Abstract:While Ag nanoparticles hold great promise for broad spectrum antibacterial activity, the potential risks of Ag nanoparticles (NPs) on human health remain a challenge. In this study, Ag/Fe3 O4 composites have been successfully prepared and characterized by transmission electron microscopy, X-ray powder diffraction, and Fourier-transform infrared spectroscopy, and their magnetic and antibacterial properties have been assessed. In vivo results show that the antibacterial effect of 500 μg/mL Ag/Fe3 O4 nanocomposites was significantly higher than that of 1000 μg/mL AgNPs after 72 h of treatment (p < 0.01). Hematoxylin and eosin (HE) staining showed that squamous epithelium and dermis collagen fibers formed in the Ag/Fe3 O4 group after 8 days treatment. Wound closure was significantly better for the Ag/Fe3 O4 group than for the AgNPs group. On the other hand, there was less Ag in blood, liver, and kidney in the Ag/Fe3 O4 group, as more Ag was retained in the wound. According to lactate dehydrogenase, γ-glutamyl transpeptidase, and reactive oxygen species results, Ag/Fe3 O4 nanocomposites caused less unwanted side-effects. This work presents a new paradigm to reduce the unwanted side-effects of AgNPs and improve their antibacterial activity, providing a new avenue for wound healing. © 2017 Wiley Periodicals, Inc. Abstract:Bacterial infection, complex wound microenvironment and persistent inflammation cause delayed wound healing and scar formation, thereby disrupting the normal function and appearance of skin tissue, which is one of the most problematic clinical issues. Although Ag NPs have a strong antibacterial effect, they tend to oxidize and form aggregates in aqueous solution, which reduces their antibacterial efficacy and increases their toxicity to tissues and organs. Current research on scar treatment is limited and mainly relies on growth factors and drugs to reduce inflammation and scar tissue formation. Therefore, there is a need to develop methods that effectively combine drug delivery, antimicrobial and anti-inflammatory agents to modulate the wound microenvironment, promote wound healing, and prevent skin scarring.

Silver Nanoparticle Loaded Collagen/Chitosan Scaffolds Promote Wound Healing Via Regulating Fibroblast Migration and Macrophage Activation

Abstract:Treatment of full-thickness skin defects poses significant clinical challenges including risk of infection and severe scaring. Silver nanoparticle (NAg), an effective antimicrobial agent, has provided a promising therapeutic method for burn wounds. However, the detailed mechanism remains unknown. Hence, we constructed a metallic nanosilver particles-collagen/chitosan hybrid scaffold (NAg-CCS) and investigated its potential effects on wound healing. In vitro scratch assay, immunofluorescence staining and antibacterial activity of the scaffold were all studied. In vivo NAg-CCS was applied in full-thickness skin defects in Sprague-Dawley (SD) rats and the therapeutic effects of treatment were evaluated. The results showed that NAg at a concentration of 10 ppm accelerated the migration of fibroblasts with an increase in expression of α-smooth muscle actin (α-SMA). Furthermore, in vivo studies showed increased levels of pro-inflammatory and scar-related factors as well as α-SMA, while markers for macrophage activation were up-regulated. On day 60 post transplantation of ultra-thin skin graft, the regenerated skin by NAg-CCS had a similar structure to normal skin. In summary, we demonstrated that NAg-CCS was bactericidal, anti-inflammatory and promoted wound healing potentially by regulating fibroblast migration and macrophage activation, making it an ideal dermal substitute for wound regeneration.
Nanosilver Alleviates Foreign Body Reaction and Facilitates Wound Repair by Regulating Macrophage Polarization

