Abstract:A novel butyrate-producing strain was isolated from cow dung and identified as Clostridium butyricum SCUT620. The product inhibition and substrate spectrum tests indicated that SCUT620 could produce 10.2-13.2 g/L butyrate from a broad range of sugars due to its high butyrate tolerance (up to 20 g/L). Surprisingly, SCUT620 was able to convert raw cassava starch to butyrate directly. In addition, the optimum fermentation parameters were determined to be 37 degrees C, 5 g/L yeast extract, 80 g/L raw cassava starch, and pH 6.0. After cheap nitrogen source screening, the butyrate concentration achieved the highest value (21.72 g/L) reported so far for C. butyricum in batch fermentation using raw cassava starch and fish meal. Techno-economic analysis indicated the promise of using C. butyricum SCUT620 for butyric acid production from raw starchy substrates. For the first time, consolidated bioprocessing of starchy biomass was reported for butyrate production by C. butyricum under unsterilized conditions.
