Abstract:Loss of TGFBI, a secreted protein induced by transforming growth factor-beta, has been implicated in cell proliferation, tumor progression, and angiogenesis by in vitro studies. However, in vivo antitumor functions of TGFBI as well as the underlying molecular mechanism are not well understood. To these aims, we have generated a mouse model with disruption of TGFBI genomic locus. Mice lacking TGFBI show a retarded growth and are prone to spontaneous tumors and 7,12-dimethylbenz(a)anthracene-induced skin tumors. In relation to wild-type (WT) mouse embryonic fibroblasts (MEF), TC7FBI(-/-) MEFs display increased frequencies of chromosomal aberration and micronuclei formation and exhibit an enhanced proliferation and early S-phase entry. Cyclin D1 is up-regulated In TGFBI(-/-) MEFs, which correlates with aberrant activation of transcription factor cyclic AMP-responsive element binding protein (CREB) identified by chromatin immunoprecipitation and luciferase reporter assays. TGFBI reconstitution in TGFBI(-/-) cells by either retroviral infection with WT TGFBI gene or supplement with recombinant mouse TGFBI protein in the culture medium leads to the suppression of CREB activation and cyclin D1 expression, and further inhibition of cell proliferation. Cyclin D1 up-regulation was also identified in most of the tumors arising from TGFBI(-/-) mice. Our studies provide the first evidence that TGFBI functions as a tumor suppressor in vivo. Abstract:PL02-02 Myeloid immune suppressor cells (MISCs), of myeloid macrophage/dendritic cell lineage, are significantly over-produced in the bone marrow and spleens of tumor-bearing mice as well as in peripheral blood of cancer patients. Interestingly, VEGF, the potent angiogenic factor and produced in most solid tumors, induces MISC production in vivo. In mice they express CD11b, a marker for myeloid cells of the macrophage lineage, and a marker for granulocytes, Gr-1. MISCs have been demonstrated to be immune suppressive since the 1980s. In this report, we demonstrate a novel tumor-promoting role of myeloid immune suppressor cells. These cells constitute 5% of total cells in solid tumors. Tumor cells coinjected with MISCs exhibited increased vascularization and decreased necrosis, as well as enhanced tumor growth. These immune cells produce high levels of MMP9. Deletion of MMP9 in these cells completely abolishes their tumor-promoting ability. MISCs were also found to directly incorporate into tumor endothelium. Consistent with this observation, MISCs acquire endothelial cell properties in tumor microenvironment and proangiogenic culture conditions. Furthermore, MISCs promote tumor metastasis through enhanced invasion. MISCs infiltrate into the invasive front with enhanced production and function of MMPs. Mice with 4T1 tumors co-injected with MISCs had significantly higher recurrent tumors at the injection site following surgical removal. 4T1 cells co-cultured with MISCs showed significantly increased invasion in vitro. Treatment with an MMP inhibitor significantly attenuated this process, suggesting a strong role of MISCs in tumor cell invasion through MMPs. In summary, MISCs are over-produced in tumor-bearing hosts, they can be re-programmed in multiple ways to promote tumor progression including angiogenesis, tumor growth and metastasis. Cellular and molecular targeting of MISC production should not only improve host immunosurveillance but also inhibit tumor metastasis, having the effect of “killing two birds with one stone.”

Abstract:Background TGFβ-induced (TGFBI/βig-H3) is a protein inducible by TGFβ1 and secreted by many types of cells. It binds to collagen, forms part of the extracellular matrix (ECM), and interacts with integrins on cell surfaces. In this study, we investigated the role of TGFBI in tumorigenesis and the underlying mechanisms. Methods Patient serum TGFBI levels were determined by ELISA. TGFBI transgenic and gene knockout mice and TGFBI-overexpressing liver cells were used for mechanistic studies. Results We demonstrated that patients with cholangiocarcinomas, hepatic carcinomas or gastric carcinomas presented significantly elevated serum TGFBI levels, and the excess TGFBI was derived from the tumor masses. TGFBI overexpression in mice resulted in increased incidence of spontaneous tumors and N,N-diethylnitrosamine (DEN)-induced liver tumor nodules, compared to that in wild type (WT) mice, while TGFBI knockout mice were comparable to WT controls in these 2 aspects. TGFBI promoted the survival of Aml-12 liver cells with DNA damage after irradiation, and augmented their post-irradiation proliferation. It activated the FAK/AKT/AKT1S1/PRS6/EIF4EBP pathway, which is known to modulate cell survival and proliferation. Conclusions Our data suggest that TGFBI functions as a promoter of certain gastrointestinal tract cancers. It provides a survival advantage to cells with DNA damage. Over a long time span, this advantage could translate into increased tumor risks.

TGFBI Protein High Expression Predicts Poor Prognosis in Colorectal Cancer Patients.

