Abstract:Based on the differences between auto-fluorescence spectra of plaque and dental tissue, a quantitative analysis method of plaque using auto-fluorescence imaging was presented. Auto-fluorescence spectrum of tooth samples excited by 405 nm was measured, on the basis of analyzing the correlation between color parameters of fluorescence and dental plaque content, R/B of fluorescence image was proposed as quantitative assessment parameter of dental plaque content, meanwhile long-wavelength pass filter with central wavelength of 490 nm was used to filter fluorescence spectral bands. Finally, experimental setup was built to obtain the auto-fluorescence image. Quantitative analysis results of three different levels of plaque content and distribution was obtained by using R/B value as segmentation thresholds, which confirmed with visual examination results.

Abstract:The determination of fluorine, the lightest element in halogens, suffers from high ionization potential and spectral interference from water molecules in mass spectrometry. Herein, we introduced a liquid nitrogen cooling unit into the laser ablation and ionization source for the first time to construct a cryogenic laser ablation and ionization time-of-flight mass spectrometry (Cryo-LAI-TOFMS) system. With this system, the interference of water-related species at m/z 19 was effectively eliminated, and fluorine atomization and ionization efficiency could reach 6.3%. A direct quantitative analysis method was developed to determine fluorine contents in phosphate rock, copper ore, industrial byproduct gypsum, stream sediment, and soil. Considering the simplicity, high sensitivity, and low spectral interference of this technique, it can be extended to the determination of fluorine content as low as μg/g in complex solid samples.

Abstract:Laser induced autofluorescence spectroscopy of enamel and teeth at different artificial demineralization stage irradiated by 403 nm semiconductor laser were investigated. The quantitative evaluation parameter of mineral content is built on the basis of the fluorescence characteristic spectrum of 443 nm and 526 nm ratio to the total fluorescence integral. In the end, the dependence of characteristic spectrum on the excitation light power was experimentally investigated. The results reveal that at least 10 mW power is necessary to stimulate the featured spectrum.

Abstract:A ratiometric near-infrared fluorescence probe based on IR-780 is developed and applied to fluoride anion (F-) detection in potable water and white flour as well as fluorescence imaging in living cells and mice. The proposed probe not only displays a linear ratiometric (F740/F690 nm) fluorescence response but also possesses near-infrared wavelengths to F- with a detection limit of 0.2 μM. Moreover, the designed probe displays high selectivity toward F-, which makes it feasible for F- detection in potable water and white flour. More importantly, applied to monitor F- in living HepG2 cells and male BALB/c mice, the probe indicates good biocompatibility and low cytotoxicity. However, no study for F- detection has been reported by a ratiometric NIR fluorescent probe so far. We expect that this probe with superior properties has great potential for use in F- detection in biological systems and in vivo studies.

Abstract:Using 6-hydroxyl-quinoline-2-benzothiazole (HQB) as a two-photon fluorophore and tert-butyldiphenylsilyl as a recognition domain for F(-), a ratiometric two-photon fluorescent fluoride probe, QF, was synthesized and fully characterized. QF displays both one- and two-photon ratiometric responses towards fluoride ions in aqueous solution. QF was enabled to detect exogenous fluoride ions in living cells by a ratiometric method. Two-photon microscopic imaging of fluoride ions in living HeLa cells and zebrafish has also been achieved. QF has been demonstrated to be an excellent fluorescent probe with high selectivity, low cytotoxicity and good photostability.

Abstract:A quantitative method to discriminate caries lesions for a fluorescence imaging system is proposed in this paper. The autofluorescence spectral investigation of 39 teeth samples classified by the International Caries Detection and Assessment System levels was performed at 405 nm excitation. The major differences in the different caries lesions focused on the relative spectral intensity range of 565-750 nm. The spectral parameter, defined as the ratio of wavebands at 565-750 nm to the whole spectral range, was calculated. The image component ratio R/(G + B) of color components was statistically computed by considering the spectral parameters (e. g. autofluorescence, optical filter, and spectral sensitivity) in our fluorescence color imaging system. Results showed that the spectral parameter and image component ratio presented a linear relation. Therefore, the image component ratio was graded as < 0.66, 0.66-1.06, 1.06-1.62, and > 1.62 to quantitatively classify sound, early decay, established decay, and severe decay tissues, respectively. Finally, the fluorescence images of caries were experimentally obtained, and the corresponding image component ratio distribution was compared with the classification result. A method to determine the numerical grades of caries using a fluorescence imaging system was proposed. This method can be applied to similar imaging systems.

Abstract:A reaction-based two-photon (TP) ratiometric fluorescence probe Z2 has been developed and successfully applied to detect and image fluoride ion in living cells and tissues. The Z2 probe was designed designed to utilize an ICT mechanism between n-butylnaphthalimide as a fluorophore and tert-butyldiphenylsilane (TBDPS) as a response group. Upon addition of fluoride ion, the Si-O bond in the Z2 would be cleaved, and then a stronger electron-donating group was released. The fluorescent changes at 450 and 540 nm, respectively, made it possible to achieve ratiometric fluorescence detection. The results indicated that the Z2 could ratiometrically detect and image fluoride ion in living cells and tissues in a depth of 250 μm by two-photon microscopy (TPM).

