Abstract:Anaerobic ammonium oxidation (Anammox) has been proposed as one of the most promising alternatives to achieve an energy-efficient urban wastewater treatment plant (WWTP). Over the past few decades, anammox-based processes have been successfully implemented in sidestream treatment. However, the evolution of anammox bacteria, cell biology, and new anammox microorganisms is still not clear. The engineering applications of anammox at low temperatures and achieving a promising way to supply nitrite for anammox in mainstream treatments are still limited. Additionally, the function and contribution of anammox bacteria to the nitrogen cycles of different ecosystems need to be determined. In order to promote the application of anammox and to further explore the connection between anammox and other microorganisms, our Research Topic contains articles on the studies of anammox and nitrogen removal. This Research Topic includes five articles that are innovative and interesting and report achievements in the process of anammox and efficient biological nitrogen removal strategies from the perspectives of metabolomics, metagenomics, and microbial ecology. Anammox is an important process in the nitrogen cycle, widely occurring in terrestrial, freshwater, and marine ecosystems. Partial nitritation, as a promising way to support nitrite for anammox, has received a lot of attention in this Research Topic. In order to explore the diversity and function of bacteria involved in nitrogen removal, Xu et al.analyzed the microbiota compositions of different kinds of sludge at a full-scale partial nitritation-anammox (PN/A) plant. This investigation uncovered the partition mechanism of anammox and nitrifiers in a full-scale PN/A plant. It is the first assessment of the microbiota compositions of three different types of sludge in a full-scale sidestream PN/A plant, providing a contribution for guiding the replication and development of fullscale PN/A systems. In this PN/A plant, biofilm sludge exhibited a higher anammox functional activity, and suspended sludge presented a higher partial nitritation activity. The highest numbers of anammox bacteria were detected in biofilm sludge, and ammonia-oxidizing bacteria (AOB) were mainly present in suspended sludge. OPEN ACCESS

Abstract:Anaerobic ammonia oxidation (Anammox) is one of the important discoveries in the field of environmental microbiology, and it plays an indispensible role in the nitrogen removal from wastewaters and the biogeochemical nitrogen cycle. Through review research progress in anaerobic ammonia oxidation, an Anammox special issue is published so as to find problems, explore applications and outlook developments. The special issue consists of reviews and original papers, mainly involving in the following aspects: i) enrichment of Anaerobic ammonia oxidation bacteria (AnAOB); ii) community analysis of AnAOB; iii) preservation of granular AnAOB sludge; iv) effect of organic matter on Anammox; v) application of Anammox process, etc.
The increasing interest of ANAMMOX research in China: bacteria, process development, and application.

Abstract:Nitrogen pollution created severe environmental problems and increasingly has become an important issue in China. Since the first discovery of ANAMMOX in the early 1990s, this related technology has become a promising as well as sustainable bioprocess for treating strong nitrogenous wastewater. Many Chinese research groups have concentrated their efforts on the ANAMMOX research including bacteria, process development, and application during the past 20 years. A series of new and outstanding outcomes including the discovery of new ANAMMOX bacterial species (Brocadia sinica), sulfate-dependent ANAMMOX bacteria (Anammoxoglobus sulfate and Bacillus benzoevorans), and the highest nitrogen removal performance (74.3-76.7 kg-N/m(3)/d) in lab scale granule-based UASB reactors around the world were achieved. The characteristics, structure, packing pattern and floatation mechanism of the high-rate ANAMMOX granules in ANAMMOX reactors were also carefully illustrated by native researchers. Nowadays, some pilot and full-scale ANAMMOX reactors were constructed to treat different types of ammonium-rich wastewater including monosodium glutamate wastewater, pharmaceutical wastewater, and leachate. The prime objective of the present review is to elucidate the ongoing ANAMMOX research in China from lab scale to full scale applications, comparative analysis, and evaluation of significant findings and to set a design to usher ANAMMOX research in culmination.

Abstract:Anaerobic ammonium oxidation (ANAMMOX) is a high-efficiency biotechnology for nitrogen removal from wastewaters under anoxic conditions.It has been successfully applied in ammonium-rich wastewaters,with a high nitrogen removal rate of 9.5 kgN/(m3·d).However,due to the very low growth rate and low cellular yield of ANAMMOX bacteria,the start-up and the operation of ANAMMOX process are difficult.Therefore,the engineering application is limited.In China,the researches about the ANAMMOX process are mainly performed in laboratories,and there is a lack of pilot test and full-scale application of ANAMMOX engineering.Consequently,aiming to apply the full-scale ANAMMOX process,the problems and the corresponding control strategies about the bacteria,process optimizations and equipments of ANAMMOX process are discussed.Based on the discussions,the application mode of ANAMMOX process and the industrialization measures are proposed.
The Anammox Process at Typical Feast-Famine States: Reactor Performance, Sludge Activity and Microbial Community

