Abstract:This paper reports a novel fluorescent aptasensor for the detection of acetamiprid pesticide with high sensitivity and selectivity based on the inner filter effect (IFE) of gold nanoparticles (AuNPs) toward fluorescent carbon dots (CDs). The aptasensor employs S-18 aptamer as the specific target recognition molecule and CDs as the signal transmission element. Free S-18 aptamer sequences can wrap the surfaces of AuNPs and enable their dispersion state even in the presence of high amounts of Tris-HCl salt, which can effectively quench the fluorescence of the CDs through the IFE. Upon adding acetamiprid pesticide, the free S-18 aptamer sequences are firstly exhausted to form some complexes; thus, the AuNPs tend to aggregate and their absorption spectrum no longer overlaps with the fluorescence emission spectrum of the CDs, which leads to obvious fluorescence recovery of the aptasensor. The properties of the CDs were extensively characterized, and the fluorescence quenching effects of the AuNPs toward the CDs were fully investigated. Additionally, the effects of some vital parameters, such as the type and amount of AuNPs, on the fluorescent aptasensor were also investigated. The proposed aptasensor has a detection limit as low as 1.08 g L-1 with a linear range of 5 to 100 g L-1 and shows high selectivity for acetamiprid over other control pesticides. Moreover, the aptasensor displays excellent accuracy and recovery in the detection of spiked real samples, suggesting that this fluorescent aptasensor can play important roles in the fields of food analysis and environmental detection.
Aptamer-based fluorescent screening assay for acetamiprid via inner filter effect of gold nanoparticles on the fluorescence of CdTe quantum dots

Abstract:This paper reports a novel aptamer-based fluorescent detection method for small molecules represented by acetamiprid based on the specific binding of aptamers with acetamiprid, and the inner filter effect (IFE) of gold nanoparticles (AuNPs) on the fluorescence of CdTe quantum dots (CdTe QDs). When CdTe QDs were mixed with AuNPs, the fluorescence of CdTe QDs was significantly quenched via IFE. The IFE efficiency could be readily modulated by the absorption and the aggregation state of AuNPs. The presence of salt could easily induce the aggregation of AuNPs, resulting in the fluorescence recovery of the quenched QDs. Acetamiprid-binding aptamer (ABA) could adsorb on the negatively charged AuNPs through the coordination interaction to protect AuNPs from salt-induced aggregation, so the fluorescence of CdTe QDs would be quenched by the IFE of AuNPs. However, the specific binding of ABA with acetamiprid could release the ABA from the surfaces of AuNPs and decrease the salt tolerance of AuNPs, so the IFE-decreased fluorescence of CdTe QDs was regained with the presence of acetamiprid, and the fluorescence enhancement efficiency was driven by the concentration of acetamiprid. Based on this principle, the aptamer-based fluorescent method for acetamiprid has been established and optimized. The assay exhibited excellent selectivity towards acetamiprid over its analogues and other pesticides which may coexist with acetamiprid. Under the optimum experiment conditions, the established method could be applied for the determination of acetamiprid with a wide linear range from 0.05 to 1.0 μM, and a low detection limit of 7.29 nM (3σ). Furthermore, this IFE-based method has been successfully utilized to detect acetamiprid in six types of vegetables, and the results were in full agreement with those from HPLC and LC-MS. The proposed method displays remarkable advantages of high sensitivity, rapid analysis, excellent selectivity, and would be suitable for the practical application of target screening in real samples. Abstract:<p>A syringe-aided apta-nanosensing method is reported for the colorimetric determination of acetamiprid. The method employs double-stranded (ds) DNA-conjugated gold agarose beads, i.e., as peroxidase-mimicking composite probes, in which the aptamer is indirectly attached to the AuNP surface through its hybridization with complementary DNA (cDNA). Upon contact with the acetamiprid target, the probes can give perceptible color change due to the possible conformation switch from dsDNA's brush-like to cDNA's 'pancake' regime. An "air-spaced pumping" procedure using a syringe equipped with ring magnets as the operation platform was proposed to facilitate the magnetic separation of the sensing probes. Therefore, the analytical steps can be easily accomplished in a syringe, including probe loading, acetamiprid capture and magnetic separation from crude samples, chromogenic reagent loading and colorimetric visualization. Acetamiprid concentration down to 3.3 ppb can be easily identified by the naked eye. The final solution also can be transferred for quantitative measurement. Under spectrometer, the ratio of the absorbance at 652 nm in the presence and absence of acetamiprid (A/A<sub>0</sub>) is linearly related to the acetamiprid concentration in the 0.4-4.5 ppb range. The limit of detection is calculated to be 0.24 ppb. Moreover, satisfactory recoveries ranging from 90.90 to 91.82 % with relative standard deviations of ≤ 2.96 % were obtained in analyzing real spiked samples.</p>