Abstract:Fluoride ions (F-), as a strongest electron withdrawing anion of the smallest size, have played an indispensable role in biological systems. Here a mitochondria-targeted near-infrared (NIR) fluorescent probe (Mito-FP) is developed for selective detection of F- in living cells. Mito-FP is designed using a MR hemicyanine as a fluorophore and tert-butyldimethylsilyl (TBDMS) as a recognition site which is linked to the fluorophore via a 4-hydroxylbenzyl alcohol self-immolative linker. Mito-FP is essentially nonfluorescent. F- can specifically cleave the Si-O bond which triggers the elimination of the quinone methide intermediate and generates the initial NIR hemicyanine fluorophore with distinct intramolecular charge transfer and activated fluorescence signal. The Mito-FP probe is responsive to the concentrations of F- in the range of 10-120 mu M with an estimated limit of detection of 3.2 mu M. Mito-FP also exhibits high selectivity toward F- with good stability in the physiological pH conditions. Mito-FP is successfully introduced to detect and image F- in HeLa cells. Moreover, Mito-FP is also demonstrated to exhibit excellent mitochondrial-targeting ability. This NIR fluorescence probe could provide a useful tool for studying F- in mitochondria.

Abstract:AbstractFluorine (19F) is a promising moiety for quantitative magnetic resonance imaging (MRI). It possesses comparable magnetic resonance (MR) sensitivity to proton (1H) but exhibits no tissue background signal, allowing specific and selective assessment of the administrated 19F‐containing compounds in vivo. Additionally, the MR spectra of 19F‐containing compounds exhibited a wide range of chemical shifts (>200 ppm). Therefore, both MR parameters (e.g., spin–lattice relaxation rate R1) and the absolute quantity of molecule can be determined with 19F MRI for unbiased assessment of tissue physiology and pathology. This article reviews quantitative 19F MRI applications for mapping tumor oxygenation, assessing molecular expression in vascular diseases, and tracking labeled stem cells. Abstract:The purpose of this study was to investigate compositional changes in human enamel irradiated with the free electron laser (FEL). The exposure on dental enamel at the wavelength of 9.64 μm was observed with the Beijing free electron laser. The distribution of elements in the irradiated or non-irradiated enamel was measured by scanning electron microscope (SEM) with energy-dispersive spectroscopy and synchrotron radiation X-ray fluorescence (SRXRF) in Beijing Synchrotron Radiation Facility (BSRF). The results showed that the P/Ca ratio in the ablation region of enamel at the maximum wavelength of infrared absorption was obviously smaller than that at the non-maximum wavelength. In the ablation region the ratios of P/Ca and Ca/Sr were smaller than those in the non-ablation region. The distributions of P, Ca and Sr in the ablation region were heterogeneous due to the element change caused by FEL irradiation.

Abstract:There are significant differences in the nature of fluorination and the mechanisms of laser sample coupling between IR and UV lasers. Their applications to oxygen isotope analysis are also different. The accuracy and precision of CO 2 laser fluorination analysis are influenced by the following factors: the grain size, the volume of sample gas, the efficiency of adsorption of molecule sieve, the adsorbing water in laser system, and the δ 17 O values affected by 14 N 19 F +. The δ 18 O values of fine grain quartz are lower than its true values because of the grain size effect. The CO 2 laser fluorination technique, using rapid heating and defocused beam, allows both high accuracy and high precision determination of oxygen isotope ratios for quartz, feldspar and whole rock. The inaccurate results that are caused by micro volume sample gas can be calibrated by an empirical curve. The declining of molecule sieve's adsorption can also produce oxygen isotope fractionation. Therefore, it is important to determine the life time of molecule sieve. Failure to make pre fluorination can result in the production of significant amounts of HF, which frosts the BaF 2 window and even reacts with some minerals.

Abstract:Dental caries has attracted widespread concern because of its universality. However, the current diagnosis of dental caries depends largely on naked eyes and dentists' experience, which causes considerable randomness and inaccuracy in the diagnosis. In this study, temporally shaped double pulse (DP) femtosecond laser-induced breakdown spectroscopy (LIBS) has been used for tooth composition detection for the first time. Under the laser fluence of 20 J/cm2, the spectra intensity for DP LIBS with a pulse delay of 200 ps was 13 times higher than that of single pulse. For further prediction, the detection model based on DP LIBS and partial least squares discrimination analysis could discriminate dentin, enamel, and caries with a prediction accuracy of above 90%. This method may provide rapid and accurate feedback to dentists for real-time diagnosis in the future.

Abstract:Femtosecond (fs) lasers have high intensity and ultrashort pulse duration. Tunneling ionization occurs for molecules subject to such intense laser fields. We have studied the mass spectra of a variety of molecules irradiated by intense fs laser pulses. These molecules include some typical volatile organic compounds contained in human breath and in the atmosphere. The results demonstrate that all of these molecules can be ionized by intense fs laser pulses. Dominant parent ion and some characteristic ionic fragments are observed for each molecule. The degree of fragmentation can be controlled by adjusting the laser intensity. Moreover, saturation ionization can occur for each molecule by increasing the laser intensity. These features indicate that fs laser mass spectrometry can be a sensitive tool to identify and quantify volatile organic compounds in human breath.

