Abstract:Enantiomeric analysis is of great significance in chemistry, chemical biology and pharmaceutical research. We herein propose a chiral 19F NMR tag containing an amino reactive NHS group to discriminate the enantiomeric amino acids under physiological conditions by NMR spectroscopy. The chiral 19F NMR tag readily forms stable amide products with the amino acids in aqueous solution. The stereospecific chemistry of enantiomeric amino acids is discriminated by a stereogenic carbon bonded with a 19F atom and is therefore decoded by the 19F reporter and manifested in the distinct 19F chemical shift. The chemical shift difference (ΔΔδ) of the chiral 19F NMR tag derived enantiomeric amino acids variants has a broad chemical shift range between -1.13 to 1.68 ppm, indicating the high sensitivity of the chiral 19F NMR tag to the stereospecific environment surrounding the individual amino acids. The distinguishable chemical shift produced by the chiral 19F NMR tag permits simultaneous discrimination and quantification of the enantiomeric amino acids in a mixture. The high fidelity of the chiral 19F NMR tag affords high-accuracy determination of the enantiomeric composition of amino acids by simple 1D NMR under physiological conditions.

Abstract:Chiral aliphatic amine compounds exhibit a range of physiological activities, making them highly sought-after in the pharmaceutical industry and biological research. One notable obstacle in studying these compounds stems from the pronounced steric hindrance surrounding the nitrogen atom. This characteristic often leads to a weak affinity of acyclic secondary amines for molecular probes, making their chiral discrimination intricate. In response to this challenge, our research has unveiled a novel 19F-labeled probe adept at recognizing and distinguishing between enantiomers of these acyclic secondary amines. By strategically incorporating a single fluorine atom as the 19F label, we have managed to diminish the steric hindrance at the binding site. This alteration bolsters the probe's affinity toward bulkier analytes. As a testament to its effectiveness, we have successfully employed our probe in the chiral analysis of relevant pharmaceuticals, accurately determining their enantiocomposition.

Abstract:Chiral differentiation is critical in diverse fields ranging from pharmaceutics to chiral synthesis. While surface-enhanced Raman scattering (SERS) offers molecule-specific vibrational information with high detection sensitivity, current strategies rely on indirect detection using additional selectors and cannot exploit SERS' key advantages for univocal and generic chiral differentiation. Here, we achieve direct, label-free SERS sensing of biologically important enantiomers by synergizing asymmetric nanoporous gold (NPG) nanoparticles with electrochemical-SERS to generate enantiospecific molecular fingerprints. Experimental and in silico studies reveal that chiral recognition is two pronged. First, the numerous surface atomic defects in NPG provide the necessary localized asymmetric environment to induce enantiospecific molecular adsorptions and interaction affinities. Concurrently, the applied potential drives and orients the enantiomers close to the NPG surface for maximal analyte-surface interactions. Notably, our strategy is versatile and can be readily extended to detect various enantiomers. Furthermore, we can achieve multiplex quantification of enantiomeric ratios with excellent predictive performance. Our combinatorial approach thus offers an important paradigm shift from current approaches to achieve label-free chiral SERS sensing of various enantiomers.

Abstract:Chiral amino-group compounds are of significance for human health, such as biogenic amino acids (AAs), dipeptides, and even various drugs. Enantiospecific discrimination of these chiral compounds is vital in diagnosing diseases, identifying pathological biomarkers and enhancing pharmaceutical chemistry research. Here, we report a simple and rapid 19F NMR-based strategy to differentiate chiral AAs, dipeptides, and amines, that were derivatized with (R)-2-(2-fluorophenyl)-2-hydroxyacetic acid ((R)-2FHA). As a result, 19 proteinogenic AAs (37 isomers) as well as Gly could be concurrently resolved. Moreover, various mirror-image dipeptides, such as Ser-His, Leu-Leu, and Ala-Ala, were commendably recognized. Intriguingly, we found that the absolute configuration of AAs in the N-terminus of dipeptides decided the relative 19F chemical shifts between two enantiomers. Besides, the ability of this method for enantiodiscrimination was further demonstrated by non-AA amines, including aromatic and aliphatic amines, and even amines having chiral centers several carbons away from the amino-group. The structurally similar antibiotics, amoxicillin and ampicillin, were well discriminated. Furthermore, this method accurately determines the de or dr values of non-racemic mixtures. Therefore, our strategy provides an effective approach for 19F NMR-based enantiodiscrimination and diastereomeric purity determination of amino-group compounds.

