Abstract:fNIRS was used to collect spontaneous hemodynamic fluctuations in children with ASD and TD children. Based on the HbO/Hb coupling, AUC > 0.80 when the time duration was longer than 1.5 min, but longer than 4 min, AUC value (~0.87) hardly varied, implying the maximal discrimination ability reached. Functional near‐infrared spectroscopy was used to record spontaneous hemodynamic fluctuations form the bilateral temporal lobes in 25 children with autism spectrum disorder (ASD) and 22 typically developing (TD) children. The coupling between oxygenated hemoglobin (HbO) and deoxygenated hemoglobin (Hb) was calculated by Pearson correlation coefficient, showing significant difference between ASD and TD, thus the coupling could be a characteristic feature for ASD. To evaluate the discrimination ability of the feature obtained in different acquisition times, the receiver operating characteristic curve (ROC) was constructed and the area under curve (AUC) was calculated. The results showed AUC > 0.8 when the time duration was longer than 1.5 min, but longer than 4 min, AUC value (~0.87) hardly varied, implying the maximal discrimination ability reached. This study demonstrated the coupling could be one of characteristic features for ASD even acquired in a short measurement time.

Abstract:We review a relatively new method for studying the developing brain in children and infants with Autism Spectrum Disorder (ASD). Despite advances in behavioral screening and brain imaging, due to paradigms that do not easily allow for testing of awake, very young, and socially-engaged children—i.e., the social and the baby brain—the biological underpinnings of this disorder remain a mystery. We introduce an approach based on functional near-infrared spectroscopy (fNIRS), which offers a noninvasive imaging technique for studying functional activations by measuring changes in the brain's hemodynamic properties. This further enables measurement of brain activation in upright, interactive settings, while maintaining general equivalence to fMRI findings. We review the existing studies that have used fNIRS for ASD, discussing their promise, limitations, and their technical aspects, gearing this study to the researcher who may be new to this technique and highlighting potential targets for future research.

Abstract:Hemispheric asymmetry in the power spectrum of low-frequency spontaneous hemodynamic fluctuations has been previously observed in autism spectrum disorder (ASD). This observation may imply a specific narrow-frequency band in which individuals with ASD could show more significant alteration in resting-state functional connectivity (RSFC). To test this assumption, we evaluated narrowband RSFC at several frequencies for functional near-infrared spectroscopy signals recorded from the bilateral temporal lobes on 25 children with ASD and 22 typically developing (TD) children. In several narrow-frequency bands, we observed altered interhemispheric RSFC in ASD. However, in the band of 0.01–0.02 Hz, more mirrored channel pairs (or cortical sites) showed significantly weaker RSFC in the ASD group. Receiver operating characteristic analysis further demonstrated that RSFC in the narrowband of 0.01–0.02 Hz might have better differentiation ability between the ASD and TD groups. This may indicate that the narrowband RSFC could serve as a characteristic for the prediction of ASD.

Abstract:To investigate the relationship between short-range and homotopic long-range resting state functional connectivity (RSFC) in children with autism spectrum disorder (ASD) and typically developing (TD) children, we analyzed functional near-infrared spectroscopy (fNIRS) RSFC in 25 children with ASD and 22 age-matched TD children. The resting state fNIRS signals, including spontaneous fluctuations in the oxygenated hemoglobin (HbO2) and deoxygenated hemoglobin (Hb) concentrations, were recorded from the bilateral temporal lobes. We found that (1) there was no difference in the short-range RSFC between the left and right hemisphere in either ASD or TD group; (2) both the short-range and homotopic long-range RSFC were weaker in the ASD than TD group; and (3) the short-range RSFC was stronger than the homotopic long-range RSFC in the ASD group, whereas no such difference was observed in the TD group. These observations might be helpful for a better understanding of the underlying cortical mechanism in ASD.