Aptamer-based Colorimetric Sensing of Acetamiprid in Soil Samples: Sensitivity, Selectivity and Mechanism.

Abstract:A simple and selective aptamer-based colorimetric method was developed for highly sensitive detection of acetamiprid, taking advantages of the sensitive target-induced colour changes that arisen from the interparticle plasmon coupling during the aggregation of Au nanoparticles (NPs). The results showed that the established method could be applied to detect acetamiprid in the linear range between 75 nM to 7.5 μM, with a low detection limit of 5 nM. Meanwhile, by employing an "artificial antibody" acetamiprid-binding aptamer (ABA) as recognition element, highly selective and specific colorimetric visualization of acetamiprid was obtained. It indicated that pesticides which may coexist with acetamiprid could not interfere with the detection of acetamiprid even that had similar structure with acetamiprid, such as imidacloprid and chlorpyrifos. Mechanism study suggested that it could be attributed to the specific supramolecular interaction between ABA and acetamiprid, as well as the resulted target-binding event induced conformation changes of ABA from random coil to hairpin structure. The practical application of the colorimetric method was realized for detecting acetamiprid in real soil samples and monitoring its natural degradation process.

Abstract:This study presents a novel colorimetric aptasensor, which seamlessly integrates gold nanoparticles (AuNPs) with the amplification potential of the hybridization chain reaction (HCR) for enhanced detection of acetamiprid. The aptamer, hybridized with a partially complementary strand that is covalently linked to AuNPs, serves as the recognition element for acetamiprid. The free end sequence of the aptamer, distal from the AuNP surface, functions as the initiating strand for the HCR, triggering the amplification process. In the absence of acetamiprid, the HCR efficiently occurs, conferring robust salt tolerance to the AuNPs and maintaining their characteristic red coloration. However, in the presence of acetamiprid, the aptamer preferentially binds to its target, disrupting the double-stranded structure and leading to the dissociation of the aptamer from the AuNPs. This dissociation results in a decrease in the HCR product, subsequently diminishing the salt tolerance of AuNPs and triggering a colorimetric transition from red to gray. This integration enhances sensitivity to 3.14 nM. Additionally, carbon quantum dots (CQDs) transduce colorimetric signals to fluorescent ones, further boosting the sensitivity to 0.24 nM. The aptasensor exhibits excellent selectivity and robustness. Real-world testing on tomato, peach, and lettuce shows recoveries of 98.50% to 100.36% with low standard deviations, validating its utility for pesticide residue analysis and food safety. This study provides a powerful tool for rapid and accurate pesticide detection, crucial for food safety.

