Abstract:AbstractMonomethylated selenium (MM-Se) forms that are precursors of methylselenol, such as methylseleninic acid (MSeA), differ in metabolism and anticancer activities in preclinical cell and animal models from seleno-methionine that had failed to exert preventive efficacy against prostate cancer in North American men. Given that human prostate cancer arises from precancerous lesions such as high-grade prostatic intraepithelial neoplasia (HG-PIN), which frequently have lost phosphatase and tensin homolog (PTEN) tumor suppressor permitting phosphatidylinositol-3-OH kinase (PI3K)–protein kinase B (AKT) oncogenic signaling, we tested the efficacy of MSeA to inhibit HG-PIN progression in Pten prostate-specific knockout (KO) mice and assessed the mechanistic involvement of p53-mediated cellular senescence and of the androgen receptor (AR). We observed that short-term (4 weeks) oral MSeA treatment significantly increased expression of P53 and P21Cip1 proteins and senescence-associated-β-galactosidase staining, and reduced Ki67 cell proliferation index in Pten KO prostate epithelium. Long-term (25 weeks) MSeA administration significantly suppressed HG-PIN phenotype, tumor weight, and prevented emergence of invasive carcinoma in Pten KO mice. Mechanistically, the long-term MSeA treatment not only sustained P53-mediated senescence, but also markedly reduced AKT phosphorylation and AR abundance in the Pten KO prostate. Importantly, these cellular and molecular changes were not observed in the prostate of wild-type littermates which were similarly treated with MSeA. Because p53 signaling is likely to be intact in HG-PIN compared with advanced prostate cancer, the selective superactivation of p53-mediated senescence by MSeA suggests a new paradigm of cancer chemoprevention by strengthening a cancer progression barrier through induction of irreversible senescence with additional suppression of AR and AKT oncogenic signaling. Abstract:Proceedings: AACR 104th Annual Meeting 2013; Apr 6-10, 2013; Washington, DC Prostate cancer (PCa) arises from progression of pre-invasive lesions such as prostatic intraepithelial neoplasia (PIN) to adenocarcinoma and metastatic disease. Due to the very long latency of lesion progression, chemoprevention of prostate adenocarcinogenesis is recognized as a plausible and essential approach to win the war against PCa. Whereas the results from recent clinical trials (SWOG 9917 and SELECT) did not support the protective function for seleno-methionine (SeMet) supplement in healthy men with or without high grade PIN, we and others have shown that supra-nutritional supplement of methylseleninic acid (MSeA), which differs, both chemically and metabolically, from SeMet, suppressed tumor growth in human prostate cancer xenografts, and in the transgenic adenocarcinoma mouse prostate (TRAMP) model. However, the former models did not mimic primary prevention, and the TRAMP mice vastly differ from the etiology and adenocarcinogenesis nature of the human prostate cancers. Therefore, a mouse model that closely recapitulates the clinic pathological features of human prostate cancer will be essential and necessary to predict the preventive efficacy and mechanisms of MSeA in men. As the loss of tumor suppressor gene Pten (phosphatase and tensin homolog deleted on chromosome 10) plays a causal role in human prostate cancers, the prostate epithelial-specific Pten-knockout mouse is such a preclinical model of choice. Here we utilized Probasin-Cre (Pb-Cre) transgenic mice expressing Cre to delete Pten in prostate epithelium to investigate whether administration of MSeA inhibited prostate tumorigenesis. We treated Pten-knockout and age-matched wild type male mice from 8 weeks of age with water or MSeA at a dose of 3 mg/kg body weight five times a week (Monday through Friday) for 27 weeks and sacrificed them for analyses. Our results show that MSeA significantly suppressed prostate tumorigenesis as evidenced by (1) suppression of tumor progression to high grade PIN or/and adenocarcinoma; (2) a 50% reduction in prostate tumor weight (P<0.01); (3) a significant reduction in cellular proliferation determined by ki67 immunostaining (P<0.01); and (4) attenuation of androgen receptor (AR) signaling. Mechanistically, we found that MSeA induced a super-activation of p53, p21 and cellular senescence. Because intact p53 signaling is preserved in most early prostate lesions, our findings provide compelling rationale to consider MSeA as a bioactive dietary supplement to activate p53-p21 mediated senescence as a barrier to prevent PIN from progressing to advanced prostate cancer. Methyl-Selenium Compounds Inhibit Prostate Carcinogenesis in the Transgenic Adenocarcinoma of Mouse Prostate Model with Survival Benefit

