Abstract:Elastomeric, robust, and biocompatible hydrogels are rare, while the need for these types of biomaterials in biomedical-related uses remains high. Here, a new family of genetically engineered silk-elastin copolymers (SELPs) with encoded enzymatic crosslinking sites is developed for a new generation of stimuli-responsive yet robust hydrogels. Input into the designs is guided by simulation, and realized via genetic engineering strategies. The avoidance of gamma irradiation or chemical crosslinking during gel fabrication, in lieu of an enzymatic process, expands the versatility of these new gels for the incorporation of labile proteins and cells. In the present study, the new SELP hydrogels offers sequence dependent, reversible stimuli-responsive features. Their stiffness covers almost the full range of the elasticity of soft tissues. Further, physical modification of the silk domains provided a secondary control point to fine-tune mechanical stiffness while preserving stimuli-responsive features, with implications for a variety of biomedical materials and device needs.

Abstract:Living biological systems, ranging from single cells to whole organisms, can sense, process information, and actuate in response to changing environmental conditions. Inspired by living biological systems, engineered living cells and nonliving matrices are brought together, which gives rise to the technology of engineered living materials. By designing the functionalities of living cells and the structures of nonliving matrices, engineered living materials can be created to detect variability in the surrounding environment and to adjust their functions accordingly, thereby enabling applications in health monitoring, disease treatment, and environmental remediation. Hydrogels, a class of soft, wet, and biocompatible materials, have been widely used as matrices for engineered living cells, leading to the nascent field of engineered living hydrogels. Here, the interactions between hydrogel matrices and engineered living cells are described, focusing on how hydrogels influence cell behaviors and how cells affect hydrogel properties. The interactions between engineered living hydrogels and their environments, and how these interactions enable versatile applications, are also discussed. Finally, current challenges facing the field of engineered living hydrogels for their applications in clinical and environmental settings are highlighted.

Abstract:Biocatalysis is an established chemical synthesis technology that has by no means been restricted to research laboratories. The use of enzymes for organic synthesis has evolved greatly from early development to proof-of-concept - from small batch production to industrial scale. Different enzyme immobilization strategies contributed to this success story. Recently, the use of hydrogel materials for the immobilization of enzymes has been attracting great interest. Within this review, we pay special attention to recent developments in this key emerging field of research. Firstly, we will briefly introduce the concepts of both biocatalysis and hydrogel worlds. Then, we list recent interesting publications that link both concepts. Finally, we provide an outlook and comment on future perspectives of further exploration of enzyme immobilization strategies in hydrogels.

