Abstract:AbstractWound infections slow down the healing process and lead to complications such as septicemia, osteomyelitis, and even death. Although traditional methods relying on antibiotics are effective in controlling infection, they have led to the emergence of antibiotic‐resistant bacteria. Hydrogels with antimicrobial function become a viable option for reducing bacterial colonization and infection while also accelerating healing processes. Chitosan is extensively developed as antibacterial wound dressings due to its unique biochemical properties and inherent antibacterial activity. In this review, the recent research progress of chitosan‐based hydrogels for infected wound treatment, including the fabrication methods, antibacterial mechanisms, antibacterial performance, wound healing efficacy, etc., is summarized. A concise assessment of current limitations and future trends is presented.
Preparation and Application of Quaternized Chitosan- and AgNPs-Base Synergistic Antibacterial Hydrogel for Burn Wound Healing

Abstract:Infection is the major reason that people die from burns; however, traditional medical dressings such as gauze cannot restrain bacterial growth and enhance the healing process. Herein, an organic- and inorganic-base hydrogel with antibacterial activities was designed and prepared to treat burn wounds. Oxidized dextran (ODex) and adipic dihydrazide grafted hyaluronic acid (HA-ADH) were prepared, mixed with quaternized chitosan (HACC) and silver nanoparticles to fabricate Ag@ODex/HA-ADH/HACC hydrogel. The hydrogel, composed of nature biomaterials, has a good cytocompatibility and biodegradability. Moreover, the hydrogel has an excellent antibacterial ability and presents fast healing for burn wounds compared with commercial Ag dressings. The Ag@ODex/HA-ADH/HACC hydrogel will be a promising wound dressing to repair burn wounds and will significantly decrease the possibility of bacterial infection.

Abstract:BACKGROUND: With the development of chemical synthesis technology, the application of chitosan-based hydrogels as wound dressing has become a hotspot in current research. OBJECTIVE: To generally introduce the chitosan-based hydrogels and to review and summarize the application of chitosan-based hydrogels carrying antimicrobial agents in wound healing.METHODS: A computer retrieval of PubMed, Web of Science, Springerlink and Medline databases was performed with "chitosan hydrogel, wound healing, dressing, antimicrobial agents" as key words for relevant articles published from 2007 to 2016. Initially, 168 articles were retrieved, and finally 76 articles were included in result analysis. RESULTS AND CONCLUSION: Because of their inherent anti-inflammatory and antibacterial properties, chitosan-based hydrogels possess the ability of absorbing exudates and forming thin film between the wound and the external environment, to exert their advantages for chronic, non-healing wounds caused by infection. Furthermore, chitosan-based hydrogels carrying antibacterial agents can inhibit the growth of microorganisms in the wound, accelerating the progress of wound healing. Moreover, the antimicrobial agents, including organic antibacterial agents and inorganic antibacterial agents (especially nano-silver), show unique advantages in wound healing and microbial resistance in combination with chitosan-based hydrogels.

Abstract:Due to their biodegradability and biocompatibility, chitosan-based hydrogels have great potential in regenerative medicine, with applications such as bacteriostasis, hemostasis, and wound healing. However, toxicity and high cost are problems that must be solved for chitosan-based hydrogel crosslinking agents such as formaldehyde, glutaraldehyde, and genipin. Therefore, we developed a biocompatible yet cost-effective chitosan-based hydrogel system as a candidate biomaterial to prevent infection during wound healing. The hydrogel was fabricated by crosslinking chitosan with dialdehyde chitosan (CTS–CHO) via dynamic Schiff-base reactions, resulting in a self-healable and injectable system. The rheological properties, degradation profile, and self-healable properties of the chitosan-based hydrogel were evaluated. The excellent antibacterial activity of the hydrogel was validated by a spread plate experiment. The use of Live/Dead assay on HEK 293 cells showed that the hydrogel exhibited excellent biocompatibility. The results demonstrate that the newly designed chitosan-based hydrogel is an excellent antibacterial wound dressing candidate with good biocompatibility.

