Long Noncoding RNA LTCONS_00091578 Associated with Adipose Triglyceride Lipase (ATGL) Regulates Hepatic Lipolysis in Rainbow Trout

Abstract:Long noncoding RNA (lncRNA) has been considered a novel regulator of lipid homeostasis. However, the function of lncRNA in lipolysis remains largely unillustrated in fish. To explore the effect of lncRNA on adipose triglyceride lipase (ATGL) in fish, we conducted a 10-week feeding experiment in rainbow trout juveniles fed with fish oil (FO), linseed oil (LO), and soybean oil (SO) diets, and performed RNA sequencing. Results showed that the open reading frame of ATGL was 1422 bp and encoded 473 amino acids. Although atgl was widely expressed in all tissues in rainbow trout, the highest expression of atgl was measured in the liver. Gene expression of atgl was markedly higher in juveniles fed with LO and SO diets than in those fed with FO diets. Importantly, we identified an antisense lncRNA LTCONS_00091578 related to ATGL from the liver by RNA sequencing for the first time. LTCONS_00091578 was in the plus strand of chromosome NC_035097.1 and 1769 bp upstream of the atgl gene. Pearson correlation analysis showed that LTCONS_00091578 expression was positively correlated with atgl expression. The transcript of liver LTCONS_00091578 was only expressed in the nucleus and was markedly increased in juveniles fed with LO and SO diets compared with those fed with FO diets. In conclusion, these results revealed that antisense lncRNA LTCONS_00091578 and atgl were involved in hepatic lipolysis. Our research might provide a novel insight into lncRNA function in the lipolysis of fish.
