Abstract:Spinal cord injury (SCI) is catastrophic and can culminate in disability and death. The routine therapy employed in early stages of SCI currently entails surgical procedures combined with high doses of methylprednisolone (MP). MP is highly controversial for the lack of consensus on its true therapeutic effects. Resveratrol (RES) has recently been recognized as a potential and novel therapeutic drug in SCI. Herein, we investigated the effect of RES in a SCI rat-model and found significant improvement in Basso-Beattie-Bresnahan scores. Results obtained from histological, immunohistochemistry, and ultra-structural examinations evidenced the tremendous treatment effect of RES. On the basis of our experimental results, we hypothesize that RES could serve as an effective SCI therapeutic with prolong treatment time following injury.

Abstract:Background:Animal spinal cord injury (SCI) models have proved invaluable in better understanding the mechanisms involved in traumatic SCI and evaluating the effectiveness of experimental therapeutic interventions. Over the past 25 years, substantial gains have been made in developing consistent, reproducible and reliable animal SCI models.Study design:Review.Objective:The objective of this review was to consolidate current knowledge on SCI models and introduce newer paradigms that are currently being developed.Results:SCI models are categorized based on the mechanism of injury into contusion, compression, distraction, dislocation, transection or chemical models. Contusion devices inflict a transient, acute injury to the spinal cord using a weight-drop technique, electromagnetic impactor or air pressure. Compression devices compress the cord at specific force and duration to cause SCI. Distraction SCI devices inflict graded injury by controlled stretching of the cord. Mechanical displacement of the vertebrae is utilized to produce dislocation-type SCI. Surgical transection of the cord, partial or complete, is particularly useful in regenerative medicine. Finally, chemically induced SCI replicates select components of the secondary injury cascade. Although rodents remain the most commonly used species and are best suited for preliminary SCI studies, large animal and nonhuman primate experiments better approximate human SCI.Conclusion:All SCI models aim to replicate SCI in humans as closely as possible. Given the recent improvements in commonly used models and development of newer paradigms, much progress is anticipated in the coming years.

Abstract:BACKGROUND:It is a very difficult task to transfer from the animal model of cervical spondylosis to clinical treatment. However, the pathology of cervical spondylosis is not very clear, and the main reason is the lack of an ideal animal model of the experiment. <br> OBJECTIVE:To review the commonly used modeling methods and evaluate the advantages and disadvantages of different modeling methods through consulting literatures. <br> METHODS:The key words were“cervical spondylosis, cervical spondylotic myelopathy, cervical spondylotic radiculopathy, vertebroarterial type of cervical spondylosis, animal model”. The first author retrieved Medline and CNKI for studies published from 1991 to 2015 concerning animal experiments or clinical studies on the construction of animal model of cervical spondylosis. Final y, 41 studies were included for reviewing. <br> RESULTS AND CONCLUSION:There are many methods for the modeling of cervical spondylosis at the present stage, including implantation method, screw insertion method, bal oon compression method, mechanical compression, injection method, spinal suture method and mechanical imbalance method. Each has its advantages and disadvantages. In the study of cervical spondylosis, we should choose the appropriate animal model according to the research purpose. Further research and the establishment of a comprehensive animal model are stil the goal of efforts.
Comparison on Gene Expression Profiles Between Different Models of Spinal Cord Injury

Abstract:Background: Spinal cord injury (SCI) is a devastating disease with no clear molecular mechanisms or effective treatments. Murine models of SCI have been widely used to explore its pathogenesis. Methods: In this study, a comprehensive bioinformatic analysis using GEO datasets was performed to evaluate the characteristics of different SCI models. Results: We found that the contusion model was similar to the compression model, with inflammation and apoptosis significantly enriched, while more complex biological processes existed in hemisection and transection model. Inflammatory markers can be used as a primary evaluation index of SCI models not only in the acute and subacute phases, but also in the chronic phase. In the meantime, apoptosis markers are more suitable for evaluating mouse SCI models while inflammatory markers are more suitable for rat SCI models. In addition, SCI models with different ages, genders, injury positions, and injury levels were also analyzed. Conclusion: Our findings indicate that SCI is a heterogeneous disease and play an instructive role in model selecting. (c) 2023 The Author(s). Improved Probe on the Manufacturing Method of Spinal Cord Injury Model of Rat

