0.35 and 0.18, respectively). Our study indicated that polymorphisms in rs180067, rs1799801, rs2276466 and rs744154 may affect the risk of gastric cancer but further large sample size studies are needed to validate any association.

Abstract:Background and Aim: As an important cytokine that modulate the cell cycle, the involvement of transforming growth factor beta-1 (TGF-beta 1) in carcinogenesis has been extensively studied for many years. Literatures have demonstrated that TGF-beta 1 gene polymorphisms may alter the risk of various cancers, such as lung, prostate and breast. To investigate whether polymorphisms of the TGF-beta 1 gene can modify the risk of gastric cancer, we conduct this hospital-based, case-control study.Methods: One hundred and sixty-seven cases and 193 gender, age-matched healthy controls were enrolled in this case-control study. TGF-beta 1 polymorphisms C-509T and T + 869C were identified by PCR-RFLP and ARMS-PCR protocols, respectively.Results: Significantly different distributions of both genes were demonstrated between the case and control. Variant genotypes -509CT, -509TT, +869TC and +869CC were associated with increased risk of gastric cancer (P = 0.001, OR = 2.54; P = 0.016, OR = 2.09; P < 0.001, OR = 3.46; P < 0.001, OR = 4.04, respectively). With haplotype analysis, wild type CT (-509C and +869T) led to a lower frequency in case than that in control (P < 0.001), while haplotype TC was more frequent in case than in control (P < 0.001). Multiple logistic regression analysis revealed that individuals with haplotype TC had an increased likelihood of developing gastric cancer (OR = 3.19, 95%CI = 1.72-5.90).Conclusions: Our findings imply that -509C > T and +869T > C gene polymorphisms in TGF-beta 1 may be a critical risk factor of genetic susceptibility to gastric cancer in the Chinese population.