Abstract:Foreign body reactions induced by macrophages often cause delay or failure of wound healing in the application of tissue engineering scaffolds. This study explores the application of nanosilver (NAg) to reduce foreign body reactions during scaffold transplantation. An NAg hybrid collagen-chitosan scaffold (NAg-CCS) was prepared using the freeze-drying method. The NAg-CCS was implanted on the back of rats to evaluate the effects on foreign body reactions. Skin tissue samples were collected for histological and immunological evaluation at variable intervals. Miniature pigs were used to assess the effects of NAg on skin wound healing. The wounds were photographed, and tissue samples were collected for molecular biological analysis at different time points post-transplantation. NAg-CCS has a porous structure and the results showed that it could release NAg constantly for two weeks. The NAg-CCS group rarely developed a foreign body reaction, while the blank-CCS group showed granulomas or necrosis in the subcutaneous grafting experiment. Both matrix metalloproteinase-1 (MMP-1) and tissue inhibitor of metalloproteinase-1 (TIMP-1) were reduced significantly in the NAg-CCS group. The NAg-CCS group had higher interleukin (IL)-10 and lower IL-6 than the blank CCS group. In the wound healing study, M1 macrophage activation and inflammatory-related proteins (inducible nitric oxide synthase (iNOS), IL-6, and interferon-γ (IFN-γ)) were inhibited by NAg. In contrast, M2 macrophage activation and proinflammatory proteins (arginase-1, major histocompatibility complex-II (MHC-II), and found in inflammatory zone-1 (FIZZ-1)) were promoted, and this was responsible for suppressing the foreign body responses and accelerating wound healing. In conclusion, dermal scaffolds containing NAg suppressed the foreign body reaction by regulating macrophages and the expression of inflammatory cytokines, thereby promoting wound healing.
AgNPs loaded adenine-modified chitosan composite POSS-PEG hybrid hydrogel with enhanced antibacterial and cell proliferation properties for promotion of infected wound healing

Abstract:Wound healing is a dynamic and complex process, it's urgent to develop new wound dressings with excellent performance to promote wound healing at the different stages. Here, a novel composite hydrogel dressing composed by silver nanoparticles (AgNPs) impregnated adenine-modified chitosan (CS-A) and octafunctionalized polyhedral oligomeric silsesquioxane (POSS) of benzaldehyde-terminated polyethylene glycol (POSS-PEG-CHO) solution was presented to solve the problem of wound infection. Modification of chitosan with adenine, not only can improve the water solubility of chitosan, but also introduce bioactive substances to promote cell proliferation. CS-A and POSS-PEG-CHO were cross-linked by Schiff-base reaction to form the injectable self-healing hydrogel. On this basis, AgNPs were added into the hydrogel, which endows the hydrogel with better antibacterial activity. Moreover, this kind of hydrogel exhibits excellent cell proliferation properties. Studies demonstrated that the hydrogel can significantly accelerate the closure of infected wounds. The histological analysis and immunofluorescence staining demonstrated that the wounds treated with the composite hydrogel exhibited fewer inflammatory cells, more collagen deposition and angiogenesis, faster regeneration of epithelial tissue. Above all, adenine-modified chitosan composite hydrogel with AgNPs loaded was considered as a dressing material with great application potential for promoting the healing of infected wounds.
Synergistic antibacterial and wound-healing applications of an imidazole-based porous organic polymer encapsulated silver nanoparticles composite

Abstract:Here, a novel antibacterial agent (IM-POP-Ag) through encapsulation of Ag nanoparticles (AgNPs) into an imidazole-based porous organic polymer (IM-POP) was developed for synergistic antibacterial and wound healing. IM-POP with an ultrahigh N content of 38.42 at% was synthesized via the copolymerization of cheap and industrially mass-produced imidazole-2-carbaldehyde and melamine, via catalyst-free Schiff base chemistry, which could realize the effectively anchor of Ag on the porous skeleton (with an Ag loading of 26.01 wt%), avoiding the aggregation and reducing the toxicity of AgNPs greatly. The inherent porosity of POP realizes the effectively control on the size of AgNPs and the coordination of Ag+ changed imidazole into imidazolium, gifting the antibacterial activity to the carriers. As a result, IM-POP-Ag presented a synergistic antibacterial activity against both Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli.). For example, at the same Ag content, the survival rates of E. coli and S. aureus in response to 200 μg mL−1 IM-POP-Ag (equivalent to 50 μg mL−1 AgNPs) were 3.44 ± 0.21% and 6.34 ± 0.40%, respectively, far outperforming that of bare AgNPs (12.19 ± 0.54% and 20.20 ± 0.19%). In vivo experiments on mice testified IM-POP-Ag could significantly promote the healing rate of infected wounds which could facilitate the growth of vessel and hair follicle, accelerating epithelialization. Hence, this novel and mass-produced antibacterial agent provides a new strategy and theoretical basis for the application of AgNPs with higher efficacy and biocompatibility in the near future.

