Exploring Anticancer, Antifungal, Antibacterial, and Antioxidant Properties Colchicum luteum Root-Mediated Silver Nanoparticles with Green Synthesis

Abstract:With growing interest in improving food safety and preservation, the green synthesis of nanoparticles offers an eco-friendly and cost-effective approach to enhance the antimicrobial and antioxidant properties of food-related systems. In this study, silver nanoparticles (Ag NPs) were synthesized using Colchicum luteum root extract ( CLR -mediated Ag NPs) to explore their potential applications in food safety by inhibiting microbial growth and oxidative degradation. The synthesis process was optimized by adjusting silver nitrate concentration (2.5–20 mM), temperature (25–85 °C), and reaction time (5–60 min). The resulting nanoparticles were characterized using various techniques, including UV–Vis, XRD, FT-IR, DLS, zeta potential, EDS, FESEM, and TEM. UV–Vis confirmed the formation of Ag NPs with an absorption peak at ~ 423 nm, while TEM analysis revealed spherical nanoparticles with sizes ranging from 20 to 40 nm. The antimicrobial activity of CLR -mediated Ag NPs was particularly effective against foodborne pathogens, including Enterococcus faecalis , Staphylococcus aureus , Klebsiella pneumoniae , Proteus mirabilis , Escherichia coli , and Candida albicans , with MIC values ranging from 18.75 to 150 μg/mL. Additionally, the antioxidant potential was evaluated using the DPPH assay, where CLR -mediated Ag NPs showed 91% inhibition of DPPH radicals at 600 μg/mL, outperforming the plant extract itself. These findings indicate that CLR -mediated Ag NPs can significantly contribute to food preservation by preventing microbial contamination and oxidative spoilage, making them an asset in the food industry. Furthermore, the nanoparticles exhibited strong anticancer activity against the MCF-7 human breast cancer cell line, with an IC50 value of 16.1 μg/mL, suggesting additional biomedical applications. In conclusion, the green synthesis of silver nanoparticles using Colchicum luteum root extract provides a sustainable solution for enhancing food safety, with promising antimicrobial properties.
