Functional Characterization of Powdery Mildew Resistance-Related Genes HbSGT1a and HbSGT1b in Hevea Brasiliensis Muell. Arg.

Abstract:SGT1 is an essential factor required for R proteins to trigger disease resistance and defense responses in plants. Although its vital role have been thoroughly studied in model plant species, so far no SGT1 gene in rubber tree has been identified. In this study, HbSGT1a and HbSGT1b from rubber trees were cloned. The amino acid sequences identities between HbSGT1a and HbSGT1b is 64.82%, and both of them have conserved TPR, CS and SGS domains, which are the characteristic domains of SGT1 proteins. Expression analysis showed that the transcripts of HbSGT1b were more abundant than HbSGT1a in all tissues. Moreover, transcripts of both HbSGT1a and HbSGT1b were triggered by powdery mildew attack in rubber trees. Additionally, transcripts of HbSGT1a and HbSGT1b were significantly induced by exogenous phytohormones and abiotic stress conditions. Yeast two-hybrid assays demonstrated that both HbSGT1a and HbSGT1b interacted with a heat shock protein 90 (HbHSP90.1). Interestingly, HbSGT1a and HbSGT1b showed a strong interaction. Taken together, these results suggested that HbSGT1 and HbHSP90.1 form a functional complex and carry out essential roles in powdery mildew and stress resistance, as well as hormone signaling in rubber tree.