Abstract:Gold nanoparticles (AuNPs)@N-(4-aminobutyl)-N-ethylisoluminol (ABEI)@Titanium dioxide nanorods (TiO2NRs) were used as sensing materials to produce a unique encapsulated nanostructure aptasensor for the detection of acetamiprid residues in this work. ABEI, an analog of luminol, was extensively used as an electrochemiluminescence (ECL) reagent. The ECL mechanism of ABEI- hydrogen peroxide (H2O2) system had connections to a number of oxygen-centered free radicals. TiO2NRs improved ECL response with high electron transfer and a specific surface area. AuNPs were easy to biolabel and could catalyze H2O2 to enhance ECL signal. AuNPs were wrapped around TiO2NRs by utilizing the reduction property of ABEI to form wrapped modified nanomaterials. The sulfhydryl-modified aptamer bound to the nanomaterial by forming gold-sulfur (Au-S) bonds. The aptamer selectively bound to its target with the addition of acetamiprid, which caused a considerable decrease in ECL intensity and enabled quantitative detection of acetamiprid. The aptasensor showed good stability, repeatability and specificity with a broad detection range (1×10-2-1×103 nM) and a lower limit of detection (3 pM) for acetamiprid residues in vegetables. Overall, this aptasensor presents a simple and highly sensitive method for ECL detecting acetamiprid, with potential applications in vegetable safety monitoring.

Abstract:Acetamiprid is mainly used to control pests. However, the residues of acetamiprid have resulted in significant negative effects on mammals. Many researchers are working on developing acetamiprid detection methods. In our study, four truncated aptamers were designed based on the secondary structure of the original aptamer. Molecular docking simulation of truncated aptamers confirmed that all of the truncated aptamers bind to acetamiprid, and their binding sites were identified. The affinity, sensitivity, and specificity of these truncated aptamers were measured by a spectrophotometric method with gold nanoparticles (AuNPs). Acetamiprid aptamers with 17-mer (Apt-J2) and 16-mer (Apt-J4) were successfully obtained, which show higher detection sensitivity and satisfactory specificity. Additionally, a spectrophotometric detection route superior to the original aptamer was explored, and the linear ranges were from 1 to 70 mu M and 0.5 to 40 mu M, respectively. The truncated aptamers were applied to the analysis of lake water, indicating that the aptamers have potential for practical analysis.

Determination of Acetamiprid by a Colorimetric Method Based on the Aggregation of Gold Nanoparticles

Abstract:A method was developed for the detection of the insecticide acetamiprid based on the strong interaction of the cyano group of acetamiprid with gold nanoparticles (AuNPs). The interaction results in the aggregation of gold nanoparticles and is accompanied by a color change from red to purple. The concentration of acetamiprid can be determined qualitatively and quantitatively by visually monitoring the color change or by using a spectrometer. Transmittance electron microscopy and UV-vis spectroscopy have been used to characterize the process. The experimental parameters were optimized with regard to the size of the AuNPs, pH, and incubation time. Under optimal experimental conditions, linear relationships between the logarithm of the concentration of acetamiprid and the absorbance were found over the range of 0.66 to 6.6 μM for AuNPs with diameters of 22.0 ± 1.0 nm and of 6.6–66 μM for AuNPs with diameters of 15.0 ± 1.0 nm. This method was successfully applied to detect acetamiprid in vegetables.
Aptamer Contained Triple-Helix Molecular Switch for Rapid Fluorescent Sensing of Acetamiprid.

Abstract:In this study, an aptamer-based fluorescent sensing platform using triple-helix molecular switch (THMS) was developed for the pesticide screening represented by acetamiprid. The THMS was composed of two tailored DNA probes: a label-free central target specific aptamer sequence flanked by two arm segments acting as a recognition probe; a hairpin-shaped structure oligonucleotide serving as a signal transduction probe (STP), labeled with a fluorophore and a quencher at the 3′ and 5′-end, respectively. In the absence of acetamiprid, complementary bindings of two arm segments of the aptamers with the loop sequence of STP enforce the formation of THMS with the “open” configuration of STP, and the fluorescence of THMS is on. In the presence of target acetamiprid, the aptamer-target binding results in the formation of a structured aptamer/target complex, which disassembles the THMS and releases the STP. The free STP is folded to a stem loop structure, and the fluorescence is quenched. The quenched fluorescence intensity was proportional to the concentration of acetamiprid in the range from 100 to 1200nM, with the limit of detection (LOD) as low as 9.12nM. In addition, this THMS-based method has been successfully used to test and quantify acetamiprid in Chinese cabbage with satisfactory recoveries, and the results were in full agreement with those from LC-MS. The aptamer-based THMS presents distinct advantages, including high stability, remarkable sensitivity, and preservation of the affinity and specificity of the original aptamer. Most importantly, this strategy is convenient and generalizable by virtue of altering the aptamer sequence without changing the triple-helix structure. So, it is expected that this aptamer-based fluorescent assay could be extensively applied in the field of food safety inspection.
Visual Aptasensor Based on PtNPs/g-C3N4 and DNase I for the Field Detection of Acetamiprid

