Abstract:Germination and physical field treatments were effective methods to change the content of active substances in flaxseed, but their combined effects have been seldom investigated. The present study characterizes the influence of germination on lignan, tocopherol, phytosterol, fatty acids, and total fat in flaxseed. It was observed that 18-24 h of germination caused increases in.-tocopherol, campesterol, stigmasterol, and sitosterol contents by 24.47, 12.66, 16.72, and 9.73%, respectively. With the noticeable reduction in total fat content, linolenic acid, oleic acid, and linoleic acid significantly decreased at 24 h germination. The secoisolariciresinol diglucoside peaked at 10.44 +/- 1.00 mg g(-1) dry weight after 6 h germination, which was an increase of 20.78% compared to nongerminated flaxseed. Furthermore, the combined effects of ultrasound or microwave and germination on the lignan and antioxidant activity were explored. The diphenyl-2-picrylhydrazyl free-radical scavenging rate of the lignan extract was significantly improved by the ultrasonic pretreatment at 60 W for 10 min or microwave at 130 W for 10 s in germinated flaxseed. The results revealed the positive impact of short-term germination on the active substances in flaxseed and indicated that the combined effect of germination and physical field treatments were effective approaches to improve the nutritional quality of flaxseed.

Abstract:The changes in chemical composition and antioxidant activities of brown and golden flaxseeds (Linum usitatissimum L.) during 5-day germination were investigated. Flaxseeds germinated in a dark and closed incubator at the temperature of 25 degrees C. With noticeable reduction in fat content, germinated flaxseeds were determined higher percentage of linolenic acid. The total phenolic content and antioxidant activities increased with germination time. The highest amount of secoisolarlciresinol diglucoside (SDG) was found in 2-day germinated brown and golden flaxseeds, which increased 1.04-fold and 1.09-fold, respectively, and germinated flaxseed oil was found rich in SDG. Intriguingly, germination increased the alpha-tocopherol content from 0 to 4.58mg per 100g and 0 to 7.83mg per 100g in brown and golden flaxseeds while gamma-tocopherol decreased. Germination process also reduced the total content of cyanogenic glucosides, the content of linustatin, neolinustatin and lotaustralin decreased significantly, but caused an increase in the content of linamarin. Therefore, germination can be utilised as an effective method to improve the nutritional value of flaxseed.

Abstract:SummaryFlaxseed (Linum usitatissimum L.) is well known for its health benefits and chemical composition. This study aimed to evaluate the dynamic changes caused by germination in vitamin C, phenolic compounds and antioxidant activities of flaxseed. It was observed that eight‐day germination of flaxseed sprouts caused increases in phytochemical contents, that is 22.1‐fold increase in vitamin C, 2.67‐fold increase in phenolics and 5.48‐fold increase in flavonoid contents as compare to initial concentration in flaxseeds on dry weight basis. The stimulation was also noted in the three identified phenolic acid ingredients including caffeic acid, p‐coumaric acid and ferulic acid. Similarly, the total PSC and ORAC values of flax sprouts at day 8 increased by 3‐ to 4‐folds compared to nongerminated flaxseeds. It is revealed that comparatively, 8‐day germination of flax sprouts leads to a significant increase in vitamin C, phenolic constituents and total antioxidant activities than those of flaxseeds.
Effect of flaxseed germination on nutritional quality

Abstract:In order to study the quality changes in the process flaxseeds' germination and achieve the high value-added utilization of the new resource product-flaxseed sprout.By measuring the content of protein,fat,amino acids,vitamins,minerals and cyanide,the contents of nutrient composition of flax seed germination before and after were analyzed.The results showed that after germination the cyanide content reduced by 8.95％,the fat content decreased by 23.60％,the protein content increased by 57.77％,the total number of amino acid increased by 61.67％,the total amount of essential amino acids increased by 62.77％.And the content of vitamins increased substantially,up to 77.37 mg/100 g.And minerals including Ca,Mg,K,Zn and Cu increased by 126％.Germination could improve the nutritional value of flaxseed.

