Unraveling Sulfur, Nitrogen Metabolism and Microbial Community Diversity in a High-Temperature Offshore Oilfield

Abstract:The diversity and composition of microbial communities are important for effective strategies of oil production, operational, and environmental issues. Microorganisms are a significant key involved and drove metabolism, especially sulfur (S) and nitrogen (N) metabolism, but related studies are still limited. Here, combined 16S rDNA gene amplicon pyrosequencing and quantitative real-time PCR analysed the abundance of bacteria, archaea, the gene related to S (dsrA, dsrB, aprA and sqr gene) and N (narG, narH and nosZ genes) metabolism in produced water sample form a high-temperature offshore oilfield. The bacteria were dominated by Proteobacteria, Firmicutes and Thermotogae at the phylum level, the archaea were predominated Euryarchaeota and Nanoarchaeaeota in all samples. At the genus level, the Thermodesulforhabdus, Desulfonauticus, Candidatus_Desulforudis and Thermodesulfobacterium related to sulfate reducing bacteria (SRB) and thermophilic bacteria (Thermolithobacter, Thermovirga and Thermosipho) were prevalent, and the preponderant archaea were Methanogen (Methermicoccus, Methanothermobacter, Methanohalophilus and Methanosaeta), Woesearchaeia, Thermococcus and Archaeoglobus. Most of them were thermophile and directly or indirectly involved in sulfate reduction, and the abundance of archaea was higher than bacteria that opposite of previous studies. The numbers of SRB genes (dsrA, dsrB and aprA) had the similar trend with the abundance of microbial communities, were richer abundance in A03L, A03S and A05 samples, and the gene related to N (narG, narH and nosZ genes) metabolism was higher counts in A09, A08, B0 and A02 samples. This work provides a detailed data for unravelling the diversity and composition of bacteria and archaea, the abundance of gene related to S and N metabolism, and suitable management of souring and corrosion in offshore petroleum reservoir.
