Abstract:There are three hepatitis B virus (HBV) envelope glycoproteins collectively known as hepatitis B virus surface antigen (HBsAg), including the large (LHBs), middle (MHBs), and small (SHBs) surface proteins. Among them, LHBs has been retained in a large proportion of liver tissues in HBV-associated hepatocellular carcinoma (HCC) patients and shown oncogenic function in transgenic mice. However, the concrete functions of LHBs in carcinogenesis of HBV-associated HCC remain elusive. In this study, we investigated the molecular and functional roles of LHBs in HBV-associated hepatocarcinogenesis. Tumor xenograft experimental data in nude mice indicated that LHBS increased tumor formation of hepatoma cells. Moreover, LHBs expression, but neither MHBs nor SHBs expression could promote colony formation and cellular proliferation of hepatoma and hepatic cells in vitro. These effects of LHBs on hepatoma cells were due to Src/PI3K/Akt signal instigation mediated by PKC /Raf1 activation. Furthermore, stable LHBs expression could promote cellular proliferation, elevate colony formation, induce G1-S cell cycle progression, and confer apoptosis resistance via Src tyrosine kinase activation in hepatoma cells. Specific Src inhibitor Saracatinib administration could reverse cellular proliferation instigation and tumor formation promotion conferred by LHBs expression in vitro and in vivo. Finally, the positive correlations between increased staining of pSrc(Y416), pAkt(S473), and proliferative marker Ki67 with LHBs expression were observed in hepatoma tissues from HCC patients. Abstract:A high incidence of tumor recurrence and metastasis has been reported in hepatocellular carcinoma (HCC) patients with chronic hepatitis B virus (HBV) infection. Although the pathological relevance and significance of hepatitis B virus X protein (HBx) in HBV-associated hepatocarcinogenesis attracted much attention in recent years, the role and molecular mechanism for HBx in hepatoma invasion and metastasis remains poorly understood. In the present study, we found that HBx expression could induce epithelial-mesenchymal transition in hepatoma and hepatic cells. This effect was shown due to stabilized Snail protein through activating the phosphatidylinositol 3-kinase/protein kinase B/glycogen synthase kinase-3β (PI3K/AKT/GSK-3β) signal pathway by HBx expression. Functional studies revealed that HBx expression could enhance hepatoma cell migration and invasion in vitro. Moreover, stable HBx expression could also facilitate intrahepatic and distant lung metastasis of HCC in a nude mice tumor metastasis model in vivo. The correlation between increased PI3K/AKT/GSK-3β signaling with elevated Snail protein level was also observed in HCC tumor tissues with intrahepatic metastasis or chronic HBV infection. These results revealed a novel function of HBx in promoting epithelial-mesenchymal transition through Snail protein stabilization by activating PI3K/AKT/GSK-3β signaling, thus facilitating tumor invasion and metastasis during HCC progression. This could provide a putative molecular mechanism for tumor recurrence and metastasis in HBV-associated HCC patients.

Abstract:e15115 Background: Hepatitis B virus (HBV) X protein (HBx) and pre-S2 deletion mutant large surface antigens have been recognized as oncoproteins that induce hepatocellular carcinoma (HCC). The interaction of these two oncoproteins in the hepatocytes and its significance in tumorigenesis remains to be elucidated. Methods: In this study, a newly developed monoclonal antibody designed specifically against HBx was used for immunohistochemical and double-labeled immunofluorescence studies and the expression of HBx and surface antigens were evaluated. In vitro studies and transgenic mice models were used to evaluate the interaction and oncogenic effects of these two viral oncoproteins. Results: We observed the co-expression of HBx with surface antigens in ground glass hepatocytes in 5 of 20 hepatitis B surface antigen positive livers. In vitro, hepatocytes co-expressing HBx and pre-S2 mutant showed an enhanced expression of vascular endothelial growth factor-A (VEGF-A), phosphorylated Akt1/2/3 (p-Akt1/2/3), phosphorylated extracellular signal-regulated kinase 1/2 (p-ERK1/2) and phosphorylated mammalian target of rapamycin (p-mTOR) signals. Transgenic mice harboring the double HBx and pre-S2 mutant construct plasmids developed HCCs at an average of 15.1 months, earlier than that of HBx (16.9 months) or pre-S2 mutant (24.5 months) alone. Interestingly, the oncogenic signals of VEGF-A, p-Akt1/2/3, p-ERK1/2, and p-mTOR were sequentially and differentially activated at different stages in the progression of tumorigenesis. Phosphorylated mTOR was consistently activated in transgenic and human HCCs. Conclusions: We conclude that ground glass hepatocytes co-expressing HBx and surface antigens may exhibit enhanced oncogenic effects and tumorigenesis in chronic HBV infections. The mTOR signal cascade may represent the key regulator in HBV tumorigenesis and is implicated for the design of HCC therapy.