Abstract:A visual aptasensor based on the combination of graphitic carbon nitride loaded with platinum nanoparticles (PtNPs/g-C3N4) and deoxyribonuclease I (DNase I) was developed for detecting acetamiprid. The prepared PtNPs/g-C3N4 exhibited excellent peroxidase (POD)-like activity, demonstrating the capacity to oxidize the colourless o-phenylenediamine (OPD) to produce the coloured product diaminophenazine (DAP) in the presence of hydrogen peroxide (H2O2). The DNA aptamer of acetamiprid can adsorb onto the surface of PtNPs/g-C3N4, thereby inhibiting its POD activity. The binding of acetamiprid to its aptamer results in the aptamer desorbing from the surface of PtNPs/g-C3N4 and subsequent digestion by DNase I. Ascribed to the synergistic effect of these factors, the aptasensor can achieve the rapid on-site detection of acetamiprid based on the acetamiprid-induced the colour change of the solution just with the smartphone. Furthermore, due to the remarkable fluorescence signal of DAP at 570 nm, the aptasensor under fluorescence mode enables highly sensitive and quantitative detection of acetamiprid with a linear range of 1 ng/mL to 4 µg/mL and a detection limit as low as 0.31 ng/mL. The aptasensor has successfully realized the detection of acetamiprid in river water and cucumber samples, offering a novel perspective for the rapid assessment of environmental pollution and food safety risks associated with acetamiprid residues.

Abstract:Accurate monitoring of trace pesticides in complex matrix remains a challenge in food safety supervision. Herein, we designed a facile zeolitic imidazolate framework (ZIF)-8/aptamer-based assay for the sensitive detection of acetamiprid. ZIF-8 efficiently adsorbs 6-carboxyfluorescein-labeled complementary DNA (cDNA-FAM) via electrostatic interaction, hydrogen bonding and Zn 2+ coordination, which contributed to resistance to cDNA-FAM displacement by biological ligands. ZIF-8 serves as an "ion pump" that contains lots of Zn 2+ who boosts cDNA-FAM adsorption and triggers the photoinduced electron transfer (PET) effect from FAM to ZIF-8, improving the sensing sensitivity. Acetamiprid could trigger the change in the adsorption state of cDNA-FAM, further tuning the PET effect and causing fluorescence conversion. The fluorescence assay showed a high sensitivity for monitoring acetamiprid with a detection limit of 0.05 ng mL −1 in the apple sample. This ZIF/DNA-based analytical platform provides a powerful tool for facile and low-cost screening of pesticide residues, with promising applications in food safety monitoring.

Abstract:As a common chlorinated nicotinic pesticide with high insecticidal activity, acetamiprid has been widely used for pest control. However, the irrational use of acetamiprid will pollute the environment and thus affect human health. Therefore, it is crucial to develop a simple, highly sensitive, and rapid method for acetamiprid residue detection. In this study, the capture probe (Fe3O4@Pt-Aptamer) was connected with the signal probe (Au@DTNB@Ag CS-cDNA) to form an assembly with multiple SERS-enhanced effects. Combined with magnetic separation technology, a SERS sensor with high sensitivity and stability was constructed to detect acetamiprid residue. Based on the optimal conditions, the SERS intensity measured at 1333 cm-1 is in relation to the concentration of acetamiprid in the range 2.25 × 10-9-2.25 × 10-5 M, and the calculated limit of detection (LOD) was 2.87 × 10-10 M. There was no cross-reactivity with thiacloprid, clothianidin, nitenpyram, imidacloprid, and chlorpyrifos, indicating that this method has good sensitivity and specificity. Finally, the method was applied to the detection of acetamiprid in cucumber samples, and the average recoveries were 94.19-103.58%, with RSD < 2.32%. The sensor can be used to analyse real samples with fast detection speed, high sensitivity, and high selectivity.
Aptasensor based on entropy-driven catalytic amplification system for sensitive detection of acetamiprid in Chinese herbal medicine