Abstract:Chemoprevention of prostate cancer by second-generation selenium compounds in reference to selenomethionine holds strong promise to deal with the disease at the root. Here we used the transgenic adenocarcinoma mouse prostate (TRAMP) model to establish the efficacy of methylseleninic acid (MSeA) and methylselenocysteine (MSeC) against prostate carcinogenesis and to characterize potential mechanisms. Eight-week-old male TRAMP mice (C57B/6 background) were given a daily oral dose of water, MSeA, or MSeC at 3 mg Se/kg body weight and were euthanized at either 18 or 26 weeks of age. By 18 weeks of age, the genitourinary tract and dorsolateral prostate weights for the MSeA- and MSeC-treated groups were lower than for the control (P < 0.01). At 26 weeks, 4 of 10 control mice had genitourinary weight >2 g, and only 1 of 10 in each of the Se groups did. The efficacy was accompanied by delayed lesion progression, increased apoptosis, and decreased proliferation without appreciable changes of T-antigen expression in the dorsolateral prostate of Se-treated mice and decreased serum insulin-like growth factor I when compared with control mice. In another experiment, giving MSeA to TRAMP mice from 10 or 16 weeks of age increased their survival to 50 weeks of age, and delayed the death due to synaptophysin-positive neuroendocrine carcinomas and synaptophysin-negative prostate lesions and seminal vesicle hypertrophy. Wild-type mice receiving MSeA from 10 weeks did not exhibit decreased body weight or genitourinary weight or increased serum alanine aminotransferase compared with the control mice. Therefore, these selenium compounds may effectively inhibit this model of prostate cancer carcinogenesis.
Superior in vivo inhibitory efficacy of methylseleninic acid against human prostate cancer over selenomethionine or selenite.

Abstract:Methylselenol has been implicated as an active anticancer selenium (Se) metabolite. However, its in vivo efficacy against prostate cancer (PCa) has yet to be established. Here, we evaluated the growth inhibitory effects of two presumed methylselenol precursors methylseleninic acid (MSeA) and Se-methylselenocysteine (MSeC) in comparison with selenomethionine (SeMet) and selenite in DU145 and PC-3 human PCa xenografts in athymic nude mice. Each Se was given by a daily single oral dose regimen starting the day after the subcutaneous inoculation of cancer cells. We analyzed serum, liver and tumor Se content to confirm supplementation status and apoptosis indices and tumor microvessel density for association with antitumor efficacy. Furthermore, we analyzed lymphocyte DNA integrity to detect genotoxic effect of Se treatments. The data show that MSeA and MSeC exerted a dose-dependent inhibition of DU145 xenograft growth and both were more potent than SeMet and selenite, in spite of less tumor Se retention than in the SeMet-treated mice. Selenite treatment increased DNA single-strand breaks in peripheral lymphocytes, whereas the other Se forms did not. Terminal deoxynucleotidyl transferase biotin-dUTP nick end labeling (TUNEL) and cleaved caspase-3 indices (apoptosis) from MSeC-treated tumors were higher than tumors from control mice or MSeA-treated mice, whereas the microvessel density index was lower in tumors from MSeA-treated mice. In the PC-3 xenograft model, only MSeA was growth inhibitory at a dose of 3 mg/kg body wt. In summary, our data demonstrated superior in vivo growth inhibitory efficacy of MSeA over SeMet and selenite, against two human PCa xenograft models without the genotoxic property of selenite.
Methylseleninic Acid Suppresses Pancreatic Cancer Growth Involving Multiple Pathways

Abstract:As a potential novel agent for treating pancreatic cancer, methylseleninic acid (MSeA) was evaluated in cell culture and xenograft models. Results showed that MSeA induced G(1) cell cycle arrest and apoptosis in a majority of human and mouse pancreatic cancer cell lines, but G(2) arrest in human PANC-1 and PANC-28 cell lines. In contrast to our previous finding in human prostate cancer LNCaP cells having a lack of P53 activation by MSeA, induction of G(2) arrest in PANC-1 cells was accompanied by increased mutant P53 Ser15 phosphorylation, upregulation of P53-targets P21Cip1 and GADD45 and G(2) checkpoint kinase (Chk2) activation, suggestive of DNA damage responses. A rapid inhibition of AKT phosphorylation was followed by reduced mTOR signaling and increased autophagy in PANC-1 cells attenuating caspase-mediated apoptosis execution. Furthermore, daily oral treatment with MSeA (3mg Se/kg body weight) significantly suppressed growth of subcutaneously inoculated PANC-1 xenograft in SCID mice. Immunohistochemical analyses detected increased p-Ser15 P53, P21Cip1, pS139-H2AX (DNA damage responses), and caspase-3 cleavage and decreased pSer473AKT and Ki67 proliferative index and reduced intratumor vascular density in MSeA-treated xenograft. These results provide impetus for further research of MSeA in the therapy and/or chemoprevention of pancreatic cancer.