Abstract:BACKGROUND:Functional near-infrared spectroscopy (fNIRS) was used to investigate spontaneous hemodynamic fluctuations in the bilateral temporal cortices for typically developing (TD) children and children with autism spectrum disorder (ASD). NEW METHOD:This paper proposed an approach to estimate the global time-varying behavior of brain activity through the measurement on change in first-order statistical properties directly from fNIRS time series. Then, a deep learning model combining the long-short term memory (LSTM) and convolutional neural network (CNN) was constructed based on the integration strategy with improved bagging algorithm, with the purpose to explore the potential patterns of temporal variation for ASD identification. RESULTS:Based on the theory of stationarity, analysis on the global time-varying behavior of hemodynamic fluctuations in oxy-hemoglobin (HbO2) and deoxy-hemoglobin (Hb) demonstrated that children with ASD showed weaker internal logic, but stronger memory and persistence to random shocks than TD children. Differentiating between ASD and TD with the proposed deep learning approach resulted in high accurate classification with sensitivity of 97.1 % and specificity of 94.3 %. COMPARISON WITH EXISTING METHODS:Using fNIRS time series of Hb from single optical channel, we achieved a better classification accuracy of 95.7 % that was about 8 % higher than previous methods with similar data. CONCLUSIONS:The characterization on time-varying behavior of brain activity holds promise for better understanding the underlying causes to ASD. And the deployed deep learning framework with an integration manner has the potential for screening children with risk of ASD.

Abstract:The accurate diagnosis of autism spectrum disorder (ASD) is of great practical significance in clinical practice. The spontaneous hemodynamic fluctuations were collected by functional near-infrared spectroscopy (fNIRS) from the bilateral frontal and temporal cortices of typically developing (TD) children and children with ASD. Since traditional machine learning and deep learning methods cannot make full use of the potential spatial dependence between variable pairs, and require a long time series to diagnose ASD. Therefore, we use adaptive spatiotemporal graph convolution network (ASGCN) and short time series to classify ASD and TD. To capture spatial and temporal features of fNIRS multivariable time series without the pre-defined graph, we combined the improved adaptive graph convolution network (GCN) and gated recurrent units (GRU). We conducted a series of experiments on the fNIRS dataset, and found that only using 2.1 s short time series could achieve high precision classification, with an accuracy of 95.4%. This suggests that our approach may have the potential to detect pathological signals in autism patients within 2.1 s. In different brain regions, the left frontal lobe has the best classification effect, and the abnormalities occur more frequently in left hemisphere and frontal lobe region. Moreover, we also found that there were correlations between multiple channels, which had different degrees of influence on the classification of ASD. From this, we can also generalize to a wider range, there may be potential correlations between different brain regions. This may help to better understand the cortical mechanism of ASD.

Abstract:Autism spectrum disorder is a neuro-developmental disorder characterized by abnormalities of neural synchronization. In this study, functional near infrared spectroscopy (fNIRS) is used to study the difference in functional connectivity in left and right inferior frontal cortices (IFC) and temporal cortices (TC) between autistic and typically developing children between 8-11 years of age. 10 autistic children and 10 typical ones were recruited in our study for 8-min resting state measurement. Results show that the overall interhemispheric correlation of HbO was significantly lower in autistic children than in the controls. In particular, reduced connectivity was found to be most significant in TC area of autism. Autistic children lose the symmetry in the patterns of correlation maps. These results suggest the feasibility of using the fNIRS method to assess abnormal functional connectivity of the autistic brain and its potential application in autism diagnosis.

Abstract:Functional near-infrared spectroscopy (fNIRS) was used to investigate spontaneous hemodynamic activity in the temporal cortex for typically developing (TD) children and children with autism spectrum disorder (ASD). Forty-seven children participated in the experiments including twenty-five with ASD. Compared with TD children, children with ASD showed weaker bilateral resting-state functional connectivity (RSFC), but much stronger fluctuation magnitude in terms of oxy-hemoglobin (HbO2) and deoxy-hemoglobin (Hb). Differentiating between ASD and TD based on a support vector machine (SVM) model including bilateral RSFC and the fluctuation power of HbO2 and Hb as variables could achieve high accurate classification with sensitivity of 81.6% and specificity of 94.6%. This study demonstrates optical brain imaging has the potential for screening children with risk of ASD.