Abstract:Aim: To investigate a simple method that can establish a reliable rat model of spinal cord injury. Methods: Moderate and severe spinal cord injury models were established by using a modified weight drop device. The pathological and functional changes of the rats in two groups were observed by means of BBB scoring and staining with hematoxylin-eosin(HE) so as to estimate the method was reliable. Results: The BBB scoring showed that the moter function of the rats in two groups recovered gradually over time after SCI. However, BBB scores in moderate SCI group were significantly higher than in severe SCI group(P<0.05). Histological and pathological experiments showed that the structure of spinal cord was damaged and the glial scar as well as cavity were formed after SCI, and the lesion area of the spinal cord in moderate SCI group was less than that in severe SCI group(P<0.05). Conclusion: The rat models of impact SCI established in this way could distinguish the graded injury, and the behavioral outcome corresponded with pathological outcome in these models, which indicated that the rat models of SCI possessed good stability, reproducibility and consistency, and could be used in the experimental study of SCI.

Abstract:Using minimally invasive methods to model spinal cord injury (SCI) can minimize behavioral and histological differences between experimental animals, thereby improving the reproducibility of the experiments. These methods need two requirements to be fulfilled: clarity of the surgical anatomical pathway and simplicity and convenience of the laboratory device. Crucially for the operator, a clear anatomical pathway provides minimally invasive exposure, which avoids additional damage to the experimental animal during the surgical procedures and allows the animal to maintain a consistent and stable anatomical morphology during the experiment. In this study, the use of a novel integrated platform called the SCI coaxial platform for spinal cord injury in small animals to expose the T9 level spinal cord in a minimally invasive way and stabilize and immobilize the vertebra of mice using a vertebral stabilizer is researched, and, finally, a coaxial gravity impactor is used to contuse the spinal cord of mice to approach different degrees of T9 spinal cord injury. Finally, histological results are provided as a reference for the readers.
Establishment and evaluation of a rat model of chronic cervical spinal cord compression

Abstract:BACKGROUND: Establishing a stable model of cervical spondylotic myelopathy is essential for the study of cervical spondylotic myelopathy pathogenesis. OBJECTIVE: To clarify the mechanism of cervical spondylotic myelopathy by establishing a cervical spondylotic myelopathy model and observing the pathophysical changes of this model. METHODS: A total of 30 Sprague-Dawley rats were divided into three groups: control group, mild compression group and severe compression group. Water-absorbing compression materials of different size were implanted below the C 5 - 7 to prepare chronic cervical spinal cord compression models. Control group was implanted nothing. RESULTS AND CONCLUSION: The normal morphology in the control group and varying degrees of compression on the spinal cord and spinal canal stenosis in the two compression groups were shown in MRI. The lower amplitude and longer latency of motor evoked potential after spinal cord compression was found in the two compression groups as compared with the control group (P < 0.05). Immunofluorescence staining showed that that a great number of healthy-shaped neurons were found in the control group. But, the number of neurons in the two compression groups was significantly decreased, and the neurons shrunk and demyelination was obvious. The difference of neuron number among the three groups was statistically significant (P < 0.05). Apoptosis in the segment of compressed spinal cord was found in the two compression groups rather than in the control group. In conclusion, rat models of chronic spinal cord compression are in line with the pathological changes of cervical spondylotic myelopathy. The surgical procedure is simple and the rate of infection and mortality is lower. Neuronal injury, demyelination and apoptosis are involved in the process of chronic spinal cord compression in rats.