Abstract:Within pharmaceutical research, ensuring the enantiomeric purity of chiral compounds is critical. Specifically, chiral amines are a crucial category of compounds, due to their extensive therapeutic uses. However, the enantiomeric analysis of these compounds, particularly those with significant steric hindrance, remains a challenge. To address this issue, our research introduces a novel chiral 19F-tagged NNO palladium pincer probe, strategically engineered with an open binding site to accommodate bulky amines. This probe facilitates the enantiodifferentiation of such amines, as evidenced by the distinct 19F NMR signals generated by the enantiomers. Moreover, our findings highlight the probe's applicability in the chiral discrimination of various psychoactive substances, underscoring its potential for the identification of illegal stimulant use and contributing to forensic investigations.

Abstract:Chiral discrimination has played significant roles in food science, pharmaceutical science, medical science, and biology. As such, design of electrochemical chiral sensors with high enantiomeric discrimination ability and service life is highly desirable. Herein, helicoid Au nanoparticles with intrinsic chirality were synthesized and utilized to construct an electrochemical chiral sensor for the first time. Notably, nearly 6 times difference of peak currents was observed for tyrosine (Tyr) enantiomers by differential pulse voltammetry (DPV) method. To the best of our knowledge, it is the best result reported so far in electrochemical enantiomers detection, which is sufficient to enable an accurate determination of the enantiomeric purity and composition of the Tyr analytes. The obtained chiral electrodes were also applied to determine the L-Tyr contents in L-Tyr capsules, beer and milk. These results highlight the tremendous potential of using Au nanoparticles with intrinsic chirality to build chiral recognition interface for enantiomers recognition and offer the promise of practical application of elec-trochemical chiral sensors.

Abstract:Chiral substances occur naturally in abiotic and living systems. The recognition and detection of chiral substances in the natural environment or their analysis and detection in biological systems are crucial. Chiral recognition is a research hotspot in clinical medicine, pharmacology, biochemistry, and other fields. Indeed, many researchers have developed various sensors with different functionalized materials for detecting and analyzing enantiomers. Supramolecular systems have important applications in the development of molecular recognition technologies, and the development of supramolecular chemistry is closely related to research on molecular devices. Therefore, this review summarizes the principle of chiral supramolecular sensors for the detection of enantiomers from the perspective of various sensor types, including optical, electrochemical, electrochemical luminescence, photoelectric, and supramolecular chemical sensors. This review also summarizes the relevant reports on chiral supramolecular sensors in the last five years. Finally, we highlight the prospects of supramolecular chiral sensors in future research.

Abstract:Chirality is ubiquitous in natural world. Although with similar physical and chemical properties, chiral enantiomerscould play different roles in biochemical processes. Discrimination of chiral enantiomers is extremely important in biochemical, analytical chemistry, and pharmaceutical industries. Conventional chiroptical spectroscopic methods are disadvantageous at a limited detection sensitivity because of the weak signals of natural chiral molecules. Recently, superchiral fields were proposed to effectively enhance the interaction between light and molecules, allowing for ultrasensitive chiral detection. Intensive theoretical and experimental works have been devoted to generation of superchiral fields based on artificial nanostructures and their application in ultrasensitive chiral sensing. In this review, we present a survey on these works. We begin with the introduction of chiral properties of electromagnetic fields. Then, the optical chirality enhancement and ultrasensitive chiral detection based on chiral and achiral nanostructures are discussed respectively. Finally, we give a short summary and a perspective for the future ultrasensitive chiral sensing.
Nanoscale chiral imaging under complex optical field excitation with controllable oriented chiral dipole moment

Abstract:Since chirality is a fundamental building block of nature, the identification of the chiral specimen's structure is of great interest, especially in applications involving the modification and utilization of proteins. In this work, by exploiting photoinduced force exerted on an achiral tip placed in the vicinity of a reciprocal chiral sample, a novel technique is proposed to detect the sample's chirality in nanoscale spatial resolution. Under separate excitation of focal field carrying chiral dipole moment with opposite handedness, there is a differential optical force.F exerted on the tip apex, which is connected to the enantiomer type and quasi-linearly depends on specific component of the sample's chirality parameter. With the help of time-reversal approach, we prove that the required excitation can be derived by radiation fields from the superposition of parallel electric and magnetic dipoles. Through adjusting the orientation of the chiral dipole moment, all the diagonal components of the sample's chirality can be exclusively retrieved. In addition, the sensitivity of the proposed technique is demonstrated to enantiospecify nanoscale chiral samples with chirality parameter on the order of 0.001. The proposed technique may open new avenue for wide applications in biomedicine, material science and pharmaceutics. Abstract:The ability of olfaction to distinguish odors is based on many different properties deriving from the molecular structure, including chirality. Even if the electronic nose (e-nose) concept has been widely used in strict analogy with biological systems to implement sensor arrays that recognize and distinguish complex odor matrices, the fabrication of an enantioselective e-nose remains a challenge. This paper introduces an array of quartz microbalances (QMB) functionalized with sensitive materials made of a combination of achiral receptors and silica nanohelices grafted by chiral and achiral porphyrins. In this combination, nanohelices provide a chiral template for the spatial arrangement of porphyrins, while porphyrins act as receptors that can interact differently with analytes. Remarkably, even if single sensors show scarce enantioselectivity, the signals of the overall array achieve recognition of the chiral identity of the five diverse enantiomeric pairs tested when the data are processed with proper multivariate algorithms. Such an innovative and generalizable approach is expected to enable the formation of an extensive library of readily integrable chiral receptors in enantioselective sensor arrays, potentially revolutionizing diverse fields such as agrochemicals, medicine, and environmental sciences.

