Abstract:Covalent organic frameworks (COFs) have generated interest as promising tools for reactive oxygen species (ROS) -mediated therapy owing to designable structures and diversified modification sites. However, the ordered porous structure of COFs and their multiple catalytic sites on the enhancement of ROS-mediated therapy of cancers have attached little attention. Here, we synthesized a nanosonosensitizer named FeTPD@GOx, consisting of the ferriporphyrin-based COF (Fe-COF) loaded with glucose oxidase (GOx) for pore-enhanced ROS-mediated therapy in which GOx catalyzed glucose to H2O2 while Fe3+/Fe2+ cycle catalyzed H2O2 to O-2 and OH. The H2O2/O-2 self-supply cycle inside the pores of COFs enhanced the generation of abundant ROS in situ upon ultrasound (US) irradiation. The mechanism examination revealed that the opening pore channels of COFs not only could catch O-2 with the rapid exchange for O-2 and O-1(2), but also provided more accessible Fenton reaction sites to iron ions for more OH generation, enhancing the efficacy of sonodynamic therapy (SDT) and chemodynamic therapy (CDT). In vivo antitumor evaluation revealed that FeTPD@GOx can not only inhibit the growth of the proximal tumor effectively but also display superior antitumor efficiency to the distant tumor upon US irradiation. Our work, which utilizes the porous structure, multiple catalytic sites, and GOx loading of COFs to hold O-2, thereby promoting gas exchange, realizing sequential catalytic reactions, amplifying ROS generation, and enhancing synergistic effect of SDT and CDT for ROS-mediated therapeutic efficacy, offers prospects for applications of COFs in biomedical fields.

Abstract:Herein, we report a composite COF material loaded with a Pt nanoenzyme and an organic photosensitizer BODIPY, synthesized via a stepwise post-synthetic modification. The obtained Pt@COF-BDP nanoparticles can efficiently and continuously convert H 2 O 2 to O 2 , thereby increasing the efficiency of single-linear oxygen production and achieving efficient tumor inhibition.

Abstract:Chemodynamic therapy (CDT), which induces cell death by decomposing high levels of H2O2 in tumor cells into highly toxic ·OH, is recognized as a promising antineoplastic approach. However, current CDT approaches are often restricted by the highly controlled and upregulated cellular antioxidant defense. To enhance ·OH-induced cellular damage by CDT, a covalent organic framework (COF)-based, ferrocene (Fc)- and glutathione peroxidase 4 (GPX4) inhibitor-loaded nanodrug, RSL3@COF–Fc (2b), is fabricated. The obtained 2b not only promotes in situ Fenton-like reactions to trigger ·OH production in cells, but also attenuates the repair mechanisms under oxidative stress via irreversible covalent GPX4 inhibition. As a result, these two approaches synergistically result in massive lipid peroxide accumulation, subsequent cell damage, and ultimately ferroptosis, while not being limited by intracellular glutathione. It is believed that this research provides a paradigm for enhancing reactive oxygen species-mediated oncotherapy through redox dyshomeostasis and may provide new insights for developing COF-based nanomedicine.

Abstract:Reactive oxygen species (ROS)-based cancer treatments have attracted much attention in recent years. However, most patients respond poorly to the monotypic ROS during these treatments. In this work, a multiple ROS-based cancer immunotherapy synergistic strategy has been developed to enhance the therapeutic effect of cancer. We prepare a three-dimensional covalent organic framework (3D COF-TATB), and embed copper ions (Cu2+) into the skeleton to obtain multifunctional nanomaterial, 3D Cu@COF-TATB. In this system, porphyrins in 3D COF-TATB serve not only as the photosensitizer for photodynamic process to produce singlet oxygen(1O2), but also as the binding sites to complex with Cu2+. Cu2+ can be reduced by the GSH to generate Cu+ to produce hydroxyl radical (•OH) through the Fenton-like reaction. Moreover, the generated multiple types of ROS induce the immunogenic cell death (ICD) of cancer cells to improve the immunogenicity and further activate an immune response for attacking the tumor. Combining with the immunoblocking inhibitor (aPD-1), 3D Cu@COF-TATB can effectively inhibit the tumor growth. This work will provide a guidance for multimodal cancer therapy in future clinical treatment settings.

