Reviews of Environmental Contamination and Toxicology

Abstract:From 2019 to 2060, the amount of plastic accumulated in the aquatic environment is expected to increase from 140 million tons to 493 million tons. The continuous release of microplastics (MPs) into the environment has negatively impacted aquatic ecosystems. MPs tend to develop biofilms on their surface in natural waters, and this new micro-ecosystem created by man-made plastic pollution is called plastisphere. The biofilm modifies the migration, transformation and fate, biological effects, and degradation processes of MPs. Although there have been numerous studies on MP biofilm, most focus on microbial diversity and structure. In addition, there are relatively few comprehensive descriptions of biofilm formation and research methods. In this paper, we review the recent works on microbes in the plastisphere, describe microbial interactions in the plastisphere, and examine the research methods, their benefits, and drawbacks concerning MP biofilms, and the four primary environmental factors (nutrient conditions, water temperature, the flow state of water, salinity) that influence microbial colonization. Next, we illustrate the whole process of how microorganisms colonize the MP surface. Finally, we study the microbial community of plastisphere in freshwater, marine, and wetland environments, and provide an outlook for future biofilm research on MPs.

Abstract:The plastisphere, which comprises the microbial community on plastic debris, rivals that of the built environment in spanning multiple biomes on Earth. Although human-derived debris has been entering the ocean for thousands of years, microplastics now numerically dominate marine debris and are primarily colonized by microbial and other microscopic life. The realization that this novel substrate in the marine environment can facilitate microbial dispersal and affect all aquatic ecosystems has intensified interest in the microbial ecology and evolution of this biotope. Whether a 'core' plastisphere community exists that is specific to plastic is currently a topic of intense investigation. This Review provides an overview of the microbial ecology of the plastisphere in the context of its diversity and function, as well as suggesting areas for further research.

Abstract:The pollution of plastic waste has become an increasingly serious environmental crisis. Recently, plastic has been detected in various kinds of environments, even in human tissues, which is an increasing threat to the ecosystems and humans. In the ocean, the plastic waste is eventually fragmentized into microplastics (MPs) under the disruption of physical and chemical processes. MPs are colonized by microbial communities such as fungi, diatoms, and bacteria, which form biofilms on the surface of the plastic called "plastisphere". In this review, we summarize the studies related to microorganisms in the plastisphere in recent years and describe the microbial species in the plastisphere, mainly including bacteria, fungi, and autotrophs. Secondly, we explore the interactions between MPs and the plastisphere. The depth of MPs in the ocean and the nutrients in the surrounding seawater can have a great impact on the community structure of microorganisms in the plastisphere. Finally, we discuss the types of MP-degrading bacteria in the ocean, and use the "seed bank" theory to speculate on the potential sources of MP-degrading microorganisms. Challenges and future research prospects are also discussed.

Homogenization of bacterial plastisphere community in soil: a continental-scale microcosm study

Abstract:Abstract Microplastics alter niches of soil microbiota by providing trillions of artificial microhabitats, termed the “plastisphere.” Because of the ever-increasing accumulation of microplastics in ecosystems, it is urgent to understand the ecology of microbes associated with the plastisphere. Here, we present a continental-scale study of the bacterial plastisphere on polyethylene microplastics compared with adjacent soil communities across 99 sites collected from across China through microcosm experiments. In comparison with the soil bacterial communities, we found that plastispheres had a greater proportion of Actinomycetota and Bacillota, but lower proportions of Pseudomonadota, Acidobacteriota, Gemmatimonadota, and Bacteroidota. The spatial dispersion and the dissimilarity among plastisphere communities were less variable than those among the soil bacterial communities, suggesting highly homogenized bacterial communities on microplastics. The relative importance of homogeneous selection in plastispheres was greater than that in soil samples, possibly because of the more uniform properties of polyethylene microplastics compared with the surrounding soil. Importantly, we found that the degree to which plastisphere and soil bacterial communities differed was negatively correlated with the soil pH and carbon content and positively related to the mean annual temperature of sampling sites. Our work provides a more comprehensive continental-scale perspective on the microbial communities that form in the plastisphere and highlights the potential impacts of microplastics on the maintenance of microbial biodiversity and ecosystem functioning.