Abstract:Objective. Feature extraction and recognition in brain signal processing is of great significance for understanding the neurological mechanism of autism spectrum disorder (ASD). Resting-state (RS) functional near-infrared spectroscopy measurement provides a way to investigate the possible alteration in ASD-related complexity of resting-state (RS) functional near-infrared spectroscopy (fNIRS) signals and to explore the relationship between brain functional connectivity and complexity. Approach. Using the multiscale entropy (MSE) of fNIRS signals recorded from the bilateral temporal lobes (TLs) on 25 children with ASD and 22 typical development (TD) children, the pattern of brain complexity was assessed for both the ASD and TD groups. Main results. The quantitative analysis of MSE revealed the increased complexity in RS-fNIRS in children with ASD, particularly in the left temporal lobe. The complexity in the RS signal and resting state functional connectivity (RSFC) were also observed to exhibit negative correlation in the medium magnitude. Significance. These results indicated that the MSE might serve as a novel measure for RS-fNIRS signals in characterizing and understanding ASD.

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Objective</h3><p>To classify children with autism spectrum disorder (ASD) and typical development (TD) using short-term spontaneous hemodynamic fluctuations and to explore the abnormality of inferior frontal gyrus and temporal lobe in ASD.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Methods</h3><p>25 ASD children and 22 TD children were measured with functional near-infrared spectroscopy located on the inferior frontal gyrus and temporal lobe. To extract features used to classify ASD and TD, a multi-layer neural network was applied, combining with a three-layer convolutional neural network, a layer of long and short-term memory network (LSTM) and a layer of LSTM with Attention mechanism. In order to shorten the time of data collection and get more information from limited samples, a sliding window with 3.5s width was utilized after comparisons, and numerous short (3.5s) fNIRS time series were then obtained and used as the input of the multi-layer neural network.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3><p>A good classification between ASD and TD was obtained with considerably high accuracy by using a multi-layer neural network in different brain regions, especially in the left temporal lobe, where sensitivity of 90.6% and specificity of 97.5% achieved.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusions</h3><p>The "CLAttention" multi-layer neural network has the potential to excavate more meaningful features to distinguish between ASD and TD. Moreover, the temporal lobe may be worth further study.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Significance</h3><p>The findings in this study may have implications for rapid diagnosis of children with ASD and provide a new perspective for future medical diagnosis.</p>

Neural Biomarkers Distinguish Severe from Mild Autism Spectrum Disorder among High-Functioning Individuals.

Abstract:Several previous studies have reported atypicality in resting-state functional connectivity (FC) in autism spectrum disorder (ASD), yet the relatively small effect sizes prevent us from using these characteristics for diagnostic purposes. Here, canonical correlation analysis (CCA) and hierarchical clustering were used to partition the high-functioning ASD group (i.e., the ASD discovery group) into subgroups. A support vector machine (SVM) model was trained through the 10-fold strategy to predict Autism Diagnostic Observation Schedule (ADOS) scores within the ASD discovery group (r = 0.30, P < 0.001, n = 260), which was further validated in an independent sample (i.e., the ASD validation group) (r = 0.35, P = 0.031, n = 29). The neuroimage-based partition derived two subgroups representing severe versus mild autistic patients. We identified FCs that show graded changes in strength from ASD-severe, through ASD-mild, to controls, while the same pattern cannot be observed in partitions based on ADOS score. We also identified FCs that are specific for ASD-mild, similar to a partition based on ADOS score. The current study provided multiple pieces of evidence with replication to show that resting-state functional magnetic resonance imaging (rsfMRI) FCs could serve as neural biomarkers in partitioning high-functioning autistic individuals based on their symptom severity and showing advantages over traditional partition based on ADOS score. Our results also indicate a compensatory role for a frontocortical network in patients with mild ASD, indicating potential targets for future clinical treatments.

Abstract:To distinguish between children with autism spectrum disorder (ASD) and typically developing (TD) children, we have uncovered a new discriminative feature, hemoglobin coupling. Functional near‐infrared spectroscopy (fNIRS) was used to record resting‐state hemodynamic fluctuations in the bilateral temporal lobes in 25 children with ASD and 22 TD children, in which the coupling between low frequency oxygenated hemoglobin (HbO) and deoxygenated hemoglobin (Hb) fluctuations was evaluated by Pearson correlation coefficient. The results showed significantly weak coupling in children with ASD in both the left and right, and throughout the whole temporal cortex. To explain this observation, a simulation study was performed using a balloon model, in which we found four related parameters could impact the coupling. This study suggested that hemoglobin coupling might be applied as a new cerebral hemodynamic characteristic for ASD screening or diagnostics. This article is protected by copyright. Resting-state Functional Brain Connectivity: Lessons from Functional Near-Infrared Spectroscopy.

