Abstract:Mechanoreceptors of the insect play a vital role for the insect to sense and monitor the environmental parameters, like flow, tactile pressure, etc. This paper presents the studies made on the morphology of the mechanoreceptor of the insect Blattella asahinai (scientific name of cockroach) that is a hair-like structure known as trichoid sensilla, by scanning electron microscope and confocal laser microscope. The scanned images show the details of sensilla components in which the hair is embedded in the sockets, which are connected with the cuticle and joint membrane, where the dendrite touches at the base of the hair passing through the cuticle layers. The images also show that the tubular bodies and microtubules are tightly compacted inside the dendrite. This paper presents the details of how the sensilla work when an external stimulus act on them. The hair deflects with the disturbance of the cuticle and joint membrane, and this deformed hair leans on the dendrite, which is attached at the base of the hair that in turn presses the tubular bodies and microtubules, which develop negative ions passing down through the dendrite to the neuron, which provides information as an electric signal to the brain of the insect so that it responds for necessary action. Based on the morphological studies, sensing mechanism, material properties of the components, and design principles will be evolved for the development of an artificial bio-inspired sensor. A solid works model of the sensilla is also presented.
Identification and Characterization of Chemosensory Genes in the Antennal Transcriptome of Spodoptera Exigua

Abstract:Chemical senses are crucial for insect behaviors such as host preference, mate choice and oviposition site selection. Various protein families are involved in these processes, including odorant receptors (ORs), ionotropic receptors (IRs), gustatory receptors (GRs), chemosensory proteins (CSPs), odorant binding proteins (OBPs) and sensory neuron membrane proteins (SNMPs). To better understand the olfactory mechanism in Spodoptera exigua, we conducted transcriptome analysis of adult antennae and identified a total of 157 candidate chemosensory genes encoding 51 ORs, 20 IRs, 7 GRs, 32 CSPs, 45 OBPs and 2 SNMPs. Quantitative real time PCR (qPCR) analysis of the tissue- and sex-specific expression profiles of ORs, GRs and IRs revealed that these genes could be detected in at least one tissue tested. SexiOR6, 11, 13 and 16, which were predicted to be pheromone receptors based on phylogenetic analysis, exhibited male-specific antennae expression. SexiOR18, 26, 28, 30, 34, 39, and 40 exhibited female-biased expression. SexiGR1, SexiGR2 and SexiGR3, are predicted carbon dioxide receptors, and the former was expressed specifically in antennae, and the latter two were expressed both in antennae and labial palps. SexiIRs had diverse expression profiles. SexiIR8a and SexiIR25a were quite conserved and expressed at high levels. This work will greatly facilitate the understanding of olfactory system in S. exigua and provides valuable information for further functional studies of the chemoreception mechanism in Lepidopteran moths.
Olfactory Optogenetics: Light Illuminates the Chemical Sensing Mechanisms of Biological Olfactory Systems

Abstract:The mammalian olfactory system has an amazing ability to distinguish thousands of odorant molecules at the trace level. Scientists have made great achievements on revealing the olfactory sensing mechanisms in decades; even though many issues need addressing. Optogenetics provides a novel technical approach to solve this dilemma by utilizing light to illuminate specific part of the olfactory system; which can be used in all corners of the olfactory system for revealing the olfactory mechanism. This article reviews the most recent advances in olfactory optogenetics devoted to elucidate the mechanisms of chemical sensing. It thus attempts to introduce olfactory optogenetics according to the structure of the olfactory system. It mainly includes the following aspects: the sensory input from the olfactory epithelium to the olfactory bulb; the influences of the olfactory bulb (OB) neuron activity patterns on olfactory perception; the regulation between the olfactory cortex and the olfactory bulb; and the neuromodulation participating in odor coding by dominating the olfactory bulb. Finally; current challenges and future development trends of olfactory optogenetics are proposed and discussed.