Abstract:Objective To establish rat model of impact spinal cord injury, observe the pathological changes of the model and assess its stability, reproducibility and consistency. Methods Moderate and severe spinal cord injury (SCI) models were established by using modified weight drop device. The pathological and functional changes after SCI were observed by means of BBB scoring, electrophysiology,immunohistochemistry and electronic microscopy so as to estimate the reproducibility of rat models and their consistency with severity of SCI. Results Locomotion and nerve impulse transduction along the spinal cord measured by motorial and sensory evoked potentials recovered gradually over time after SCI.However, the recovery rate of moderate SCI group was better than that of severe SCI group. Histological and immunohistochemical experiments showed that the glial scar as well as cavity were formed after SCI.Whereas, compared with moderate SCI group, the injury of severe SCI group was severer, with less spared tissue. Electronic microscopic observation displayed that hemorrhage, edema, neutrophilic granulocytic infiltration and chromatin margination of glia arose at day 1 after SCI. Vacuolization of mitochondria, degeneration of axon with edema could be seen at 2 weeks after injury. Degeneration of myelin and deposition of collagen fibril emerged at 8 weeks postinjury. Conclusions The rat models of impact SCI established in this study can distinguish the graded injury, and significantly correlate with the behavioral,electrophysiological and pathological outcomes, which indicates that the models possess good stability, reproducibility and consistency. Glial scar with cavity marked by GFAP or Vimentin is the pathological hallmark after SCI, and thereby GFAP or Vimentin can be used as a marker for demarcate the border of glial scar.
Experimental modelling of human spinal cord injury: a model that crosses the species barrier and mimics the spectrum of human cytopathology

Abstract:Study design: Literature review and presentation of an experimental model of human spinal cord injury, (SCI). Objectives: Experimental designs seek to mimic and model the physical processes by which human SCI occurs and replicate the variety of chronic pathologies that characterize its long term effects. The variations in biological processes that are present between species have contributed to recent difficulties in generalizing experimental findings to the human condition. In this review, one finds: (1) a discourse on the pathological nature of the chronic human lesion, (2) a consideration of how the physical properties of soft tissue injury result in acute and chronic changes in the spinal substance, (3) a description of a device (ESCID) that is able to replicate and dynamically monitor physical indices of SCI as they take place in experimental models, and (4) a summary of how use of this device in different species has allowed the biomechanical descriptors of such injuries to be easily compared even in murine models. Setting: Ohio State University, Ohio, USA. Results: Careful attention to the details of injury device design has finally allowed a direct comparison of contusion-type injury models in the rat and mouse. Biomechanical outcomes with predictive capabilities have evolved that allow the investigator to create the range of pathologies seen in the human lesion even in these small vertebrates. The predictive cytopathology and our ability to manipulate the mouse genome will allow the testing of specific hypotheses related to cause and effect in experimental spinal cord injuries. Since the biomechanics, pathology, and chronic outcomes appear to be similar to those seen in the human, these animal models should facilitate rapid progress in the design of human therapeutics. Conclusions: Biomechanics of certain elements of experimental spinal injury are surprisingly accurate descriptors of acute and chronic pathologies in the spinal cord. This tenet applies across species and has often allowed more accurate design of clinical trials in the past few decades. As molecular approaches to this problem evolve, the use of species with known genomes appear warranted. Models that take advantage of these approaches are likely to produce innovations that quicken the pace of human trial strategies.
A Cervical Spinal Cord Hemi-Contusion Injury Model Based on Displacement Control in Non-Human Primates (Macaca fascicularis)

Abstract:Non-human primate (NHP) spinal cord injury (SCI) models can be informative in the evaluation of treatments that show promise in rodent models, prior to translation to humans. In the present study, we aimed to establish a cervical spinal hemi-contusion model with controlled displacement and evaluate the abnormalities in behavior, electrophysiology, histology and MRI. Twelve adult NHPs were divided into an SCI group (n=8, 24 and 48 weeks) and a control group (n=4). An impactor (=4 mm) was driven to compress the left C5 cord at 800 mm/s. The contusion displacement and peak force was 4.08±0.17 mm and 19.8±4.6 N. The behavioral assessment showed a consistent dysfunction below the wrist and spontaneous recovery of limb function after injury. Lesion length and lesion area at the epicenter based on T2 hyperintensity were 5.68±0.47 mm and 5.99±0.24 mm2 at 24 weeks-post-injury (WPI), and 5.29±0.17 mm and 5.95±0.24 mm2 at 48 WPI. The spared spinal cord area immuno-positive for glial fibrillary acidic protein was significantly reduced while the staining intensity increased at 24 WPI and 48 WPI compared to the sham group. Ipsilateral somatosensory and motor evoked potentials were dynamic, increasing in latency and decreasing in amplitude compared to pre-operative values or the contralateral values, and correlated to varying degrees with behavioral outcomes. A shift in size-frequency distribution of sensory neurons of the DRG was consistent with a loss of large-diameter cells. The present study demonstrated that the NHP SCI model resulted in consistent unilateral limb dysfunction and potential plasticity in the face of loss of spinal cord and DRG tissue.

