Abstract:The "shell biorefinery," which valorizes the shell waste chitin into fine chemicals, has developed rapidly in recent years. Herein, we present a novel base-free heteropolyacid-catalyzed oxidation method for the transformation of chitin biomass into organic acid. After a series of optimization experiments, a 5.93 % yield of formic acid and 25.09 % yield of acetic acid were achieved in the presence of 0.5 equivalent of Mo-V-P heteropolyacids (H 4 PMo 11 VO 40 ·2H 2 O) and air at 180 °C under hydrothermal conditions for 4 h. Meanwhile, we have demonstrated that the Keggin-type heteropolyacid catalysts are capable of efficiently converting microcrystalline chitin into organic acids. The synthesized heteropolyacids are well characterized with FT-IR, XRD, ICP-AES, and TGA. The possible reaction pathway was speculated accordingly. This method offers several advantages, including readily available raw materials, simple operation, and relatively higher yield.

Abstract:Chitin is an abundant and sustainable biomass from oceanic waste. Herein, a simple, mild, and effective method was developed to convert chitin biomass into valuable organic acids at room temperature using a common base and oxidant in water. In dilute sodium hydroxide (NaOH) solution, similar to 72.3% yield of organic acid products including acetic acid (AA), formic acid (FA), glyceric acid (GlyA), and so forth was obtained from the chitin monomer N-acetyl-D-glucosamine under ambient air. Meanwhile, tunable product selectivity can be achieved by employing different oxidative reagents. FA was predominantly formed in 57.1% yield using hydrogen peroxide (H2O2), while AA and GIyA were the major products in 41.2% yield with oxygen (O-2) gas. Divergent reaction pathways were presumably involved when using different oxidants. Compared to the conventional methods at high temperatures and/or pressures, the established approach requires minimal energy inputs, inexpensive equipment with reduced carbon emission, capital costs, and safety risks.

Abstract:The "shell biorefinery" has been proposed and developed rapidly in recent years, which valorizes the underestimated chitin biopolymer from oceanic waste to complement the lignocellulosic biomass for renewable chemicals. Herein, we exploited a simple and effective method to convert chitin biomass into acetic acid (AA) by using vanadium pentoxide (V2O5) and oxygen gas (O-2) in base-free water, which makes the process more environmentally and economically favorable than previous methods. Under optimal conditions, the highest AA yield was obtained in 33.4% and 30.0% from the chitin monomer N-acetyl-D-glucosamine (NAG) and ball milled chitin (BM chitin) polymer. The V2O5 catalyst has multifunctionally facilitated the hydrolysis, deacetylation, and subsequent oxidation into AA. FTIR and XRD analyses were conducted for the solid residues after the reaction. The FTIR spectra of the solid residues highly resembled that of chitin, inferring that chitin hydrolysis into NAG probably happened prior to deacetylation. Besides, XRD data demonstrated that the reaction system could effectively destruct the crystalline regions during the reaction. The study demonstrated a new catalytic approach for chitin biorefinery to renovate the shell waste into valuable chemicals.

Abstract:This paper describes chitin liquefaction in ethylene glycol (EG) under the catalysis of sulfuric acid for the first time. Up to 75% of chitin was liquefied at 165 degrees C within 90 min by using 8 wt % of acid (refer to the mass of EG). The major products (30% yield) were identified to be hydroxyethyl-2-amino-2-deoxyhexopyranoside (HADP) and hydroxyethyl-2-acetamido-2-deoxyhexopyranoside (HAADP) by GC-MS and confirmed by NMR Kinetic studies were conducted based on which a plausible mechanism for product formation was proposed. HADP was dominant during the reaction, whereas HAADP formed fast at an initial stage and then most of it was hydrolyzed to HADP. Unreacted chitin residues with different reaction times were characterized by Fourier transform infrared spectroscopy (FTIR), X-ray powder diffraction (XRD), and solid state NMR. The FTIR results showed that negligible deacetylation reaction occurred, supporting the assumption that HADP was produced from the hydrolysis of HAADP rather than directly from chitin polymer chains. The XRD analysis revealed the gradual decrease in crystallinity with the increase in reaction time, indicating the damage of the crystalline domain by the liquefaction process. The simple, cheap, and efficient liquefaction of chitin opens up a new route to produce chemicals and materials from waste in the fishing industry.