Abstract:Flaxseed is one of the richest seed sources of lignan and other bioactive compounds. The present study characterized lignan biosynthesis and potential health benefits in flaxseeds during 10-day germination. The transcription levels of lignan biosynthesis, along with variation of bioactivities, including cellular antioxidant activity and antiproliferative activity of flaxseed sprouts during germination, were studied. The results showed that 8-day germination brought about a 6.3-fold increase in secoisolariciresinol diglucoside and 4.5-fold increase in secoisolariciresinol compared to ungerminated flaxseeds. The highest amount of total phenolics and total flavonoids were found in 10-day geminated flaxseed, namely a 5.6-fold and 55-fold increase compared to ungerminated flaxseed. Transcription analysis revealed that five key-encoding genes in the lignan biosynthetic pathway were up-regulated during germination. Furthermore, the highest antioxidant and antiproliferative activities were found on day 10. These findings suggest that germination for 8-10 days leads to optimal lignan production and potential health benefits if incorporated into the human diet. (C) 2016 Elsevier Ltd. All rights reserved.

Abstract:This study evaluated the effect of ultrasonic pretreatment of flaxseeds for 0–10 min on the biosynthesis of tocopherols, tocotrienols, and carotenoids in flax sprouts. The results showed that tocopherol, tocotrienol and carotenoid contents were significantly increased (up to 130%) after 8-min ultrasonic pretreatment in LT001 flax sprouts as compared with control group. The similar increasing trend of tocopherols and tocotrienols was occurred in fiber flax sprouts after ultrasonic pretreatment, while carotenoid contents showed a decreasing trend. The expression levels of genes involved in the biosynthesis of tocopherols, tocotrienols, and carotenoids were analyzed and the results could partly explain the changing trends of tocopherols, tocotrienols and carotenoids in two different varieties of flax sprouts responding to ultrasonic pretreatments. Present study could be useful guidance for ultrasonic pretreatment to enhance micronutrients and biofortify nutritional values in flax sprouts.

Abstract:This study evaluated different ultrasonic treatments for lignan biosynthesis in two varieties of flax sprouts. Results showed that lignans in flax sprouts significantly raised with ultrasonic pretreatment. Secoisolariciresinol diglucoside dramatically increased by about 6-fold at the flax sprouts. Ultrasonic pretreatment could also affect the accumulation of caffeic acid and p-coumaric acid in flax sprouts. Moreover, it is suggested that fiber flax sprout was more sensitive to ultrasonic pretreatment. The expression levels of genes involved in the biosynthesis of lignan were analyzed and the results could partly explain the accumulation of these compounds. The contents of secoisolariciresinol diglucoside were clustered with ferulic acid, which indicated that the accumulation of ferulic acid might be the key factor during flax sprout maturation for lignan accumulation. Present study could be useful guidance for ultrasonic pretreatment in the promotion of lignan accumulation and the fortification of nutritional values in flax sprouts as a functional vegetable.
Comprehensive analysis of oxidative stability and nutritional values of germinated linseed and sunflower seed oil

Abstract:Germination of seeds is known to affect the nutritional composition of cold-pressed oils. This study focused on the effects of germination on the antioxidants and oxidative stability of linseed and sunflower seed oil. As hypothesized, germination led to increased antioxidant activities and tocopherol, chlorophyll and carotenoid content. Analysis revealed a 37.2 ± 3.5-fold and 11.6 ± 1.5-fold increase in polyphenol content in linseed and sunflower seed oil from germinated seeds, respectively. Using LC-HRMS/MS, profiles with up to 69 polyphenolic substances were identified in germinated seed oils for the first time. Germination promoted lipid hydrolysis, as evidenced by NMR, with overall significant decreases in triacylglycerol content leading to increased diacylglycerol and free fatty acid values. Rancimat measurements predicted a 4.10 ± 0.52-fold longer shelf-life for germinated linseed oil. This study successfully demonstrated the potential of germination to develop PUFA-rich oils with enhanced antioxidant capacity and oxidative stability.