Abstract:Spinal cord injury (SCI) is a physically and psychologically devastating clinical condition. The typical treatment regimens of decompressive surgery and rehabilitation therapy still leave many patients with permanent disability. The development of new therapies and devices can be accelerated if relevant translational animal models are more effectively used in pre-clinical stages. Swine is a highly relevant model for SCI research, especially with respect to spine and spinal cord anatomy, spine vasculature, immune responses to injury, and functional assessments. Several spine injury models have recently been developed for swine and are beginning to be used to evaluate new therapies. Swine models of SCI offer tremendous advantages for efficient translation of pre-clinical discoveries and the development of new therapies and devices. Future swine models will also be enhanced by advances in gene-editing technology to further elucidate the complex pathophysiology associated with SCI and provide a means to engineer specific spinal pathologies.

Abstract:Spinal cord injury (SCI) due to traumatic injuries such as car accidents and falls is associated with permanent spinal cord dysfunction. Creation of contusion models of spinal cord injury by impacting the spinal cord results in similar pathologies to most spinal cord injuries in clinical practice. Accurate, reproducible, and convenient animal models of spinal cord injury are essential for studying spinal cord injury. We present a novel automated spinal cord injury contusion device for mice, the Guangzhou Jinan University smart spinal cord injury system, that can produce spinal cord injury contusion models with accuracy, reproducibility, and convenience. The system accurately produces models of varying degrees of spinal cord injury via laser distance sensors combined with an automated mobile platform and advanced software. We used this system to create three levels of spinal cord injury mice models, determined their Basso mouse scale (BMS) scores, and performed behavioral as well as staining assays to demonstrate its accuracy and reproducibility. We show each step of the development of the injury models using this device, forming a standardized procedure. This method produces reproducible spinal cord injury contusion mice models and reduces human manipulation factors via convenient handling procedures. The developed animal model is reliable for studying spinal cord injury mechanisms and associated treatment approaches.

Construction of rat spinal cord injury model based on Allen’s animal model

Abstract:The aim of this study is to explore the construction of rat spinal cord injury model guided by Allen's model. Methods: Male rats aged 4–5 weeks and weighing about 250 g are selected as subjects in the Animal Laboratory Center of XX Hospital. Rats are divided into two groups, which are experimental group 1 and experimental group 2, respectively, so as to construct spinal cord injury model in rats. The first group is given 300 g.cm hitting force of T10 spinal cord, and the second group is given 500 g.cm hitting force of T10 spinal cord. Within 25 days after spinal cord injury in Allen's rats, the survival, neurological function, diet, motor ability, tactile ability and auditory ability of the two groups are monitored and evaluated daily. Results: In terms of survival, the survival rate of rats in group 1 is 85%, while that of rats in group 2 is 21%, and there is a concentrated death phenomenon in group 2. In terms of neurological function recovery, experimental group 1 is stable and gets 7 points and experimental group 2 is stable and gets 3 points. In terms of diet, the experimental group 1 is stable and gets 5 points and the experimental group 2 is stable and gets 2 points. In terms of motor ability, the experimental group 1 is stable and gets 5 points and the experimental group 2 is stable and gets 2 points. In tactile sense, experimental group 1 is stable and gets 17 points and experimental group 2 is stable and gets 12 points. It can be seen that the post-operative recovery ability of the experimental group 1 is better than that of the experimental group 2. Conclusion: Under the guidance of Allen's model, compared with the group 2, the experimental group 1 of the rat spinal cord injury model has better recovery in each index. It can be seen that the smaller impact strength is more beneficial to the recovery of rats after spinal cord injury surgery.