Abstract:Chitin adsorbents with different degrees of deacetylation (DDA) were prepared from crab shells waste and applied in adsorptive removal of dye Congo Red (CR). The adsorbents were characterized by thermal gravimetric analysis, Fourier transform infrared spectroscopy, X-ray diffraction, and elemental analysis. The effects of solid-liquid ratio, initial dye concentration, contact time, temperature, pH value, salt concentration, and DDA on adsorption were studied. It was indicated from orthogonal experiments that the removal of CR reached 99.28% under the optimum condition (DDA = 17.85%, pH = 5.40, and time = 20 h) at 300 K. The equilibrium data were plotted by Langmuir and Freundlich isotherms and described better with Langmuir model (Q(m) = 366.3 mg/g, 300 K) as the temperature increased and the adsorption kinetics was well fitted by pseudo-second-order equation. It was verified from the thermodynamic parameters (Delta G, Delta H, and Delta S) that the adsorption was spontaneous, exothermic and entropy decreasing. Functional groups including amino, acetyl amino, O-glycosides, and -O- on the pyranose ring of chitin were involved in adsorption as active sites through van der Waals force and hydrogen-bonding with the adsorbate. Moreover, CR adsorption on chitin with different DDA further revealed that the crystallinity of chitin played a more important role than amino group content.

Abstract:A large number of hydrogen bonds is the main reason for hindering the dissolution and reaction of chitin, and a mild and green deacetylation method to prepare chitosan for a wider range of applications is urgent. As a non-toxic and degradable green solvent, the deep eutectic solvent can effectively interfere with the hydrogen bond network of chitin, making chitin more susceptible to other solvents. Therefore, a NADES system consisting of betaine and glycerol was proposed for application in the deacetylation reaction of chitin to facilitate further attack of N-acetyl groups by low concentrations of NaOH. After optimizing the reaction conditions, chitosan with 83.77 % deacetylation was prepared, requiring only a concentration of 25 wt% NaOH. The analysis of the product chitosan showed that NADES could not only effectively improve the degree of deacetylation, but also reduce the degree of damage to the molecular weight by alkali. In addition, the potential mechanisms involved in the deacetylation process by NADES were explored. The nature of the reaction was verified by FT-IR, XRD and theoretical calculations as the process of opening intra/intermolecular hydrogen bonds of chitin by NADES. More importantly, experimental and in-depth theoretical studies provide a reference for the green preparation of chitosan.

Abstract:Chitin is the second most abundant renewable polysaccharide on Earth. The degradation of chitin into soluble and bioactive N -acetyl chitooligosaccharides (NCOSs) and N -acetyl-D-glucosamine (GlcNAc) has emerged as a pivotal step in the efficient and sustainable utilization of chitin resources. However, because of its dense structure, high crystallinity, and poor solubility, chitin typically needs pretreatment via chemical, physical, and other methods before enzymatic conversion to enhance the accessibility between substrates and enzyme molecules. Consequently, there has been considerable interest in exploring the direct biological degradation of crystalline chitin as a cost-effective and environment-friendly technology. This review endeavors to present several biological methods for the direct degradation of chitin. We primarily focused on the importance of chitinase containing chitin-binding domain (CBD). Additionally, various modification strategies for increasing the degradation efficiency of crystalline chitin were introduced. Subsequently, the review systematically elucidated critical components of multi-enzyme catalytic systems, highlighting their potential for chitin degradation. Furthermore, the application of microorganisms in the degradation of crystalline chitin was also discussed. The insights in this review contribute to the explorations and investigations of enzymatic and microbial approaches for the direct degradation of crystalline chitin, thereby fostering advancements in biomass conversion.

