The Compatible Effects of RNA Interference Targeting Laccase2 with Biocontrol in Henosepilachna Vigintioctopunctata

Abstract:Henosepilachna vigintioctopunctata is a notorious defoliator of many solanaceous and cucurbitaceous plants. The conservation and augmentation of indigenous natural enemies and the application of dsRNA pesticides are two poten-tial environment-friendly pest control strategies for H. vigintioctopunctata. In order to improve the compatibility of the two promising approaches, dsRNA pesticides may be designed as antifeedants and slower-acting insecticides that can cause delayed death to attract and arrest predators and parasitoids. Laccase 2 (Lac2), an enzyme withp-diphenol:dioxygen oxido-reductase activity, exerts a conserved function in cuticle tanning in insects. Independent RNAi experiments were performed using two dsRNA delivery methods of injection and dietary introduction to silence lac2 at the third-and fourth-instar larval stages of H. vigintioctopunctata to detect whether it can cause antifeedant and delayed death. The results revealed that RNAi of Hvlac2 caused untanned cuticles, especially in the head capsule, mouthpart and leg claws. Remarkably, knock-down of Hvlac2 decreased larval fresh weight and inhibited food consumption. As a result, the fat bodies were smaller and the development was compromised. Although depletion of Hvlac2 led to 100% mortality, most of the resultant beetles kept alive for more than a week before death. Taken together, the data will enable to develop a novel control strategy through a combination of dsRNA-based pesticide and biocontrol in H. vigintioctopunctata.