Abstract:Objective:To set up a practical and reliable animal model of spinal cord injury.Methods:Spinal cord was hemi-sected C 5～7 and T 9～11 level.At different times after injury,the behavior and the histological change of lesioned cord was observed and evaluated.Results:Its showedthat the hSCI's lateral cord and substantia grisea was degenerated to necrosis under microscopy.It had significance the CBS assessment of study group than that of sham-operated group( P 0.05).The rat's spinal cord ventral horn's motor neuron had great changes of the motor and feel function.Conclusion:Hemi-sected of C 5～7 spinal cord has the advanges of low mortality and it is safe.

Abstract:Animal models are necessary to identify pathological changes and help assess therapeutic outcomes following spinal cord injury (SCI). Small animal models offer value in research in terms of their easily managed size, minimal maintenance requirements, lower cost, well-characterized genomes, and ability to power research studies. However, despite these benefits, small animal models have neurologic and anatomical differences that may influence translation of results to humans and thus limiting the success of their use in preclinical studies as a direct pipeline to clinical studies. Large animal models, offer an attractive intermediary translation model that may be more successful in translating to the clinic for SCI research. This is largely due to their greater neurologic and anatomical similarities to humans. The physical characteristics of pig spinal cord, gut microbiome, metabolism, proportions of white to grey matter, bowel anatomy and function, and urinary system are strikingly similar and provide great insight into human SCI conditions. In this review, we address the variety of existing porcine injury models and their translational relevance, benefits, and drawbacks in modeling human systems and functions for neurophysiology, cardiovascular, gastrointestinal and urodynamic functions.
Animal Models for Treating Spinal Cord Injury Using Biomaterials-Based Tissue Engineering Strategies

Abstract:The aim of the study is to provide an up-to-date review of studies that used preclinical animal models for the evaluation of tissue engineering treatments for spinal cord injury (SCI), which involved the use of biomaterials with or without the addition of cells or biomolecules. Electronic search of the PubMed, Web of Science, and Embase databases was performed for relevant studies published between January 2009 and December 2019. In total, 1579 articles were retrieved, of which 58 studies were included for analysis. Among the included studies, rats were the most common species used for animal models of SCI, while complete transection was the most commonly used injury pattern. Immediate intervention after injury was conducted in the majority of studies, and 8 weeks was the most common final time point of outcome assessment. A wide range of natural and synthetic biomaterials with different morphologies were used as a part of tissue engineering treatments for SCI, including scaffolds, hydrogels, and particles. Experimental parameters in studies using SCI animal models to evaluate tissue engineering treatments should be carefully considered to match the purpose of the study. Biomaterials that have functional modifications or are applied in combination with cells and biomolecules can be effective in creating a permissive environment for SCI repair in preclinical animal models. Impact statement This review provides an up-to-date summary of the preclinical landscape where tissue engineering treatments involving biomaterials were tested in animal models of spinal cord injury (SCI). Using studies published within the last 10 years, novel perspectives were presented on the animal species used, injury pattern, timing of intervention and outcome measurement, and biomaterials selection, as well as a summary of the individual findings of each study. This review provides unique insight into biomaterials-based tissue engineering strategies that have progressed to testing in animal models of SCI, which will help shape future research in the field and propel the clinical translation of discoveries.

Abstract:Spinal cord injuries (SCIs) are difficult to treat. The first animal SCI model (featuring the dropping of a weight) was established by Allen in 1911, and other animal models have been developed since then. Most animal studies have focused only on the molecular features of SCIs, which remain disputed. Recently, it has become clear that SCI may trigger mental and cognitive disorders, however, and brain changes secondary to SCI are under investigation. No consensus on an optimal animal model for cerebral research has emerged. We discuss the appropriate SCI models for studying secondary brain changes.

Abstract:As with all tissues of the central nervous system, the low regeneration ability of spinal cord tissue after injury decreases the potential for repair and recovery. Initially, in spinal cord injuries (SCI), often the surgeon can only limit further damage by early surgical decompression. However, with the development of basic science, especially the development of genetic engineering, molecular biology, tissue engineering, and materials science, some promising progress has been made in promoting the repair of central nervous system injuries. For example, transplantation of neural stem cells (NSCs), olfactory ensheathing cells (OECs), and gene- mediated transdifferentiation to repair central nervous system injury. This paper summarizes the progress and prospects of SCI repair with tissue engineering scaffold and cell transdifferentiation from an extensive literatures.
