Abstract:Herein we report that few-atom silver nanoclusters (Ag NCs) can be effective energy acceptors for upconversion phosphors (UCPs). A luminescence resonance energy transfer (LRET) probe for biothiols was constructed by decorating UCPs with dithiol-stabilized Ag NCs. Owing to the unique properties of ultrasmall NCs, properties which bridge the gap between those of small molecules and those of nanoparticles, the use of approximately 1.9 nm Ag NCs as energy acceptors endows the probe with high energy-transfer efficiency, good biocompatibility, and flexibility. The UCP-Ag NC nanoprobe enables rapid and robust target assay in solutions. It was also uploaded into living cells and used to detect intracellular biothiol levels with high discrimination. Moreover, the probe shows transportability in vivo and can be used for tissue imaging. The facile growth of few-atom metal NCs on diverse templates may enable the development of various nanoprobes combining UCPs and metal NCs.

Abstract:The enthusiasm for research on lanthanide-doped upconversion nanoparticles is driven by both a fundamental interest in the optical properties of lanthanides embedded in different host lattices and their promise for broad applications ranging from biological imaging to photodynamic therapy. Despite the considerable progress made in the past decade, the field of upconversion nanoparticles has been hindered by significant experimental challenges associated with low upconversion conversion efficiencies. Recent experimental and theoretical studies on upconversion nanoparticles have, however, led to the development of several effective approaches to enhancing upconversion luminescence, which could have profound implications for a range of applications. Herein we present the underlying principles of controlling energy transfer through lanthanide doping, overview the major advances and key challenging issues in improving upconversion luminescence, and consider the likely directions of future research in the field.

Abstract:Lanthanide-doped upconversion nanoparticles (UCNPs) have attracted considerable interest due to their superior physicochemical features, such as large anti-Stokes shifts, low autofluorescence background, low toxicity and high penetration depth, which make them extremely suitable for use as alternatives to conventional downshifting luminescence bioprobes like organic dyes and quantum dots for various biological applications. A fundamental understanding of the photophysics of lanthanide-doped UCNPs is of vital importance for discovering novel optical properties and exploring their new applications. In this review, we focus on the most recent advances in the development of lanthanide-doped UCNPs as potential luminescent nano-bioprobes by means of our customized lanthanide photophysics measurement platforms specially designed for upconversion luminescence, which covers from their fundamental photophysics to bioapplications, including electronic structures (energy levels and local site symmetry of emitters), excited-state dynamics, optical property designing, and their promising applications for in vitro biodetection of tumor markers. Some future prospects and efforts towards this rapidly growing field are also envisioned.

Abstract:Rare earth ions doped upconversion nanoparticles (UCNPs) are capable of stepwisely converting two or more lower energy near-infrared (NIR) excitation photons into a higher energy visible and ultraviolet emission photon, resulting in a large anti-Stokes shift of several hundred nanometers. Because the f-electron configurations of lanthanide ions have abundant energy levels, and many of them possess long lifetimes, UCNPs exhibit unique optical properties including optical tunability over emission wavelengths and lifetimes, better photo-stability and improved monochromatic color-purity and spatial resolution. However, the low luminescent efficiency, largely owing to their substantially weak and narrow band absorptions, has greatly hampered the translation of UCNPs-based technologies from the academic stage to real world products. Recently, increasing efforts have been paid on boosting the upconversion luminescence brightness and efficiency, including inert shells coating, high concentration of activators doping, organic dyes modification, and so on. Many strategies even aimed to explore high quality UCNPs with some more unique optical properties, such as orthogonal emissions or encoding luminescent signals, and broaden their applications.
A Hybrid Strategy to Enhance Small-Sized Upconversion Nanocrystals

Abstract:Upconversion nanoparticles (UCNPs) are characterized by high photostability, narrow spectral bands, excellent tuneability, and low biotoxicity, facilitating a broad range of biomedical applications. However, the small size required in many biological applications implies a lower luminescent brightness, as large surface-to-volume ratio is always accompanied with severe surface quenching. Herein, we introduce a strategy to overcome the surface quenching by incorporating an acceptor dye, sulforhodamine B (SRB) to surpass energy relaxation on long-lived lanthanide excited states. The surface modification of SRB led to up to 98.8% energy transfer efficiency, accompanied with the emergence of an intense SRB emission, with four orders of magnitude of change in the SRB/UCNPs emission ratio. The further structural optimisation led to an 8-fold upconversion emission enhancement. Moreover, the system exhibits excellent photostability, with only a 25% reduction over two hours under intense irradiation. By incorporating a pH responsive 5-carboxytetramethylrhodamine (5-TAMRA) to the UCNPs, we achieved a self-referencing protochromic sensor that are specific to protons and resistant to interference from various metal ions. This work provides a facile method for enhancing small-sized nanocrystals for biomedical sensing and imaging applications.