Robust biocatalytic single enzyme nanogels by versatile strategies

Abstract:Biocatalysis has been considered as an important and cost-effective technology for the manufacture of fine chemicals, synthesis of pharmaceuticals, biosensors, bioremediation, protein digestion in proteomic analysis, and biofuel cells. A facile in-situ aqueous polymerization procedure to fabricate single enzyme nanogels as robust nanometer scale biocatalysis was studied. Using versatile modification strategies, e.g., acryloylation of lysine group, introducing vinyl group to glutamic acid and asparagine, control of modification pH, active site protection by inhibitors, different single enzyme nanogels were successfully prepared. These nanogels showed greatly improved stability to temperature, organic solvent, and low pH solution without the compromise of catalytic capability. These nanogels were quite suitable to biocatalytic applications in harsh condition, which are constrained by small size or high mass-transfer rating reaction or exposed for extreme pH and temperature. Abstract:Hydrogels provide a versatile platform for biomedical material fabrication that can be structurally and mechanically fine‐tuned to various tissues and applications. Applications of hydrogels in biomedicine range from highly dynamic injectable hydrogels that can flow through syringe needles and maintain or recover their structure after extrusion to solid‐like wound‐healing patches that need to be stretchable while providing a selective physical barrier. In this study, a toolbox is designed using thermo‐responsive poly(N‐isopropylacrylamide) (PNIPAM) polymeric matrices and nanocelluloses as reinforcing agent to obtain biocompatible hydrogels with altering mechanical properties, from a liquid injectable to a solid‐like elastic hydrogel. The liquid hydrogels possess low viscosity and shear‐thinning properties at 25°C, which allows facile injection at room temperature, while they become viscoelastic gels at body temperature. In contrast, the covalently cross‐linked solid‐like hydrogels exhibit enhanced viscoelasticity. The liquid hydrogels are biocompatible and are able to delay the in vitro release and maintain the bioactivity of model drugs. The antimicrobial agent loaded solid‐like hydrogels are effective against typical wound‐associated pathogens. This work presents a simple method of tuning hydrogel mechanical strength to easily adapt to applications in different soft tissues and broaden the potential of renewable bio‐nanoparticles in hybrid biomaterials with controlled drug release capabilities. Abstract:Abstract Biocatalysis is increasingly replacing traditional methods of manufacturing fine chemicals due to its green, mild, and highly selective nature, but biocatalysts, such as enzymes, are generally costly, fragile, and difficult to recycle. Immobilization provides protection for the enzyme and enables its convenient reuse, which makes immobilized enzymes promising heterogeneous biocatalysts; however, their industrial applications are limited by the low specific activity and poor stability. Herein, we report a feasible strategy utilizing the synergistic bridging of triazoles and metal ions to induce the formation of porous enzyme-assembled hydrogels with increased activity. The catalytic efficiency of the prepared enzyme-assembled hydrogels toward acetophenone reduction is 6.3 times higher than that of the free enzyme, and the reusability is confirmed by the high residual catalytic activity after 12 cycles of use. A near-atomic resolution (2.1 Å) structure of the hydrogel enzyme is successfully analyzed via cryogenic electron microscopy, which indicates a structure–property relationship for the enhanced performance. In addition, the possible mechanism of gel formation is elucidated, revealing the indispensability of triazoles and metal ions, which guides the use of two other enzymes to prepare enzyme-assembled hydrogels capable of good reusability. The described strategy can pave the way for the development of practical catalytic biomaterials and immobilized biocatalysts.

Abstract:In this article, we report a "smart" hydrogel system, which can be remodeled into multiple architectures through dynamic covalent adaptable networks. The topological changes in hydrogel structures yield dynamically tunable properties through the reformation of covalent chemical linkages via amine-thiol scrambling, thiol-thiol exchange, decoupling reaction, and disulfide formation. The stiffness of the hydrogels can be regulated via dynamic covalent bonding, with some hydrogels displaying selfhealing and shear thinning properties, as demonstrated by rheological measurements. Significantly, the dramatic structural transformations are achieved under neutral aqueous conditions at room temperature. These "smart" hydrogels show good biocompatibility, which can induce cell growth in two-dimensional cell culture and effectively serve as a scaffold for encapsulating and releasing human mesenchymal stem cells in three-dimensional cell culture. Thus, the developed "smart" hydrogel system holds great potential in biomedical applications such as tissue engineering and cell therapy.
Engineering enzyme conformation within liquid-solid hybrid microreactors for enhanced continuous-flow biocatalysis

Abstract:The artificial engineering of an enzyme's structural conformation and dynamic properties to promote its catalytic activity and stability outside cellular environments is highly pursued in industrial biotechnology. Here, we describe an elegant strategy of combining the rationally designed liquid-solid hybrid microreactor with a tailor-made polyethylene glycol functional ionic liquid (PEG-IL) microenvironment to exercise a high level of control over the configuration of enzymes for practical continuous-flow biocatalysis. As exemplified by a lipase driven kinetic resolution reaction, the obtained system exhibits a 2.70 to 30.35-fold activity enhancement compared to their batch or traditional IL-based counterparts. Also, our results demonstrate that the thermal stability of encapsulated lipase can be significantly strengthened in the presence of PEG groups, showcasing a long-term continuous-flow stability even up to 1000 h at evaluated temperature of 60 oC. Through systematic experiment and molecular dynamics simulation studies, the conformational changes of the active site cavity in the modified lipases are correlated with enzymatic properties alteration, and the pronounced effects of PEG-groups in stabilizing enzyme's secondary structures by delaying unfolding at elevated temperatures are identified. We believe that this study will guide the design of high-performance enzymatic systems, promoting their utilization in real-world biocatalysis applications.