Abstract:Resting-state functional near-infrared spectroscopy (R-fNIRS) is an active area of interest and is currently attracting considerable attention as a new imaging tool for the study of resting-state brain function. Using variations in hemodynamic concentration signals, R-fNIRS measures the brain's low-frequency spontaneous neural activity, combining the advantages of portability, low-cost, high temporal sampling rate and less physical burden to participants. The temporal synchronization of spontaneous neuronal activity in anatomically separated regions is referred to as resting-state functional connectivity (RSFC). In the past several years, an increasing body of R-fNIRS RSFC studies has led to many important findings about functional integration among local or whole-brain regions by measuring inter-regional temporal synchronization. Here, we summarize recent advances made in the R-fNIRS RSFC methodologies, from the detection of RSFC (e.g., seed-based correlation analysis, independent component analysis, whole-brain correlation analysis, and graph-theoretical topological analysis), to the assessment of RSFC performance (e.g., reliability, repeatability, and validity), to the application of RSFC in studying normal development and brain disorders. The literature reviewed here suggests that RSFC analyses based on R-fNIRS data are valid and reliable for the study of brain function in healthy and diseased populations, thus providing a promising imaging tool for cognitive science and clinics.

Abstract:Objective: This paper aims to explore whether a single functional near-infrared spectroscopy (fNIRS) channel can adequately distinguish Autism spectrum disorder (ASD) from typically developing (TD) and independently obtain complete feature information, so as to determine the characteristics of ASD. Method: Using the fluctuation entropy of fNIRS from 25 children with ASD and 22 TD children, the pattern of diversity and self-similarity on the time-series was objectively discovered and quantified. By using this characteristic feature and a network cluster classification model consisting of support vector machine (SVM), Gaussian Naive Bayes (GNB), decision tree (DT), multi-layer perceptron (MLP) and logistic regression (LR) to diagnose ASD. Result: The fluctuation entropy of TD is greater than that of ASD. The finding, which comes from a mathematical point of view, may reflect the ASD characteristics of higher self-similarity in the spontaneous activity. The classification between ASD and TD was achieved with an accuracy of 97.8% by using the fNIRS time series from one single optical channel. Conclusion: The fluctuation entropy of fNIRS could accurately distinguish ASD, even using the time-series from a single optical channel. Significance: The fluctuation entropy in this study is a very sensitive index for differentiating between ASD and TD, which might have the penitential to be a fNIRS-based biomarker in the future for the prediction of ASD. In addition, we believe that single channel can provide accurate ASD and TD identification results, which has potential application value in ASD screening in the future. (C) 2020 Elsevier Ltd. Abstract:OBJECTIVES:To assess the possibility of distinguishing autism spectrum disorder (ASD) based on the characteristic of spontaneous hemodynamic fluctuations and to explore the location of abnormality in the brain.METHODS:Using the sample entropy (SampEn) of functional near-infrared spectroscopy (fNIRS) from bilateral inferior frontal gyrus (IFG) and temporal cortex (TC) on 25 children with ASD and 22 typical development (TD) children, the pattern of mind-wandering was assessed. With the SampEn as feature variables, a machine learning classifier was applied to mark ASD and locate the abnormal area in the brain.RESULTS:The SampEn was generally lower for ASD than TD, indicating the fNIRS series from ASD was unstable, had low fluctuation, and high self-similarity. The classification between ASD and TD could reach 97.6% in accuracy.CONCLUSIONS:The SampEn of fNIRS could accurately distinguish ASD. The abnormality in terms of the SampEn occurs more frequently in IFG than TC, and more frequently in the left than in the right hemisphere.SIGNIFICANCE:The results of this study may help to understand the cortical mechanism of ASD and provide a fNIRS-based diagnosis for ASD.