Nanoporous BODIPY-Based Cationic Porous Organic Polymer Composites as Photocontrolled NO-Releasing Platforms for Wound Healing and Antimicrobial Applications

Abstract:Here, an intelligent theranostic therapeutic platform based on a nanoporous ionic porous organic polymer (POP) hybrid (BG-SNP) endowed with photodriven nitric oxide (NO) release capacity was facilely fabricated as a four-mode synergistic antimicrobial agent. The BG-SNP nanoporous composite incorporated the motifs of guanidinium, BODIPY, and sodium nitroprusside (SNP) and, concurrently, could realize the combinational photothermal, photodynamic, cationic, and NO gas antibacterial therapy. This specific hybrid is constructed by in situ ion exchange, in which the negatively charged nitroprusside anion was bonded with the positively charged POP, forming a composite with controllable carbon monoxide generation behavior. Both the release amount and speed are highly controlled in response to photoirradiation. Upon laser irradiation (638 nm, 1.0 W cm–2) for 10 min, a BG-SNP nanoporous composite (250 μg mL–1) could rapidly generate massive quantities of NO, exerting a multimode synergistic antibacterial effect with bacteria killing >99% toward both the Staphylococcus aureus and Escherichia coli. Meanwhile, BG-SNP featured good biosafety according to blood and cell compatibility experiments. In vivo open-wound-healing assay revealed that the BG-SNP nanoporous composite also featured enhanced antibacterial and antiinflammatory capabilities in the infected wounds, thereby prominently accelerating the regeneration of skin tissue and closure of wounds. This unique “all-in-one” POP composite provides a safe but reliable multimode synergistic antibacterial strategy for the rapid repair of infected wounds.

Preparation of nano-silver containing black phosphorus based on quaternized chitosan hydrogel and evaluating its effect on skin wound healing

Abstract:Hydrogels with favorable biocompatibility and antibacterial properties are essential in postoperative wound hemorrhage care, facilitating rapid wound healing. The present investigation employed electrostatic adsorption of black phosphorus nanosheets (BPNPs) and nano‑silver (AgNPs) to cross-link the protonated amino group NH3+ of quaternized chitosan (QCS) with the hydroxyl group of hyaluronic acid (HA). The electrostatic interaction between the two groups resulted in the formation of a three-dimensional gel network structure. Additionally, the hydrogel containing AgNPs deposited onto BPNPs was assessed for its antibacterial properties and effects on wound healing. Hydrogel demonstrated an outstanding drug-loading capacity and could be employed for wound closure. AgNPs loaded on the BPNPs released silver ions and exhibited potent antibacterial properties when exposed to 808 nm near-infrared (NIR) radiation. The ability of the hydrogel to promote wound healing in an acute wound model was further evaluated. The BPNPs were combined with HA and QCS in the aforementioned hydrogel system to improve adhesion, combine the photothermal and antibacterial properties of the BPNPs, and promote wound healing. Therefore, the reported hydrogels displayed excellent biocompatibility and hold significant potential for application in the field of tissue engineering for skin wound treatment.
Simultaneous Biofilm Disruption, Bacterial Killing, and Inflammation Elimination for Wound Treatment Using Silver Embellished Polydopamine Nanoplatform

