Abstract:The growth and development of plants require 14 essential mineral nutrient elements, such as nitrogen, phosphorus, potassium, sulfur, iron, etc. Nevertheless, the bioavailability of these elements varies greatly in the different types of soils. In order to adapt to nutrient-deficient environment, plants must sense the changes of external and internal mineral nutrient concentrations, and generate physiological and morphological responses via a series of signal transduction events, to facilitate the nutrient uptake and utilization. Ethylene is an essential gaseous plant hormone, and plays pivotal roles in many aspects of the plant life cycle. In recent years,accumulating studies have shown that ethylene also plays an important part in regulating plant responses to various nutrient deficiency stresses. Here, we comprehensively summarized the molecular mechanisms of ethylene regulating different nutritional stress responses directly or synergistically with other plant hormones/chemical signaling molecules, and gave an outlook for the future research.

Abstract:Background Iron (Fe) deficiency in crops is a worldwide agricultural problem. Plants have evolved several strategies to enhance Fe acquisition, but increasing evidence has shown that the intrinsic plant-based strategies alone are insufficient to avoid Fe deficiency in Fe-limited soils. Soil micro-organisms also play a critical role in plant Fe acquisition; however, the mechanisms behind their promotion of Fe acquisition remain largely unknown.Scope This review focuses on the possible mechanisms underlying the promotion of plant Fe acquisition by soil micro-organisms.Conclusions Fe-deficiency-induced root exudates alter the microbial community in the rhizosphere by modifying the physicochemical properties of soil, and/or by their antimicrobial and/or growth-promoting effects. The altered microbial community may in turn benefit plant Fe acquisition via production of siderophores and protons, both of which improve Fe bioavailability in soil, and via hormone generation that triggers the enhancement of Fe uptake capacity in plants. In addition, symbiotic interactions between micro-organisms and host plants could also enhance plant Fe acquisition, possibly including: rhizobium nodulation enhancing plant Fe uptake capacity and mycorrhizal fungal infection enhancing root length and the nutrient acquisition area of the root system, as well as increasing the production of Fe3+ chelators and protons.

Abstract:Plants take up metals, including the essential micronutrients [iron (Fe), copper (Cu), zinc (Zn), and manganese (Mn)] and the toxic heavy metal cadmium (Cd), from soil and accumulate these metals in their edible parts, which are direct and indirect intake sources for humans. Multiple transporters belonging to different families are required to transport a metal from the soil to different organs and tissues, but only a few of them have been fully functionally characterized. The transport systems (the transporters required for uptake, translocation, distribution, redistribution, and their regulation) differ with metals and plant species, depending on the physiological roles, requirements of each metal, and anatomies of different organs and tissues. To maintain metal homeostasis in response to spatiotemporal fluctuations of metals in soil, plants have developed sophisticated and tightly regulated mechanisms through the regulation of transporters at the transcriptional and/or posttranscriptional levels. The manipulation of some transporters has succeeded in generating crops rich in essential metals but low in Cd accumulation. A better understanding of metal transport systems will contribute to better and safer crop production. Research Progresses on Functional Roles and Regulation Mechanisms of Nutrient Element Transporters in Plants

Abstract:Essential nutrient elements for plants, including nitrogen (N), phosphorus (P), potassium (K), and zinc (Zn), are involved in regulation of many physiological and biochemical processes in plants, which are critical factors directly determining yield and quality of crops. However, excessive subcellular accumulation of these essential nutrient elements will be toxic for plant cells or the whole plant. Thus, uptake and transport processes of these nutrients must be tightly regulated at the cellular level. Focused on Arabidopsis thaliana and Oryza sativa, this review briefly summarized the types, physical and chemical properties of nutrient element transporters, and elaborated their biological functions and action mechanisms in higher plants, including N transporter families (NPF, NRT2, CLC, SLAC/SLAH, and AMT), P transporter families (PHT1, PHT2, PHT3, PHT4, and PHT5), K transporter families (KT/HAK/KUP, HKT, CHX, and KEA), and Zn transporter families (ZIP, CDF, and P1B-type ATPase). Endocytosis from the plasma membrane (PM) is an important route for uptake of extracellular materials and regulation of PM-resident receptors or transporters, so we introduced potential roles of transporter endocytosis in uptake and transport of nutrient elements in plant cells. In the end, we made prospects of future studies on functional roles and regulation mechanisms of transporters, providing new ideas and strategies for efficient utilization of nutrients and genetic improvement of quality in crops.