Abstract:Upconverting luminescent nanoparticles are photostable, nonblinking, and low chemically toxic fluorophores that are emerging as promising fluorescent probes at the single molecule level. High luminescence intensity upconversion nanoparticles (UCNPs) have previously been achieved by doping with high amounts of rare-earth ions using high excitation power (>2.5 MW/cm(2)). However, such particles are inadequate for in vitro live-cell imaging and single-particle tracking, as high excitation power can cause photodamage. Here, we compared UCNP luminescence intensities with different dopant concentrations and presented more efficient (about seven times) UCNPs at low excitation power by increasing the concentrations of Yb3+ and Tm3+ dopants (NaYF4: 60% Yb3+, 8% Tm3+) and adding a core-shell structure.

Abstract:Nanoprobes based on lanthanide-doped upconversion nanoparticles (UCNPs) exhibit promising potential in bioimaging and biosensing due to their unique optical properties. However, conventional UCNP nanoprobes based on the dye quenching effect are still limited in biosensing due to their low upconversion efficiency. The advent of dye-sensitized upconversion has resulted in nanoprobes with significantly enhanced efficiency; however, these still suffer from a high initial emissive background. In view of this, herein, we have constructed a dye-quenched/sensitized switching upconversion nanoprobe for high-contrast imaging of the pH-related tumor microenvironment. Under normal conditions, the luminescence of the nanoprobe at 540 nm (UCL 540 ) was significantly quenched by the employed dye. Upon being triggered by an acid, the dye would switch to its fluorescent form to sensitize the luminescence of UCNPs, affording a significant enhancement of UCL 540 . The switching from dye-quenched UCL to dye-sensitized UCL jointly enables the detection of a high signal-to-background ratio (high up to 50), allowing for high-contrast mapping of the tumor specific acidic microenvironment in vivo . We believe that this nanoplatform holds considerable promise for acid-related sensing.

Abstract:Upconversion nanocrystals (UCNCs) hold promise for bioimaging, solar cells, photocatalysis and volumetric displays. However, their upconversion luminescence intensities are usually low due to the weak and narrowband near-infrared absorption of lanthanide ions. Herein, we introduce and validate a strategy to hugely enhance upconversion luminescence intensity by using an organic near-infrared dye as an antenna to sensitize core/shell UCNCs. The dye can increase absorptivity and broaden the absorption spectrum of the UCNCs. Such dye sensitization, in combination with a core/shell structure, can tremendously enhance the upconversion luminescence (UCL) intensity of the UCNCs. The UCL intensity of dye-sensitized UCNCs excited at 820 nm is 800-folds higher than that of pure UCNCs excited at 980 nm. Further enhancement can be obtained by optimization of the dye emission and UCNC absorption spectral overlap. Moreover, the proposed approach can be extended to cover any part of the solar spectrum by using a set of dyes. This work provides new insights into the efficient enhancement of upconversion luminescence of the UCNCs and facilitates their applications.

Abstract:Overdose of Cu2+ is associated with multiple diseases, such as Wilson disease, Parkinson disease, and Alzheimer disease. Therefore, detections of Cu2+ in vivo and in vitro are meaningful. Near-infrared fluorescent probes are commonly applied for the detection of Cu2+ in real time. With mono detection signal provided, these probes are not persuasive when applied to complicated biological environments, such as cells and animals. In this report, we conjugated the organic fluorescent probe CYDAC16 with the UCNPs (lanthanide-doped upconversion nanoparticles) to develop a ratiometric luminescent probe for Cu2+. The composite material UCNPs-CYDAC16 provides a ratiometric signal based on an upconversion luminescence 660 and 800 nm. With good sensitivity and selectivity for Cu2+, it works well in vivo and in vitro for the detection of Cu2+. We believe it is promising in further biological applications.