Abstract:Molecular chirality recognition plays a pivotal role in chiral generation and transfer in living systems and makes important contribution to the development of diverse applications spanning from chiral separation to soft nanorobots. To detect chirality recognition, most of the molecular sensors described to date are based on the design and preparation of the host-guest complexation with chromophore or fluorophore at the reporter unit. Nevertheless, the involved tedious procedures and complicated chemical syntheses hamper their practical applications. Here, we report the plasmonically chiroptical detection of molecular chirality recognition without the need for a chromophore or fluorophore unit. This facile methodology is based on plasmonic nanotransducers that can convert molecular chirality recognitions occurring at nanoscale interfaces into asymmetrically amplified plasmonic circular dichroism readouts, enabling enantiospecific recognition and quantitative determination of the enantiomeric excess of small amino acids. Importantly, such a plasmon-based chirality sensing shows 10<sup>2</sup>-10<sup>3</sup> amplification in the plasmonic circular dichroism signals from the detections of racemate and near-racemate of molecular analysts, demonstrating an extraordinary sensitivity to the host-guest enantioselective interactions. Furthermore, with advantages of easy-processing, cost-effective, and specific to interfacial molecular chirality, our chiroptical sensing scheme could hold considerable promise toward applications of enantioselective high-throughput screening in biology, stereochemistry, and pharmaceutics.

Abstract:Enantioselective identification of chiral molecules holds immense significance in the fields of biological processes, chemical reactions, and drug development due to the optical rotation properties of enantiomers. Moreover, the chiral nanocomposite plays an important role in capable of selectively interacting with specific chiral molecules. By utilizing the chiral surfaces of metal polyphenol framework, an enantioselective identification chiral nanocomposite is fabricated on polyphenol colloidal spheres (PCS). The multivariate metal-based nanocomposite use (L)-tartaric acid (TA) and copper as a chiral recognition selector and signal corrector, aminothiophenol (PATP) anchoring Ag NPs as artificial traction skeleton. Applying (L)-/ (D)-cysteine as patterns enantiomers, the chiral recognition selector Cu- (L)-TA framework perform stereoselective recognition of target molecules through strong affinity and hydrogen interaction. The discriminative SERS signal and CT-related totally symmetric (a1) value of Cu- PCS@Ag NPs and PATP molecules is regarded as discriminating part and internal standard to correct the signal drifting. The put forward method has a good linearity in the concentration range of 1 nM similar to 10 mM and detection limits as low as 0.37 pM and 0.30 pM for L/D-Cys. This work provides specific method for the chiral molecule recognition and determination in complex matrix sample.

Abstract:Chiral recognition, especially rendering specificity in biomolecular recognition, is a basic property of many biomolecules. Being closely related to the chirality of biomolecules, it has been regarded as one of the most important areas in biological and medical sciences due to the different effects in biological systems. Based on the possible interactions between chiral selectors and the enantiomers, various methods including chromatographic techniques such as gas or liquid chromatography, electromigration techniques such as capillary electrophoresis and so on were developed for the chiral separation and recognition of different optical isomers. Recently, chemical sensors and biosensors have been gradually designed and developed for the analysis of chiral compounds. Based on the difference in electrical response to different isomers, chiral identifications can be successfully implemented. Major successes in enantiomer recognition based on electrochemical analysis are reviewed. The research data available for highly enantio -selective recognition are categorized into several subgroups according to specific topics and critically discussed for the period since 1994, and the latest techniques for electrochemical chiral recognition of enantiomers were also reviewed. Simultaneously, a brief conclusive summary and future perspectives are presented, and the challenges and scientific prospects of the newest generation of electrode modifiers in electrochemical sensing are also proposed.