Abstract:The molecular engineering of dissipative energy to construct the hyperelastic hydrogels with outstanding swelling resistance, good biocompatibility, excellent hydrophilicity and skin‐like elastic moduli is introduced. Noticeable progress has been made in mechanically strong and tough of polymer gels, whereas this consistency between mechanical robustness and perfect elasticity remains a challenge. Here, a simple method is conducted for fabricating hyperelastic hydrogels based on the proposed molecular engineering of dissipative energy taking advantage of two distinct interactions with great difference of binding energy. This molecular engineering of dissipative energy refers to the complementarity at the molecular level between strong and weak interactions: the strong interaction provides the mechanical strength, the weak interaction eliminates the yielding and dense chain entanglements are further implanted, resulting in the perfect hyperelasticity. Such a complementarity in reformation time and binding energy of two interactions triggers a wide range of controllable window for regulating mechanical performances, filling the current gap in the design of hyperelasticity using only high‐order structures. The resultant hyperelastic hydrogels exhibit remarkable tensile strength and linear elastic deformation, but also outstanding swelling resistance, biocompatibility, hydrophilicity, and skin‐like elastic moduli. The work thus provides a promising concept for designing elastic and robust polymer gels and brings a new perspective in the combination of mutually incompatible mechanical properties, which will likely promote innovative applications in bioengineering and bioelectronics.

Abstract:An enzyme‐assisted surface activation approach is presented to create reactive hydrogel microparticles (rHMPs) composed of thiolated hyaluronic acid and hyperbranched poly(β‐hydrazide esters). This approach improves the accessibility of active elements within HMPs, promoting initial cell adhesion and spreading, and facilitating its subsequent application as bioprinting inks and tissue engineering scaffolds. Advancing the integration of nonliving and living components relies heavily on functional hydrogel materials with biocompatibility and customizability. In this study, an enzyme‐assisted surface activation method is developed to produce reactive hydrogel microparticles (HMPs) comprising thiolated hyaluronic acid and hyperbranched poly(β‐hydrazide esters). Fluorescence labeling analysis reveals an over six‐fold increase in surface‐active functional group density on the hydrogels and three‐fold on HMPs after enzyme activation. This enhancement improves accessibility of active elements, facilitating post‐functionalization and optimizing their capacity to support initial cell adhesion and spreading as carriers for cell cultures. Additionally, by utilizing the exposed reactive double bonds on the HMP surfaces post‐enzymatic treatment, thiolated HMPs are produced through a thiol‐ene coupling reaction with thiolated polymers. These thiolated HMPs bond together spontaneously, resulting in the formation of annealed granular hydrogels with interconnected large‐pore networks and a tunable storage modulus (G') from tens to hundreds of pascals, compatible with most soft tissues. Integrated with 3D bioprinting, this hydrogel ink generates prints that foster cell adhesion, migration, growth, and network formation. Moving forward, integrating various granular hydrogel scaffold systems with the enzyme‐assisted activation technique holds significant promise for enhancing performance and expanding applications in regenerative medicine and innovative living materials.

Abstract:Abstract Hydrogels have been designed to react to many different stimuli which find broad applications in tissue engineering and soft robotics. However, polymer networks bearing mechano-responsiveness, especially those displaying on-demand self-stiffening and self-softening behavior, are rarely reported. Here, we design a mechano-controlled biocatalytic system at the molecular level that is incorporated into hydrogels to regulate their mechanical properties at the material scale. The biocatalytic system consists of the protease thrombin and its inhibitor, hirudin, which are genetically engineered and covalently coupled to the hydrogel networks. The catalytic activity of thrombin is reversibly switched on by stretching of the hydrogels, which disrupts the noncovalent inhibitory interaction between both entities. Under cyclic tensile-loading, hydrogels exhibit self-stiffening or self-softening properties when substrates are present that can self-assemble to form new networks after being activated by thrombin or when cleavable peptide crosslinkers are constitutional components of the original network, respectively. Additionally, we demonstrate the programming of bilayer hydrogels to exhibit tailored shape-morphing behavior under mechanical stimulation. Our developed system provides proof of concept for mechanically controlled reversible biocatalytic processes, showcasing their potential for regulating hydrogels and proposing a biomacromolecular strategy for mechano-regulated soft functional materials.