Abstract:Bacterial infection is one of the most challenging tasks in biomedical fields and still remains a serious threat to human lives today. Antibacterial hydrogel has attracted increasing scholarly interest as a promising wound dressing material. The antibacterial properties of ultrasmall gold nanoparticles (UsAuNPs) have also been widely recognized. Developing a tensile, self-healable, biocompatible composite hydrogel with desirable antibacterial properties to facilitate wound recovery is essential to improve treatment outcomes. However, UsAuNPs selfaggregate easily, resulting in decreased antibacterial effectiveness and limited clinical application. To address this problem, a composite material, UsAuNPs and carboxymethyl chitosan (UsAuNPs@CMCS) was designed. CMCS limit the self-aggregation of gold nanoparticles (AuNPs) at the size of 1-3 nm and attracts bacteria carrying negative charge on the surface, which renders the high antibacterial efficacy of the composite material. When the composite was crosslinked with the highly compatible polyacrylic acid (PAA) backbone of the hydrogel, the resulting PAA-CMCS-Au hydrogel displayed desirable antibacterial properties and facilitated wound recovery in vivo. This research establishes a new strategy to prevent the self-aggregation of UsAuNPs, and provides new insights in treating wounds with antibacterial composite hydrogels.

Abstract:The management of wound healing represents a significant clinical challenge due to the complicated processes involved. Chitosan has remarkable properties that effectively prevent certain microorganisms from entering the body and positively influence both red blood cell aggregation and platelet adhesion and aggregation in the bloodstream, resulting in a favorable hemostatic outcome. In recent years, chitosan-based hydrogels have been widely used as wound dressings due to their biodegradability, biocompatibility, safety, non-toxicity, bioadhesiveness, and soft texture resembling the extracellular matrix. This article first summarizes an overview of the main chemical modifications of chitosan for wound dressings and then reviews the desired properties of chitosan-based hydrogel dressings. The applications of chitosan-based hydrogels in wound healing, including burn wounds, surgical wounds, infected wounds, and diabetic wounds are then discussed. Finally, future prospects for chitosan-based hydrogels as wound dressings are discussed. It is anticipated that this review will form a basis for the development of a range of chitosan-based hydrogel dressings for clinical treatment.

Abstract:Bacterial infection treatment for chronic wounds has posed a major medical threat and challenge. Bacteria at the wounded sites can compete with the immune system and subsequently invade live tissues, leading to more severe tissue damage. Therefore, there is an urgent demand for wound dressings with antibacterial and anti-inflammatory properties. Considering the concept of moist healing, hydrogels with a three-dimensional (3D) network structure are widely used as wound dressings due to their excellent hydrophilicity, water retention properties, and biocompatibility. Developing antibacterial hydrogels for the treatment of infected wounds has been receiving extensive attention recently. This article categorizes antibacterial hydrogels according to their materials and antibacterial modes, and introduces the recent findings and progress regarding their status. More importantly, with the development of emerging technologies, new therapies are utilized to prepare antibacterial hydrogels such as nanoenzymes, photothermal therapy (PTT), photodynamic therapy (PDT), metal-organic frameworks (MOFs), and other external stimuli-responsive methods. Therefore, this review also examines their progress, challenges, and future trends as wound dressings. In the following studies, there will still be a focus on antibacterial hydrogels that have a high performance, multi-functions, and intelligence, especially biocompatibility, a high and long-lasting antibacterial property, responsiveness, and on-demand therapeutic ability.

Abstract:S. aureus (SA) and P. aeruginosa (PA) contaminations in thermally hurt patients have controlled invasive illness producing humanity. Gold nanoparticles (AuNPs) engaged in behaviour have limits owed to their oxidative damage. Current examinations demonstrated the biosynthesis of highly biocompatible AuNPs from rich antioxidant of aqueous cutting of Aerva javanica (A. javanica). The cell viability of the biosynthesised AuNPs was examined by cancerous cell lines (MCF-7 and MDA-MB-231) and non-cancerous cell lines (HUVECs), which displayed the high biocompatibility in all the cell lines. We developed the encapsulation with chitosan hydrogels to the AuNPs for the burn wound infections created by the Sprague Dawley rats. Remarkable burn wound reduction in the chitosan hydrogels was found. The histological analysis also suggested the burn wound contracting without causing any damages. The outcomes indicated that AuNPs incorporate to the chitosan hydrogels are promising burn wound dressing resources for the treatment for wound healing.

Abstract:Wound dressing is an important tool for wound management. Designing wound dressings by combining various novel materials and drugs to optimize the peri-wound environment and promote wound healing is a novel concept. Hydrogels feature good ductility, high water content, and favorable oxygen transport, which makes them become some of the most promising materials for wound dressings. In addition, nanomaterials exhibit superior biodegradability, biocompatibility, and colloidal stability in wound healing and can play a role in promoting healing through their nanoscale properties or as carriers of other drugs. By combining the advantages of both technologies, several outstanding and efficient wound dressings have been developed. In this paper, we classify nano-based hydrogel dressings into four categories: hydrogel dressings loaded with a nanoantibacterial drug; hydrogel dressings loaded with oxygen-delivering nanomedicines; hydrogel dressings loaded with nanonucleic acid drugs; and hydrogel dressings loaded with other nanodelivered drugs. The design ideas, advantages, and challenges of these nano-based hydrogel wound dressings are reviewed and analyzed. Finally, we envisaged possible future directions for wound dressings in the context of relevant scientific and technological advances, which we hope will inform further research in wound management.

