Effect of Heat Treatment on Structure and in Vitro Digestion of Water-Soluble Protein from Mussel (mytilus Edulis)

Abstract:This study aimed to investigate the effect of different heat treatment (HT) temperatures (30, 40, 50, 60, 70, 80, 90, and 100 °C) on the structure and digestibility of mussel water-soluble proteins (MWSP). The SDS-PAGE results showed that the high molecular weight bands of MWSP increased significantly after HT, suggesting that the HT promoted protein aggregation. Moreover, HT significantly increased the particle size, carbonyl content, and surface hydrophobicity of MWSP and decreased the solubility. The secondary structure results showed that HT reduced the α-helix content and increased the β-fold and β-turn content. In addition, the fluorescence intensity of MWSP decreased with increasing temperature, suggesting that the HT changed the tertiary structure. Nile fluorescence intensity results showed that HT promoted the formation of MWSP aggregates. In vitro digestion results showed that the digestibility of MWSP increased with increasing temperature at lower heating temperatures (30-40 °C), while protein aggregation and tighter structure at higher heating temperatures (> 50 °C) inhibited the digestion of MWSP. The present study has provided a reference for the effects of heat treatment on the structure and digestive behavior of shellfish protein.