Abstract:This study aimed at determining the changes in physicochemical characteristics and lipid concomitant of rapeseed oil during germination. Rapeseeds of two varieties, namely Shanyou 28 (Sy 28) and Qinyou 10 (Qy 10), were germinated. Oil was obtained by roasting, pressing and moderate refinement. The germination of 12-48 h led to a reduction in acid value, and 24-72 h led to a decrease in peroxide value. alpha-Tocopherol content increased consistently throughout the entire period, increasing to 2.96- and 2.50-fold at the end of germination. In the germination process of 36-72 h, beta-sitosterol, campesterol and brassicasterol content increased gradually. At 72 h, the total phytosterol content rose modestly (1884.02-2071.55 and 1743.96-2691.72 mg kg(-1)). Carotenoid content increased dramatically (5.81-12.68 and 2.54-12.35 mg kg(-1)). An increase occurred in unsaturated fatty acids from 24 to 72 h, whereas saturated fatty acids decreased. These findings indicated that germination enhanced alpha-tocopherol, phytosterol and carotenoid, as well as the fatty acid composition, thereby improving the nutritional value and edible quality of oil. Among various constituents, alpha-tocopherol in oil could be considered as a reference marker to distinguish the germination stage and estimate the nutritional value of rapeseed oil.

Abstract:The composition and structure changes of three energy substances (protein, lipid, and sugar) and minerals during flaxseed germination were investigated. Na, Ca, Fe, and total free amino acids fluctuating increased and peaked at 7 d. Oil and alpha-linolenic acid contents increased initially and reached the maximal increment by 14.8 % and 1.4 % (p < 0.05) at 2 d, after which it declined. Soluble sugar mainly consisted of sucrose (50.47 %-72.77 %), glucose, and fructose during germination. Semi-cylindrical depression was enhanced on flaxseed granule surface, and oil bodies distribution from relatively uniform toward cell wall during 0-2 d. Protein order and stability were varied firstly, then grew steadily at 4-7 d and peaked at 7 d. Metabolic sequence (sugar, protein, and lipid) and related tricarboxylic acid pathway were proposed. Conclusively, germinated flaxseed at 2 and 4 d had higher physicochemical and structural properties, which could serve as high-quality resources for lipid and protein processing respectively.

Abstract:Flaxseed Lignan Macromolecules (FLM) is an important polyphenol in flaxseed with interfacial adsorption behavior. However, FLM is easily degraded during thermal treatment in emulsions which further influence its interfacial properties and application. In this work, the interfacial properties of FLM between the oil-water was evaluated by compression isotherms and interfacial tension to investigated the regulation mechanism of FLM and its heat-treated products on the stability of O/W emulsions. Furthermore, the improvement mechanism of FLM heat-treated products on the physicochemical stability of flaxseed oil emulsion was clarified. Studies showed that thermal degradation occurred on the terminal phenolic acids in FLM when treated under 100 and 150 °C (FLM-100 and FLM-150) without antioxidative activity decreasing. FLM-100 and FLM-150 improved the physicochemical stability of sunflower lecithin (S90) stabilized flaxseed oil emulsion and reduced the concentration of hydroperoxides and TBARS by 26.7 % and 80 % (p<0.05) during storage. It was due to the high interfacial anchoring of FLM-100 and FLM-150, which further strengthened the interface of oil droplets and improved the interfacial antioxidative effect of FLM. This implies that FLM-100 and FLM-150 could act as new efficient antioxidants to be applied in food emulsions.

Abstract:Flaxseed is considered as a major raw material for producing functional vegetable oil. To promote the extraction process of flaxseed oil, effects of radio frequency (RF) pretreatment at investigated temperatures (70, 90, 110, 130, and 150 degrees C) on yield, physicochemical property and sensory quality of flaxseed oil were determined. The results showed that the yield of flaxseed oil increased from 31.29 to 38.23% after RF pretreatment at 150 degrees C (p < 0.05). The total phenolic, lignan, antioxidant capacity, chlorophyll, and carotenoid contents of flaxseed oil increased significantly with the increase of RF pretreatment temperature (p < 0.05). Nonetheless, the higher RF pretreatment temperatures caused a slight increase on oil oxidation indices (free fatty acid, peroxide value, conjugated diene, conjugated trienes, and p-anisidine value). These results provide theoretical guidance for practical applications of RF pretreatment in the production and processing of edible vegetable oil.
A Novel Type of Sprout Food Development: Effects of Germination on Phytic Acid, Glucosinolates, and Lipid Profiles in Rapeseed