Abstract:Insects use olfaction to detect ecologically relevant chemicals in their environment. To maintain useful responses over a variety of stimuli, olfactory receptor neurons are desensitized to prolonged or high concentrations of stimuli. Depending on the timescale, the desensitization is classified as short-term, which typically spans a few seconds; or long-term, which spans from minutes to hours. Compared with the well-studied mechanisms of desensitization in vertebrate olfactory neurons, the mechanisms underlying invertebrate olfactory sensitivity regulation remain poorly understood. Recently, using a large-scale functional screen, a conserved critical receptor phosphorylation site has been identified in the model insect Drosophila melanogaster, providing new insight into the molecular basis of desensitization in insects. Here, we summarize the progress in this area and provide perspectives on future directions to determine the molecular mechanisms that orchestrate the desensitization in insect olfaction.

Abstract:Background Most animals, including humans and insects, consume foods based on their senses. Feeding is mostly regulated by taste and smell. Recent insect studies shed insight into the cross-talk between taste and smell, sweetness and temperature, sweetness and texture, and other sensory modality pairings. Five canonical tastes include sweet, umami, bitter, salty, and sour. Furthermore, other receptors that mediate the detection of noncanonical sensory attributes encoded by taste stimuli, such as Ca 2+ , Zn 2+ , Cu 2+ , lipid, and carbonation, have been characterized. Deorphanizing receptors and interactions among different modalities are expanding the taste field. Methods Our study explores the taste system of Drosophila melanogaster and perception processing in insects to broaden the neuroscience of taste. Attractive and aversive taste cues and their chemoreceptors are categorized as tables. In addition, we summarize the recent progress in animal behavior as affected by the integration of multisensory information in relation to different gustatory receptor neuronal activations, olfaction, texture, and temperature. We mainly focus on peripheral responses and insect decision-making. Conclusion Drosophila is an excellent model animal to study the cellular and molecular mechanism of the taste system. Despite the divergence in the receptors to detect chemicals, taste research in the fruit fly can offer new insights into the many different taste sensors of animals and how to test the interaction among different sensory modalities.

Abstract:Insect biogenic amines play important roles in mediating behavioral and physiological processes as neurotransmitters, neuromodulators and neurohormones. They activate specific G-protein coupled receptors on the cell surfaces to induce downstream signaling pathways. These biogenic amine receptors show different pharmacological properties from their vertebrate counterparts and therefore become potential targets for pest control. However, their physiological functions are not well understood. We summarize our recent progress on insect biogenic amine receptors, and emphasize the characterizations of novel receptors and their functions in behavior and immunity.

The Power of Drosophila Genetics in Studying Insect Toxicology and Chemical Ecology

Abstract:Insect toxicology and chemical ecology are inherently interconnected disciplines, both dedicated to unraveling the intricate relationships between insects and the diverse array of chemical compounds that pervade their surroundings. Drosophila melanogaster , owing to its genetic and physiological similarities to other insects, serves as a robust model system in the study of insect toxicology. Moreover, state-of-the-art techniques in Drosophila neurobiology have extensively probed the chemosensory system of insects, providing significant insights into their adaptation to chemical environments. In this review, we emphasize the advancements achieved through the application of Drosophila genetics in investigations spanning both of these fields, significantly enhancing our understanding of the mode of action and resistance mechanisms of insecticides, as well as unraveling the molecular and cellular mechanisms underlying insect chemosensation and associated behaviors. The profound insights derived through this tiny fly not only enrich our understanding of the broader world of insects but also hold the potential to develop more effective and sustainable strategies for pest management.

Abstract:Nociception refers to the process of encoding and processing noxious stimuli, which allow animals to detect and avoid potentially harmful stimuli. Several types of stimuli can trigger nociceptive sensory transduction, including thermal, noxious chemicals, and harsh mechanical stimulation that depend on the corresponding nociceptors. In view of the high evolutionary conservation of the mechanisms that govern nociception from Drosophila melanogaster to mammals, investigation in the fruit fly Drosophila help us understand how the sensory nervous system works and what happen in nociception. Here, we present an overview of currently identified conserved genetics of nociception, the nociceptive sensory neurons responsible for detecting noxious stimuli, and various assays for evaluating different nociception. Finally, we cover development of anti-pain drug using fly model. These comparisons illustrate the value of using Drosophila as model for uncovering nociception mechanisms, which are essential for identifying new treatment goals and developing novel analgesics that are applicable to human health.