Abstract:Hydrogels have considerably emerged in a variety of fields, but their weak mechanical properties severely restrict the wide range of implementation. Herein, we propose a multiscale hydrogen bonding toughening strategy using saccharides-based materials to optimize the hydrogel network. The monosaccharide (glucose) at molecular scale and polysaccharide (cellulose nanofibrils) at nano/micro scale can effectively form hydrogen bonds across varied scales within hydrogel network, leading to significantly enhanced mechanical properties. Besides, the toughened hydrogels present excellent environmental resilience and bad solvent resistance, allowing them to retain their performance under various severe environments. Notably, after exchanging with a bad solvent ethanol, the alcogel exhibits strain-depended vivid interference color, allowing it to function as mechano-optical sensors. Finally, this strategy has been shown to be adaptable across multiple material systems, and the resulting hydrogels have potential as the bioelectronic interface for long-term stable recording of physiological signals, highlighting the promise of sustainable biomaterials for designing high-quality hydrogels for advanced applications.

Rationally Designed Synthetic Protein Hydrogels with Predictable and Controllable Mechanical Properties

Abstract:A key challenge in biomaterials research is to produce synthetic hydrogels that can replicate the diverse mechanical properties of the naturally occurring tissues for various biomedical applications, including tissue engineering, stem cell and cancer research, cell therapy, and immunomodulation. However, currently, the methods that can be used to control the mechanical properties of hydrogels are very limited and are mainly focused only on the elasticity of hydrogels. In this work, combining single molecule force spectroscopy, protein engineering and theoretical modeling, we show that synthetic protein hydrogels with predictable mechanical properties can be rationally designed using protein building blocks with known mechanical properties at the molecular level. We have successfully tuned the elasticity, extensibility, toughness and self-healing of hydrogels using this approach [1]. We have also demonstrated the engineering of physically crosslinked muscle-mimic hydrogels which are strong, tough, elastic and able to function under significant load and spontaneously self-heal when damaged [1]. Next, by using light-responsive proteins as building blocks, we have been able to engineer hydrogels whose mechanical properties are reversibly tunable by light illumination [2]. These hydrogels can capture the dynamic mechanical environments of naturally occurring extracellular matrices and are potentially suitable for cell culture and tissue engineering. Finally, we show that the hydrogel network structure is also important for the mechanical properties of hydrogels. The hydrogels made of the same protein building blocks but with different network structures can have distinct mechanical responses. These studies provide a biomechanical basis for the rational design of synthetic proteins hydrogels with predictable and controllable mechanical properties.

Abstract:We report a strategy to create photodegradable poly(ethylene glycol)–based hydrogels through rapid polymerization of cytocompatible macromers for remote manipulation of gel properties in situ. Postgelation control of the gel properties was demonstrated to introduce temporal changes, creation of arbitrarily shaped features, and on-demand pendant functionality release. Channels photodegraded within a hydrogel containing encapsulated cells allow cell migration. Temporal variation of the biochemical gel composition was used to influence chondrogenic differentiation of encapsulated stem cells. Photodegradable gels that allow real-time manipulation of material properties or chemistry provide dynamic environments with the scope to answer fundamental questions about material regulation of live cell function and may affect an array of applications from design of drug delivery vehicles to tissue engineering systems.

