Visual Investigation of the Effects of Pfoa on Phospholipid in the Zebrafish (danio Rerio) Tissues Level Using Maldi-Tof-Ims

Abstract:Perfluorooctanoic acid (PFOA) has been listed as an emerging persistent organic pollutant. Numerous studies showed that PFOA can produce various toxicities to organs, but there was no study yet to directly visualize the effects of PFOA on phospholipids. Phospholipids play multiple and essential roles in organisms. To this end, matrix-assisted laser desorption/ionization time of flight imaging mass spectrometry (MALDI-TOF-IMS) was used to visualize changes of phospholipids in the different tissues of zebrafish after exposure to PFOA. This study found that the major perturbed phospholipids were identified as phosphatidylcholine (PC), diacylglycerol (DG), phosphatidic acid (PA), phosphatidylglycerol (PG), sphingomyelin (SM), and triacylglycerol (TG). The perturbed phospholipids were generally more sensitive in the low-exposure group during exposure period. Some phospholipids changes were reversible in some tissues(liver, gill, and brain)and irreversible in others(such as the high exposure of intestine). Four phospholipids were identified as the common potential biomarkers. Moreover, the spatial distribution of perturbed phospholipids was mainly located around the margins or center of tissue, which implied that distinct biochemical events take place in certain tissue regions. This will provide novel insights into mechanisms of PFASs-induced ecotoxicity in the future.
