Recovery of Polyphenols from Pomegranate Peels by Ultrasound-Assisted Accelerated Penetration Extraction

Abstract:Pomegranate peels (PPs) contain rich polyphenols but are tricky to be disposed of. For this purpose, the recovery of PPs polyphenols is an attractive strategy for disposing PPs and feeding the demands of polyphenols-based functional food. However, emerging techniques should be developed to elevate efficiency. Our previous study presented a new accelerated penetration extraction (APE) that added ions to precipitate polyphenols in extraction. Here, we have developed a new extraction concept and a novel method based on ultrasound and APE – ultrasound-assisted accelerated penetration extraction (UAAPE) to extract polyphenols from PPs, in which precipitation effects of polyphenols and ultrasound-induced degradation effects of cell walls simultaneously contribute to the dissolution of PPs polyphenols from the perspective of mass transfer theory. We employed COSMOtherm to help select solvents and concentrations, and 50% aqueous ethanol is chosen as a solvent for subsequent experiments. The maximum yield of UAAPE is predicted by 152.58 mg/g and validated by 158.52 mg/g at optimized conditions (0.67 mg/mL of NaOH concentration, 10 mg/mL of CaCl2 concentration, 458.33 W and 30.56 min) by Box-Behnken Design (BBD). Fick’s law, Modified Fick’s law, and second-order rate law were fitted in extraction kinetics among which Fick’s law possesses the most precise prediction of the polyphenol contents. Extraction kinetics have been accelerated showing that UAAPE can not only mitigate the reduction trends of concentration gradients of polyphenols but also improve the extraction rate coefficient which is thought to be relevant to ultrasound-induced degradation of PPs thus further enhancing the extraction process. Such suppose is verified by SEM. We also elaborately compared different extraction methods including CSE, APE, and UAAPE. Consequently, UAAPE presents a high potential ability in polyphenols extraction and treatments of PPs.