Abstract:Antibacterial wound dressings can effectively avoid the residual of antibacterial nanomaterials for injection in vivo, reduce their biological toxicity to normal cells and tissues, making them be widely applied in biomedical field. Herein, an approach of combining ion-crosslinking, in-situ reduction and microwave-assisted methods was employed to prepare chitosan-copper-gallic acid nanocomposites (CS-Cu-GA NCs) with dual-functional nano-enzyme characteristics (oxidase- and peroxidase-like functions). The oxidase-like activity of CS-Cu-GA NCs can facilitate the production of hydrogen peroxide (H2O2) when it contacted with physiologically relevant antioxidants (AH2) in bacteria. Subsequently, H2O2 was catalyzed to generate hydroxyl radicals (OH) under the peroxidase-like activity of CS-Cu-GA NCs. Furthermore, CS-Cu-GA NCs integrate the inherent antibacterial properties of chitosan, Cu NPs and Cu2+. Animal experiments revealed that the antibacterial dressing incorporating CS-Cu-GA NCs exhibited its effective promotion of S. aureus-infected wounds healing, as well as no damage to normal tissues. Besides, the antibacterial dressing was prepared to a band aid with excellent water swelling and antibacterial properties, which was further fixed in a medical tape to construct a portable antibacterial product that can be applied to the surface of human skin and showed excellent waterproof performance, providing a new insight for the construction of clinical antibacterial wound healing products.

Abstract:Wounds continue to be a serious medical concern due to their increasing incidence from injuries, surgery, burns and chronic diseases such as diabetes. Delays in the healing process are influenced by infectious microbes, especially when they are in the biofilm form, which leads to a persistent infection. Biofilms are well known for their increased antibiotic resistance. Therefore, the development of novel wound dressing drug formulations and materials with combined antibacterial, antibiofilm and wound healing properties are required. Nanomaterials (NM) have unique properties due to their size and very large surface area that leads to a wide range of applications. Several NMs have antimicrobial activity combined with wound regeneration features thus give them promising applicability to a variety of wound types. The idea of NM-based antibiotics has been around for a decade at least and there are many recent reviews of the use of nanomaterials as antimicrobials. However, far less attention has been given to exploring if these NMs actually improve wound healing outcomes. In this review, we present an overview of different types of nanomaterials explored specifically for wound healing properties combined with infection control.
Novel Chitosan Hydrogels Reinforced by Silver Nanoparticles with Ultrahigh Mechanical and High Antibacterial Properties for Accelerating Wound Healing.

Abstract:Poor mechanical properties of chitosan hydrogels limit their applications as the wound dressing. To overcome this drawback, we abandoned the traditional glacial acetic acid method but adopted a LiOH/urea solvent system to synthesize chitosan hydrogel. Then reductive Ag nanoparticles were integrated into the chitosan hydrogel networks, aiming at reinforcing the mechanical properties and improving the antibacterial properties of chitosan hydrogel. The synthesized hydrogels were subsequently characterized using the FTIR, XRD, SEM, and TEM. In addition, swelling characteristics, mechanical properties, antibacterial abilities as well as wound healing efficacy on Sprague-Dawley rats were evaluated. The results showed that the novel hydrogel exhibited porous three-dimensional network and ultrahigh mechanical properties due to the inter-molecular and intra-molecular interactions. The compressive strength was 15.95 +/- 1.95 MPa, >100 times stronger than that of the control group. Meanwhile, the hydrogels still remained structural integrity even if the strain exceeded 90%. Furthermore, compared with the controls, the hydrogels exhibited more excellent antibacterial performance and significantly (p < 0.05) increased the rate of the re-epithelialization and collagen deposition, effectively accelerating the wound healing. Therefore, the synthesized hydrogel with ultrahigh mechanical properties will be found potential applications in the fields of biomedicine. (C) 2018 Elsevier B.V. All rights reserved.

