Abstract:Objective: To set up a three-parameter method for cell cycle analysis by two-laser flow cytometer, which can detect two types of cyclin plus DNA content in one measurement, and that analyze unscheduled expression of cyclins. Methods: Three-color fluorescence was used for analysis of two types of cyclins and DNA content simultaneously in individual cells by two-laser flow cytometry. MOLT-4 cells were used to study the expression of major cyclins in mammalian cells. A Triton-X100 permeabilization procedure was optimized for detection of two types of cyclins. One cyclin was stained directly with a FITCconjugated monoclonal antibody (mAb), and the other, indirectly with RPE-Cy5-conjugated secondary antibody, while DNA was stained with the fluorochrome DAPI. mAMSA and mimosine treated MOLT-4cells were used to test this three-parameter method. Results: Permeabilization with 0.5% Triton-X100 in PBS containing 1% BSA for 5 min on ice provided optimal conditions for the simultaneous labelling of two cyclins plus DNA in single cells. It was found that the emission spectrum of the three dyes (DAPI, FITC and RPE-Cy5) could be measured with no compensation. Based on cyclinA/cyclinE/DNA flow cytometric analysis, asynchronously growing MOLT-4 cells could be dividedinto 6 compartments (G1o, G1e, G1l,S, G2, and M) simultaneously, allowing for analysis of cell cycle phase specific perturbations without the necessity of cell synchronization. Unscheduled cyclin B1 expression was observed in G1 cells treated with mimosine and cyclin E in G2 cells treated with mAMSA. We found that unscheduled cyclin expression paralleled expected cyclin expression. Conclusion: Thus, three-color FCM analysis of cells may not only be applied to measure unscheduled vs. expected cyclin expression but may also be used to estimate the fraction of cycling cells in up to 6 cell populations.

Abstract:AbstractMetabolic activities are often accompanied by cell-cycle progression, yet known connections between these two processes remain limited. Here, we identified the isocitrate dehydrogenase 3β (IDH3β) as a novel substrate of anaphase-promoting complex/cyclosome (APC/C)-CDH1 and an important regulator of the cell cycle. In esophageal squamous cell carcinoma (ESCC), IDH3β was posttranslationally upregulated in late G1 phase, and overexpression of IDH3β accelerated G1–S transition, contributing to the promotion of cell proliferation in vitro and in vivo. α-Ketoglutarate (α-KG), a crucial metabolite in tricarboxylic acid (TCA) cycle, was dependent on IDH3β level and partially accounted for IDH3β-mediated cell growth. IDH3β expression increased PFKFB3 protein levels and enhanced glucose uptake, and high expression of IDH3β correlated with poor survival in patients with ESCC, suggesting a potential application of IDH3β in prognosis. Overall, our results highlight a new molecular connection between cell-cycle regulation and the TCA cycle in ESCC.Significance:These findings show that IDH3β is an APC/C-CDH1 substrate and is expressed in a cell-cycle–dependent manner, highlighting novel molecular cross-talk between the TCA cycle and cell cycle in cancer cells.

Abstract:Cellular processes such as cell cycle progression, mitosis, apoptosis, and cell migration are characterized by well-defined events that are modulated as a function of time. Measuring these events in the context of time and its perturbation by small molecule compounds and RNAi can provide mechanistic information about cellular pathways being affected. We have used impedance-based time-dependent cell response profiling (TCRP) to measure and characterize cellular responses to antimitotic compounds or siRNAs. Our findings indicate that small molecule perturbation of mitosis leads to unique TCRP. We have further used this unique TCRP signature to screen 119 595 compound library and identified novel antimitotic compounds based on clustering analysis of the TCRPs. Importantly, 113 of the 117 hit compounds in the TCRP antimitotic cluster were confirmed as antimitotic based on independent assays, thus establishing the robust predictive nature of this profiling approach. In addition, potent and novel agents that induce mitotic arrest either by directly interfering with tubulin polymerization or by other mechanisms were identified. The TCRP approach allows for a practical and unbiased phenotypic profiling and screening tool for small molecule and RNAi perturbation of specific cellular pathways and time resolution of the TCRP approach can serve as a complement for other existing multidimensional profiling approaches.
