Abstract:A gold-catalyzed regioselective [3 + 2] cycloaddition of alkynyl triazenes with 1,2,4-dioxazoles was developed. The triazene group in the products could be replaced to obtain iodo-oxazoles, providing potential transformations to diverse oxazole structures. This protocol features readily available starting materials, mild reaction conditions and scalability. A plausible mechanism involving a nitrene transfer process was proposed.

Abstract:Herein we report an electrochemical strategy for the construction of polysubstituted oxazoles from easily available ketones and acetonitrile under room temperature. The method shows high efficiency, a broad substrate scope, and requires no external chemical oxidant. Mechanistic investigations suggest that the reaction proceeds through a Ritter-type reaction/oxidative cyclization using acetonitrile as the reactant and solvent. X-ray crystallographic analysis reveals that the reaction involves the attack of acetonitrile on the carbonyl carbon instead of the α-carbon.

Abstract:Herein, an atom-economical and eco-friendly electrochemical oxidation/cyclization of glycine derivatives through intramolecular Shono-type oxidative coupling is disclosed, leading to a variety of 2-substituted benzoxazoles in 51-85% yields. This oxidative cyclization proceeded in transition metal- and oxidant-free conditions and generated H-2 as only a byproduct. Additionally, gram-scale reactions and a broad substrate scope demonstrated the synthetic usefulness of this protocol.

Abstract:On account of the broad utilities of organophosphorus compounds, the development of highly efficient and concise phosphination methods is significantly important and urgent. Herein, we disclose a novel method for the synthesis of phosphorylated heterocycles: versatile intermediate propargylamines serving as a new type of radical acceptors incorporated in P-radicals via a photocatalytic strategy. This reaction proceeds through a cascade phosphinoylation/ cyclization/oxidation/aromatization pathway using readily available starting materials under mild conditions of light with excellent atom economy, catalyzed by AgOAc or fac-Ir(ppy)(3). One of the phosphorylated quinolines was selected, as an example, as an electron-transporting material for fabricating phosphorescence organic light-emitting diodes displaying excellent electroluminescence performances with a maximum external quantum efficiency of 21.9% with negligible efficiency roll-off ratios.

Abstract:The 1,3,4‐oxadiazole is a widely encountered motif in the areas of pharmaceuticals, materials, and agrochemicals. This work has established a mild, mediated electrochemical synthesis of 2,5‐disubstituted 1,3,4‐oxadiazoles from N‐acyl hydrazones. Using DABCO as the optimal redox mediator has enabled a mild oxidative cyclisation, without recourse to stoichiometric oxidants. In contrast to previous methods, this indirect electrochemical oxidation has enabled a broad range of substrates to be accessed, with yields of up to 83%, and on gram scale. The simplicity of the method has been further demonstrated by the development of a one‐pot procedure, directly transforming readily available aldehydes and hydrazides into valuable heterocycles.

Abstract:We herein report a direct electrochemical dehydrogenative CH phosphonylation of thiazoles derivatives with H2 evolution. Employing electricity as the green and sole oxidant, cheap metal as electrode, the anodic oxidation together with cathodic hydrogen evolution process provides a green and efficient strategy for CH phosphonylation. A diverse range of phosphorus products were constructed under external metal and oxidant-free conditions at ambient temperature, featuring atom economy, simple operation and wide reaction scope.

Abstract:The oxazolidine-2,4-dione motif is found frequently in biologically important compounds. A tandem phosphorus-mediated carboxylative condensation of primary amines and alpha-ketoesters/base-catalyzed cyclization reaction have been developed. These processes provide a novel and convenient access to various oxazolidine-2,4-diones in a one-pot fashion using atmospheric carbon dioxide and readily available substrates under very mild and transition-metal-free conditions.

Abstract:An effective method for the synthesis of benzazoles through the electrochemical oxidative cyclization of o‐aminophenol/o‐aminothiophenol/o‐phenylenediamine and aldehydes was developed. A series of benzazoles were efficiently synthesized by using this method in good yields. Furthermore, this method was applied to the synthesis of drug thiabendazole in a gram‐scale, which proved the practicality of the method. This approach avoids using catalysts and oxidants, and has the advantages of wide substrate range, mild reaction conditions and good tolerance of functional groups. Finally, the possible reaction mechanism was proposed and supported by cyclic voltammetry (CV) and the control experiments.

Abstract:A metal- and oxidizing reagent-free electrochemical dehydrogenative annulation of enamides with O -nucleophiles has been developed, leading to a series of 4-oxo-oxazolines under environmentally friendly conditions. The present method demonstrates broad substrate scope and wide functional group tolerance. In addition to water, alcohols and carboxylic acids were also competent reaction partners. The merit of this electrochemical protocol has also been demonstrated by the gram-scale reaction and the synthesis of 2-(3,4-dimethoxyphenyl)-5-methyl-4,5-dihydrooxazol-4-ol (an antitumor nature product precursor). The mechanistic results suggest that the formation and homolytic cleavage of the N–Br bond is key to the success of this electrochemical dehydrogenative annulation reaction.

Abstract:Background The ubiquitous oxazoles have attracted more and more attention in both industrial and academic fields for decades. This interest arises from the fact that a variety of natural and synthetic compounds which contain the oxazole substructure exhibit significant biological activities and antiviral properties. Although various synthetic methodologies for synthesis of oxazols have been reported, the development of milder and more general procedure to access oxazoles is still desirable. Results In this manuscript, a novel method for synthesis of polysubstituted oxazoles was developed from metal-free decarboxylative cyclization of easily available primary α-amino acids with 2-bromoacetophenones. Conclusions The method was simple, and this reaction could be carried out smoothly under mild and metal-free conditions. By virtue of this method, various polysubstituted oxazoles were obtained from the primary α-amino acids with moderate yields.

Abstract:This review presents an outline of current advancements in the photochemical and electrochemical synthesis of oxazoles and isoxazoles. Oxazole and isoxazole are important building blocks for a variety of medicinally useful compounds. Therefore, the synthesis of these heterocycles via sustainable technologies is worth demanding. Photochemical and electrochemical are two among the few sustainable technologies. This review covers a brief discussion on reaction parameters like catalysts, substrates scope, reaction temperature, solvents, electrodes, electrolytes (in case of electrochemical process), light source (in case of photochemical process) for individual reaction. Detailed discussion on mechanistic insight for each reaction is also presented. Finally, summary, and future direction toward the development of effective electrochemical and photochemical methods for the reaction of oxazoles and isoxazoles is also presented.

Abstract:Pyrazolines and pyrazoles are common and important motifs of pharmaceutical agents and agrochemicals. Herein, the first electrochemical approach for their direct synthesis from easily accessible hydrazones and dipolarophiles up to decagram scale is presented. The application of a biphasic system (aqueous/organic) even allows for the conversion of highly sensitive alkenes, wherein inexpensive sodium iodide is employed in a dual role as supporting electrolyte and mediator. In addition, mechanistic insight into the reaction is given by the isolation of key step intermediates. The relevance of the presented reaction is underlined by the synthesis of commercial herbicide safener mefenpyr-diethyl in good yields.

Abstract:A robust and regioselective copper(II)-catalyzed cycloaddition of internal alkynes and nitrites providing an effective access to oxazole fragments is reported herein. Water substantially participated in this reaction. Control experiments proposed that this transformation may proceed via an enamide intermediate.