Abstract:A novel AgNPs and PDAs‐based nano platform (Ag/PDAs) offering broad‐spectrum antibacterial activity and biofilm disruption under 808 nm laser irradiation. Ag/PDAs modulate inflammation by affecting macrophage polarization, promoting tissue healing in biofilm‐infected wounds in mice. Due to the presence of spatial barriers, persistent bacteria, and excessive inflammation in bacteria biofilm‐infected wounds, current nanoplatforms cannot effectively address these issues simultaneously during the therapeutic process. Herein, a novel biomimetic photothermal nanoplatform integrating silver and polydopamine nanoparticles (Ag/PDAs) that can damage biofilms, kill bacterial persisters, and reduce inflammation for wound treatment is presented. These findings reveal that Ag/PDAs exhibit a broad‐spectrum antimicrobial activity through direct damage to the bacterial membrane structure. Additionally, Ag/PDAs demonstrate a potent photothermal conversion efficiency. When combined with near‐infrared (NIR) irradiation, Ag/PDAs effectively disrupt the spatial structure of biofilms and synergistically eradicate the resident bacteria. Furthermore, Ag/PDAs show remarkable anti‐inflammatory properties in counteracting bacterium‐induced macrophage polarization. The in vivo results confirm that the topical application of Ag/PDAs significantly suppress Staphylococcus aureus biofilm‐infected wounds in murine models, concurrently facilitating wound healing. This research provides a promising avenue for the eradication of bacterial biofilms and the treatment of biofilm‐infected wounds.

Abstract:As a broad-spectrum antimicrobial material, ZnO nanoparticles (NPs) offer a novel approach to infected wounds in the clinic; however, it is very necessary to improve the antimicrobial performance of ZnO to satisfy the clinic requirements. In this work, a new antimicrobial hybrid ZnO@PDA/GL13K was prepared by attaching the antimicrobial GL13K peptide on the surface of ZnO NPs via polydopamine (PDA) as a covalent bonding agent, which can enhance the reactive oxygen species (ROS) production and damage the cell wall, showing superior antimicrobial efficacy. Besides, the ZnO@PDA/GL13K hybrid can promote the healing of bacterial infected wounds, with the wound area only remaining 1.8% on day 9, about 2 times smaller than that of the ZnO group. Moreover, ZnO@PDA/GL13K also showed good biocompatibility, and no side effect on normal tissue and organs was found, implying that the antimicrobial hybrid may possess prospective application in biomedical fields.
Hydrogel Loaded with Peptide-Containing Nanocomplexes: Symphonic Cooperation of Photothermal Antimicrobial Nanoparticles and Prohealing Peptides for the Treatment of Infected Wounds

Abstract:Current treatment for chronic infectious wounds is limited due to severe drug resistance in certain bacteria. Therefore, the development of new composite hydrogels with nonantibiotic antibacterial and pro-wound repair is important. Here, we present a photothermal antibacterial composite hydrogel fabricated with a coating of Fe2+ cross-linked carboxymethyl chitosan (FeCMCS) following the incorporation of melanin nanoparticles (MNPs) and the CyRL-QN15 peptide. Various physical and photothermal properties of the hydrogel were characterized. Cell proliferation, migration, cycle, and free-radical scavenging activity were assessed, and the antimicrobial properties of the hydrogel were probed by photothermal therapy. The effects of the hydrogel were validated in a model of methicillin-resistant Staphylococcus aureus (MRSA) infection with full-thickness injury. This effect was further confirmed by changes in cytokines associated with inflammation, re-epithelialization, and angiogenesis on the seventh day after wound formation. The MNPs demonstrated robust photothermal conversion capabilities. The composite hydrogel (MNPs/CyRL-QN15/FeCMCS) promoted keratinocyte and fibroblast proliferation and migration while exhibiting high antibacterial efficacy, effectively killing more than 95% of Gram-positive and Gram-negative bacteria. In vivo study using an MRSA-infected full-thickness injury model demonstrated good therapeutic efficacy of the hydrogel in promoting regeneration and remodeling of chronically infected wounds by alleviating inflammatory response and accelerating re-epithelialization and collagen deposition. The MNPs/CyRL-QN15/FeCMCS hydrogel showed excellent antibacterial and prohealing effects on infected wounds, indicating potential as a promising candidate for wound healing promotion.
Photoactivated Release of Nitric Oxide and Antimicrobial Peptide Derivatives for Synergistic Therapy of Bacterial Skin Abscesses