Single-shot Non-destructive Quantum Sensing for Gaseous Samples with Hundreds of Chiral Molecules

Abstract:Chiral discrimination that is efficient to tiny amounts of chiral substances, especially at the single-molecule level, is highly demanded. Here, we propose a single-shot nondestructive quantum sensing method addressing such an issue. Our scheme consists of two steps. In the first step, the two enantiomers are prepared in different rotational states via microwave enantio-specific state transfer. Then, the chiral discrimination is transferred to quantum hypothesis testing. In the second step, we for the first time introduce a non-destructive quantum-state detection technique assisted with a microwave resonator to chiral discrimination, through which the molecular chirality is determined by the sign of the output signals. Using a typical chiral molecule, 1,2-propanediol, and an experimentally feasible model based on spherical Fabry-Pérot cavity, we show that the molecular chirality of slowly moving enantiopure gaseous samples with $10^2 - 10^3$ molecules can be highly credibly distinguished in a single-shot detection. By further trapping chiral molecules, it is promising to achieve chiral discrimination at the single molecule level by using our approach.

Abstract:Non-biaryl atropisomers and their stereochemistry have attracted much attentions in the past years. However, application of the non-biaryl atropisomers as chiral solvating agents is yet to be explored. In this work, four aromatic amide-derived atropisomeric phosphine ligands (hosts) were used as chiral solvating agents to recognize various mandelic acid derivatives (guests) in 1 H nuclear magnetic resonance (NMR) spectroscopy. It is found that chiral center configurations of the four hosts have different effects on the enantiorecognition to the used guests. In addition, the host and guest interaction was further investigated by determination of the host-guest complex stoichiometry using the Job's method and density functional theory calculation, respectively. Moreover, chiral analysis accuracy of these hosts was evaluated through relationship between enantiomeric excess values of 4-chloromandelic acid provided by NMR and gravimetry, respectively.

Abstract:Chirality, as the symmetric breaking of molecules, plays an essential role in physical, chemical and especially biological processes, which highlights the accurate distinction among heterochiralities as well as the precise preparation for homochirality. To this end, the well-designed structure-specific recognizer and catalysis reactor are necessitated, respectively. However, each kind of target molecules requires a custom-made chiral partner and the dynamic disorder of spatial-orientation distribution of molecules at the ensemble level leads to an inefficient protocol. In this perspective article, we developed a universal strategy capable of realizing the chirality detection and control by the external symmetry breaking based on the alignment of the molecular frame to external stimuli. Specifically, in combination with the discussion about the relationship among the chirality (molecule), spin (electron) and polarization (photon), i.e., the three natural symmetry breaking, single-molecule junctions were proposed to achieve a single-molecule/event-resolved detection and synthesis. The fixation of the molecular orientation and the CMOS-compatibility provide an efficient interface to achieve the external input of symmetry breaking. This perspective is believed to offer more efficient applications in accurate chirality detection and precise asymmetric synthesis via the close collaboration of chemists, physicists, materials scientists, and engineers.
Chiral Metafilms and Surface Enhanced Raman Scattering For Enantiomeric Discrimination of Helicoid Nanoparticles

Abstract:Chiral nanophotonic platforms provide a means of creating near fields with both enhanced asymmetric properties and intensities. They can be exploited for optical measurements that allow enantiomeric discrimination at detection levels greater than 6 orders of magnitude than is achieved with conventional chirally sensitive spectroscopic methods based on circularly polarized light. The optimal approach for exploiting nanophotonic platforms for chiral detection would be to use spectroscopic methods that provide a local probe of changes in the near field environment induced by the presence of chiral species. Here we show that surface enhanced Raman spectroscopy (SERS) is such a local probe of the near field environment. We have used it to achieve enantiomeric discrimination of chiral helicoid nanoparticles deposited on left and right-handed enantiomorphs of a chiral metafilm. Hotter electromagnetic hotspots are created for matched combinations of helicoid and metafilms (left-left and right-right), while mismatched combinations leads to significantly cooler electromagnetic hotspots. This large enantiomeric dependency on hotspot intensity is readily detected using SERS with the aid of an achiral Raman reporter molecule. In effect we have used SERS to distinguish between the different EM environments of the plasmonic diastereomers produced by mixing chiral nanoparticles and metafilms. The work demonstrates that by combining chiral nanophotonic platforms with established SERS strategies new avenues in ultrasensitive chiral detection can be opened.