Abstract:Anaerobic ammonium oxidation (Anammox) is a chemolithotrophic bioprocess which has been widely applied in the treatment of different concentrations of ammonium-containing wastewaters. However, there is less attention on the problem that the instantaneous growth rate (or metabolic rate) and equilibrium growth rate were un-synchronous for anammox bacteria due to their long generation time and self-immobilization of the granular sludge which could lead to the inaccurate estimation. In this study, the anammox process was firstly divided to four typical feast-famine (starvation, satiation, tolerance and poison) states based on the combination of both off-site and in-situ anammox reaction kinetics. Then, four respective lab-scale bioreactors were operated at each state for over a year to achieve stable anammox performance. The results showed that the nitrogen removal rates of bioreactors were 0.53, 2.24, 9.30 and 12.96 kg N/(m(3).d); and the specific anammox activities of granular sludge were 188.94 (48%), 313.29 (79%), 397.50 (100%) and 198.60 (50%) mg N/(g VSS.d) which could reflect the reactivity of each feast-famine state. The stable microbial communities of bioreactors were cultured and analyzed, whose species diversity went down with the decrease of Shannon and ACE index. The relative abundance of anammox bacteria increased from 11% to 57% from starvation to poison state. Candidatus Brocadia/Nitrospira, Candidatus Kuenenia and Brocadiaceae unclassified were revealed to be the distinctive functional bacteria, which could serve as the indicator of each state. The setting up of the typical feast-famine states could be regarded as the landmark to help the design, control and optimization of anammox process.

Abstract:Anammox process is a new biotechnology for nitrogen removal from wastewater. It has become one of the research hotspots and has been put into wide used in wastewater treatment engineering. Anammox reactor based on granular sludge bed is a high-rate bioreactor. The biophase in the form of granular sludge is the key factor that determines the reactor performance. It can be divided into three structures from macroscale to microscale: sludge bed, granular sludge and bacterial cell. The morphological and functional characteristics of three structures and their relation with reactor performance are reviewed so as to give guidance for online optimization and process control.

Abstract:Anaerobic ammonium oxidation (Anammox) is a new and promising biological nitrogen removal technology. It is a burning issue due to its high-rate and cost-effectiveness for nitrogen removal from wastewater. However,Anammox bacteria presents a very long doubling time and low cellular yield,and is susceptible to environmental conditions. Therefore,it is necessary to take careful methods to cultivate Anammox biomass. Based on the characteristics of the Anammox enrichment and the seeding sludge sources,the enrichment techniques are introduced and discussed in this paper. It is proposed that the enrichment course of Anammox culture can be accelerated with useful controlling and enhancing strategies.

Abstract:Anammox process is a new biotechnology for nitrogen removal from wastewaters. It was favored in the field of environmental engineering once it came out and now it has become the upgrading technology. Anammox bacteria (AnAOB) are the function source of Anammox process and Anammox granular sludge (AnGS) formed by AnAOB is the vital pillar of Anammox granular sludge bed system. However, due to the slow growth of AnAOB and their sensitivity to the change of environmental conditions, Anammox nitrogen removal system not only starts up slowly, but runs easily to be unstable and even collapses. Fortunately, AnAOB can select, combine and fix the functional bacterial community freely to form AnGS, thus ensuring the continuous work of the Anammox nitrogen removal system for its excellent gravity settling property and high efficient substrate conversion property. In this paper, the taxonomy and characteristic of AnAOB, compopsition, structure, and function of AnGS are reviewed so as to give guidance for the optimization and expansion of Anammox process.

Abstract:Full-scale anaerobic ammonium oxidation (anammox) engineering applications are vastly limited by the sensitivity of anammox bacteria to the complex mainstream ambience factors. Therefore, it is of great necessity to comprehensively summarize and overcome performance-related challenges in mainstream anammox process at the macro/micro level, including the macroscopic process variable regulation and microscopic biological metabolic enhancement. This article systematically reviewed the recent important advances in the enrichment and retention of anammox bacteria and main factors affecting metabolic regulation under mainstream conditions, and proposed key strategies for the related performance optimization. The characteristics and behavior mechanism of anammox consortia in response to mainstream environment were then discussed in details, and we revealed that the synergistic nitrogen metabolism of multi-functional bacterial genera based on anammox microbiome was conducive to mainstream anammox nitrogen removal processes. Finally, the critical outcomes of anammox extracellular electron transfer (EET) at the micro level were well presented, carbon-based conductive materials or exogenous electron shuttles can stimulate and mediate anammox EET in mainstream environments to optimize system performance from a micro perspective. Overall, this review advances the extensive implementation of mainstream anammox practice in future as well as shedding new light on the related EET and microbial mechanisms.