Abstract:It is of paramount importance to develop novel approaches for combating bacterial resistance and the integration of different antibacterial mechanisms is essential to achieve synergistic bactericidal efficiency while reducing the associated side effects. Herein, amphiphilic antimicrobial copolymers derived from poly-l-lysine (PLL), black phosphorus quantum dots (BPQDs) as near-infrared (NIR) sensitizer, and S-nitrosocysteamine (SNO) as nitric oxide (NO) donor, are assembled into PELI@BPQD-SNO nanoparticles through electrostatic interactions. Amphiphilic copolymers with isopentanyl grafts on PLL at a ratio of 50% achieve an optimal balance between antibacterial activity and hemolysis rate. Photothermal effect of BPQDs leads to NIR-responsive release of NO and the combination with amphiphilic copolymers mutually enhances long-term inhibition of bacterial growth. In an S. aureus-infected subcutaneous abscess model, the bactericidal rate of PELI@BPQD-SNO/NIR treatment reaches nearly 99.6%, which is significantly higher than those without NO release (38%) or amphiphilic copolymers (24%) or NIR irradiation (17%). PELI@BPQD-SNO/NIR treatment shows full recovery of infected wounds, efficient retardation of inflammatory cells, and reconstruction of blood vessels similar to those of healthy skin. Therefore, the electrostatic assembly demonstrates a promising strategy to deliver charged therapeutic agents and the photoactivated release of NO and amphiphilic copolymers achieves synergistic antibacterial efficacy without using any antibiotics.

Abstract:Introduction The efficacy of wound-healing treatments can be significantly enhanced through innovative combination therapies. This research investigates the wound-healing properties of a combination therapy involving silver nanoparticles (AgNPs) synthesized using Delphinium denudatum (Dd), bovine tendon collagen (BTC), and the antibiotic doxycycline (DOX) in Wistar albino rats. Each component has known therapeutic benefits: AgNPs possess antimicrobial properties, BTC aids in tissue regeneration, and DOX is an effective antibiotic. The synergy between these components is hypothesized to enhance wound closure, reduce inflammation, and promote scar-free healing. Methods The synthesis of DdAgNPs was carried out using Dd. The presence of AgNPs was confirmed by ultraviolet-visible (UV-Vis) spectroscopy and high-resolution transmission electron microscopy (HRTEM). The study was conducted on Wistar albino rats following ethical guidelines for animal research. The rats were divided into different groups to receive various treatments: DdAgNPs alone, BTC alone, DOX alone, combinations of two components, and the triple combination of DdAgNPs: BTC: DOX. Wound closure rates, epithelialization, and collagen deposition were monitored and recorded over time. Tissue samples from the wound sites were collected for histological analysis. Hematoxylin and eosin (H&E) staining was used to evaluate epithelialization and overall tissue architecture. Collagen deposition was assessed using Masson's trichrome staining. Additionally, the expression of cyclooxygenase-2 (COX-2) was measured as an indicator of inflammation. Results UV-Vis spectroscopy provided the characteristic surface plasmon resonance peak indicative of AgNPs, while HRTEM revealed the morphology and size of the nanoparticles, showing spherical particles with an average size of 35±10.42 nm. The combination therapy of DdAgNPs: BTC: DOX significantly enhanced wound closure compared to individual and dual-component treatments. This was evidenced by faster epithelialization and increased collagen deposition. The histological analysis showed that the triple combination treatment resulted in more organized tissue architecture and denser collagen fibers. Furthermore, the treatment led to a marked decrease in COX-2 expression, indicating reduced inflammation and potential for lower scar formation. Conclusion The synergistic application of DdAgNPs, BTC, and DOX presents a promising strategy for advanced wound healing and tissue regeneration. The combination therapy not only accelerates wound closure but also enhances the quality of healing by promoting epithelialization and collagen deposition while reducing inflammation. These findings offer a potential pathway for developing effective, scar-free healing solutions, highlighting the benefits of integrating multiple therapeutic agents in wound care.
One-step Construction of Silver Nanoparticles Immersed Hydrogels by Triple-Helix Β-Glucans and the Application in Infectious Wound Healing