Abstract:Microplastic (MP), an emerging contaminant, is globally prevalent and poses potential environmental threats and ecological risks to both aquatic and terrestrial ecosystems. When MPs enter into natural environments, they may serve as artificial substrates for microbial colonization and plastisphere formation, providing new ecological niches for microorganisms. Recent studies of the plastisphere have focused on aquatic ecosystems. However, our understanding of the soil plastisphere e.g. its formation process, microbial ecology, co-transport of organic pollutants and heavy metals, and effects on biogeochemical processes is still very limited. This review summarizes latest methods used to explore the soil plastisphere, assesses the factors influencing the microbial ecology of the soil plastisphere, and sheds light on potential ecological risks caused by the soil plastisphere. The formation and succession of soil plastisphere communities can be driven by MP characteristics and soil environmental factors. The soil plastisphere may affect a series of ecological processes, especially the co-transport of environmental contaminants, biodegradation of MPs, and soil carbon cycling. We aim to narrow the knowledge gap between the soil and aquatic plastisphere, and provide valuable guidance for future research on the soil plastisphere in MP-contaminated soils.
Deciphering the diversity and functions of plastisphere bacterial communities in plastic-mulching croplands of subtropical China

Abstract:Considering the inhomogeneity of plastisphere and surrounding soil, it is plausible that the microbial community colonizing it also varies, affecting soil services and sustainability. Herein, we analyzed the soil and film residue from fifty-five plastic-mulching croplands in the subtropical areas of China. Based on the outcomes of this analysis, we explored the diversity and functions of the associated bacterial communities. Alpha-diversity and phylogenetic diversity of the plastisphere bacterial community was significantly lower than the surrounding soil. The average net relatedness and net nearest taxa indices of samples were less than zero. Four phyla and twenty genera were enriched in the plastisphere compared to the surrounding soil. Ecological networks of the plastisphere community showed multiple nodes, but fewer interactions, and the members of Bradyrhizobium, Rhodospirillaceae, and Bacillus were indicated as the hub species. Predicted pathways related to human disease, as well as the metabolisms of cofactors, vitamins, amino acids, and xenobiotic biodegradation, were reinforced in the plastisphere, and meanwhile, accompanied by an increase in abundance of genes related to carbon, nitrogen, and phosphorus cycles. These results demonstrated the diversity and functions of the plastisphere microbiome and highlighted the necessity for exploring the ecological and health risks of plastic residue in croplands.

Abstract:Marine microplastic pollution is a growing problem for ecotoxicology that needs to be resolved. In particular, microplastics may be carriers of "dangerous hitchhikers," pathogenic microorganisms, i.e., Vibrio . Microplastics are colonized by bacteria, fungi, viruses, archaea, algae and protozoans, resulting in the biofilm referred to as the "plastisphere." The microbial community composition of the plastisphere differs significantly from those of surrounding environments. Early dominant pioneer communities of the plastisphere belong to primary producers, including diatoms, cyanobacteria, green algae and bacterial members of the Gammaproteobacteria and Alphaproteobacteria . With time, the plastisphere mature, and the diversity of microbial communities increases quickly to include more abundant Bacteroidetes and Alphaproteobacteria than natural biofilms. Factors driving the plastisphere composition include environmental conditions and polymers, with the former having a much larger influence on the microbial community composition than polymers. Microorganisms of the plastisphere may play key roles in degradation of plastic in the oceans. Up to now, many bacterial species, especially Bacillus and Pseudomonas as well as some polyethylene degrading biocatalysts, have been shown to be capable of degrading microplastics. However, more relevant enzymes and metabolisms need to be identified. Here, we elucidate the potential roles of quorum sensing on the plastic research for the first time. Quorum sensing may well become a new research area to understand the plastisphere and promote microplastics degradation in the ocean.