Abstract:Upconversion nanocrystals that converted near-infrared radiation into emission in the ultraviolet spectral region offer many exciting opportunities for drug release, photocatalysis, photodynamic therapy, and solid-state lasing. However, a key challenge is the development of lanthanide-doped nanocrystals with efficient ultraviolet emission, due to low conversion efficiency. Here, we develop a dye-sensitized, heterogeneous core–multishelled lanthanide nanoparticle for ultraviolet upconversion enhancement. We systematically study the main influencing factors on ultraviolet upconversion emission, including dye concentration, excitation wavelength, and dye-sensitizer distance. Interestingly, our experimental results demonstrate a largely promoted multiphoton upconversion. The underlying mechanism and detailed energy transfer pathway are illustrated. These findings offer insights into future developments of highly ultraviolet-emissive nanohybrids and provide more opportunities for applications in photo-catalysis, biomedicine, and environmental science.

Abstract:Lanthanide-doped upconversion nanoparticles (UCNPs) have recently aroused enormous attention, due to large anti-Stokes shifts, weak autofluorescence, less photodamage and photobleaching, as well as deeper tissue penetration. Owing to their advantageous photophysical properties, UCNPs have been widely applied in analytical and biomedical research fields, such as sensing, imaging, and therapy. This review mainly focuses on recent advances in chromophore-assembled upconversion nanoprobes (UC nanoprobes) for chemo/biosensing. We first demonstrate the principle for constructing UC nanoprobes, and then give a detailed summary of chromophore loading strategies. Furthermore, recent achievements on the construction and application of chromophore-loaded UC nanoprobes are summarized. Also, the current challenges and future prospects of UC nanoprobes are discussed as a conclusion. We expect this review will offer valuable guidance on the selection of chromophores and their loading strategies to further facilitate the fabrication and applications of chromophore-assembled UC nanoprobes.

Abstract:Near-infrared (NIR) dye-sensitized upconversion nanoparticles (UCNPs) can broaden the absorption range and boost upconversion efficiency of UCNPs. Here, we achieved significantly enhanced upconversion luminescence in dye-sensitized core/active shell UCNPs via the doping of ytterbium ions (Yb(3+)) in the UCNP shell, which bridged the energy transfer from the dye to the UCNP core. As a result, we synergized the two most practical upconversion booster effectors (dye-sensitizing and core/shell enhancement) to amplify upconversion efficiency. We demonstrated two biomedical applications using these UCNPs. By using dye-sensitized core/active shell UCNP embedded poly(methyl methacrylate) polymer implantable systems, we successfully shifted the optogenetic neuron excitation window to a biocompatible and deep tissue penetrable 800 nm wavelength. Furthermore, UCNPs were water-solubilized with Pluronic F127 with high upconversion efficiency and can be imaged in a mouse model.

Abstract:Owing to their unique photo-physical properties, rare-earth ions-doped upconversion nanoparticles (UCNPs) have attracted extensive attention in recent years. UCNPs have many special merits, such as a long luminescence lifetime, narrow emission band widths, high quantum yields and low toxicity, which allows their potential applications in bio-medical field, biological luminescent labels and drug delivery carriers. Compared with traditional fluorescence labels exited by UV (ultraviolet), such as organic dyes and quantum dots, UCNPs can transfer near-infrared (NIR) light into visible light, which is commonly called upconversion luminescence (UCL). Abstract:Lanthanide-doped upconversion nanomaterials that convert low energy excitation wavelength to high energy emission have attracted considerable attention due to the advantages of high penetration depth and high resistance to autofluorescence interference capability. This review highlights the literature of the past several years covering recent advances of upconversion nanoprobes for biodetection. We begin by discussions of the key points in fabrication of lanthanide-activated nanoprobes. We further conduct a careful evaluation of various detection applications of upconversion nanoprobes, including temperature, ions, small molecules, DNA, enzyme, virus, and etc. We finish the discussion by assessing current challenges and future prospects of upconversion nanoprobes for fundamental studies and clinical applications.