Abstract:Selenomethionine (SeMet) did not prevent prostate cancer in the SELECT trial and in two hormone-driven rat models. However, we have shown that daily oral bolus administration of next-generation selenium forms, methylseleninic acid (MSeA) and Se-methylselenocysteine (MSeC) at 3 mg Se/kg body weight, inhibits prostate carcinogenesis in the TRAMP and pten-deficient mouse models and In Vivo growth of human prostate cancer cells. Here, we determined whether these Se forms prevent prostate cancer in a chemically induced-androgen promoted carcinogenesis rat model in which SeMet was not preventive. WU rats were treated with methylnitrosourea, and one week later, slow-release testosterone implants when they were randomized to groups fed AIN-93M diet supplemented with 3 ppm selenium as MSeA or MSeC or control diet. Mean survival, tumor incidence in all accessory sex glands combined (dorsolateral and anterior prostate plus seminal vesicle) and the incidence of tumors confined to dorsolateral and/or anterior prostate were not statistically significantly different among the groups. Thus, MSeA and MSeC feeding was not preventive in this model. The contrast with the inhibitory effects of MSeA and MSeC in mouse models may be due to differences in carcinogenic mechanisms, selenium dosage, delivery mode, and pharmacokinetics or fundamental rat-mouse differences in selenium metabolism.

Abstract:A127 Inhibition of cancer cell proliferation through the induction of G1 cell cycle arrest by methylselenol may, to some extent, account for the cancer chemopreventive activity of selenium. Previous studies have documented the induction of cyclin-dependent kinase (CDK) inhibitor proteins p21cip1 and/or p27kip1 and G1 arrest by the methylselenol precursor methylseleninic acid (MSeA) in prostate and other cancer cells. However, whether these CDK inhibitors play a critical mediator role in G1 arrest by MSeA has not been addressed. In the present work, we show exposure of p53-mutant DU145 prostate cancer (PCa) cells to sub-apoptotic concentrations of MSeA induced p21cip1 mRNA (3 h) and protein (6 h) much faster than p27kip1 mRNA (12 h) and protein (12 h). Immunoprecipitation detected increased binding of p21cip1 to CDK2 and CDK6 in MSeA-exposed DU145 cells. Knocking down of p21cip1 by siRNA completely abolished G1 arrest induction by MSeA in DU145 cells, yet si-p27kip1 had no effect. G1 arrest was observed in p53-wild type LNCaP PCa cells and p53-null PC-3 PCa cells by MSeA in close association with p21cip1 induction and si-p21cip1 attenuated G1 arrest in these cells. In terms of in vivo relevance, we observed an induction of p21cip1 by a chemopreventive level of MSeA intake as early as 72 h after treatment in DU145 xenograft model. Our data support p53-independent induction of p21cip1 by MSeA to mediate the G1 arrest effect in PCa cells. The universality of p21cip1-mediated G1 cell cycle arrest by MSeA in other organ sites and cell types awaits further investigation. Abstract:Because the Selenium (Se) and Vitamin E Cancer Prevention Trial (SELECT) failed to show the efficacy of selenomethionine for prostate cancer prevention, there is a critical need to identify safe and efficacious Se forms for future trials. We have recently shown significant preventive benefit of methylseleninic acid (MSeA) and Se-methylselenocysteine (MSeC) in the transgenic adenocarcinoma mouse prostate (TRAMP) model by oral administration. The present work applied iTRAQ proteomic approach to profile protein changes of the TRAMP prostate and to characterize their modulation by MSeA and MSeC to identify their potential molecular targets. Dorsolateral prostates from wild-type mice at 18 weeks of age and TRAMP mice treated with water (control), MSeA, or MSeC (3 mg Se/kg) from 8 to 18 weeks of age were pooled (9-10 mice per group) and subjected to protein extraction, followed by protein denaturation, reduction, and alkylation. After tryptic digestion, the peptides were labeled with iTRAQ reagents, mixed together, and analyzed by two-dimensional liquid chromatography/tandem mass spectrometry. Of 342 proteins identified with >95% confidence, the expression of 75 proteins was significantly different between TRAMP and wild-type mice. MSeA mainly affected proteins related to prostate functional differentiation, androgen receptor signaling, protein (mis)folding, and endoplasmic reticulum–stress responses, whereas MSeC affected proteins involved in phase II detoxification or cytoprotection, and in stromal cells. Although MSeA and MSeC are presumed precursors of methylselenol and were equally effective against the TRAMP model, their distinct affected protein profiles suggest biological differences in their molecular targets outweigh similarities. Abstract:We have shown that, in contrast to selenomethionine (SeMet) or selenite, methylseleninic acid (MSeA) and Se-methylselenocysteine (MSeC) exert prostate cancer (PCa) inhibitory effect in preclinical models. Here we investigated the prostate proteome signatures of mice treated with each selenium (Se) form for hypothesis generation concerning their potential in vivo molecular targets and cancer risk modification. Nude mice bearing subcutaneous PC-3 xenografts were treated daily with each Se form (3 mg Se/kg) orally for 45 days. Five prostates were pooled from each group. Their proteomes were profiled by LC-MS/MS with iTRAQ labeling. Of the 1,088 proteins identified, 72 were significantly modulated by one or more Se forms. MSeA and MSeC each induced separate sets of tumor suppressor proteins and suppressed different onco-proteins. Proteins induced by selenite and shared with MSeC were related to energy metabolism (e.g., fatty-acid synthase), and those induced by SeMet included vimentin and heat-shock protein-70, favoring cancer growth. While proteome changes induced by MSeA were associated with PCa risk reduction, desirable risk-reducing signatures induced by MSeC were counterbalanced by risk-promoting patterns shared with selenite and SeMet. We propose that the balance of oncogenic vs. suppressor protein patterns in the prostate may impact the direction of PCa risk modification by a given selenium.

Abstract:Programmed death-ligand 1 (PD-L1)-mediated resistance has become a great challenge for tumor treatment. Cisplatin increased tumor PD-L1 expression, promoted chemotherapy resistance. Interferon-γ (IFN-γ)-induced PD-L1 expression might facilitate immunotherapy resistance. Methylseleninic acid (MSeA), a selenium (Se) compound, offered superior cancer chemo-preventive activities and enhanced tumor sensitivity to diverse chemotherapeutic drugs. This study explored the effects of MSeA on the PD-L1-mediated resistance using both in vitro and in vivo models. Results showed that MSeA substantially attenuated cisplatin-induced PD-L1 expression via inhibiting protein kinase B phosphorylation, thereby potentiated cisplatin cytotoxicity in prostate and lung cancer cell models. In lung cancer xenograft model, MSeA significantly suppressed cisplatin-induced PD-L1 expression, consequently enhanced T-cell immunity, ultimately improved the therapeutic efficacy of cisplatin. Moreover, IFN-γ-induced tumor PD-L1 expression was remarkably reduced by MSeA, with correlated reductions in janus kinase 2 and signal transducer and activator of transcription 3 (STAT3) phosphorylation in prostate and lung cancer cell models. Our findings, for the first time, demonstrated that MSeA is a potential agent to overcome PD-L1-mediated chemotherapy and immunotherapy resistance. Such information might have potential clinical implications for prostate and lung cancer treatment.

Abstract:Purpose: To test whether and how selenium enhances the apoptosis potency of selected chemotherapeutic drugs in prostate cancer (PCA) cells.Experimental Design: DU145 and PC3 human androgen-independent PCA cells were exposed to minimal apoptotic doses of selenium and/or the topoisomerase I inhibitor 7-ethyl-10-hydroxycamptothecin (SN38), the topoisomerase II inhibitor etoposide or the microtubule inhibitor paclitaxel/taxol. Apoptosis was measured by ELISA for histone-associated DNA fragments, by flow cytometric analysis of sub-G, fraction, and by immunoblot analysis of cleaved poly(ADP-ribose)polymerase. Pharmacologic inhibitors were used to manipulate caspases and c-Jun-NH2-terminal kinases (JNK).Results: The methylselenol precursor methylseleninic acid (MSeA) increased the apoptosis potency of SN38, etoposide, or paclitaxel by several folds higher than the expected sum of the apoptosis induced by MSeA and each drug alone. The combination treatment did not further enhance JNK1/2 phosphorylation that was induced by each drug in DU145 cells. The JNK inhibitor SP600125 substantially decreased the activation of caspases and apoptosis induced by MSeA combination with SN38 or etoposide and completely blocked these events induced by MSeA/paclitaxel. The caspase-8 inhibitor zIETDfmk completely abolished apoptosis and caspase-9 and caspase-3 cleavage, whereas the caspase-9 inhibitor zLEHDfmk significantly decreased caspase-3 cleavage and apoptosis but had no effect on caspase-8 cleavage. None of these caspase inhibitors abolished JNK1/2 phosphorylation. A JNK-independent suppression of survivin by SN38 and etoposide, but not by paclitaxel, was also observed. In contrast to MSeA, selenite did not show any enhancing effect on the apoptosis induced by these drugs.Conclusions: MSeA enhanced apoptosis induced by cancer therapeutic drugs in androgen-independent PCA cells. In DU145 cells, the enhancing effect was primarily through interactions between MSeA and JNK-dependent targets to amplify the caspase-8-initiated activation cascades. The results suggest a novel use of methyl selenium for improving the chemotherapy of PCA.