Abstract:The increasing number of plastic particles in the environment gives rise to a unique ecological niche called the plastisphere, which is characterized by a dynamic assemblage of microorganisms comprising autotrophs, heterotrophs, predators, and pathogens. This paper reviews the formation, characteristics, and factors influencing the terrestrial and aquatic plastisphere. The terrestrial and aquatic plastisphere forms when hitchhiking microorganisms, often bacteria, in the environment adhere to the surfaces of plastic particles and alter the surface properties of the particles for subsequent colonization of increasingly tightly clinging microorganisms. The terrestrial plastisphere is not as mobile as the aquatic plastisphere and is defined by the characteristics of the soil anchoring it. The microorganisms on the plastisphere are often defined by the dominant microorganisms in the surroundings, particularly those of the phyla Proteobacteria, Actinobacteria, Bacteroidota, Firmicutes, Chloroflexi, Acidobacteria, Cyanobacteria, Ascomycota, Basidiomycota, Chytridiomycota, Ciliophora, Ochrophyta, and Chlorophyta. However, the compositions and dominant microbial species on different plastic surfaces vary widely as they change with time, plastic properties, and biotic and abiotic environmental factors. Temporal changes in the plastisphere are due to succession. Plastic properties, particularly the type, surface properties, size, color, degree of aging, and chemical leaching, also affect the composition of the plastisphere. Biotic environmental factors comprising the dominant ambient microorganisms and interspecies interactions, together with the abiotic ones, including pH, temperature, nutrient availability, soil properties, salinity, and light, significantly shape the plastisphere. This review provides insight into the biodiversity of the plastisphere, and its roles in spreading pathogens and degrading plastics.

Abstract:The plastisphere, a unique microbial biofilm community colonizing plastic debris and microplastics (MPs) in aquatic environments, has attracted increasing attention owing to its ecological and public health implications. This review consolidates current state of knowledge on freshwater plastisphere, focussing on its biodiversity, community assembly, and interactions with environmental factors. Current biomolecular approaches revealed a variety of prokaryotic and eukaryotic taxa associated with plastic surfaces. Despite their ecological importance, the presence of potentially pathogenic bacteria and mobile genetic elements (i.e., antibiotic resistance genes) raises concerns for ecosystem and human health. However, the extent of these risks and their implications remain unclear. Advanced sequencing technologies are promising for elucidating the functions of plastisphere, particularly in plastic biodegradation processes. Overall, this review emphasizes the need for comprehensive studies to understand plastisphere dynamics in freshwater and to support effective management strategies to mitigate the impact of plastic pollution on freshwater resources.

Abstract:Fungi play important roles in biofilms, are very versatile in their ecological role, and are considered as plastic degraders. Here we aim to increase the resolution of the fungal members of the Plastisphere, to understand fungal substrate specificities and related potential ecological impacts. Fifteen-month-old fungal Plastisphere communities were investigated on 9 different plastic types and glass in seawater from the North Sea. By integrating scanning electron microscopy (SEM) imaging, ITS-based fingerprinting, and re-evaluated 18S rRNA gene sequence data through a fungal-specific phylogeny-based pipeline, we observed fungal Plastispheres and identified specific characteristics based on morphotypes, phylogeny, and biodiversity across different substrate types. Plastic types selected for specific fungal communities with polyolefine communities indicating significantly higher diversity compared to all other plastic types. Furthermore, specific plastic types may select for specific fungal taxa and their potential hosts, highlighting the complexity of marine biofilm food webs, and related ecological implications.

Abstract:The plastisphere is a new ecological niche. Compared to the surrounding water, microbial community composition associated with the plastisphere is known to differ with functional consequences. Here, this study characterized the bacterial and fungal communities associated with four types of plastisphere (polyethylene, polystyrene, polypropylene and polyvinyl chloride) in an estuarine habitat; assessed ecological functions including carbon, nitrogen, phosphorus and sulfur cycling, and determined the presence of antibiotic resistance genes (ARGs) and human pathogens. Stochastic processes dominated the community assembly of microorganisms on the plastisphere. Several functional genera related to nutrient cycling were enriched in the plastisphere. Compared to surrounding water and other plastisphere, the abundances of carbon, nitrogen and phosphorus cycling genes (cdaR, nosZ and chpy etc.) and ARGs (aadA2-1, cfa and catB8 etc.) were significantly increased in polyvinyl chloride plastisphere. In contrast, the polystyrene plastisphere was the preferred substrate for several pathogens being enriched with for example, Giardia lamblia 18S rRNA, Klebsiella pneumoniae phoE and Legionella spp. 23S rRNA. Overall, this study showed that different plastisphere had different effects on ecological functions and health risk in estuaries and emphasizes the importance of controlling plastic pollution in estuaries. Data from this study support global policy drivers that seek to reduce plastic pollution and offer insights into ecological functions in a new ecological niche of the Anthropocene.