Abstract:The traditional method of acetamiprid residue detection is difficult to operate, time-consuming and laborious, and requires high professional knowledge of the detection personnel, which can no longer meet the rapid detection in the field. For this reason, a colorimetric aptasensor based on an entropy-catalyzed amplification system was developed for the ultrasensitive and selective determination of acetamiprid. In the absence of acetamiprid, the aptamer and cDNA build a double-stranded structure. The formed hemin/G-quadruplex mimicking DNAzyme can catalyze the substrate ABTS to generate the colored ion ABTS- with the help of H2O2, and the solution turns blue-green. On the contrary, the presence of acetamiprid triggers the release of cDNA, which in turn initiates the entropy-driven system, resulting in the inability to form DNAzyme and therefore no blue-green production of the solution. The quantity of acetamiprid determined the color period. Under the optimal experimental conditions, the method showed a linear correlation (R2=0.9837) for the detection of acetamiprid over the concentration range of 0.1 ~ 100 ng/mL, with a limit of detection of 0.06 ng/mL.The method was used for the determination of acetamiprid in spiked Coix lacryma and bitter almond, with the recoveries in the range of 90.3% ~ 110.3%. The proposed enzyme-free and label-free assay can be developed into a simple, sensitive and rapid detection platform.

Abstract:A highly sensitive aptamer electrochemical sensor based on hydroxylated black phosphorus /thionine/poly-L-lysine (hBP/TH/PLL) nanocomposite and Au nanoparticles (Au NPs) was herein introduced for detection of Acetamiprid. The hBP/TH/PLL nanocomposite was prepared through the capping effect of thionine and adhesion property of poly-L-lysine (PLL). The nanocomposite can maintain excellent electrical conductivity of black phosphorus and effectively prevent oxidation of black phosphorus. The sensor was based on hBP/TH/PLL composite and AuNPs, and used highly specific aptamers as sensor probes. It has advantages such as low detection limit, high selectivity, great repeatability and stability. Under optimum conditions, the linear range of aptamer electrochemical sensor is 10 fM to 1 μ M and detection limit is 9.407 fM. The aptamer electrochemical sensor has great selectivity and anti-interference, and has a potential application prospect in the field of rapid trace detection of pesticide residues.

Abstract:The work was based on N-(4-Aminobutyl)-N-ethylisoluminol (ABEI)-functionalized Fe-MIL-101 and gold nanoparticles (AuNPs) as sensing materials, and an electrochemiluminescence (ECL) aptasensor was constructed for detecting acetamiprid. As a metal-organic framework (MOF) material, Fe-MIL-101, was renowned for its unique three-dimensional network structure and efficient catalytic capability. ABEI, a common ECL reagent, was widely applied. ABEI was introduced into the Fe-MIL-101 structure as a luminescence functionalization reagent to form Fe-MIL-101@ABEI. This approach avoided limitations on the loading capacity of luminescent reagents imposed by modification and encapsulation methods. With character of excellent catalytic activity and ease of bioconjugation, AuNPs offered significant advantages in biosensing. Leveraging the reductive properties of ABEI, AuNPs were reduced around Fe-MIL-101@ABEI, resulting in the modified luminescent functionalized material denoted as Fe-MIL-101@ABEI@AuNPs. An aptamer was employed as a recognition element and was modified accordingly. The aptamer was immobilized on Fe-MIL-101@ABEI@AuNPs through gold-sulfur (Au-S) bonds. After capturing acetamiprid, the aptamer induced a decrease in the ECL signal intensity within the ABEI-hydrogen peroxide (H 2 O 2 ) system, enabling the quantitative detection of acetamiprid. The aptasensor displayed remarkable stability and repeatability, featured a detection range of 1×10 -3 -1×10 2 nM, and had a limit of detection (LOD) of 0.3 pM (S/N=3), which underscored its substantial practical application potential.