Abstract:Bacterial infections have always been a major threat to human health. Skin wounds are frequently exposed to the external environment, and they may become contaminated by bacteria derived from the surrounding skin, the local environment, and the patient's own endogenous sources. Contaminated wounds may enter a state of chronic inflammation that impedes healing. Urgent development of antibacterial wound dressings capable of effectively combating bacteria and overcoming resistance is necessary. Nanotechnology and nanomaterials present promising potential as innovative strategies for antimicrobial wound dressings, owing to their robust antibacterial characteristics and the inherent advantage of avoiding antibiotic resistance. Therefore, this review provides a concise overview of the antimicrobial mechanisms exhibited by low-dimensional nanomaterials. It further categorizes common low-dimensional antimicrobial nanomaterials into zero-dimensional (0D), one-dimensional (1D) and two-dimensional (2D) nanomaterials based on their structural characteristics, and gives a detailed compendium of the latest research advances and applications of different low-dimensional antimicrobial nanomaterials in wound healing, which could be helpful for the development of more effective wound dressings.

Abstract:The complicated wound repair process caused by microbial infection is still a clinical problem due to antibiotic resistance. Therefore it is necessary to employ the incorporating bioactive molecules in the dressing to solve this problem. Herein, a multifunctional nanocomposite hydrogel (CS-HCA-Icps) with the pathological pH-responsive drug release has been developed to promote the infection-impaired wound healing. CS-HCA-Icps nanocomposite hydrogel composed of catechol-grafted chitosan (CS-HCA) and a curcumin-Fe 3+ coordination nanoparticles (Icps, CurFe 3+ ) exhibits the favorable activities in free radical scavenging, anti-bacterial and anti-inflammatory. The favorable biocompatibility is also demonstrated both in vitro and in vivo experiments. These demonstrate the promoting efficacy of hydrogel in wound healing. In this study, Chitosan (CS) shows excellent biocompatibility and antibacterial properties for tissue repair. After functional modification with HCA, the catechol groups are beneficial to improve antioxidant capacity for wound repair, Moreover, Icps nanomedicine are able to enhance the loaded Cur release in response to the pathological acidic microenvironment at the inflammatory stage of wounds. Thus, the pathological pH-responsive hydrogel integrating anti-bacterial, antioxidant, and anti-inflammatory functions may represent a promising strategy for safe and efficient wound healing, in particular for potential clinical use.

Abstract:Bacterial infection and the ever-increasing bacterial resistance have imposed severe threat to human health. And bacterial contamination could significantly menace the wound healing process. Considering the sophisticated wound healing process, novel strategies for skin tissue engineering are focused on the integration of bioactive ingredients, antibacterial agents included, into biomaterials with different morphologies to improve cell behaviors and promote wound healing. However, a comprehensive review on anti-bacterial wound dressing to enhance wound healing has not been reported. In this review, various antibacterial biomaterials as wound dressings will be discussed. Different kinds of antibacterial agents, including antibiotics, nanoparticles (metal and metallic oxides, light-induced antibacterial agents), cationic organic agents, and others, and their recent advances are summarized. Biomaterial selection and fabrication of biomaterials with different structures and forms, including films, hydrogel, electrospun nanofibers, sponge, foam and three-dimension (3D) printed scaffold for skin regeneration, are elaborated discussed. Current challenges and the future perspectives are presented in this multidisciplinary field. We envision that this review will provide a general insight to the elegant design and further refinement of wound dressing.

Photothermal enhanced antibacterial chitosan-based polydopamine composite hydrogel for hemostasis and burn wound repairing

Abstract:Bleeding and bacterial infection are common problems associated with wound treatment, while effective blood clotting and vessel regeneration promotion are the primary considerations to design the wound dressing materials. This research presents a chitosan-based hydrogel with grafted quaternary ammonium and polyphosphate (QCSP hydrogel) as the antibacterial hemostatic dressing to achieve burn wound treatment. The tissue adhesion of the hydrogel sealed the blood flow and the polyphosphate grafted to the chitosan promoted the activation of coagulation factor V to enhance the hemostasis. At the same time, the grafted quaternary ammonium enhanced the antibacterial ability of the biodegradable hydrogel wound dressing. In addition, the polydopamine as a photothermal agent was composited into the hydrogel to enhance the antibacterial and reactive oxygen scavenging performance. The in vivo hemostasis experiment proved the polyphosphate enhanced the coagulation property. Moreover, this photothermal property of the composite hydrogel enhanced the burn wound repairing rate combined with the NIR stimulus. As a result, this hydrogel could have potential application in clinic as dressing material for hemostasis and infection prone would repairing.
Spreadable Thermosensitive Nanocomposite Hydrogel Dressing with Ultrasound-Responsive Bactericidal/repair-Promoting Regulation and Cascade Antioxidantion for Infected Burn Wound Repair