Abstract:The applicability and potential advantages of femtosecond laser-induced ionization microscopy in examining biological samples are demonstrated. High-resolution section images of the inner structures and the topographical surface profiles of a piece of unstained onion skin and a human oral epithelium specimen are obtained by collecting the plasma emission signals generated either inside the samples or from air in the vicinity. It is concluded that the high material discrimination capability and the minimum damage effect give this laser-induced ionization imaging technique great potentials in the area of biological imaging and medical analyses.

Abstract:In view of the few reports of the near-infrared emissive probe for fluorine ions, we herein designed and synthesized a new easy-to-get colorimetric and near-infrared emissive fluorescent probe (IS-NR-F) with a large Stokes shift (>127 nm). Based on specific F- triggered desilylation reaction induced enhanced ICT strategy involving the donor phenolate anion and the acceptor malononitrile, the probe exhibited dual colorimetric and fluorescent turn-on responses, and provided excellent selectivity for fluoride ions. The fluorescent response at 665 nm displayed very good linear relationship in the wide concentration range and deduced a low detection limit of 0.09 ppm. The detection mechanism was confirmed by H-1 NMR, ESIMS, and TLC calculation. Moreover, probe IS NR F has been successfully employed to detect F - in tap water, toothpaste samples, and fluorescent imaging of F- in HeLa cells. (C) 2019 Elsevier Ltd. All rights reserved.

Abstract:Fluorine (F) is a pivotal element in the formation of human dental and skeletal tissues, and the consumption of water and tea constitutes a significant source of fluoride intake. However, prolonged ingestion of water and tea with excessive fluoride content can lead to fluorosis, which poses a serious health hazard. In this manuscript, a novel turn-on fluorescent probe DCF synthesized by bis-coumarin and tert-butyldiphenylsilane (TBDPS) was introduced for detecting F- in potable water and tea infusions. By leveraging the unique chemical affinity between fluoride and silicon, F- triggers the silicon-oxygen bond cleavage in DCF, culminating in a conspicuous emission of yellow fluorescence. Validated through a succession of optical tests, this probe exhibits remarkable advantages in terms of superior selectivity, a low detection limit, a large Stokes shift, and robust interference resistance when detecting inorganic fluoride. Moreover, it can serve as portable test strips for on-site real-time identification and quantitative analysis of F-. Furthermore, the application of DCF for in-situ monitoring and imaging of F- in zebrafish and soybean root tissues proved its significant value for F- detection in both animal and plant systems. This probe potentially functions as an efficient instrument for delving into the toxic mechanisms of fluoride in physiological processes.

Abstract:Herein, a ratiometric fluorescence sensing strategy coupled with smartphone imaging-based sensing platform was proposed for the on-site determination of fluoride ion (F-) with high sensitivity and accuracy. The principle of sensing strategy is based on the fluoride-promoted Si-O bond cleavage of 2-(tert-butyldiphenylsilyloxy)phenol (2-TBDPSP) to release 2-hydroxyphenolate, which rapidly auto-oxidized to ortho-quinone. As excellent electron acceptor, these quinone species covalently bonded on the surface of dual-emission amino-modified quantum dots (QDs) nanohybrid via a Michael's type adduction, quenching the fluorescence of green-emitting QDs on the surface of the nanohybrid, while not affecting the fluorescence of red-emitting QDs embedding silica nano spheres. Upon exposure to different amounts of F-, the variations of dual emission intensity ratios display continuous color changes from green to red, which could be directly observed by naked eyes. Then a smartphone imaging-based sensing platform was constructed by 3D-printing technology. The smartphone camera acquired the images of fluorescence derived from samples, and the Color Picker APP installed in smartphone continued to read out the Red, Green and Blue (RGB) channel values of these images. There was a linear relationship between the ratio of Red and Green (R/G) and F- concentration in the range of 0-70.0 mu M. The limit of detection (LOD) was estimated to be 2.0 mu M, much lower than the allowable level of F- (similar to 63.16 mu M) in drinking water set by World Health Organization. This methodology reported here is low-cost, portable, easy-operation, and thus potentially attractive for F- determination without the need of elaborate equipment.

Abstract:The cystea mine-capped CdTe quantum dots (QDs) undergo spontaneous self-aggregation via NH-N hydrogen bonds. In the presence of F, the aggregates disassembled due to the substitution of NH-F hydrogen bonds for NH-N hydrogen bonds. Thus, the hydrodynamic size of the QDs aggregates was decreased gradually as the concentration of F-increased. Here a one-step sensitive method using dynamic light scattering(DLS) was introduced for directly detection of F-. The detection limit of this method was ca. 20 nmol/L. It was ca. 2 orders of magnitude lower than that of fluorscence methods and F- selective electrode, as reported previously.