Abstract:Hepatitis B x antigen (HBxAg) is a trans-activating protein that contributes to liver cancer, in part, by altering the expression of cellular genes. However, few natural effectors of HBxAg have been identified. Hence, HBxAg positive and negative HepG2 cells were prepared and analyzed by PCR select cDNA subtraction. The results identified elevated vascular endothelial growth factor receptor-3 short form splice variant (VEGFR-3(s)) expression in HBxAg positive compared to negative cells. Normally, VEGFR-3 activates Akt signaling in lymphatic endothelial cells, resulting in lymphangiogenesis. In contrast, the results here show that the expression of VEGFR-3(s) is up-regulated in > 75% of HBxAg positive hepatocellular carcinoma (HCC) nodules. VEGFR-3(s) up-regulation correlates with the expression of HBxAg, is associated with decreased survival in tumor bearing patients, and when over-expressed in HepG2 cells, strongly stimulated cell growth in culture, in soft agar, and accelerated tumor formation in a ligand independent manner. VEGFR-3(s) siRNA partially blocked the ability of HBxAg to promote hepatocellular growth. In conclusion, HBxAg may short circuit VEGFR-3(s) signaling in liver cancer. Blocking VEGFR-3(s) signaling may be effective in preventing tumor development and/or prolonging survival in tumor bearing patients.

Abstract:Hepatitis B x antigen (HBxAg) is a trans-activating protein that may be involved in hepatocarcinogenesis, although few natural effectors of HBxAg that participate in this process have been identified. To identify additional effectors, whole cell RNA isolated from HBxAg-positive and HBxAg-negative HepG2 cells were compared by polymerase chain reaction select cDNA subtraction, and one clone, upregulated gene, clone 11 (URG11), was chosen for further characterization. Elevated levels of URG11 mRNA and protein were observed in HBxAg-positive compared to HBxAg-negative HepG2 cells. Costaining was observed in infected liver (P<.01). URG11 stimulated cell growth in culture (P<.01), anchorage-independent growth in soft agar (P<.001), and accelerated tumor formation (P<.01), and yielded larger tumors (P<.02) in SCID mice injected subcutaneously with HepG2 cells. These data suggest that URG11 is a natural effector of HBxAg that may promote the development of hepatocellular carcinoma.

Abstract:Hepatitis B virus (HBV) is an aetiological factor for liver cirrhosis and hepatocellular carcinoma (HCC). Despite current antiviral therapies that successfully reduce the viral load in patients with chronic hepatitis B, persistent hepatitis B surface antigen (HBsAg) remains a risk factor for HCC. To explore whether intrahepatic viral antigens contribute directly to hepatocarcinogenesis, we monitored the mitotic progression of HBV-positive cells. Cytokinesis failure was increased in HBV-positive HepG2.2.15 and 1.3ES2 cells, as well as in HuH-7 cells transfected with a wild-type or X-deficient HBV construct, but not in cells transfected with an HBsAg-deficient construct. We show that expression of viral large surface antigen (LHBS) was sufficient to induce cytokinesis failure of immortalized hepatocytes. Premitotic defects with DNA damage and G2 /M checkpoint attenuation preceded cytokinesis in LHBS-positive cells, and ultimately resulted in hyperploidy. Inhibition of polo-like kinase-1 (Plk1) not only restored the G2 /M checkpoint in these cells, but also suppressed LHBS-mediated in vivo tumourigenesis. Finally, a positive correlation between intrahepatic LHBS expression and hepatocyte hyperploidy was detected in >70% of patients with chronic hepatitis B. We conclude that HBV LHBS provokes hyperploidy by inducing DNA damage and upregulation of Plk1; the former results in atypical chromatin structures, and the latter attenuates the function of the G2 /M DNA damage checkpoint. Our data uncover a mechanism by which genomic integrity of hepatocytes is disrupted by viral LHBS. These findings highlight the role of intrahepatic surface antigen as an oncogenic risk factor in the development of HCC. Copyright © 2018 Pathological Society of Great Britain and Ireland. Abstract:Hepatitis B virus X protein (HBx) plays a crucial role in the development of hepatocellular carcinoma. Here, we sought to identify the mechanisms by which HBx mediates liver cell proliferation. We found that HBx upregulated the levels of cyclooxygenase-2 (COX-2), 5-lipoxygenase (5-LOX) and phosphorylated extracellular signal-regulated protein kinases 1/2 (p-ERK1/2) in liver cells. HBx-induced p-ERK1/2 was abolished by inhibition of Gi/o proteins, COX or LOX. In addition, HBx increased the amounts of prostaglandin E2 (PGE2) and leukotriene B4 (LTB4) released from cell lines derived from hepatocytes. Moreover, these released arachidonic acid metabolites were able to activate ERK1/2. Interestingly, activated ERK1/2 could upregulate the expression of COX-2 and 5-LOX in a positive feedback manner. In conclusion, HBx enhances and maintains liver cell proliferation via a positive feedback loop involving COX-2, 5-LOX, released arachidonic acid metabolites, Gi/o proteins and p-ERK1/2.