Abstract:Hydrogels composed of polysaccharides and silver nanoparticles (AgNPs) are widely recognized for their applications in wound dressings, particularly for healing wounds prone to infection. Traditional methods for preparing AgNPs immersed hydrogels are often complex, costly, and may lead to sustained cytotoxicity. To address these challenges, we developed a biocompatible, one-step green reduction strategy to generate AgNPs within hydrogels using a triple-helix β-glucan (PCPA) derived from Poria cocos, a renowned Chinese traditional herb. PCPA serves as a reducing agent, converting silver ions into AgNPs while its triple-helix conformation prevents AgNPs aggregation. The resulting hydrogel (PAg-G) is injectable and contains uniformly distributed AgNPs. PAg-G exhibits broad-spectrum antimicrobial activity and enhanced bioactivity. The in vivo study on S.aureus-infected SD rats demonstrated that PAg-G can accelerate wound healing within 12 days by down-regulating inflammatory factors such as IL-6 and TNF-α, and up-regulating VEGF and CD31 expression, promoting neovascularization in wound tissues. This innovative one-step construction of AgNPs immersed hydrogels offers a promising approach for the development of antimicrobial hydrogels, especially for treating bacterial-infected wounds.

Abstract:Hydrogels loaded with antimicrobial agents have been widely used for treating infected wound defects. However, hydrogels derived from a porcine dermal extracellular matrix (PADM), containing silver nanoparticles (AgNPs), have not yet been studied. Therefore, we investigated the therapeutic effect of an AgNP-impregnated PADM (AgNP–PADM) hydrogel on the treatment of infected wounds. An AgNP–PADM hydrogel was synthesized by embedding AgNPs into a PADM hydrogel. We examined the porosity, moisture retention, degradation, antibacterial properties, cytotoxicity, antioxidant properties, and ability of the PADM and AgNP–PADM hydrogels to treat infected wounds in animals. The PADM and AgNP–PADM hydrogels were pH sensitive, which made them flow dynamically and solidify under acidic and neutral conditions, respectively. The hydrogels also exhibited porous network structures, satisfactory moisture retention, and slow degradation. Additionally, the AgNP–PADM hydrogel showed a slow and sustained release of AgNPs for at least 7 days without the particle size changing. Thus, the AgNPs exhibited adequate antibacterial ability, negligible toxicity, and antioxidant properties in vitro. Moreover, the AgNP–PADM hydrogel promoted angiogenesis and healed infected skin defects in vivo. The AgNP–PADM hydrogel is a promising bioderived antibacterial material for clinical application to infected wound dressings.

Abstract:Remedying a multidrug-resistant (MDR) bacteria wound infection is a major challenge due to the inability of conventional antibiotics to treat such infections against MDR bacteria. Thus, developing wound dressings for wound care, particularly against MDR bacteria, is in huge demand. Here, we present a strategy in designing wound dressings: we use a small molecule (6-aminopenicillanic acid, APA)-coated gold nanoparticles (AuNPs) to inhibit MDR bacteria. We dope the AuNPs into electrospun fibers of poly(ε-caprolactone) (PCL)/gelatin to yield materials that guard against wound infection by MDR bacteria. We systematically evaluate the bactericidal activity of the AuNPs and wound-healing capability via the electrospun scaffold. APA-modified AuNPs (Au_APA) exhibit remarkable antibacterial activity even when confronted with MDR bacteria. Meanwhile, Au_APA has outstanding biocompatibility. Moreover, an in vivo bacteria-infected wound-healing experiment indicates that it has a striking ability to remedy a MDR bacteria wound infection. This wound scaffold can assist the wound care for bacterial infections.