Abstract:<p>Autism spectrum disorder (ASD) patients are often reported altered patterns of functional connectivity (FC) on resting-state functional magnetic resonance imaging (rsfMRI) scans. However, the results in similar brain regions were inconsistent. In this study, we first investigated statistical differences in large-scale resting-state networks (RSNs) on 192 healthy controls (HCs) and 103 ASD patients by using independent component analysis (ICA). Second, an image-based meta-analysis (IBMA) was applied to discover the consistency of spatial patterns from different sites. Last, utilizing these patterns as features, we used Support Vector Machine (SVM) classifier to identify whether a subject was suffering from ASD or not. As a result, six RSNs were obtained with ICA. In each RSN, we identified altered functional connectivity between ASD and HC across the multi-site data. We calculated the area under the receiver operating characteristic curve plots (AUC) to determine the classification performance. The AUC value of classification reaches 0.988. In conclusion, the present study indicates that intrinsic connectivity patterns produced from rsfMRI data could yield a possible biomarker of ASD and contributed to the neurobiology of ASD.</p>

Abstract:Functional near-infrared spectroscopy (fNIRS) is a neuroimaging technique that has been gaining increasing interest as a method to investigate the brain function of individuals on the autism spectrum. It is a non-invasive, portable and relatively motion-tolerant method of measuring haemodynamic activity in the brain. fNIRS can be particularly effective for quantifying brain function in challenging clinical populations. In light of this, there is a growing body of fNIRS literature focusing on individuals on the autism spectrum. The aim of this review is to evaluate and summarise key studies from the literature and discuss their implications for the field. Potential limitations of the fNIRS approach and resolution of these issues based on emerging fNIRS research are also discussed.

Abstract:Objective: Based on functional near-infrared spectroscopy (fNIRS), the use of graph neural networks has become an effective solution for autism spectrum disorder (ASD) identification. However, previous graph neural network studies only considered stable long-term spatial correlations, ignoring the dynamic brain functional connectivity. Meanwhile, most studies only considered one fNIRS attribute, but fNIRS is based on the neurovascular coupling mechanism that requires the co-variation of multiple attributes to be reflected. Methods: We propose a Dynamic Multi-attribute Spatio-temporal Graph Neural Network (DMSTGNN) model to identify ASD. We simultaneously model stable long-term and dynamic short-term spatial correlations in brain functional connectivity. Extract spatial and temporal correlations using graph convolution and dilated inception convolution respectively. Spatio-temporal correlations are extracted from multiple attributes and fused. Results: Data are collected using fNIRS from the bilateral inferior frontal gyrus and temporal lobes of 22 typically developing (TD) and 25 ASD children. Experimental results show achieving 96.8% classification accuracy using only the short-time series of 3.5 s. We find that the inferior frontal gyrus is strongly correlated with ASD children and in bilateral inferior frontal gyrus and temporal lobe, ASD children have weaker resting-state long-range functional connectivity. Conclusion: DMSTGNN can obtain good classification results. The generated spatial graphs can visually reflect the dynamic brain functional connectivity network. Significance: This study may discover the cerebral cortex regions most associated with ASD children and the brain functional connectivity differences between ASD and TD children. These findings provide new perspectives for future ASD diagnosis and pathological explanation.

Abstract:Autism spectrum disorder (ASD) is a neuro-developmental disorder, characterized by impairments in one's capacity for joint attention. In this study, functional near-infrared spectroscopy (fNIRS) was applied to study the differences in activation and functional connectivity in the prefrontal cortex between children with autism spectrum disorder (ASD) and typically developing (TD) children. 21 ASD and 20 TD children were recruited to perform joint and non-joint attention tasks. Compared with TD children, children with ASD showed reduced activation and atypical functional connectivity pattern in the prefrontal cortex during joint attention. The atypical development of left prefrontal cortex might play an important role in social cognition defects of children with ASD.

Abstract:The functional Near-infrared Spectroscopy (fNIRS) has been more and more widely used to measure the activation state of prefrontal cortex when performing function-related tasks among children with various developmental disorders. Children with autism spectrum disorder (ASD) and attention deficit hyperactivity disorder (ADHD) have shown obvious executive function defects. We aimed to summarize the studies with fNIRS, to summarize the activation patterns of prefrontal cortex (PFC) of participants with ASD or ADHD in performing functional tasks. We selected 630 articles according to PRISMA guidelines, and the eligibility criteria were: 6–16 years old individuals diagnosed with ASD or ADHD by DSM-4 or 5, using fNIRS, having executive function (EF) task, typical development (TD) control, and between-group comparison of PFC activation. Nine studies were finally included in the quantitative analysis, and compared to TD, ASD and ADHD showed the opposite PFC activation patterns during n-back tasks. We discussed the task-specific PFC activation in young participants with ASD and ADHD, and provided some new ideas on that issue.