Abstract:Transforming growth factor-beta-induced (TGFBI) serves as a linker protein and plays a role in the activa- tion of morphogenesis, cell proliferation, adhesion, migration, differentiation and inflammation. High expression levels of the human TGFBI gene are correlated with numerous human malignancies. In order to explore the roles of TGFBI in the tumor progression of colorectal cancer, colorectal cancer specimens from 115 patients with strict follow-up were selected for the analysis of TGFBI by immunohistochemistry. The correlations between TGFBI expres- sion and the clinicopathological features of colorectal cancers were evaluated. In the colorectal cancer tissues, TGFBI was mainly localized in the cytoplasm and stroma and scarcely in the nucleus. TGFBI expression in the cyto- plasm and stroma was not found to be associated with age, gender, tumor histopathological grading, PT category and tumor location (P > 0.05 for each). However, high TGFBI expression in the cytoplasm and stroma correlated with lymph node metastasis, distant metastasis and Dukes stage (P < 0.05 for each). The survival rate was significantly lower in patients with high TGFBI expression than in those with low TGFBI expression. Furthermore, we found that tumor node metastasis (TNM) staging (HR: 2.963; 95% CI: 1.573-1.664; P = 0.000), differentiation (HR: 1.574; 95% CI: 1.001-2.476; P = 0.049) and high TGFBI cytoplasmic expression (HR: 3.332; 95% CI: 1.410-7.873; P = 0.000) proved to be independent prognostic factors for survival in colorectal cancer. In conclusion, TGFBI plays an important role in the progression of colorectal cancers and it is an independent poor prognostic factor for colorectal cancer patients.

Abstract:The human TGFB1 gene is polymorphic, and genetic variants are associated with altered cancer risk. However, human genetic association studies have had variable outcomes because TGFbeta1 action is context-dependent. We used the murine skin model of chemical carcinogenesis in genetic linkage analysis of three independent Mus musculus NIH/Ola x (Mus spretus x M. musculus NIH/Ola)F1 backcrosses, to identify a skin tumor susceptibility locus, Skts14, on proximal chromosome 7. Tgfb1 maps at the peak of linkage. The mouse Tgfb1 gene is polymorphic, resulting in cis-regulated differential allelic mRNA expression between M. spretus and M. musculus in F1 mouse skin. This phenomenon is reflected in differential phospho-SMAD2 levels, downstream of TGFbeta signaling, between these two mouse species. In normal F1 mouse skin, the Tgfb1SPR allele is expressed at higher levels than the Tgfb1NIH allele, and this differential is accentuated by phorbol 12-myristate 13-acetate treatment. In benign F1 papillomas, this imbalance is reversed, possibly by selection against expression of a hyperactive Tgfb1SPR allele in TGFbeta growth-responsive tumors. We demonstrate that skin tumor susceptibility is altered by Tgfb1 gene dosage, but that manifestation of Tgfb1-linked skin tumor susceptibility in M. musculus NIH/Ola x (M. spretus x M. musculus NIH/Ola)F1 backcross mice depends on interactions with another unlinked tumor modifying locus, Skts15, that overlaps Tgfbm3 on chromosome 12. These findings illustrate the power of complex genetic interactions in determining disease outcome and have major implications to the assessment of disease risk in individuals harboring variant TGFB1 alleles.

Abstract:Abstract Alterations in TGFβ signaling have significant effects on tumor initiation and progression. TGFβ functions both as a tumor suppressor and as a tumor promoter. Most work so far to dissect mechanisms underlying this dual role of TGFβ has been largely focused on differential regulation of signaling pathways by tumor autonomous TGFβ signaling. Studies from our lab demonstrate that TGFβ significantly regulate inflammation in tumor microenvironment and host immune response. When TGFβ R2 receptor is deleted in mammary cancer epithelial cells, there was an increased inflammation and recruitment of Gr-1+CD11b+ myeloid cells, which leads to increased tumor invasion and metastasis. This infiltration of Gr-1+CD11b+ cells also results in increased abundance of TGFβ1 and metalloproteinase activity in tumors with Tgfbr2 deletion. Interestingly, Tumor-infiltrating Gr-1+CD11b+ cells exhibited significant elevated TGFß signaling including increased expression of RI and RII receptor, TGFß1 and TGFß3 ligands, as well as latent transforming growth factor ß binding protein when compared with normal myeloid cells. Genetic deletion of Tgfbr2 in myeloid cells significantly attenuated 4T1 breast carcinoma tumor growth and metastasis. This is accompanied with significantly increased IFNγ production in the lungs of tumor bearing mice. These data suggest that TGFß, the tumor suppressor, also functions as an inflammation and immune suppressor. TGFß signaling in host derived myeloid cells is an important mediator in tumor immune surveillance. This new understanding of TGFß in inflammation and immune response in the interface of tumor and host may provide insight information for TGFß switch from a tumor suppressor to a tumor promoter.