Abstract:We aimed to elucidate the mechanism by which hepatitis B virus X (HBx) mutations increase the occurrence of hepatocellular carcinoma (HCC) and identify novel putative therapeutic targets. Wild-type HBx (WT-HBx) and four HBx mutants (M1, A1762T/G1764A; M2, T1674G+T1753C+A1762T/G1764A; M3, C1653T+T1674G+A1762T/G1764A; and Ct-HBx, carboxylic acid-terminal truncated HBx) were delivered into Sleeping Beauty ( SB ) mouse models. The HCC incidence was higher in the M3-HBx- and Ct-HBx-injected SB mice. M3-HBx had a stronger capacity of upregulating inflammatory cytokines than other HBx variants. Ectopic expression of M3-HBx and Ct-HBx significantly increased proliferation and S phase proportion of HepG2 and HeLa cells, compared to WT-HBx. Plasminogen activator inhibitor-1 (PAI1) and cell division cycle 20 (CDC20) were identified as novel effectors by cDNA microarray analysis. M3-HBx and Ct-HBx significantly upregulated the expression of PAI1 and CDC20 in HepG2 and HeLa cells as well as the livers of SB mice. Silencing PAI1 attenuated the effects of M3-HBx and Ct-HBx on the growth of HepG2 and HeLa cells. PAI1, an important player bridging the HBx mutants and HCC, should be a promising candidate as a prognostic biomarker and therapeutic target in HBV-related HCC.

Abstract:Hepatocellular carcinoma (HCC) is one of the most common and deadly malignant neoplasms worldwide. Chronic hepatitis B virus (HBV) infection is closely associated with the occurrence of HCC. The HBV genome encodes a ubiquitous transactivator, termed the HBV X protein (HBx), that is essential for HBV replication in vivo. HBx is involved in multiple steps of carcinoma development. Even in the preneoplastic stage, HBx acts as a tumor promoter. HBx participates in several mechanisms that have been linked to cell proliferation, including gene transcription, cell cycle regulation, and several signaling pathways. Moreover, HBx mutants, especially those with mutations in the COOH-terminal end, have been implicated in hepatocarcinogenesis. Therefore, therapeutic strategies targeting HBx could be effective at multiple stages of HCC development, even as early as the preneoplastic stage.
Small hepatitis B virus surface antigen promotes malignant progression of hepatocellular carcinoma via endoplasmic reticulum stress-induced FGF19/JAK2/STAT3 signaling

Abstract:Chronic hepatitis B virus (HBV) infection is one of the major global health problems. Although the small protein of hepatitis B virus surface antigen (HBsAg), SHBs, is the most abundant HBV viral protein, its pathogenic role and molecular mechanism in malignant progression of HBV-related hepatocellular carcinoma (HCC) remain largely unknown. Here we reported that SHBs expression induced epithelial-mesenchymal transition (EMT) process in HCC cells and significantly increased their migratory and invasive ability as well as metastatic potential. Mechanistically, SHBs expression in HCC cells induced endoplasmic reticulum (ER) stress that activated the activating transcription factor 4 (ATF4) to increase the expression and secretion of fibroblast growth factor 19 (FGF19). The autocrine released FGF19 in turn activated JAK2/STAT3 signaling for induction of EMT process in HCC. Notably, SHBs was positively correlated with the expression of mesenchymal markers, the phosphorylation status of JAK2 and STAT3 as well as FGF19 levels in human HCC samples. HCC patients with SHBs positive had a more advanced clinical stage and worse prognosis. These results suggest an important role of SHBs in the metastasis and progression of HCC and may highlight a potential target for preventive and therapeutic intervention of HBV-related HCC and its malignant progression.
