Fabrication of Magnesium Phosphate Bone Cement with Enhanced Osteogenic Properties by Employing Zeolitic Imidazolate Framework-8

Abstract:In this work, zeolitic imidazolate framework-8 containing Zn in favor of bone regeneration was first introduced into magnesium phosphate bone cement (MPCs). The physical properties of ZIF-8/MPCs (ZIFMPCs) including microstructures, setting time, compressive strength and degradation in vitro were studied. The ZIFMPCs material was cultured with mouse bone marrow stem cells to evaluate the osteogenesis activities. It was found that the setting times of MPCs were prolonged from 8.2 ± 0.3 min to 13.6 ± 0.2 min (5.0ZIFMPC), and the exothermic temperatures were reduced from 45.8 ± 0.4 °C (MPC) to 38.0 ± 0.2 °C (3.0ZIFMPC). The ZIFMPC with 1 wt% ZIF-8 had the highest compressive strength up to 38.61 MPa as compared with MPC. In comparison with MPC, the ZIFMPCs could significantly drive proliferation and differentiation of osteoblastic mBMSCs as well as elevate expression of osteogenic related proteins. Hence, ZIF-8 had a significant effect on the osteogenesis of mBMSCs. These findings suggested that the ZIFMPCs with enhanced mechanical performance, excellent cytocompatibility and osteogenic activity could serve as potential candidates to enhance the functionality of synthetic bone graft material.