Abstract:Plastics are offering a new niche for microorganisms colonizing their surface, the so-called "plastisphere," in which diversity and community structure remain to be characterized and compared across ocean pelagic regions. Here, we compared the bacterial diversity of microorganisms living on plastic marine debris (PMD) and the surrounding free-living (FL) and organic particle-attached (PA) lifestyles sampled during the Tara expeditions in two of the most plastic polluted zones in the world ocean, i.e., the North Pacific gyre and the Mediterranean Sea. The 16S rRNA gene sequencing analysis confirmed that PMD are a new anthropogenic ocean habitat for marine microbes at the ocean-basin-scale, with clear niche partitioning compared to FL and PA lifestyles. At an ocean-basin-scale, the composition of the plastisphere communities was mainly driven by environmental selection, rather than polymer types or dispersal effect. A plastisphere "core microbiome" could be identified, mainly dominated by Rhodobacteraceae and Cyanobacteria . Predicted functions indicated the dominance of carbon, nitrogen and sulfur metabolisms on PMD that open new questions on the role of the plastisphere in a large number of important ecological processes in the marine ecosystem.

Abstract:The microbial community colonized on microplastics (MPs), known as the 'plastisphere', has attracted extensive concern owing to its environmental implications. Coastal salt marshes, which are crucial ecological assets, are considered sinks for MPs. Despite their strong spatial heterogeneity, there is limited information on plastisphere across diverse environments in coastal salt marshes. Herein, a 1-year field experiment was conducted at three sites in the Yancheng salt marsh in China. This included two sites in the intertidal zone, bare flat (BF) and Spartina alterniflora vegetation area (SA), and one site in the supratidal zone, Phragmites australis vegetation area (PA). Petroleum-based MPs (polyethylene and expanded polystyrene) and bio-based MPs (polylactic acid and polybutylene succinate) were employed. The results revealed significant differences in bacterial community composition between the plastisphere and sediment at all three sites examined, and the species enriched in the plastisphere exhibited location-specific characteristics. Overall, the largest difference was observed at the SA site, whereas the smallest difference was observed at the BF site. Furthermore, the MP polymer types influenced the composition of the bacterial communities in the plastisphere, also exhibiting location-specific characteristics, with the most pronounced impact observed at the PA site and the least at the BF site. The polybutylene succinate plastisphere bacterial communities at the SA and PA sites were quite different from the plastispheres from the other three MP polymer types. Co-occurrence network analyses suggested that the bacterial community network in the BF plastisphere exhibited the highest complexity, whereas the network in the SA plastisphere showed relatively sparse interactions. Null model analyses underscored the predominant role of deterministic processes in shaping the assembly of plastisphere bacterial communities across all three sites, with a more pronounced influence observed in the intertidal zone than in the supratidal zone. This study enriches our understanding of the plastisphere in coastal salt marshes.

Abstract:The microbial colonization profiles on microplastics (MPs) in marine environments have recently sparked global interest. However, many studies have characterized plastisphere microbiomes without considering the ecological processes that underly microbiome assembly. Here, we carried out a three-timepoint exposure experiment at 1-, 4-, and 8-week and investigated the colonization dynamics for polyethylene, polypropylene, polystyrene, polyvinyl chloride, and acrylonitrile-butadiene-styrene MP pellets in natural coastal water. Using high-throughput sequencing of 16S rRNA, we found diversity and evenness were higher (p < 0.05) in the plastisphere communities than those in seawater, and microorganisms colonizing were co-influenced by environmental factors, polymer types, and exposure duration. Functional potential and co-occurrence network analysis revealed that MP exposure enriched the xenobiotic biodegradation potential and reduced the complexity of the MP microbial network. Simultaneously, null-model analyses indicated that stochastic processes contributed a bigger role than deterministic processes in shaping plastisphere microbial community structure with dispersal limitations contributing to a greater extent to microbial succession trajectories. These results implied the plastic surface had a more important role as a raft onto which microbes attach rather than selectively recruiting plastic-specific microbial colonizers. Our work strengthened the understanding of the ecological mechanisms by which microbial community patterns are controlled during colonization by plastic-associated microbes.
Plastisphere Microbiomes Respiring Persistent Organohalide Pollutants