Abstract:The neural substrates that the fruitfly Drosophila uses to sense smell, taste and light share marked structural and functional similarities with ours, providing attractive models to dissect sensory stimulus processing. Here we focus on two of the remaining and less understood prime sensory modalities: graviception and hearing. We show that the fly has implemented both sensory modalities into a single system, Johnston's organ, which houses specialized clusters of mechanosensory neurons, each of which monitors specific movements of the antenna. Gravity- and sound-sensitive neurons differ in their response characteristics, and only the latter express the candidate mechanotransducer channel NompC. The two neural subsets also differ in their central projections, feeding into neural pathways that are reminiscent of the vestibular and auditory pathways in our brain. By establishing the Drosophila counterparts of these sensory systems, our findings provide the basis for a systematic functional and molecular dissection of how different mechanosensory stimuli are detected and processed.
Recent Insights into Insect Olfactory Receptors and Odorant-Binding Proteins

Abstract:Human and insect olfaction share many general features, but insects differ from mammalian systems in important ways. Mammalian olfactory neurons share the same overlying fluid layer in the nose, and neuronal tuning entirely depends upon receptor specificity. In insects, the olfactory neurons are anatomically segregated into sensilla, and small clusters of olfactory neurons dendrites share extracellular fluid that can be independently regulated in different sensilla. Small extracellular proteins called odorant-binding proteins are differentially secreted into this sensillum lymph fluid where they have been shown to confer sensitivity to specific odorants, and they can also affect the kinetics of the olfactory neuron responses. Insect olfactory receptors are not G-protein-coupled receptors, such as vertebrate olfactory receptors, but are ligand-gated ion channels opened by direct interactions with odorant molecules. Recently, several examples of insect olfactory neurons expressing multiple receptors have been identified, indicating that the mechanisms for neuronal tuning may be broader in insects than mammals. Finally, recent advances in genome editing are finding applications in many species, including agricultural pests and human disease vectors.

Non-synaptic ion channels in insects--basic properties of currents and their modulation in neurons and skeletal muscles

Abstract:Insects are favoured objects for studying information processing in restricted neuronal networks, e.g. motor pattern generation or sensory perception. The analysis of the underlying processes requires knowledge of the electrical properties of the cells involved. These properties are determined by the expression pattern of ionic channels and by the regulation of their function, e.g. by neuromodulators. We here review the presently available knowledge on insect non-synaptic ion channels and ionic currents in neurons and skeletal muscles. The first part of this article covers genetic and structural informations, the localization of channels, their electrophysiological and pharmacological properties, and known effects of second messengers and modulators such as neuropeptides or biogenic amines. In a second part we describe in detail modulation of ionic currents in three particularly well investigated preparations, i.e. Drosophila photoreceptor, cockroach DUM (dorsal unpaired median) neuron and locust jumping muscle. Ion channel structures are almost exclusively known for the fruitfly Drosophila, and most of the information on their function has also been obtained in this animal, mainly based on mutational analysis and investigation of heterologously expressed channels. Now the entire genome of Drosophila has been sequenced, it seems almost completely known which types of channel genes--and how many of them--exist in this animal. There is much knowledge of the various types of channels formed by 6-transmembrane--spanning segments (6TM channels) including those where four 6TM domains are joined within one large protein (e.g. classical Na+ channel). In comparison, two TM channels and 4TM (or tandem) channels so far have hardly been explored. There are, however, various well characterized ionic conductances, e.g. for Ca2+, Cl- or K+, in other insect preparations for which the channels are not yet known. In some of the larger insects, i.e. bee, cockroach, locust and moth, rather detailed information has been established on the role of ionic currents in certain physiological or behavioural contexts. On the whole, however, knowledge of non-synaptic ion channels in such insects is still fragmentary. Modulation of ion currents usually involves activation of more or less elaborate signal transduction cascades. The three detailed examples for modulation presented in the second part indicate, amongst other things, that one type of modulator usually leads to concerted changes of several ion currents and that the effects of different modulators in one type of cell may overlap. Modulators participate in the adaptive changes of the various cells responsible for different physiological or behavioural states. Further study of their effects on the single cell level should help to understand how small sets of cells cooperate in order to produce the appropriate output.
["Gating-Spring" Model and Molecular Basis of Mechanotransduction in Drosophila Melanogaster].

