Abstract:Now, Peng-Fei Xu, Hao Wei and colleagues report a one-pot ring expansion–contraction sequence that enables a carbon-to-nitrogen transmutation in arenols to yield N -heteroarenes (pictured). The researchers started their study by using methylphenanthren-9-ol as the model substrate and diphenylphosphate, N -bromosuccinimide and tetrabutylammonium-azide as reagents to generate the azido ketone intermediate I . The presence of an iron salt was important to catalyse the subsequent ring expansion via formation of a metal−nitrene species that undergoes intramolecular 1,2-aryl migration to generate intermediate II . Moreover, the iron catalyst is also involved in the ring contraction that follows. The latter includes a ring opening step to intermediate III , which goes through cyclization and dehydration to afford the N -heteroarene product. Reaction yields were improved using tricyclohexylphosphine oxide as ligand. Catalysts based on ruthenium, copper, cobalt and rhodium were less effective in operating this transformation. After having established the optimal reaction conditions, the substrate scope was explored, revealing that diverse polycyclic compounds such as naphthol, phenanthrol, anthranol, benzo(a)anthranol, benzo[c]phenanthrol and fused heteroarenols are compatible with this reaction. The elegance of this study lies in the strategic selection of the starting materials, enabling a cascade reaction that combines three transformations in one pot. This provides direct access to nitrogen-containing heterocyclic motifs that are prevalent in many biologically active compounds via swapping of a carbon for a nitrogen atom in the ring system, providing an alternative to the de novo synthesis of resulting complex polycyclic compounds. This synthetic method might also be useful for the investigation of structure–activity relationships in medicinal chemistry.