Performance of ANAMMOX process with different operation strategies based on substrate concentration

Abstract:The performance of ANAMMOX(anaerobic ammonium oxidation) process with different operation strategies was investigated in two lab-scale upflow bioreactors.The results showed that the nitrogen removal rate of 3.78 kg·m-3·d-1 was achieved when the bioreactor was operated at high substrate concentrations with recirculation ratio of 1.07.However,it went up to 25.04 kg·m-3·d-1,which was 6.62 times higher than that above-mentioned,when the bioreactor was operated at relatively low substrate concentrations without recirculation.The operation strategy to run the bioreactor at relatively low substrate concentrations was far better than that to run it at high substrate concentrations.In the operation,ammonia could be a little excess over nitrite since the toxicity of ammonia was weaker than that of nitrite.The nitrite loading rate should be determined according to the capacity of nitrite conversion so as to avoid the nitrite inhibition from the overloading.When nitrogen-containing wastewater with a high concentration is treated,effluent recirculation can alleviate the nitrite inhibition or influent dilution can reduce the nitrite concentrations below the threshold value.
Anammox-based technologies: A review of recent advances, mechanism, and bottlenecks

Abstract:The removal of nitrogen via the ANAMMOX process is a promising green wastewater treatment technology, with numerous benefits. The incessant studies on the ANAMMOX process over the years due to its long start-up and high operational cost has positively influenced its technological advancement, even though at a rather slow pace. At the moment, relatively new ANAMMOX technologies are being developed with the goal of treating low carbon wastewater at low temperatures, tackling nitrite and nitrate accumulation and methane utilization from digestates while also recovering resources (phosphorus) in a sustainable manner. This review compares and contrasts the handful of ANAMMOX -based processes developed thus far with plausible solutions for addressing their respective bottlenecks hindering full-scale implementation. Ultimately, future prospects for advancing understanding of mechanisms and engineering application of ANAMMOX process are posited. As a whole, technological advances in process design and patents have greatly contributed to better understanding of the ANAMMOX process, which has greatly aided in the optimization and industrialization of the ANAMMOX process. This review is intended to provide researchers with an overview of the present state of research and technological development of the ANAMMOX process, thus serving as a guide for realizing energy autarkic future practical applications.

Abstract:Anammox achieves chemoautotrophic nitrogen removal under anaerobic and anoxic conditions and is a low-carbon wastewater biological nitrogen removal process with broad application potential. However, the physiological limitations of AnAOB often cause problems in engineering applications, such as a long start-up time, unstable operation, easily inhibited reactions, and difficulty in long-term strain preservation. Exogenous additives have been considered an alternative strategy to address these issues by retaining microbes, shortening the doubling time of AnAOB and improving functional enzyme activity. This paper reviews the role of carriers, biochar, intermediates, metal ions, reaction substrates, redox buffers, cryoprotectants and organics in optimizing anammox. The pathways and mechanisms of exogenous additives, which are explored to solve problems, are systematically summarized and analyzed in this article according to operational performance, functional enzyme activity, and microbial abundance to provide helpful information for the engineering application of anammox.

Abstract:Abstract Conventional nitrogen removal in wastewater treatment requires a high oxygen and energy input. Anaerobic ammonium oxidation (anammox), the single-step conversion of ammonium and nitrite to nitrogen gas, is a more energy and cost effective alternative applied extensively to sidestream wastewater treatment. It would also be a mainstream treatment option if species diversity and physiology were better understood. Anammox bacteria were enriched up to 80%, 90% and 50% relative abundance, from a single inoculum, under standard enrichment conditions with either stepwise-nitrite and ammonia concentration increases (R1), nitric oxide supplementation (R2), or complex organic carbon from mainstream wastewater (R3), respectively. Candidatus Brocadia caroliniensis predominated in all reactors, but a shift towards Ca. Brocadia sinica occurred at ammonium and nitrite concentrations > 270 mg NH 4 –N L −1 and 340 mg NO 2 –N L −1 respectively. With NO present, heterotrophic growth was inhibited, and Ca. Jettenia coexisted with Ca. B. caroliniensis before diminishing as nitrite increased to 160 mg NO 2 –N L −1 . Organic carbon supplementation led to the emergence of heterotrophic communities that coevolved with Ca. B. caroliniensis. Ca. B. caroliniensis and Ca. Jettenia preferentially formed biofilms on surfaces, whereas Ca. Brocadia sinica formed granules in suspension. Our results indicate that multiple anammox bacteria species co-exist and occupy sub-niches in anammox reactors, and that the dominant population can be reversibly shifted by, for example, changing nitrogen load (i.e. high nitrite concentration favors Ca. Brocadia caroliniensis). Speciation has implications for wastewater process design, where the optimum cell immobilization strategy (i.e. carriers vs granules) depends on which species dominates.