Abstract:As a newly emerging crystalline porous materials, covalent organic frameworks (COFs) possess fascinating structures and some specific features such as modularity, crystallinity, porosity, stability, versatility, and biocompatibility. Besides adsorption/separation, sensing, catalysis and energy, COFs recently show a new trend in biomedical application. This contribution provides an overview of recent developments of COF-based medicines in cancer therapeutics, including drug delivery, photodynamic therapy (PDT), photothermal therapy (PTT), and combined therapy. Furthermore, major challenges and developing trend in this field are also discussed. These recent developments are summarized and discussed to help encourage further contributions in this emerging and promising field.

Abstract:Developing effective photosensitizers to initiate the generation of singlet oxygen (1O2) is of great significance in both chemistry and physiology. Herein, linking the photoactive porphyrin moieties by in situ-formed robust imidazole groups, a covalent organic framework (COF), PyPor-COF, was successfully designed and synthesized. Detailed characterizations reveal that it not only possesses high crystallinity, permanent porosity, and robust stability but also shows a semiconductive photoresponse activity. As demonstrated by electron paramagnetic resonance experiments, the COF can initiate the generation of 1O2 efficiently under visible-light irradiation, the efficiency of which is higher than that of the pristine porphyrin-based reactant and even higher than some commonly used commercially available photosensitizing agents. Anticancer experiments prove that it can efficiently trigger the production of 1O2 in a physiological environment. This work demonstrates that the imidazole-linked porphyrin-incorporated COF is a highly promising photosensitizer that can even be applied in photodynamic therapy.

Abstract:Due to the limited concentration of hydrogen peroxide (H 2 O 2 ) in the tumor microenvironment, peroxidase (POD)-mimicking nanozyme-mediated catalytic therapy usually cannot completely eliminate tumor issues. In this study, we report an H 2 O 2 -responsive nanozyme constructed by loading 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) into POD-like Fe-porphyrin covalent organic frameworks (COFs) (Fe-DhaTph). The resulting ABTS@Fe-DhaTph can function as a multifunctional therapeutic nanoagent for combination therapy of catalytic therapy, photothermal therapy (PTT), and photodynamic therapy (PDT). In the tumor microenvironment, ABTS@Fe-DhaTph can catalyze the decomposition of endogenous H 2 O 2 to produce highly cytotoxic hydroxyl radicals (·OH) for in situ tumor catalytic therapy. Meanwhile, the loaded ABTS is oxidized by H 2 O 2 with the assistance of ABTS@Fe-DhaTph, and the resulting oxABTS, with strong near-infrared absorbance, can enable the H 2 O 2 -dependent PTT. Besides Fe-DhaTph-induced PDT, the generated oxABTS with glutathione (GSH)-depletion ability can synergistically enhance the generation of reactive oxygen species, thereby improving catalytic therapy and PDT. To the best of our knowledge, ABTS@Fe-DhaTph is the first COF-based nanozyme to serve as a cascade-amplified synergistic therapeutic nanoagent for cancer treatment.

Abstract:The clinical practice of photodynamic therapy of cancer (PDT) is mostly limited to superficial types of skin cancer. The major reason behind this limited applicability is the need for light in the photogeneration of ROS, and in particular singlet oxygen. In order to circumvent this major roadblock, we designed and synthesized naphthalene‐derived endoperoxides with mitochondria targeting triphenylphosphonium moieties. Here, we show that these compounds release singlet oxygen by thermal cycloreversion, and initiate cell death with IC50 < 10 μM in cancer cell cultures. The mouse 4T1 breast tumor model study, where the endoperoxide compound was introduced intraperitoneally, also showed highly promising results, with negligible systemic toxicity. Targeted delivery of singlet oxygen to cancer cell mitochondria could be the breakthrough needed to transform Photodynamic Therapy into a broadly applicable methodology for cancer treatment by keeping the central tenet and discarding problematic dependencies on oxygen or external light.