Abstract:Treating severe burn wounds poses significant challenges, including considerable cell loss, excessive inflammation, and a high susceptibility to bacterial infections. Ideal burn dressings should exhibit excellent antibacterial properties, anti-inflammatory effects, and promote cell proliferation. Additionally, they need facilitate painless dressing changes and be user-friendly. Herein, we synthesized a thermosensitive hydrogel by crosslinking poly (N-isopropylacrylamide-co-allyloxybenzaldehyde) (PNA) and amino-terminated Pluronic F127 (APF) through a Schiff base reaction. It exhibited reversible gel-sol transition and spreadability. By incorporating piezoelectric gold nanoparticle-modified barium titanate (Au@BaTiO3) and cascade antioxidant MOF-818, a nanocomposite hydrogel dressing with diverse bioactive functionalities was developed. Results demonstrated that the nanocomposite hydrogel possessed gel-sol transition properties, maintained a stable gel state within a broad temperature range, and desirable self-healing property. Au@BaTiO3 exhibited good piezoelectric properties and ROS generation upon ultrasound stimulation, while MOF-818 displayed highly efficient cascade nanozyme activity. The combination of Au@BaTiO3 and MOF-818 promoted fibroblast proliferation and migration, reduced intracellular ROS levels, and induced anti-inflammatory polarization of macrophages under ultrasound stimulation. In vitro and in vivo antibacterial results disclosed that the nanocomposite hydrogel had excellent antibacterial activity under high-intensity ultrasound stimulation. When applied to infected burn wounds, the nanocomposite hydrogel can rapidly sterilize the wound upon initial high-intensity ultrasound, and then reduce inflammation and promote M2 macrophage polarization by the following low-intensity ultrasound stimulation, and thus accelerating the healing by improving granulation tissue formation, angiogenesis, and collagen deposition.

Abstract:The ever-growing challenges of traditional antibiotic therapy and chronic wound healing have created a hot topic for the development and application of new antimicrobial agents. Silver nanoclusters (Ag NCs) with ultrasmall sizes (<2 nm) and antibacterial effects are promising candidates for next-generation antibiotics, particularly against multi-drug resistant strains. However, the biosafety in the clinical application of Ag NCs remains suboptimal despite some existing studies of Ag NCs for biomedical applications. Considering this, an ultrasmall Ag NC with excellent water solubility was synthesized by a two-phase ligand-exchange method, which exhibits broad-spectrum antibacterial performance. The minimum inhibitory concentrations of Ag NCs against MRSA , S. aureus , P. aeruginosa and E. coli were evaluated as 50, 80, 5 and 5 μg mL −1 , respectively. Furthermore, a carbomer hydrogel was prepared to be incorporated into the Ag NCs for achieving excellent biocompatibility and biosafety. In vitro experiments demonstrate that the Ag NC-gel exhibits good antibacterial properties with lower cytotoxicity. Finally, in vivo experiments suggest that this ultrasmall Ag NC functionalized with the hydrogel can serve as an effective and safe antimicrobial agent to aid in wound healing.

Abstract:Bacterial infection remarkably impedes wound healing, with antibiotics traditionally serving as the primary therapeutic intervention. However, the escalating misuse of antibiotics and the emergence of bacterial resistance present substantial treatment challenges for infected wounds. Consequently, the development of antibiotic-free antimicrobial dressings holds pertinent research and clinical relevance. To this end, this study aimed to introduce an all-natural hydrogel dressing, amalgamating polyphenols and polysaccharides, exhibiting pronounced antibacterial and antioxidant properties without relying on antibiotics. First, we constructed curcumin-tannic acid‐zinc ion nanospheres (CTZN) through self-assembly. Our experimental results showed that the nanospheres had excellent biocompatibility, antioxidant, and antimicrobial abilities. Subsequently, we prepared carboxymethylated chitosan/oxidized sodium alginate hydrogels via Schiff base reactions. Incorporation of CTZN into the hydrogel system not only improves the inherent qualities of the hydrogel but also confers multifunctional properties, including antimicrobial, antioxidant, and anti-inflammatory abilities. In this study, we enhanced the physicochemical properties and biological activity of hydrogels by introducing natural material nanospheres, offering a novel approach that could pave the way for the development of purely natural biomaterial dressings.