Abstract:Plant mineral nutrition is critical for agricultural productivity and for human nutrition; however, the availability of mineral elements is spatially and temporally heterogeneous in many ecosystems and agricultural landscapes. Nutrient imbalances trigger intricate signalling networks that modulate plant acclimation responses. One signalling agent of particular importance in such networks is phytomelatonin, a pleiotropic molecule with multiple functions. Evidence indicates that deficiencies or excesses of nutrients generally increase phytomelatonin levels in certain tissues, and it is increasingly thought to participate in the regulation of plant mineral nutrition. Alterations in endogenous phytomelatonin levels can protect plants from oxidative stress, influence root architecture, and influence nutrient uptake and efficiency of use through transcriptional and post-transcriptional regulation; such changes optimize mineral nutrient acquisition and ion homeostasis inside plant cells and thereby help to promote growth. This review summarizes current knowledge on the regulation of plant mineral nutrition by melatonin and highlights how endogenous phytomelatonin alters plant responses to specific mineral elements. In addition, we comprehensively discuss how melatonin influences uptake and transport under conditions of nutrient shortage.

Abstract:In nature, land plants as sessile organisms are faced with multiple nutrient stresses that often occur simultaneously in soil. Nitrogen (N), phosphorus (P), sulfur (S), zinc (Zn), and iron (Fe) are five of the essential nutrients that affect plant growth and health. Although these minerals are relatively inaccessible to plants due to their low solubility and relative immobilization, plants have adopted coping mechanisms for survival under multiple nutrient stress conditions. The double interactions between N, Pi, S, Zn, and Fe have long been recognized in plants at the physiological level. However, the molecular mechanisms and signaling pathways underlying these cross-talks in plants remain poorly understood. This review preliminarily examined recent progress and current knowledge of the biochemical and physiological interactions between macro- and micro-mineral nutrients in plants and aimed to focus on the cross-talks between N, Pi, S, Zn, and Fe uptake and homeostasis in plants. More importantly, we further reviewed current studies on the molecular mechanisms underlying the cross-talks between N, Pi, S, Zn, and Fe homeostasis to better understand how these nutrient interactions affect the mineral uptake and signaling in plants. This review serves as a basis for further studies on multiple nutrient stress signaling in plants. Overall, the development of an integrative study of multiple nutrient signaling cross-talks in plants will be of important biological significance and crucial to sustainable agriculture.

Abstract:Phosphate is one of the indispensable elements for plant growth and development. The uptake of phosphate from soils and translocation between organs and subcellular compartments require a series of phos-phate transporters with phosphate transport activity. In recent years, with the rapid development of molecular biology and functional genomics technology, researchers have made great progress in clarifying the function of phosphate transporters and its molecular regulation mechanism. In this paper, focused on Arabidopsis and rice, we reviewed the recent progress on the discovery of phosphate transporters, their functions and molecular regu-lation mechanisms. It would provide reference for improving phosphorus absorption and utilization efficiency in crop breeding.
Latent Function of Microorganisms on Plant Iron Acquisition

Abstract:Iron deficiency in plants is a worldwide agricultural problem. It has been proved that soil microorganisms have a beneficial effect on the iron acquisition of some plants, but the underlying mechanism is still unclear. Based on last ten years' research results, possible mechanism on how the microorganisms favor plant iron acquisition was discussed in this review. Iron-deficiency-induced root exudates, such as phenolic and flavin compounds, acting as inhibitors and carbon sources, affected the architecture of microorganisms in the rhizosphere and induced a formation of a specified microorganism community. Those specified microorganisms in turn benefited the plant iron acquisition through the secretion of siderophore which could improve the bioavailability of iron in soil. In addition, the symbiosis between some microorganisms and plants could also improve plant iron nutrient status. The underlying processes may involve: the formation of rhizobium nodulation enhanced plant iron-deficient response, and mycorrhizal fungi infected root which improved the iron nutrient status through enlarging absorption area and excreting the materials which chelated iron of plant.