A Catalase‐Like Metal‐Organic Framework Nanohybrid for O 2 ‐evolving Synergistic Chemoradiotherapy

Abstract:Tumor hypoxia, the "Achilles' heel" of current cancer therapies, is indispensable to drug resistance and poor therapeutic outcomes especially for radiotherapy. Here we propose an in situ catalytic oxygenation strategy in tumor using porphyrinic metal-organic framework (MOF)-gold nanoparticles (AuNPs) nanohybrid as a therapeutic platform to achieve O2 -evolving chemoradiotherapy. The AuNPs decorated on the surface of MOF effectively stabilize the nanocomposite and serve as radiosensitizers, whereas the MOF scaffold acts as a container to encapsulate chemotherapeutic drug doxorubicin. In vitro and in vivo studies verify that the catalase-like nanohybrid significantly enhances the radiotherapy effect, alleviating tumor hypoxia and achieving synergistic anticancer efficacy. This hybrid nanomaterial remarkably suppresses the tumor growth with minimized systemic toxicity, opening new horizons for the next generation of theranostic nanomedicines.

Abstract:Covalent organic frameworks (COFs) have attracted much attention for various applications, while developing COFs with biomedical functions remains a big challenge. Most COFs are built purely from organic molecules, no carbon dots (CDs)‐based COFs have been exploited to date. In this work, diamine molecules p‐phenylenediamine and BODIPY are selected as model monomers to react with aldehyde‐containing CDs and construct nanoscale COFs, named CCOF‐1 and CCOF‐2, respectively. After the modification with poly(ethylene glycol) (PEG), the formed CCOF‐1@PEG and CCOF‐2@PEG are stable and well‐dispersible in aqueous media. The merits of CCOF‐2@PEG containing favorable physiological stability, good biocompatibility, and strong reactive oxygen species generation ability enable it to act as an excellent photodynamic therapy (PDT) agent for tumor treatment. The high PDT therapeutic efficiency for inhibiting cell proliferation and tumor growth was well shown by in vitro and in vivo experiments. This work demonstrates a new strategy for fabricating functional COF‐based platforms for biomedical applications.

A singlet oxygen-storing covalent organic framework for "Afterglow" photodynamic therapy

Abstract:Photodynamic therapy (PDT) is an emerging treatment but often restricted by the availability of oxygen. Enhancing the lifespan of singlet oxygen (1O2) by fractionated generation is an effective approach to improve the efficacy of PDT. Herein, an imine-based nanoscale COF (TpDa-COF) has been synthesized and functionalized with a pyridone-derived structure (Py) to create a 1O2-storing nanoplatform TpDa-COF@Py, which can reversibly capture and release 1O2. Under 660 nm laser exposure, Py interacts with 1O2 produced by the porphyrin motif in COF backbones to generate 1O2-enriched COF (TpDa-COF@Py + hv), followed by the release of 1O2 through retro-Diels-Alder reactions at physiological temperatures. The continuous producing and releasing of 1O2 upon laser exposure leads to an "afterglow" effect and a prolonged 1O2 lifespan. In vitro cytotoxicity assays demonstrates that TpDa-COF@Py + hv exhibits an extremely low half-maximal inhibitory concentration (IC50) of 0.54 µg/mL on 4T1 cells. Remarkably, the Py-mediated TpDa-COF@Py nanoplatform demonstrates enhanced cell-killing capability under laser exposure, attributed to the sustained 1O2 cycling, compared to TpDa-COF alone. Further in vivo assessment highlights the potential of TpDa-COF@Py + hv as a promising strategy to enhance phototheronostics and achieve effective tumor regression. Accordingly, the study supplies a generalized 1O2 "afterglow" nanoplatform to improve the effectiveness of PDT.
Covalent Organic Framework-Based Nanocomposite for Synergetic Photo-, Chemodynamic-, and Immunotherapies

Abstract:Cancer deaths are mainly caused by tumor metastases. However, tumor ablation therapies can only target the primary tumor but not inhibit tumor metastasis. Herein, a multifunctional covalent organic framework (COF)-based nanocomposite is designed for synergetic photo-, chemodynamic- and immunotherapies. Specifically, the synthesized COF possesses the ability to produce singlet oxygen under the 650 nm laser irradiation. After being metallized with FeCl3, p-phenylenediamine is polymerized on the surface of COF with Fe3+ as the oxidant. The obtained poly(p-phenylenediamine) can be used for photothermal therapy. Meanwhile, the overexpressed H2O2 in the tumor would be further catalyzed and decomposed into hydroxyl radicals (•OH) by the Fe3+/Fe2+ redox couple via Fenton reaction. Intriguingly, the increase of temperature caused by photothermal therapy can accelerate the production of •OH. Moreover, the tumor-associated antigen induced a robust antitumor immune response and effectively inhibited tumor metastasis in the presence of anti-PD-L1 checkpoint blockade. Facile Synthesis of a Cubic Porphyrin-Based Covalent Organic Framework for Combined Breast Cancer Therapy