Abstract:Germination, a new processing technology, is widely used to improve the nutritional value of plant seeds. Cereals and legumes sprouts are widely popular around the world. In this study, a novel form of sprouts-based rapeseed was produced. Germination promoted the degradation of phytic acid and glucosinolates (GLs) in rapeseed. On the fourth day of germination, the degradation rate of phytic acid in rapeseed was nearly 80%, and the degradation rate of GLs reached more than 90%. The changes in lipid species and content by sprouting were assessed. The crude fat content was significantly reduced during rapeseed germination. Using non-targeted lipidomics, three hundred eighty-nine lipid molecules in 4 categories and 17 subclasses were identified in rapeseed/rapeseed sprouts. Glycerides were most abundant, followed by fatty acids, glycerophospholipids, and sphingolipids. Germination affected not only the lipid content but also its composition. Some functional lipids (such as FAHFAs) were generated and enriched during germination. The content of monounsaturated fatty acids first raised and then decreased during germination. The finding will serve as a theoretical reference for developing novel sprouts based on rapeseed, furnish primary data concerning the nutritional content of rapeseed sprouts, and provide fresh ideas for rapeseed resource exploitation.
Study on the Material Changes of Flax Seeds before and after Germination

Abstract:In this experiment,the flax seed before and after germination was chosen as the raw material,determination of conventional materials such as amino acids,vitamins,reducing sugar,crude fiber,ash and other active substances such as phytic acid,γ-aminobutyric acid,ferulic acid,coumaric acid,total flavonoids,rutin,and so on.The results showed that nutritious materials and active compositions consisted of the total amino acids,vitamin B,crude fiber,phytic acid,γ-aminobutyric acid,ferulic acid increased obviously.Vitamin A,D,C,K1 were not checked out.The amount of vitamin E,reducing sugar,ash,coumaric acid and the total flavonoids reduced.The increasing or decreasing the levels of material before and after germination could provide product design ideas and directions for subsequent processing and application,thus developing targeted high value-added products.

Preparation of high‐quality concentrated fragrance flaxseed oil by steam explosion pretreatment technology

Abstract:<p>In this study, flaxseed was pretreated by steam explosion technology and subsequently pressed to prepare flaxseed oil. GC, UPLC, HPLC, and GC‐MS techniques were used to analyze the quality characteristics of the prepared flaxseed oil. These included the food safety risk indices, micronutrient components, and oxidative stability. The effects of different steam explosion pressures on the quality characteristics and relative volatile components of flaxseed oil were also investigated. The results revealed that steam explosion pretreatment technology could significantly increase the oil yield, improve micronutrient content, and strengthen the oxidation stability of the product. Moreover, the food safety risk indices (e.g., benzopyrene) were controlled within a reasonable range, while the fatty acid content remained almost unchanged. Notably, the relative pyrazine content in the total volatile components of flaxseed oil was 68.25% when the steam explosion pressure reached 1.2 MPa. This was considered as the main factor that contributed to the unique concentrated fragrance of the produced flaxseed oil. To prove the superiority of the steam explosion pretreatment, we compared this technique with traditional high‐temperature roasting and popular microwave pretreatment techniques. The results revealed that flaxseed oil prepared by steam explosion pretreatment displayed the best quality characteristics and most concentrated fragrance. Thus, steam explosion technology shows great potential for application to produce high‐quality concentrated fragrance flaxseed oil. This study provides significant reference and guidance for the preparation process of flaxseed oil.</p>

Abstract:Oilseed rape (Brassica napus L.) is one of the most important oil crops worldwide, and has a large planting area in Yangtze River basin. As one of the four major edible oils, rapeseed oil plays a fundamental role in human life activities. In addition, with the increasing global demand for rapeseed oil, raising oilseed rape production becomes an important direction of rapeseed breeding. As one of the most widely distributed secondary metabolites in nature, flavonoids play a critical part in plant growth and development. Flavonoids can not only improve stress tolerance, but also regulate various physiological activities of plants. However, the role of flavonoids in determining seed oil contents and the evolutionary mechanism of how pigments accumulate in seed coats remain elusive. In this review, we highlighted the metabolism, distribution, and function of flavonoids and its inhibiting effects on fatty acid accumulation in rapeseed. Understanding the molecular mechanism of flavonoids affecting fatty acid accumulation can provide a theoretical basis for increasing oil content of seeds by regulating the allocation of carbon sources in the future.