Abstract:Global shellfishery waste generation is from 6 to 8 million metric tons annually. Chitin, as a major component in crustacean shells, is the second most abundant biopolymer on Earth, having the potential to supplement the lignocellulosic biomass resource for renewable chemicals. Herein, we established direct transformation of chitin and raw shrimp shells into acetic acid (HAc) by a catalytic method using metal oxide and oxygen gas in basic water. The work showcased that chitin is a superior starting material to other major biomass resources for HAc production. A 38.1% yield of HAc was produced from chitin, which was more than a 2-fold increase compared with that from cellulose. Moreover, a 47.9% yield was directly obtained from crude shrimp shells. Another finding is that heterocyclic compound pyrrole was generated as the major nitrogen-containing (N-containing) product in the reaction system, which offers a potential chemical route for one-step pyrrole formation from a sustainable resource. The study opens new avenues to transform shellfishery waste into platform chemicals.
Choline Chloride-Zinc Chloride Deep Eutectic Solvent Mediated Preparation of Partial O-acetylation of Chitin Nanocrystal in One Step Reaction.

Abstract:An efficient and one step production of acetylated and esterified chitin nanocrystals q(CNCs) was successfully achieved using choline chloride-zinc chloride deep-eutectic solvent q(ChCl-ZnCl2 DES). In this method, ChClZnCl2 DES with a mole ratio of 1: 2, was used for the esterification and O-acetylation of chitin at 90 degrees C for 3 h and 6 h. This strategy consisted in using ChCl-ZnCl2 DES as a green and non-volatile solvent for chitin under heterogeneous condition that, upon addition of acetic anhydride or acetic acid, simultaneous acetylation or esterification and hydrolysis occurred. The DES act as an efficient catalysis of the functional reaction. The acetylation and hydrolysis proceeded efficiently and the yield of partial acetylated CNCs was ca. 61.6% with DS q(degree of substitution) of 0.23 under the optimized conditions. Acetic acid was used to substituted acid anhydride to produce CNCs. The yield of acetic acid induced CNCs was ca. 62.0% with higher DS of 0.34. A thorough investigation of the physicochemical characteristics changes of chitin pointed out that the main skeletal structure of obtained CNCs was intact. The thermal stability of CNCs decreased after treated by ChCl/ ZnCl2. In addition, thermal stability of CNCs functionalized with acetic acid is lower than the ones functionalized with acetic anhydride. DES showed low expenditure and toxicity. This approach provides a novel method for production of functional chitin nanocrystals.
Waste Shell Biorefinery: Sustainable Production of Organonitrogen Chemicals

Abstract:This chapter focuses on sustainable production of organonitrogen chemicals from the concept of a waste shell biorefinery. Chitin waste is naturally nitrogenous and can be converted into small molecule, high value-added organic compounds. In aqueous media or organic solvents/co-solvents, chitin can be depolymerized/deacetylated to acetylglucosamine or glucosamine. In addition, chitin can also be converted to important platform chemicals such as amino acids and furans by oxidation and dehydration. Finally, this chapter introduces conversion strategies for chitin, such as hydrothermal methods, hydrogenation reactions and condensation reactions and demonstrates the types and routes of conversion of chitin to organic nitrogenous chemicals.
Subcritical water hydrolysis of N-acetyl-D-glucosamine: Hydrolysis mechanism, reaction pathways and optimization for selective production of 5-HMF and levulinic acid

Abstract:In this study, the subcritical water hydrolysis of N-acetyl-d-glucosamine (NAG), a monomer of abundant renewable marine biopolymer chitin, for production of value-added chemicals is investigated. The enhanced ionization of water at subcritical conditions (200 °C; 50-100 bar), provides enough acidity for deacetylation of NAG resulting in 80% yield of acetic acid along with traces of formic acid, lactic acid, glucose, fructose, 5-hydroxymethylfurfural (5-HMF),etc. The significant humin formation indicates that the self-condensation and polymerisation of 5-HMF is favored in water at subcritical conditions. A catalyst p-toluenesulfonic acid (p-TsOH) was employed to selectively convert NAG to levulinic acid (LA) via ring opening of 5-HMF previously formed during hydrolysis. The maximum yield of 27.13 ± 1% and 53.46 ± 1% of 5-HMF and LA, respectively, was obtained at process conditions optimized using Box-Behnken design coupled with response surface methodology. Subcritical water enables greener conversion of NAG to platform chemicals wherein the selective production can be achieved by tuning the process conditions.
Design and Experimental Study on Closed-Loop Process of Preparing Chitosan from Crab Shells.

