Mapping the Endogenous Zn2+ in Situ During Zebrafish Embryogenesis by a Fluorogenic Sensor

Abstract:Zn2+ is one of the most critical micronutrients involved in embryonic development and growth. In situ tracking of the metabolic distribution of Zn2+ during embryogenesis can help to understand its role. However, the lack of practical fluorescence imaging tools hinders the dynamic tracking of Zn2+ changes and distribution in vivo. Here, we report a fluorogenic sensor BDA-1 based on BODIPY fluorophore, which can continuously monitor the changes and distribution of Zn2+ and achieve fluorescence mapping of labile Zn2+ in different organs during zebrafish embryonic development. The sensor BDA-1 itself exists as a tautomer and exhibits non-fluorescence due to the photo-induced electron transfer (PeT) process, while after specific binding to Zn2+, the product no longer exists as a tautomer and the fluorescence is significantly enhanced. The reversible behavior of fluores-cence can be demonstrated in living cells and zebrafish. Furthermore, BDA-1 successfully mapped the distri-bution and changes of Zn2+ at key stages of embryonic development. The sensor is expected to serve as a powerful tool to study Zn2+-related biological processes and promote the understanding of Zn2+ functions in the course of embryonic development.