Abstract:We provided a gene pool of moderate size for selecting or manipulating the candidate genes that favour the acquisition of seed dormancy, shedding light on the elevation of seed oil content in oilseed rape by blocking lipid degradation in developing seeds. In oilseed rape, the association between the germination potential of premature seeds and the final level of seed lipids, and the underlying mechanism, is elusive. Here, we investigated phenotypic differences in the germination percentage of premature seeds in a collection of oilseed rape cultivars. We compared the dynamic lipid accumulation between the deep-, moderate- and non-dormant genotypes and compared the transcriptomes of the seeds at 40 days after pollination between multiple pairs of deep- and non-dormant genotypes. We identified a wide range of differences in germination percentage of premature seeds and the association between the germination potential and the change of fatty acid content at late stage of seed maturation. The comparisons of transcriptomes between deep- and non-dormant seeds revealed the genetic basis for the dormant difference, e.g. the different expression levels of the genes involved in gibberellic and abscisic acid biosynthesis and/or signalling, fatty acid metabolic pathways, and the structure of seed cell wall. We provided a gene pool of moderate size for selecting or manipulating the candidate genes that favour the acquisition of seed dormancy, shedding light on the elevation of seed oil content in oilseed rape by blocking lipid degradation in developing seeds.
Effect of natural fat-soluble antioxidants on oxidative stability of flaxseed oil

Abstract:【Objective】This study investigated the effects of natural fat-soluble antioxidants on the oxidative stability of flaxseed oil when used alone, in combination and in augmentation, in order to improve the oxidative stability and extend the shelf life of flaxseed oil.【Method】Three natural fat-soluble antioxidants, tea polyphenols palmitate, rosemary extract and ascorbyl palmitate, were selected and the phytic acid and citric acid were used as synergists. Using the protection factor（Pf） as a measurement index, the optimal formulation of three antioxidants was optimized by response surface methodology based on the use of single antioxidants, and then the antioxidant effect was enhanced and the shelf life was predicted by using synergists.【Result】 The antioxidants alone had significant effects on the oxidative stability of flaxseed oil, but the effect was poor compared with the synthetic antioxidants（positive control group）. The optimal compound formulation was tea polyphenol palmitate of 0.21 g·kg - 1 and rosemary extract of 0.45 g·kg - 1. Under the optimal compound formulation, the Pf was 1.87 and the oxidation stability was good. The citric acid was determined to be used as synergist with 0.20 g·kg -1 . The final formulation was applied to flaxseed oil and the predicted shelf life of flaxseed oil at 25 ℃ was 843 d, which was twice as long as the addition of synthetic antioxidants（positive control group） and 10 times as long as the addition of antioxidants（blank group）.【Conclusion】The compound formulation significantly improved the oxidative stability of flaxseed oil, extended the shelf life of flaxseed oil, provided a theoretical basis for replacing synthetic antioxidants, promoted the industrial development of flaxseed oil and expanded other applications.

Abstract:Flaxseed oil is the main component of the flaxseed and has many beneficial functions to human health. In this paper, the ultrasound-assisted extraction of oil from flaxseed is reported. The effects of some operating parameters such as ultrasonic power, extraction time, extraction temperature and solvent to solid ratio on the yield of flaxseed oil have been investigated and some of the results have been compared with that of conventional method. It has been found that ultrasound-assisted extraction requires a shorter extraction time and a reduced solvent consumption. The yield of flaxseed oil has been found to increase with the increase of the ultrasonic power and to decrease as the temperature is increased. Scanning electronic microscopy analysis was carried out on the flaxseed powder after the extraction. The images are powerful evidences to show the effect of ultrasound. The fatty acid compositions of the oils extracted by the ultrasound-assisted method and the conventional method have been analyzed using gas chromatography. It has been shown that the compositions of the flaxseed oils were not affected significantly by the application of ultrasound (p>0.05). The ultrasound-assisted extraction may be an effective method for lipid production.