Abstract:Intercropping facilitates the efficient utilization of land, light, water and nutrients. It is, therefore, important to increase the biodiversity of farmland and to develop sustainable ecological agriculture in both theory and practice. Intercropping helps improve the mobilization and uptake of soil iron (Fe) and zinc (Zn) and corresponding nutritional status in the plants, thus achieving grain micronutrient biofortification. In this review, phenomena of the improvement of Fe and Zn nutrition in dicotyledonous plants as affected by intercropping with gramineous plants (e.g. maize/peanut intercropping) were summarized. Moreover, the possible mechanisms in relation to interspecific rhizosphere molecular and physiological processes, as well as the changes in interspecific root morphology and distribution and microorganisms in the rhizosphere were elucidated. The accumulation, transfer and distribution of Fe and Zn in the plants in intercropping systems were also reviewed. The possible affecting factors on nutrients of Fe and Zn were analyzed. Based on the present advances in the mobilization and acquisition of soil Fe and Zn, and their accumulation and distribution in plants as well as the related management and environment influence factors, some new research questions were pointed out. Quantitative analysis, dynamic and systemic researches and field studies on Fe and Zn transfer from soil to plant in intercropping systems should be strengthened in the future.

Abstract:Nitrogen, phosphorous and potassium are essential nutrients for plant growth and development. However, their contents in soils are limited so that crop production needs to invest a lot for fertilizer supply. To explore the genetic potentialities of crops (or plants) for their nutrient utilization efficiency has been an important research task for many years. In fact, a number of evidences have revealed that plants, during their evolution, have developed many morphological, physiological, biochemical and molecular adaptation mechanisms for acquiring nitrate, phosphate and potassium under stress conditions. Recent discoveries of many transporters and channels for nitrate, phosphate and potassium uptake have opened up opportunities to study the molecular regulatory mechanisms for acquisition of these nutrients. This review aims to briefly discuss the genes and gene families for these transporters and channels. In addition, the functions and regulation of some important transporters and channels are particularly emphasized.
MicroRNAs modulating nutrient homeostasis: a sustainable approach for developing biofortified crops

Abstract:During their lifespan, sessile plants have to cope with bioavailability of the suboptimal nutrient concentration and have to constantly sense/evolve the connecting web of signal cascades for efficient nutrient uptake, storage, and translocation for proper growth and metabolism. However, environmental fluctuations and escalating anthropogenic activities are making it a formidable challenge for plants. This is adding to (micro)nutrient-deficient crops and nutritional insecurity. Biofortification is emerging as a sustainable and efficacious approach which can be utilized to combat the micronutrient malnutrition. A biofortified crop has an enriched level of desired nutrients developed using conventional breeding, agronomic practices, or advanced biotechnological tools. Nutrient homeostasis gets hampered under nutrient stress, which involves disturbance in short-distance and long-distance cell-cell/cell-organ communications involving multiple cellular and molecular components. Advanced sequencing platforms coupled with bioinformatics pipelines and databases have suggested the potential roles of tiny signaling molecules and post-transcriptional regulators, the microRNAs (miRNAs) in key plant phenomena including nutrient homeostasis. miRNAs are seen as emerging targets for biotechnology-based biofortification programs. Thus, understanding the mechanistic insights and regulatory role of miRNAs could open new windows for exploring them in developing nutrient-efficient biofortified crops. This review discusses significance and roles of miRNAs in plant nutrition and nutrient homeostasis and how they play key roles in plant responses to nutrient imbalances/deficiencies/toxicities covering major nutrients-nitrogen (N), phosphorus (P), sulfur (S), magnesium (Mg), iron (Fe), and zinc (Zn). A perspective view has been given on developing miRNA-engineered biofortified crops with recent success stories. Current challenges and future strategies have also been discussed.

Abstract:The essentiality of 14 mineral elements so far have been reported in plant nutrition. Eight of these elements were known as micronutrients due to their lower concentrations in plants (usually ≤100 mg/kg/dw). However, it is still challenging to mention an exact number of plant micronutrients since some elements have not been strictly proposed yet either as essential or beneficial. Micronutrients participate in very diverse metabolic processes, including from the primary and secondary metabolism to the cell defense, and from the signal transduction to the gene regulation, energy metabolism, and hormone perception. Thus, the attempt to understand the molecular mechanism(s) behind their transport has great importance in terms of basic and applied plant sciences. Moreover, their deficiency or toxicity also caused serious disease symptoms in plants, even plant destruction if not treated, and many people around the world suffer from the plant-based dietary deficiencies or metal toxicities. In this sense, shedding some light on this issue, the 13 mineral elements (Fe, B, Cu, Mn, Mo, Si, Zn, Ni, Cl, Se, Na, Al, and Co), required by plants at trace amounts, has been reviewed with the primary focus on the transport proteins (transporters/channels) in plant roots. So, providing the compiled but extensive information about the structural and functional roles of micronutrient transport genes/proteins in plant roots.