Abstract:BACKGROUND:Castration-resistant progression of prostate cancer after androgen deprivation therapy remains a critical challenge in the clinical management of prostate cancer. Resurgent androgen receptor activity is an established driver of castration-resistant progression, and upregulation of androgen receptor expression has been implicated to contribute to the resurgent androgen receptor activity. We reported previously that methylselenocysteine can decrease the expression and activity of androgen receptor. Here we investigated the ability of methylselenocysteine to inhibit castration-resistant progression of prostate cancer. METHODS:The regrowth of LNCaP prostate cancer xenografts after castration was monitored. The levels of prostate-specific antigen in mouse serum were measured by ELISA. Tumor cell proliferation and apoptosis were analyzed via Ki-67 immunohistochemistry and TUNEL assay, respectively. Intratumoral angiogenesis was assessed by immunohistochemistry staining of vascular endothelial growth factor and CD31. RESULTS:We showed that methylselenocysteine delayed castration-resistant regrowth of LNCaP xenograft tumors after androgen deprivation. This was accompanied by decreased serum levels of prostate-specific antigen, inhibition of prostate cancer cell proliferation and tumor angiogenesis, as well as downregulation of androgen receptor and induction of apoptosis in the relapsed tumors. CONCLUSIONS:The present study represents the first to show the preclinical efficacy of methylselenocysteine in delaying castration-resistant progression of prostate cancer. The findings provide a rationale for evaluating the clinical application of combining methylselenocysteine with androgen deprivation therapy for the treatment of advanced prostate cancer.
Abstract 1264: Studying Senescence in Prostate of Selenium Treated Rats Undergoing Carcinogen-Induced, Androgen-Promoted Prostate Carcinogenesis

Abstract:Abstract Preclinical studies from us and others have shown that oral intake of next-generation selenium (Se) forms, especially methylseleninic acid (MSeA) and Se-methylselenocysteine (MSeC) inhibits mouse prostate carcinogenesis. Our recent study has shown that MSeA suppresses pten-deficient mouse high grade prostatic intraepithelial neoplasia progression to adenocarcinoma in association with superactivating p53-p21 mediated cellular senescence. To address the question whether the activation of senescence is induced in prostate carcinogenesis driven by different etiology, we sought to test the in vivo effect of different selenium forms fed to rats undergoing chemically induced, androgen promoted prostate carcinogenesis. Wistar-Unilever (WU) rats (10-12 wks) were sequentially treated with androgen receptor blocker flutamide for 21 days (10 mg/kg/day by gavage), followed by testosterone propionate (TP, 10 mg/kg, s.c) on day 22. Three days after TP, chemical carcinogen methylnitrosourea (MNU, 30 mg/kg/bw) was administered by i.p. injection. One week later, the rats received testosterone-containing silastic implants. Rats were randomized to groups fed the AIN-93M diet or supplemented with 3 ppm of Se as MSeA, MSeC, or selenomethionine (SeMet). Separate groups of rats were fed these diets, but did not receive the carcinogen treatment. After 16 weeks of this dietary treatment, rats were euthanized. Blood, major organs and different prostate lobes were dissected and weighed. The body weight of carcinogen-exposed rats was significantly lower than their non-carcinogen-exposed counterparts. However, total prostate complex weight was higher in carcinogen-treated groups than the non-carcinogen groups, consistent with MNU and extra-testicular androgen promoting prostate epithelial cell proliferation. There were no significant organ-to-body weight ratio differences of various prostate lobes in the different dietary groups, regardless of the carcinogen exposure status. In order to test the involvement of senescence in prostate of rats fed different Se diets, we have undertaken steps to optimize staining method for senescence-associated β-galactosidase (SA-βgal) activity in different lobes of prostate frozen sections. Together, with whole slide imaging using an Aperio Digital Pathology Scanner for quantitation, we are afforded an approach for more objective interrogation of the role of senescence in this carcinogenesis model and chemoprevention by selenium compounds. Methylseleninic acid potentiates apoptosis induced by chemotherapeutic drugs in prostate cancer cells through enhancing caspase activation