Abstract:The fabrication of lanthanide upconversion nanocomposites as probes has become a new research hotspot due to its special advantages via utilizing upconversion luminescence (UCL) as a detection signal. Herein, a hybrid organic dye modified upconversion nanophosphor is successfully developed as a nanoprobe for cysteine/homocysteine. Yolk-shell structured upconversion nanoparticles (YSUCNP) with lanthanide upconversion nanophosphor as moveable core and silica as mesoporous shell are synthesized, and a colorimetric chemodosimeter for cysteine/homocysteine is accommodated in the hollow cavities. Thus, cysteine/homocysteine can be quantitatively detected on the basis of luminescent resonance energy transfer (LRET) in a UCL turn-off pattern. The dye-loaded YSUCNP possess good dispersibility in aqueous solution; thus detection of the targeted molecule can be achieved in pure water. Cellular experiments carried out with laser-scanning upconversion luminescence microscopy further demonstrate that the dye-loaded YSUCNP can serve as an intracellular nanoprobe to detect cysteine/homocysteine. Moreover, this dye-loading protocol can be developed as a common approach to construct other chemodosimeter-modified UCNP hybrid nanoprobes, as proved by a UCL turn-on style sensor for cyanide.

Abstract:In this work, we developed a simple and selective luminescence sensing system for detecting ascorbic acid (AA) based on NaGdF4:Yb,Er@NaYF4 upconversion nanoparticles (UCNPs) and oxidase-like CoOOH nanoflakes. When p-phenylenediamine (PPD) and oxidase-like CoOOH nanoflakes were added to the UCNPs solutions, PPD, as a typical substrate of oxidase, could be oxidized to PPDox. The luminescence intensity of UCNPs was quenched because PPDox could play a role as an energy acceptor. When AA was added to the above solution mixture, the CoOOH nanoflakes were destroyed, and then, the luminescence intensity was recovered. Under the best experimental conditions, the linear range for detection of AA was 0.8 to 280 μM, and the detection limit was 0.25 μM. Furthermore, the system could be applied to real sample analysis with satisfactory results.

Abstract:Upconversion (UC) process in lanthanide-doped nanomaterials has attracted great research interest for its extensive biological applications in vitro and in vivo, benefiting from the high tissue penetration depth of near-infrared excitation light and low autofluorescence background. However, the 980 nm laser, typically used to trigger the Yb3+-sensitized UC process, is strongly absorbed by water in biological structures and could cause severe overheating effect. In this article, we report the extension of the UC excitation spectrum to shorter wavelengths, where water has lower absorption. This is realized by further introducing Nd3+ as the sensitizer and by building a core/shell structure to ensure successive Nd3+→Yb3+→activator energy transfer. The efficacy of this Nd3+-sensitized UC process is demonstrated in in vivo imaging, and the results confirmed that the laser-induced local overheating effect is greatly minimized.

Abstract:Upconversion nanoparticles (UCNPs) is a class of promising probes widely used in protein molecules imaging due to the no photobleaching and non-blinking emission. However, it is still remained challenging to synthesize a type of ultra-small but bright UCNPs. In this paper, a new class of efficient sub-5-nm upconversion nanodots (UCNDs) was elaborately designed and experimentally demonstrated. The proposed uniform UCNDs comprise two parts: a 3.5-nm Ln3+-doped ZrO2 core and 0.5-nm NaYF4 shell. The emission intensity of the proposed UCNDs was measured to be more than 10 times brighter than that of the conventional 10-nm NaYF4 UCNPs. Under 300 kW·cm−2 irradiance excitation, the 15 mol% Er3+-doped UCNDs exhibit an 18-fold enhanced intensity and one fifth emission lifetime compared to the 0.5 mol% Er3+-doped ones. Using the biocompatible UCNDs, the aptamer-mediated proteins-targeted imaging was performed and demonstrated high efficiency. These ultra-small, efficient UCNDs would have great potentials in rapid scanning and cellular imaging.

Abstract:The ability to rapidly, sensitively, and selectively detect ascorbic acid (AA) is important for medical assays and diagnoses. Here, a new cobalt oxyhydroxide (CoOOH)-functionalized up-conversion nanoparticle (UCNP) system was designed for the detection of AA. For the UCNPs, ligand-free β-NaYF4:Gd/Yb/Tm/Ho@NaYF4 offered simultaneous blue and red light emission, but CoOOH on the UCNP surface acted as a quencher that could effectively absorb the 475 nm-wavelength light produced by the luminescence of the nanoparticles. This quenching was found to be easily eliminated by the addition of AA through the FRET process. The ratio of the intensity of the up-conversion luminescence at 475 nm to that at 654 nm showed an excellent linear correlation with the concentration of AA. The nanoprobe also exhibited excellent accuracy in the quantitative analysis of AA in diluted fetal bovine serum. In addition, the probe showed an ability to be used for imaging AA in living cells, and may thus be considered as a good candidate for use in clinical diagnosis and drug screening.