Abstract:The purpose of this article is to design a green and comprehensive utilization process for preparing chitosan from crab shells. Glutamate acid was used as a decalcifying agent for crab shells, and the mixed solution of potassium hydroxide/isopropanol was used for deproteinization and deacetylation to prepare chitosan. Glutamic acid and isopropanol could be recovered for recycling. At the same time, calcium carbonate and protein in crab shells were converted into calcium hydrogen phosphate and compound fertilizer containing nitrogen, phosphorus, and potassium, respectively. The prepared chitosan was characterized by Fourier-transform infrared (FT-IR), differential scanning calorimetry (DSC), x-ray diffraction (XRD), and scanning electron microscopy (SEM), and its deacetylation degree and viscosity average molecular weight were 88.7% +/- 0.68% and 792.1 +/- 10.82 kDa, respectively. The recoveries of glutamic acid and isopropanol were 95.56% +/- 1.39% and 88.14% +/- 1.13%, respectively. The prepared chitosan has large molecular weight and deacetylation degree, controllable production cost, comprehensive utilization of crab shell components, and greatly reduced waste emissions.
Effectively inhibiting the degradation of chitin during extraction from crustacean waste via a novel deep eutectic solvent aqueous solution

Abstract:This study proposed a feasible route for effectively inhibiting degradation of chitin by introducing a novel low-viscosity gluconic acid-based deep eutectic solvents (DESs) aqueous solution. Under the optimal extraction conditions, a high-quality chitin with molecular weight of 3.75 × 105 Da, purity of 94.5 % and yield of 79.1 % was obtained from crab shell. The maximum yield and molecular weight of chitin extracted by DESs were 1.37 times and 3.02 times higher than traditional acid/alkali method, respectively. DESs showed satisfactory recycling performance and the chitin purity could still reach 82.6 % after reusing DESs four times. Mechanism analysis confirmed that introduction of amino acids enhanced the interactions in the DESs system. The main reason for inhibiting degradation of chitin was that the reagents mainly acted on calcium carbonate and protein, without appreciable effect on the structure of chitin. This study provided a green and effective utilization strategy for crustacean waste based on multifunctional DESs systems.

Abstract:The bioconversion of chitin into N-acetyl-d-glucosamine (GlcNAc) using chitinolytic enzymes is one of the important avenues for chitin valorization. However, industrial applications of chitinolytic enzymes have been limited by their poor thermostability. Therefore, it is necessary to discover thermostable chitinolytic enzymes for GlcNAc production from chitin. In this study, two chitinolytic enzyme-encoding genes CaChiT and CaHex from Caldicellulosiruptor acetigenus were identified and heterologously expressed in Escherichia coli. The purified recombinant CaChiT and CaHex showed optimal activities at 70 °C and 90 °C respectively, and exhibited good thermostability over a range of temperature below 70 °C and broad pH stability at pH range of 3.0-8.0. CaChiT and CaHex were active on colloidal chitin, pNP-(GlcNAc)2, pNP-(GlcNAc)3, and pNP-GlcNAc, pNP-(GlcNAc)2, pNP-(GlcNAc)3, pNP-Glc respectively. Besides, the chitin oligosaccharides and colloidal chitin hydrolysis profiles revealed that CaChiT degraded chitin chains through exo-mode of action. Furthermore, CaChiT and CaHex exhibited a synergistic effect in the degradation of colloidal chitin, reaching 0.60 mg/mL of GlcNAc production after 1 h incubation. These results suggested that a combination of CaChiT and CaHex have great potential for industrial applications in the enzymatic production of GlcNAc from chitin-containing biowastes.