Nitrogen Uptake in Plants: The Plasma Membrane Root Transport Systems from a Physiological and Proteomic Perspective

Abstract:Nitrogen nutrition in plants is a key determinant in crop productivity. The availability of nitrogen nutrients in the soil, both inorganic (nitrate and ammonium) and organic (urea and free amino acids), highly differs and influences plant physiology, growth, metabolism, and root morphology. Deciphering this multifaceted scenario is mandatory to improve the agricultural sustainability. In root cells, specific proteins located at the plasma membrane play key roles in the transport and sensing of nitrogen forms. This review outlines the current knowledge regarding the biochemical and physiological aspects behind the uptake of the individual nitrogen forms, their reciprocal interactions, the influences on root system architecture, and the relations with other proteins sustaining fundamental plasma membrane functionalities, such as aquaporins and H+-ATPase. This topic is explored starting from the information achieved in the model plant Arabidopsis and moving to crops in agricultural soils. Moreover, the main contributions provided by proteomics are described in order to highlight the goals and pitfalls of this approach and to get new hints for future studies.

Abstract:Currently, eight trace elements have been considered to be essential for higher plants, Fe, Zn, Mn, Cu, Ni, B, Mo, and Cl, which are termed micronutrients. Genotypes of plants vary widely in their adaptation to micronutrient deficiency stresses in soils. The adaptation of plants to micronutrient deficiency stresses appears to be associated, in most cases, with more efficiency in microniutrient acquisition from soils. The efficiency in acquisition of micronutrients from soils by plants is genetically controlled. This review briefly summarizes the current knowledge of physiological mechanisms of major micronutrient uptake by higher plants, genotypic differences in micronutrient acquisition from soils, and what is known of the genetic mechanisms of micronutrient efficiency in plants.

Abstract:The rhizosphere contains both essential nutrients and potentially harmful substances for plant growth. Plants, as sessile organisms, must efficiently absorb the necessary nutrients while actively avoiding the uptake of toxic compounds. Metalloids, which are elements that exhibit properties of both metals and nonmetals, can have different effects on plant growth, ranging from being essential and beneficial to being toxic. The toxicity of metalloids in plants arises due to either the dosage of exposure or the specific elemental type. To utilize or detoxify these elements, plants have developed various transporters that regulate their uptake and distribution in plants. Arguably, genomic sequence analysis suggests the presence of such transporter families throughout the plant kingdom, from chlorophytes to higher plants. These transporters form defined families with related transport preferences. The isoforms within these families have evolved with specialized functions regulated by defined selectivity. Hence, understanding transporters' chemistry to atomic detail is important to achieve desired genetic modifications for crop improvement. Here, we outline various adaptations in plant transport systems to deal with metalloids, including their uptake, distribution, detoxification, and homeostasis in plant tissues. Structural parallels are drawn to other nutrient transporter systems to support emerging themes of functional diversity of active sites of transporters, elucidating adaptations of plants to utilize and extrude metalloid concentrations. Considering the observed physiological importance of metalloids, this review intends to highlight the shared and disparate features in metalloid transport systems and their corresponding nutrient transporters.

Abstract:Fe is essential for plant growth.However,plants show severe iron deficiency due to low availability of Fe in soils,which affects the yield and quality of crops and ultimately affects human nutrition.Therefore,it is the hot research area to improve human nutritional status through biofortification.This review covers the research progress on the molecular mechanism of Fe uptake and translocation in plants and deposition in seeds in the past five years,and includes new discovery of combination of reduction and chelation strategies of iron uptake in Rice and the importance of YSL family in the three biological processes in plants.Furthermore,the technical ways of iron nutrition improvement and iron enrichment in seeds by genetic engineering and agronomic measures based on the studies on molecular mechanism of iron uptake and translocation in plants are discussed.

Abstract:Small RNAs including micro RNAs (miRNA) play an indispensable role in cell signaling mechanisms. Generally, miRNAs that are 20-24 nucleotides long bind to specific complementary transcripts, attenuating gene expression at the post-transcriptional level or via translational inhibition. In plants, miRNAs have emerged as the principal regulator of various stress responses, including low nutrient availability. It has been reported that miRNAs are vital for maintaining nutrient homeostasis in plants by regulating the expression of transporters that are involved in nutrient uptake and mobilization. The present review highlights the role of various miRNAs in several macro- or micronutrient deficiencies in plants. Understanding the regulation of different transporters by miRNAs will aid in elucidating the underlying molecular signal transduction mechanisms during nutritional stress. Recent findings regarding nutrient related-miRNAs and their gene regulation machinery may delineate a novel platform for improving the nutritional status of cereal grains or crop biofortification programs in the future.