Abstract:A simple fluorescence detection platform has been established for acetamiprid assay based on DNA three-way junctions (TWJs), which can triple the fluorescence signal without any other amplification. It is designed with three single-stranded DNAs (ssDNA), each of which contains one-third or two-thirds of the G-quadruplex sequence at each end. Upon the addition of acetamiprid, the conformation of the aptamer-containing double-stranded DNA (dsDNA) changes from its original conformation and releases a strand of ssDNA. This ssDNA, with the other two ssDNAs, can assemble into DNA TWJs, and the three pairs of the branched ends of the DNA TWJs are adjacent to each other, allowing them to form three units of G-quadruplexes. Hence, the fluorescence of N -methyl mesoporphyrin IX (NMM) is lighted by the nascent G-quadruplexes. Graphene oxide (GO) is then added to minimize the detection background by absorbing the free NMM and non-target-induced ssDNA. The proposed strategy can assay acetamiprid in a wide linear range of 0–500 nM with a detection limit of 5.73 nM. More importantly, this assay platform demonstrates high potential for acetamiprid assay in food control and environmental monitoring.

Abstract:A simple aptasensor for sensitive and selective detection of acetamiprid has been developed based on electrochemical impedance spectroscopy (EIS). To improve sensitivity of the aptasensor, gold nanoparticles (AuNPs) were electrodeposited on the bare gold electrode surface by cycle voltammetry (CV), which was employed as a platform for aptamer immobilization. With the addition of acetamiprid, the formation of acetamiprid–aptamer complex on the AuNPs-deposited electrode surface resulted in an increase of electron transfer resistance (Ret). The change of Ret strongly depends on acetamiprid concentration, which is applied for acetamiprid quantification. A wide linear range was obtained from 5 to 600nM with a low detection limit of 1nM. The control experiments performed by employing the pesticides that may coexist or have similar structure with acetamiprid demonstrate that the aptasensor has only specific recognition to acetamiprid, resulting in high selectivity of the aptasensor. The dissociation constant, Kd of 23.41nM for acetamiprid–aptamer complex has been determined from the differential capacitance (Cd) by assuming a Langmuir isotherm, which indicates strong interaction between acetamiprid and aptamer, further proving high selectivity of the aptasensor. Besides, the applicability of the developed aptasensor has been successfully evaluated by determining acetamiprid in the real samples, wastewater and tomatoes.

Abstract:In this work, a fluorescence/localized surface plasmon resonance (Fluorescence/LSPR) dual-readout sensing system that using DNA functionalized gold nanoparticles (AuNPs) as nanoprobe was designed for acetamiprid (ACE) and organophosphorus pesticides (OPs) detection. The ACE detection was based on the fluorescence resonance energy transfer (FRET) between fluorochrome Cy3-labeled aptamer and AuNPs of nanoprobe. The high specificity of ACE allowed Cy3-labeled aptamer to release from nanoprobe, resulting in a significantly fluorescence recovery related to the concentration of ACE. On the other hand, the strategy for OPs detection relied on the acetylcholinesterase (AChE)-mediated oxidative polymerization blocking of dopamine on AuNPs surface and the inhibition of OPs toward AChE. The degree of polymerization of dopamine depended on the concentration of OPs, which led to a tremendous LSPR spectral shift and scattering color change of nanoprobe. The dual-readout sensing system showed great sensitivity and selectivity for ACE and OPs (used chlorpyrifos as model) with a detection limit of 166.7 pg mL−1 and 0.17fgmL−1, respectively. Moreover, two distinct color changes could be visual recognized to indicate the content of ACE and OPs, which was considered to be a conceivable way for pesticides residues assay. Therefore, this work paved a new avenue for rapid detection of multi-pesticides via a Fluorescence/LSPR dual signal response and visual screening, and hold great potential for noxious substance monitoring.