Cell-Encapsulating Microfluidic Hydrogels with Enhanced Mechanical Stability

Abstract:Whilst microfluidic hydrogels find broad applications in multiple fields such as tissue engineering and regenerative medicine, it has been challenging to sustain the microfluidic structure of most hydrogels due to their insufficient mechanical properties. In this study, we presented a simple method to fabricate microfluidic hydrogels with mechanically enhanced microchannels by using interpenetrating polymer network hydrogels composed of agarose and poly(ethylene glycol) (PEG). The microchannels within the hydrogels were mechanically enhanced with additional PEG layer. Both experimental and numerical results indicated that the mechanically enhanced PEG layer along the microchannels improved the resistance to deformation under compressive loading relative to controls (i.e., without enhanced channel walls). We further assessed the diffusion properties and viability of cells encapsulated within the hydrogels, which showed no significant difference between the enhanced and control groups. The microfluidic hydrogel fabrication approach developed here holds great potential to impact a wide range of fields, such as microfluidics, tissue engineering and regenerative medicine.

Abstract:In the field of tissue engineering, the development of novel switchable cell growth scaffolds and triggered substrate surfaces is desirable for manipulating detachment of cells. In this paper, two kinds of pH-responsive C2-cyclohexane based cell scaffold biomaterials (G1 and G2) have been designed and synthesized. The highlight is that they can induce detachment of cells from material's surface by slightly reducing the pH of culture medium in an acceptable range. The abilities to culture cells on hydrogel scaffolds and guide cells to detach without proteolytic enzyme treatment are very useful for their development in various biomedical applications.

Abstract:<p>Artificial cells are a powerful platform in the study of synthetic biology and other valuable fields. They share a great potential in defining and utilizing the superiority of the living system. Here, a protein synthesis system based on thermal responsive hydrogels with porous structure is reported. The hydrogels can immobilize plasmids on the surface inside their porous structure through a volume phase transition upon 34 °C, forming an aggregation state of DNAs as in nature conditions. The artificial microgels can carry out bioreactions in cell‐free systems and exhibit a sustainable and efficient performance for protein translation. The protein synthesis level reaches a maximum of twice more than that in a conventional solution system when the plasmid concentration is 10–20 ng µL<sup>−1</sup>, along with a doubled effective interval. This is perhaps attributed to confined transcription and translation processes in the near‐surface area of hydrogels. Summarily, the research provides an easy‐handling approach in fabricating effective microgels for cell‐free synthesis and also inspirations for constructing a configurable artificial cell.</p>

Abstract:Cells' local mechanical environment can be as important in guiding cellular responses as many well-characterized biochemical cues. Hydrogels that mimic the native extracellular matrix can provide these mechanical cues to encapsulated cells, allowing for the study of their impact on cellular behaviours. Moreover, by harnessing cellular responses to mechanical cues, hydrogels can be used to create tissues in vitro for regenerative medicine applications and for disease modelling. This Primer outlines the importance and challenges of creating hydrogels that mimic the mechanical and biological properties of the native extracellular matrix. The design of hydrogels for mechanobiology studies is discussed, including appropriate choice of cross-linking chemistry and strategies to tailor hydrogel mechanical cues. Techniques for characterizing hydrogels are explained, highlighting methods used to analyze cell behaviour. Example applications for studying fundamental mechanobiological processes and regenerative therapies are provided, along with a discussion of the limitations of hydrogels as mimetics of the native extracellular matrix. The article ends with an outlook for the field, focusing on emerging technologies that will enable new insights into mechanobiology and its role in tissue homeostasis and disease.

Abstract:Cell microenvironment plays critical roles in regulating cellular activities both spatially and temporally. Engineering cell microenvironment using hydrogels has attracted increasing attention given their native-mimicking properties. In particular, developing hydrogels with specific functions has recently emerged as novel biocomposites to enable the regulation of cell microenvironment from a variety of aspects such as the biological, mechanical and electrical microenvironment. In this review, the state-of-the-art methods for the preparation and application of several novel functional hydrogels are presented, including magnetic hydrogels, photoresponsive hydrogels, conductive hydrogels, thermoresponsive hydrogels, molecule-response hydrogels, as well as tough and stretchable hydrogels. In particular, the applications of these functional hydrogels for engineering cell microenvironment are also reviewed.