Abstract:Plastics are invading nearly all ecosystems on earth, acting as emerging repositories for toxic organic pollutants and thereby imposing substantial threats to ecological integrity. The colonization of plastics by microorganisms, forming the plastisphere, has garnered attention due to its potential influence on biogeochemical cycles. However, the capability of plastisphere microorganisms to attenuate organohalide pollutants remains to be evaluated. This study revealed that the plastisphere, collected from coastal ecosystems, harbors unique microbiomes, while the natural accumulation of organohalide pollutants on plastics may favor the proliferation of organohalide-respiring bacteria (OHRB). Laboratory tests further elucidated the high potential of plastisphere microbiota to reductively dehalogenate a variety of organohalide pollutants. Notably, over 70% tested plastisphere completely debrominated tetrabromobisphenol A (TBBPA) and polybrominated diphenyl ethers (PBDEs) to nonhalogenated products, whereas polychlorinated biphenyls (PCBs) were converted to lower congeners under anaerobic conditions. Dehalococcoides, Dehalogenimonas, and novel Dehalococcoidia populations might contribute to the observed dehalogenation based on their growth during incubation and positive correlations with the quantity of halogens removed. Intriguingly, large fractions of these OHRB populations were identified in a lack of the currently known TBBPA/PBDEs/PCBs reductive dehalogenase (RDase) genes, suggesting the presence of novel RDase genes. Microbial community analyses identified organohalides as a crucial factor in determining the composition, diversity, interaction, and assembly of microbes derived from the plastisphere. Collectively, this study underscores the overlooked roles of the plastisphere in the natural attenuation of persistent organohalide pollutants and sheds light on the unignorable impacts of organohalide compounds on the microbial ecology of the plastisphere.

Global Municipal Solid Waste (MSW) landfills contain 42 % plastic waste, therefore representing one of the most significant plastispheres. MSW landfills are also the third largest anthropogenic methane sources and the important anthropogenic N2O source. Surprisingly, knowledge of microbiota and the associated microbial carbon and nitrogen cycles of landfill plastispheres is very limited. In this study, we characterized and compared the organic chemicals profile, bacterial community structure and metabolic pathway on plastisphere and the surrounding refuse in a large-scale landfill using GC/MS and 16S rRNA genes high-throughput sequencing, respectively. Landfill plastisphere and the surrounding refuse differed in organic chemicals composition. However, abundant phthalate-like chemicals were determined in both environments, implying the plastics additives leaching. Bacterial colonizing on the plastics surface had significantly higher richness than that in the surrounding refuse. Plastic surface and the surrounding refuse had distinct bacterial community composition. Genera of Sporosarcina, Oceanobacillus and Pelagibacterium were detected on the plastic surface with high abundance, while Ignatzschineria, Paenalcaligenes and Oblitimonas were rich in the surrounding refuse. Typical plastics biodegradation genus Bacillus, Pseudomonas and Paenibacillus were detected in both environments. However, Pseudomonas was dominant in plastic surface (up to 88.73 %), whereas Bacillus was rich in the surrounding refuse (up to 45.19 %). For the carbon and nitrogen cycle, plastisphere was predicted to had significant (P < 0.05) higher functional genes involved in carbon metabolism and nitrification, indicating more activated carbon and nitrogen microbial activity on the plastics surface. Additionally, pH was the main driver in shaping the bacterial community composition on plastic surface. These results indicate that landfill plastispheres serve as unique niches for microbial community habitation and function on microbial carbon and nitrogen cycles. These observations invite further study of the landfill plastispheres ecological effect.