Abstract:A cubic porphyrin-based covalent organic framework (COF) named CTP with excellent hydrophilicity was prepared with a facile method for the first time. Different from the conventional methods for the synthesis of porphyrin-based COFs, this facile strategy has greatly shortened the reaction time under mild conditions. Linking the porphyrin monomer into the COF overcame its poor solubility and biocompatibility and also narrowed the band gap owing to the formation of the π-conjugation structure. The improved biocompatibility and narrowed band gap enabled CTP to be an excellent sonosensitizer with an enhanced sonodynamic effect. Moreover, CTP could also effectively realize photothermal conversion under external irradiation due to the extended conjugated structure. This work developed a novel synthesis method for COFs and employed a COF as a sonosensitizer for the first time, which not only provided a new strategy to improve the efficiency of organic sonosensitizers but also inspired us to design more functional COFs for versatile applications.

Abstract:The development of covalent organic framework (COF) sonosensitizers with intrinsic sonodynamic effects is highly desirable. However, such COFs are generally constructed using small-molecule photosensitizers. Herein, we report that the reticular chemistry-based synthesis of COFs from two inert monomers yields a COF-based sonosensitizer (TPE-NN) with inherent sonodynamic activity. Subsequently, a nanoscale COF TPE-NN is fabricated and embedded with copper (Cu)-coordinated sites to obtain TPE-NN-Cu. Results show that Cu coordination can enhance the sonodynamic effect of TPE-NN, whereas ultrasound (US) irradiation for sonodynamic therapy can augment the chemodynamic efficacy of TPE-NN-Cu. Consequently, TPE-NN-Cu upon US irradiation shows high-performance anticancer effects based on mutually reinforced sono-/chemo-nanodynamic therapy. This study reveals the backbone-originated sonodynamic activity of COFs and proposes a paradigm of intrinsic COF sonosensitizers for nanodynamic therapy.

Abstract:The exploration of type I mechanism remains a great challenge in photodynamic therapy (PDT) due to the limited electron transfer from the photosensitizer to ground state oxygen to form superoxide anion radical (O 2 − ). In this study, we design three kinds of nanoscale covalent organic frameworks (COFs) with tunable local polarization for accelerating the generation of O 2 − . Among three COFs, the tetrakis-(4-formylphenyl)benzene-based COF (TB-COF) exhibits highly aggravated local polarization with 3.29 Debye of the dipole moment, and thus drives the photoinduced e − reduction of O 2 /O 2 − , which matches with the energy level of TB-COF. The elongated charge-separated state of TB-COF is also identified by femtosecond transient absorption spectra and time-dependent density functional theory calculation. Significantly, the inhibited electron-hole recombination of COFs further improves the O 2 − production efficiency upon protonation and thus strengthens anti-cancer effect in the acidic microenvironment of tumor cell or in vivo . The reticular polarization engineering of COFs provides a new type I PDT paradigm in biomedical applications.

Abstract:Covalent organic frameworks (COFs) have emerged as a kind of promising material for analytical and biomedical purposes. However, simultaneous cancer diagnosis and therapy with COFs remain a challenge. We report here a COF-based theranostic nanoplatform by integrating a dye-labeled oligonucleotide onto porphyrin-based COF nanoparticles for highly efficient cancer diagnosis and therapy. The fluorescence of the dye was effectively quenched by the COF through fluorescence resonance energy transfer (FRET). In the presence of biomarker survivin mRNA, more stable duplexes were formed and separated from the COF NPs, enabling the recovery of the fluorescence signal and selective cancer imaging. Under NIR laser irradiation, COF NPs generated abundant reactive oxygen species (ROS) to induce cancer cell apoptosis owing to their crystalline reticular structure.In vitroandin vivoexperiments revealed that the nanoplatform has a high specificity and inhibition effect toward cancer cells and solid tumors. Interestingly, prognostic evaluation was also realized with COF-survivin. This work will offer new insights into COF-based probes and inspire the development of more versatile tools for biomedical applications.