Abstract:The ability of anammox bacteria to utilize organic matter has drawn extensive attention. However, the metabolic discrepancies between autotrophic and mixotrophic anammox consortia need to be further explored. Here, microbial transcript and metabolomic analysis were conducted for the samples harvested in the reactors and batch assays to investigate the phenotype discrepancies and intrinsic causes in autotrophic and mixotrophic anammox consortia. Results showed that metabolically active community structures did not show significant difference between autotrophic and mixotrophic anammox consortia (C/N = 0.3). Changes in the metabolic state were the main cause for those discrepancies in virtue of the added acetate oxidized via the acetyl-CoA pathway by mixotrophic anammox bacteria. At C/N ratio of 0.3, anammox activity was obviously promoted compared to that in the autotrophic condition, due to higher levels of NADH and NAD+, as well as ATP consumption. Mixotrophic anammox consortia were found to yield more biomass, resulting from enhanced purine, pyrimidine, and putrescine synthetic pathways for regulating bacterial growth. Up-regulated amino sugar and nucleotide sugar metabolism pathways participating in regulating more extracellular polysaccharides secreted by mixotrophic anammox consortia. In adverse environment with higher COD concentration, more extracellular proteins were produced by anammox consortia to protect themselves and amino acids also accumulated in the cell. This study provides useful information to catch the optimal metabolism way of anammox consortia and accelerate anammox bacterial cultivation or reactor startup for wastewater treatment.

Abstract:<p>When looking back and wonder how we did it, I became even more aware of how my wanderings in microbiology are all linked, from the start of my PhD with Hans Veldkamp on sulfur oxidizing bacteria in chemostats. My interests broadened from obligate chemolithoautotrophic bacteria to facultative organisms and the question about the ecological niches of these different metabolic types. The sulfide oxidizing bacteria also may be used to produce elemental sulfur which can easily be removed from wastewater. This fitted in a long standing collaboration with Dimitry Sorokin on the ecophysiology and application of alkaliphilic sulfur bacteria. Then came the denitrifying sulfur oxidizing bacteria and their application to remove sulfide from wastewater, which lead to our interest in nitrate, nitrite and ammonium removal in general. The big surprise was the serendipitous discovery of the 'anammox'‐process, whereby ammonium is anaerobically oxidized to dinitrogen gas with nitrite as electron acceptor. The early days of our anammox research are the main focus of this paper which describes the struggle of growing and identifying the most peculiar bacteria we ever came across. A specialized organelle, the anammoxosome was shown to be responsible for the key ammonium oxidation, whereby a rocket fuel, hydrazine, turned out to be an intermediate. Soon after we became aware that anammox is everywhere and in the marine environment makes up a major portion of the nitrogen cycle. The intense scientific collaboration with Mike Jetten and Mark van Loosdrecht and colleagues led to our further understanding and application of this fascinating process which is briefly summarized in this paper. My broader interest in environmental microbiology and microbial ecology has been a regularly returning theme, taking me all over the world to great collaborations lasting to this very day.</p><p>This article is protected by copyright. The Organics-Mediated Microbial Dynamics and Mixotrophic Metabolisms in Anammox Consortia under Micro-Aerobic Conditions.

Abstract:The engineering applications of mainstream anaerobic ammonium oxidation (anammox) have raised increasing attention due to its energy-efficient, however, the organics-mediated microbial dynamics and mixotrophic metabolisms in anammox consortia under micro-aerobic conditions are still elusive. Here, the response of the anammox process to sodium acetate and glucose at a C/N ratio ranging from 0 to 0.5 was investigated under micro-aerobic conditions, respectively. Results showed that the additional glucose could promote the nitrogen removal efficiency (NRE) and nitrogen removal rate (NRR) of anammox processes at a low C/N ratio (0.3), representing 84.00% and 0.53 N kg.m(-3).d(-1). The introduced organics could regulate the diversity of the microbial community and simplify the microbial relationship in anammox consortia. Anammox could not benefit from the introduced sodium acetate, while glucose could effectively enhance the anammox activity and microbial interactions in anammox consortia. Glucose might also stimulate the mixotrophic mechanism of Ca. Kuenenia, further promotes the proliferation of anammox sludge under micro-aerobic conditions. This study reveals that glucose could positively mediate microbial interactions and mixotrophic metabolism in anammox consortia under micro-aerobic conditions, which raises a new horizon for the proliferation of anammox sludge for mainstream engineering applications.