Abstract:Deep eutectic solvents (DESs) have been widely applied in biomass extraction due to their low toxicity, low cost, easy degradability, and other excellent characteristics. However, there are few reports on extraction of chitin, an important part of biomass, via DESs method, let alone the study of its related mechanism. Therefore, a novel natural deep eutectic solvents (NADESs) system consisting of lactic acid and choline chloride was proposed for the extraction of chitin. After optimizing the reaction process conditions, the obtained chitin with high purity up to 99.33% was prepared, which was comparable to the results obtained by the conventional method. And analysis results of the aforementioned products showed that the prepared chitin was acylated with a degree of 0.43. Moreover, the potential mechanism in the preparation process was explored. Free H+ in NADESs was confirmed to be the main reason for decalcification. Meanwhile, by constructing covalent bond, the hydrogen bond in NADESs was able to dissolve the protein bound to chitin, and then forming a new NADESs system, which further verified the essence of the acylation phenomenon was dehydration condensation between acid and -OH of chitin. Most importantly, the experiments as well as in-deep theoretical studies provide a directional reference that preparation of chitin from marine sources can be realized by utilizing natural resources.

Abstract:Chitin treatment using different methods, including ball mill grinding, steam explosion, alkaline treatment, phosphoric acid, and ionic liquid (IL) dissolution/reprecipitation have been systematically investigated. The chitin structures were thoroughly investigated by using a series of analytical techniques, and the reactivity after each treatment was evaluated in dehydration and liquefaction reactions. The parallel studies enable direct comparisons of these methods and help to establish the structure-activity correlations. Ball mill grinding in dry mode was the most effective method, with the crystal size and the hydrogen-bond network being the two crucial factors in enhancing the reactivity. Remarkably, the yield of 3-acetamido-5-acetylfuran (3A5AF) from chitin dehydration increased to the highest amount (28.5 %) after ball mill grinding (the previous record yield was 7.5% for untreated chitin).

Abstract:Chitin, a kind of polysaccharide mainly obtained from food waste, has emerged as an important biodegradable biopolymer in composite materials. The difficulty of aldehyde modification, which greatly limited the application of chitin nanocrystals, was addressed by applying a facile route of partial deacetylation followed by periodate oxidation in this study. Deacetylation occurred on the surface of both crystalline and amorphous regions, which were significantly degraded in the following periodate oxidation due to the inevitable cleavage of chitin chains, leading to an increase in the crystallinity index of obtained di-aldehyde chitin. The degree of deacetylation and periodate addition had limited improvement in the aldehyde content of di-aldehyde chitin with a maximum value of around 0.42 mmol/g. With further 2,2,6,6-tetramethylpiperidine-1-oxyl-mediated oxidation, the carboxyl content of tri-carboxyl chitin was improved to 1.58 mmol/g, which played a critical role in the dispersion efficiency and morphology of chitin nanocrystals. The obtained rod-like chitin nanocrystals with a ζ-potential value of −42 mV and an average size of 97 nm have potential application in dye-adsorption and emulsion stabilizers.

Abstract:<p>Recently, the development of solution processing of chitin/chitosan in natural deep eutectic solvents (NADESs) is one of the most promising strategies for handling insoluble biopolymers. A facile and efficient approach including choline chloride-based and betaine-based NADESs, were designed as solvents for chitin/chitosan with various degree of deacetylation (DD). The maximum solubility of chitin/chitosan (10.2 g/100 g) was obtained in NADESs composed of choline chloride-lactic acid (ChLa). The relationships between the solubility, crystallinity index (CrI), and DD of the dissolved chitin/chitosan with different pH values of NADESs were discussed, and the results indicated that DD of chitin/chitosan dominated its solubility. Furthermore, mechanistic analysis showed that the hydrogen-bonding interactions contained NH⋯OH or OH⋯OH between chitin and NADESs was the possible reason for better solubility of chitin in ChLa. The NADESs-chitin processing platform in this text reveals the potential of NADESs for the environmentally-aware research and developing chitin-based industrial processes and products.</p>
