Effect of Acupotomy Intervention on Synovial Fibrosis in Rabbits with KOA

Abstract:Objective: To investigate the effect of acupotomy on synovial fibrosis in rabbits with knee osteoarthritis(KOA). Methods: New Zealand rabbits were randomly divided into the blank group, the model group, the acupotomy group and the electro-acupuncture(EA) group, with 8 rabbits in each group. The modified Videman left hind limb was fixed in straight position for 4 weeks to prepare the model. The blank group and the model group were not interfered, whereas the acupotomy group and the EA group were intervened for 3 weeks. Passive range of motion(PROM) was measured, the deposition of synovial collagen was observed by Sirius red staining, and the protein expressions of TGF-β1, α-SMA and COL-I in synovial tissues of knee joints were detected by Western Blot. Results: Compared with that in the model group, PROM was significantly increased in the acupotomy group and the EA group(P<0.01), and PROM of the acupotomy group was higher than that of the EA group(P<0.05). Compared with that in the model group, collagen deposition of synovial tissues was reduced in the acupotomy group and the EA group. The protein expressions of TGF-β1, α-SMA and COL-I were significantly higher in the model group than those in the blank group(P<0.01); the protein expressions of TGF-β1, α-SMA and COL-I were significantly inhibited in the acupotomy group and the EA group(P<0.01); compared with those in the EA group, the protein expressions of TGF-β1, α-SMA and COL-I were significantly decreased in the acupotomy group(P<0.05, P<0.01). Conclusion: Acupotomy can down-regulate the expressions of α-SMA and COL-I by regulating the expression of TGF-β1 to alleviate synovial fibrosis in treatment of KOA.