Abstract:The skin wound-healing under infectious conditions remains challenging owing to the lack of efficient strategies to inhibit drug-resistant bacteria growth and control inflammation. Photothermal therapy shows efficient antimicrobial effects, whereas it generates excessive heat to damage tissue and inflammation to impair tissue regeneration. Herein, we develop the multifunctional gold-based nanoflowers incorporated with photosensitizer (Ce6, for PDT) and anti-inflammatory drug (bromfenac sodium/BS). This allows for a nanosystem to combine the mild-photothermal therapy (mPTT), photodynamic therapy (PDT), and drug-controlled release anti-inflammation therapy for infectious skin regeneration. Upon laser irradiation, the local temperature increased (to a mild temperature of ∼ 45 ℃, mPTT) along with the singly linear oxygen (from PDT) for anti-infection; the release of BS was triggered for anti-inflammation. The multifunctional nanoflowers achieved 99 % antibacterial efficiencies and biofilm inhibition in vitro. They showed good biocompatibility and improved wound-healing in the animal models of subcutaneous abscess and skin wound infected with drug-resistant bacteria. In addition to the antibacterial effect from mPTT and PDT, the nanoflowers regulated the immune microenvironment by controlled releasing BS, inhibiting inflammation and promoting growth factor production, collagen deposition, and angiogenesis to improve skin wound-healing. Therefore, this study provides an advanced nano-system with photo-triggered antimicrobial and anti-inflammation activities, which improves infectious skin tissue regeneration.

Abstract:Hydrogel-based delivery systems have now emerged as a pivotal platform for addressing chronic tissue defects, leveraging their innate capacity to suppress pathogenic infections and facilitate expedited tissue regeneration. In this work, an injectable hydrogel dressing, termed AgNPs-dermal matrix hydrogel (Ag@ADMH), has been designed to expedite the healing process of wounds afflicted with methicillin-resistant Staphylococcus aureus (MRSA), featuring sustained antibacterial efficacy. The synthesis of the hydrogel dressing entailed a self-assembly process of collagen fibers within an acellular dermal matrix to construct a three-dimensional scaffold, encapsulated with plant polyphenol-functionalized silver nanoparticles (AgNPs). The Ag@ADMH demonstrated exceptional biocompatibility, and enables a sustained release of AgNPs, ensuring prolonged antimicrobial activity. Moreover, the in vitro RT-qPCR analysis revealed that compared with ADMH, Ag@ADMH diminish the expression of iNOS while augmenting CD206 expression, thereby mitigating the inflammatory response and fostering wound healing. Especially, the Ag@ADMH facilitated a reduction in M1 macrophage polarization, as evidenced by a significant decrement in the M1 polarization trend and an enhanced M2/M1 ratio in dermal matrix hydrogels laden with AgNPs, corroborated by confocal microscopy and flow cytometry analyses of macrophage phenotypes. The in vivo assessments indicated that Ag@ADMH minimized fibrous capsule formation. In a full-thickness skin defect model of MRSA infection, the formulation significantly attenuated the inflammatory response by reducing MPO and CD68 expression levels, concurrently promoting collagen synthesis and CD34 expression, pivotal for vasculogenesis, thereby accelerating the resolution of MRSA-infected wounds. These attributes underscore the injectable extracellular matrix hydrogel as a formidable strategy for the remediation and regeneration of infected wounds.

Abstract:Infected skin wounds represent a serious health threat due to the long healing process and the risk of colonization by multi-drug-resistant bacteria. Silver nanoparticles (AgNPs) have shown broad-spectrum antimicrobial activity. This study introduces a novel approach to address the challenge of infected skin wounds by employing gellan gum-dopamine (GG-DA) as a dual-functional agent, serving both as a reducing and capping agent, for the in situ green synthesis of silver nanoparticles. Unlike previous methods, this work utilizes a spray-drying technique to convert the dispersion of GG-DA and AgNPs into microparticles, resulting in nano-into-micro systems (AgNPs@MPs). The microparticles, with an average size of approximately 3 μm, embed AgNPs with a 13 nm average diameter. Furthermore, the study explores the antibacterial efficacy of these AgNPs@MPs directly and in combination with other materials against gram-positive and gram-negative bacteria. The versatility of the antimicrobial material is showcased by incorporating the microparticles into injectable hydrogels. These hydrogels, based on oxidized Xanthan Gum (XGox) and a hyperbranched synthetic polymer (HB10K-G5-alanine), are designed with injectability and self-healing properties through Shiff base formation. The resulting nano-into-micro-into-macro hybrid hydrogel emerges as a promising biomedical solution, highlighting the multifaceted potential of this innovative approach in wound care and infection management.
