A Putative Adhesin-encoding Gene AOL_s00007g5 is Involved in the Mycelial Growth and Development of Nematode-trapping Fungus Arthrobotrys Oligospora

Abstract:Arthrobotrys oligospora is one of the most commonly studied fungi that can form abundant traps to capture and consume nematodes at the parasitic stage. Three-dimensional adhesive traps play key roles in capturing nematodes and regulating the lifestyle switch from saprophytic to carnivorous type. Here, we describe the effect of the adhesin-encoding gene AOL_s00007g5 (Aog5) on the mycelial growth and trap development of A. oligospora. Deletion of Aog5 gene significantly induced the formation of more traps in A. oligospora following nematode addition. ΔAog5 mutant grew relatively slower on tryptone-yeast extact-glucose-agar and potato dextrose agar culture media compared to wild-type strain, while there was no significant influence on conidiation and conidial germination. However, the mycelial growth of ΔAog5 mutant was less sensitive to chemical stressors, indicating that it was more adaptable to the external environment. Therefore, the adhesin Aog5 is involved in the mycelial growth and trap development of A. oligospora to facilitate the nematode capture. Our findings revealed the potential biological roles of adhesion gene Aog5 in A. oligospora, which may aid in the development of biocontrol agents against nematodes.
