Knockout of Putative Arsenite Methyltransferase Gene Does Not Alter Arsenic Uptake nor Speciation in the Green Alga Chlamydomonas Reinhardtii

Abstract:In this study, arsenic (As) accumulation, distribution, and speciation in freshwater algae Chlamydomonas reinhardtii, wild type (WT) and mutant (MUT) with putative arsenic methyltransferase knockout treated with 20 μg/L arsenate, were investigated using continuous fractionation and ICP-MS/ESI-MS analyses. The As accumulation was almost identical in WT and MUT strains (p>0.2). Lipid soluble fraction accounted for the majority of intracellular As (WT 51.5%, MUT 59.1%), followed by water extractable fraction (WT 29 %, MUT 16 %). Arsenoribosides (AsSug) were major species in the hydrophilic As compounds, while an arsenoriboside containing phytol (AsSugPhytol) dominated the hydrophobic arsenicals of both C. reinhardtii strains. Together, the findings of this work demonstrate that in C. reinhardtii, As may be detoxified by forming complex hydrophilic and lipophilic As compounds. However, the mutation at gene Cre11.g467612, which was predicted to be required for methylation of As, did not seem to influence the As speciation profile in C. reinhardtii cells.