Abstract:Microorganisms can colonize to the surface of microplastics (MPs) to form biofilms, termed "plastisphere", which could significantly change their physiochemical properties and ecological roles. However, the biofilm characteristics and the deep mechanisms (interaction, assembly, and biogeochemical cycles) underlying plastisphere in wetlands currently lack a comprehensive perspective. In this study, in situ biofilm formation experiments were performed in a park with different types of wetlands to examine the plastisphere by extrinsic addition of PVC MPs in summer and winter, respectively. Results from the spectroscopic and microscopic analyses revealed that biofilms attached to the MPs in constructed forest wetlands contained the most abundant biomass and extracellular polymeric substances. Meanwhile, data from the high-throughput sequencing showed lower diversity in plastisphere compared with soil bacterial communities. Network analysis suggested a simple and unstable co-occurrence pattern in plastisphere, and the null model indicated increased deterministic process of heterogeneous selection for its community assembly. Based on the quantification of biogeochemical cycling genes by high-throughput qPCR, the relative abundances of genes involving in carbon degradation, carbon fixation, and denitrification were significantly higher in plastisphere than those of soil communities. This study greatly enhanced our understanding of biofilm formation and ecological effects of MPs in freshwater wetlands.

Abstract:The plastisphere refers to biofilm formation on the microplastic (MP) surface, but its subsequent functions, especially driving the nitrogen biogeochemical cycle, are rarely studied. Here, MPs were incubated in the pelagic water and benthic water-sediment interface of an aquaculture pond, and the two corresponding microcosms amended with incubated plastisphere were simulated. The results showed decreased ammonia concentrations and increased nitrification rates in microcosms with either pelagic or benthic plastispheres. To uncover the possible mechanisms, the community structure and function of the plastisphere were investigated. As clarified by 16S rRNA, the community diversity of the pelagic plastisphere was significantly higher than that of the corresponding hydrosphere. Plastisphere communities, especially those incubated in pelagic water, were separated from the hydrosphere. Moreover, the abundance of Proteobacteria increased while the abundance of Cyanobacteria decreased in both plastispheres. Metagenome further revealed that the abundance of amoA and annotated Nitrososphaeraceae_archaeon and hao and affiliated Nitrosomonas_europaea, which contributed to ammonia oxidation to nitrite, was higher in the benthic plastisphere. Comparing the pelagic plastisphere with the corresponding hydrosphere, however, the abundance of nxrA and annotated Nitrobacter hamburgensis and nxrB and the affiliated Nitrospira moscoviensis, which are involved in nitrite oxidation, was more abundant in the plastisphere. These findings suggest that the plastisphere might selectively enrich functional microorganisms and genes in a habitat-dependent manner to promote nitrification in aquaculture ponds.

Abstract:Upon entering the marine environment, plastics are colonized by a plethora of microorganisms to form a plastisphere, influencing the fate and transport of the plastic debris and the health of marine ecosystems. The assembly of marine plastisphere is generally believed to be dominated by stochastic processes. However, it remains elusive whether microbial interaction in the assembly of plastisphere microbial communities is conserved or not. We analyzed the plastisphere microbiomes of 137 plastic debris samples from intertidal zones at different geographical locations and habitats (seagrass, coral, mangrove, beach, and open ocean) and compared them with the surrounding sediment and seawater microbiomes. Microbial community structures of the plastisphere from different locations were more similar to each other but differed substantially from the surrounding sediment and water microbiomes, implying a common mechanism of plastisphere assembly. We used different machine learning algorithms (Multinomial Logistic Regression, Support Vector Machine, Decision Trees, Random Forest, and Artificial Neural Networks) to classify plastic debris samples with high sensitivity based on the microbiome composition. Eukaryotic and prokaryotic phototrophic organisms such as green algae, diatoms, and cyanobacteria, were found to be enriched on the plastic surfaces. Network analysis revealed the central role of the phototrophic organisms in the formation and sustenance of the plastispheres. We found that phototrophs served as core members interacting strongly with heterotrophic organisms in marine plastisphere, irrespective of the sampling location, habitats, and polymer types. This would explain the stochastic assembly of the plastisphere along with conserved properties driven by the phototrophs in the surrounding environment. Our results highlight the importance of phototrophic organisms in shaping the marine plastisphere microbial communities.
