Abstract:Background: Consumption of western diet causes obesity inducing metabolic syndrome and fatty liver.Feeding mice a high fat diet (HFD) causes fatty liver, hepatocyte ballooning, hepatic stellate cell (HSC) activation and fibrosis.Cholangiocytes are also susceptible to damage upon exposure to HFD.Histamine (HA) (i) is a trophic growth factor that stimulates biliary proliferation; (ii) induces HSC activation and fibrosis and (iii) is a regulator of leptin signaling.Our aim was to evaluate the effects of the loss of HDC on the biliary epithelium following HFD.Methods: WT (balb-c) and HDC -/- mice were fed a control diet or a high fat, trans-fat diet (HFD, 45% of calories from fat) coupled with a high fructose corn syrup equivalent (55% fructose, 45% glucose w/w) for 13 weeks.Animals were weighed weekly and food/water intake was measured.From all mice, we collected serum and liver.H&E and Oil red O staining were performed to determine liver damage and fat deposition.Intrahepatic biliary mass (IBDM) was evaluated by CK-19 immunohistochemistry. Hepatic fibrosis was detected by staining for Masson's Trichrome and hydroxyproline content was measured.In total liver we measured a-SMA, fibronectin and collagen type-1a by qPCR.To determine the specific effects of HFD on biliary epithelium, we isolated cholangiocytes from livers by laser capture microdissection (LCMD) before measuring the aforementioned parameters by qPCR.Leptin serum levels were measured by EIA and the leptin receptor, Ob-R, was evaluated by qPCR in total liver and cholangiocyte lines transfected with empty vector or HDCshRNA.Results: Both HFD WT and HDC -/- mice had increased body weight and liver/BW ratios; however, HFD HDC -/- mice had higher LW/BW and weighed 40% more compared to HFD WT.Fat deposition and lipid droplet accumulation significantly increased in both HFD WT and HDC -/- mice compared to control.In HFD WT mice IBDM and hepatocyte damage increased compared to control.Interestingly, in HFD HDC -/- mice, hepatocyte damage increased, but IBDM and biliary damage was unchanged or reduced compared to control.In both HFD WT and HDC -/- mice fibrosis increased compared to control.In cholangiocytes isolated by LCMD, fibrosis, senescence and proliferation increased in HFD WT mice compared to control, but were unchanged in HFD HDC -/- compared to control.Serum leptin levels significantly increased in HFD HDC -/- mice compared to control.Ob-R expression increased in HFD WT compared to control, but decreased in total liver from HFD HDC -/- and in cholangiocyte cell lines lacking HDC, suggesting that HA regulates leptin signaling.Conclusion: HDC -/- mice fed HFD are more susceptible to fatty liverinduced fibrosis coupled with hepatocyte damage whereas cholangiocytes are protected from HFD damage.HDC/HA/leptin signaling may be important to prevent biliary damage in obese patients.