Abstract:The sense of mechanical stimuli (e.g. force or deformation) in the environment underlies several important physiological processes, for example the perception of sound, touch, pain and acceleration. The key step in mechanosensation is to convert the extracellular mechanical stimuli into cellular electrical or chemical signals. This process is termed as mechanotransduction. Based on mechanical and electrophysiological measurements, "Gating-Spring" theory was proposed as a general model to describe the cell biological mechanism of mechanotransduction. However, despite efforts made in several model organisms, the molecular basis of the "Gating-Spring" model remains elusive. In recent years, several key progresses have been made using the mechanoreceptors of Drosophila melanogaster as the models. This article introduces the "Gating-Spring" theory and reviews the recent research progresses on the fly mechanotransduction.

Abstract:Olfaction is a critical sensory modality for invertebrates, and it mediates a wide range of behaviors and physiological processes. Like most living organisms, insects live in rhythmic environments: the succession of nights and days is accompanied by cyclic variations in light intensity and temperature, as well as in the availability of resources and the activity of predators. Responding to olfactory cues in the proper temporal context is thus highly adaptive and allows for the efficient allocation of energy resources. Given the agricultural or epidemiological importance of some insect species, understanding olfactory rhythms is critical for the development of effective control strategies. Although the vinegar fly Drosophila melanogaster has been a classical model for the study of olfaction and circadian rhythms, recent studies focusing on non-model species have expanded our understanding of insect olfactory rhythms. Additionally, recent evidence revealing receptor co-expression by sensory neurons has brought about an ongoing paradigm shift in our understanding of insect olfaction, making it timely to review the state of our knowledge on olfactory rhythms and identify critical future directions for the field. In this Review, we discuss the multiple biological scales at which insect olfactory rhythms are being analyzed, and identify outstanding questions.

Functional Interaction Between Drosophila Olfactory Sensory Neurons and Their Support Cells

Abstract:Insects detect volatile chemicals using antennae, which house a vast variety of olfactory sensory neurons (OSNs) that innervate hair-like structures called sensilla where odor detection takes place. In addition to OSNs, the antenna also hosts various support cell types. These include the triad of trichogen, tormogen, and thecogen support cells that lie adjacent to their respective OSNs. The arrangement of OSN supporting cells occurs stereotypically for all sensilla and is widely conserved in evolution. While insect chemosensory neurons have received considerable attention, little is known about the functional significance of the cells that support them. For instance, it remains unknown whether support cells play an active role in odor detection, or only passively contribute to homeostasis, e.g., by maintaining sensillum lymph composition. To investigate the functional interaction between OSNs and support cells, we used optical and electrophysiological approaches in Drosophila . First, we characterized the distribution of various supporting cells using genetic markers. By means of an ex vivo antennal preparation and genetically-encoded Ca 2+ and K + indicators, we then studied the activation of these auxiliary cells during odor presentation in adult flies. We observed acute responses and distinct differences in Ca 2+ and K + fluxes between support cell types. Finally, we observed alterations in OSN responses upon thecogen cell ablation in mature adults. Upon inducible ablation of thecogen cells, we notice a gain in mechanical responsiveness to mechanical stimulations during single-sensillum recording, but a lack of change to the neuronal resting activity. Taken together, these results demonstrate that support cells play a more active and responsive role during odor processing than previously thought. Our observations thus reveal that support cells functionally interact with OSNs and may be important for the extraordinary ability of insect olfactory systems to dynamically and sensitively discriminate between odors in the turbulent sensory landscape of insect flight.

Abstract:Insects have sophisticated olfactory systems that enable them to detect and respond to complex exogenous chemical cues. The encoding mechanisms of these chemical signals have been studied both in their peripheral and central nervous systems (CNS). While many neuromodulators have been shown to play significant roles in olfactory processing within the antennal lobes of the brain, their roles in peripheral olfactory sensory systems, such as the antennae, are less understood. This review focuses on the role of serotonin (5-HT) receptor in the locust antenna, specifically the modulatory function of the serotonin receptor2 on odour inputs. We also review recent studies on the modulation of olfaction in the peripheral nervous systems of other insects and discuss potential directions for future research on the role of neuromodulators in insect peripheral olfactory systems.