Abstract:4933 The chemotherapy of advanced hormone refractory prostate cancer is severely compromised by drug resistance and dose limiting toxicity. Here we tested whether selenium could enhance the apoptosis potency of selected chemotherapeutic drugs in DU145 and PC3 human androgen-independent prostate cancer cells. Exponentially growing cells were exposed to low doses of selenium and/or the topoisomerase I poison 7-ethyl-10-hydroxycamptothecin (SN38), the topoisomerase II inhibitor etoposide or the microtubule drug paclitaxel/taxol. Apoptosis was measured by ELISA for histone-associated DNA fragments, by flow cytometric analysis of sub-G1 fraction, and by immunobloting the caspase cleaved poly(ADP-ribose)polymerase. Our results show that the methylselenol precursor methylseleninic acid (MSeA) increased the apoptosis potency of SN38, etoposide or paclitaxel by several folds higher than the expected sum of the apoptosis induced by MSeA and each drug alone in both cell lines. Mechanistically, the combination treatment did not further enhance the phosphorylation of c-Jun N-terminal kinase (JNK)1/2 that was induced by each drug in DU145 cells. The JNK inhibitor SP600125 substantially decreased the activation of caspases and apoptosis induced by MSeA combination with the drugs. The caspase-8 inhibitor zIETDfmk completely abolished apoptosis and caspase-9 and caspase-3 cleavage, whereas the caspase-9 inhibitor zLEHDfmk significantly decreased caspase-3 cleavage and apoptosis but had no effect on caspase-8 cleavage. None of these caspase inhibitors abolished JNK1/2 phosphorylation. In contrast to MSeA, selenite did not show any enhancing effect on the apoptosis induced by these drugs. In conclusion, MSeA enhanced apoptosis induced by cancer therapeutic drugs in DU145 cells primarily through interactions between MSeA and JNK-dependent targets to amplify what appeared to be a caspase-8-initiated activation cascade. The results suggest, to our knowledge, for the first time a novel use of methyl selenium for improving the chemotherapy of prostate cancer. Abstract:Proceedings: AACR 103rd Annual Meeting 2012‐‐ Mar 31‐Apr 4, 2012; Chicago, IL Mutations of the p53 tumor suppressor gene constitute the most frequent genetic alteration known in human cancers. The vast majority (∼80%) of the mutations is missense mutations, and the DNA-binding domain is the most frequently mutable region of p53. p53 mutation leads to checkpoint defects, genomic instability, and propagation of cells with genetic damage. Developing therapeutic agents that could directly bind to the mutant p53 and reconstitute p53 function has been intensively pursued and proven to be feasible. However, the challenge remains to develop a drug that specifically reactivates mutant p53 without activating the pro-apoptotic function of wild-type p53 in normal tissues. In the present study, we show that a small-molecule drug, methylseleninic acid (CH3SeO2H, MSA), can selectively reactivate mutant p53 to induce apoptosis in cancer cells. MSA can be used at a concentration at least one order of magnitude lower than that of other mutant-p53-rescue agents to achieve the same p53-reactivating and growth-inhibitory effects. MSA reduces redox-active free thiols in mutant p53 without the requirement of other proteins, potentially through forming adducts with thiols in mutant p53. The modification restores wild-type conformation to mutant p53, leading to increased DNA binding and transactivation as well as expression of growth-suppressive and pro-apoptotic target genes in both cultured cancer cells and an orthotopic tumor model. Although MSA also redox modifies recombinant wild-type p53, the modification does not lead to the induction of growth-suppressive or pro-apoptotic target genes in normal fibroblasts. Instead, the expression of p53-regulated DNA-repair genes is significantly induced in normal fibroblasts, suggesting that MSA may selectively modify p53 for DNA repair or apoptosis depending on the level of endogenous or exogenous DNA damage. Our findings, therefore, demonstrate the effectiveness and selectivity of MSA as a novel potent mutant-p53-rescue drug. Citation Format: {Authors}. {Abstract title} [abstract].
